The dwarf river crab Liberonautes latidactylus nanoides Cumberlidge & Sachs, 1989, from Liberia--a new second intermediate host of Paragonimus uterobilateralis.

Large populations of the dwarf river crab Liberonautes latidactylus nanoides Cumberlidge & Sachs, 1989, collected from the St. Paul river at the Haindi/Mauwa focus of paragonimiasis in Liberia were examined for parasitization by the human lung fluke, Paragonimus uterobilateralis. Four specimens out of 763 L. l. nanoides proved to be positive for P. uterobilateralis (0.52%), and each of these specimens contained only 1 metacercaria. The dwarf river crab forms the basis of a seasonal commercial fishery in Liberia, and it is sold for human consumption in rural markets. However, the low levels of

Mechanism of asparagine deamidation during human senile cataractogenesis.

Previous studies have demonstrated that asparagine-143 of gammaS crystallin is preferentially deamidated in human cataractous lenses. A possible mechanism of nonenzymatic deamidation involves the initial formation of L -succinimidyl and D -succinimidyl intermediates, followed by ring cleavage and production of the respective L - and D - forms of beta-aspartate and alpha-aspartate. To determine if this mechanism occurs during cataractogenesis of the human lens, synthetic peptide standards together with reverse phase HPLC were used to purify and quantitate tryptic peptides from cataractous lense

Arg-substituted VmCT1 analogs reveals promising candidate for the development of new antichagasic agent.

VmCT1 is an antimicrobial peptide (AMP) isolated from the venom of the scorpion Vaejovis mexicanus with antimicrobial, anticancer and antimalarial activities, which the rational design with Arg-substitution has yielded AMPs with higher antimicrobial activity than VmCT1. Chagas is a neglected tropical disease, becoming the development of new antichagasic agents is urgent. Thus, we aimed to evaluate the antichagasic effect of VmCT1 and three Arg-substituted analogues, as well their action mechanism. Peptides were tested against the epimastigote, trypomastigote, amastigote forms of Trypanossoma c

Molecular and Structural Characterization of an Immunopurified Telomerase from Leishmania major and the Effect of Telomerase Inhibitors.

Leishmania major is the etiological agent of cutaneous leishmaniasis (CL) in several countries in Asia and Northern Africa. The disease is considered a zoonotic infection where rodents are the reservoirs and phlebotomine sandflies are the vectors. Once inside the human body, the parasite multiplies inside the macrophages of infected patients, but the disease eventually cures spontaneously, leaving scars where the phlebotomine bites occurred. Given the importance of the replicative forms in the parasite's cell cycle, here, we decided to study the enzyme telomerase, which has the critical role o

Inhibition of pituitary adenylate cyclase activating polypeptide induced relaxation of guinea-pig tracheal smooth muscle by charybdotoxin.

The aim of the present study was to investigate whether or not charybdotoxin (CAS 95751-30-7, ChTX), a selective and potent Ca(2+)-dependent K+ channel blocker, inhibits the relaxation of guinea-pig tracheal smooth muscle induced by pituitary adenylate cyclase activating polypeptides with 27 residues (PACAP27) and with 38 residues (PACAP38). Two forms of PACAP were discovered in hypothalamic tissues, and are known to increase the tissues cyclic AMP levels and to be independent of beta-adrenoceptors. The relaxant effects of these polypeptides were evaluated by measuring the isometric tension of

Photo-Oxidation of α-Pinene Oxidation Products in Atmospheric Waters - pH- and Temperature-Dependent Kinetic Studies.

The atmospheric α-pinene oxidation leads to three carboxylic acids: norpinonic acid (NPA), pinic acid (PA), and 3-methyl-1,2,3-butanetricarboxylic acid (MBTCA). In this study, the OH radical kinetics in the aqueous phase of these carboxylic acids were investigated at different temperatures and pH values of solutions. Activation parameters and the corresponding atmospheric lifetimes of the acids in the troposphere were derived. The overall second-order rate constants for the individual speciation forms of the acids (AH and A- for NPA; AH2, AH- and A2- for PA; and AH3, AH2-, AH2- and A3- for MBT

Modulation of fibroblast behavior by Leishmania: a pathway to understanding disease progression in cutaneous leishmaniasis.

Leishmaniasis is a parasitic infection caused by obligatory intracellular protozoa of the Leishmania genus. Macrophages are the main cell for Leishmania parasites that play a key role in immune response against it. Interestingly, both the promastigote and amastigote forms of the parasite have also been detected in fibroblasts, revealing the complex interactions during the infection. Cutaneous leishmaniasis, caused by different Leishmania species, presents different clinical outcomes, largely influenced by the interactions between the parasite and host immune cells. This study aimed to determin

The Effect of Ursolic Acid on Leishmania (Leishmania) amazonensis Is Related to Programed Cell Death and Presents Therapeutic Potential in Experimental Cutaneous Leishmaniasis.

Among neglected tropical diseases, leishmaniasis is one of the most important ones, affecting more than 12 million people worldwide. The available treatments are not well tolerated, and present diverse side effects, justifying the search for new therapeutic compounds. In the present study, the activity of ursolic acid (UA) and oleanolic acid (OA) were assayed in experimental cutaneous leishmaniasis (in vitro and in vivo). Promastigote forms of L. amazonensis were incubated with OA and UA for 24h, and effective concentration 50% (EC50) was estimated. Ultraestructural alterations in Leishmania a

In vitro evaluation of inorganic mercury and methylmercury effects on the intestinal epithelium permeability.

The mercurial forms [inorganic divalent mercury, Hg(II) and methylmercury, CH3Hg] produce neurological and immune effects as well as hematological and renal alterations. The main route of exposure is through the diet. Consequently, the gastrointestinal mucosa is exposed to these mercurial forms, though the potential toxic effects upon the mucosa are not clear. The present study evaluates the toxicity of Hg(II) and CH3Hg (0.1-2 mg/L) in an intestinal epithelium model using the differentiated and undifferentiated human Caco-2 cell line. The experiments made show the mercurial forms generate react

Hemophagocytic syndrome in children and adults.

Hemophagocytic syndrome, also known as hemophagocytic lymphohistiocytosis (HLH), is a heterogenic syndrome, which leads to an acute, life-threatening inflammatory reaction. HLH occurs both in children and adults, and can be triggered by various inherited as well as acquired factors. Depending on the etiology, HLH can be divided into genetic (i.e., primary) and acquired (i.e., secondary) forms. Among genetic HLH forms, one can distinguish between familial HLH and other genetically conditioned forms of HLH. Acquired HLH can be typically triggered by infections, autoimmune diseases, and malignanc

Analysis of differential proteomes in pathogenic and non-pathogenic Leptospira: potential pathogenic and virulence factors.

Leptospirosis is a bacterial zoonotic disease caused by spirochetes in the genus Leptospira. To date, factors determining the pathogenicity and virulence of leptospires remain unclear. We performed a gel-based proteomic analysis to evaluate differential leptospiral proteomes in the pathogenic L. interrogans (serovars Australis, Bratislava, Autumnalis, and Icterohaemorrhagiae) and the non-pathogenic L. biflexa (serovar Patoc). Quantitative proteome analysis and MS protein identification revealed 42 forms of 33 unique proteins whose levels were significantly greater in the pathogenic serovars co

Biochemical characterization of GAF domain of free-R-methionine sulfoxide reductase from Trypanosoma cruzi.

Trypanosoma cruzi is the causal agent of Chagas Disease and is a unicellular parasite that infects a wide variety of mammalian hosts. The parasite exhibits auxotrophy by L-Met; consequently, it must be acquired from the extracellular environment of the host, either mammalian or invertebrate. Methionine (Met) oxidation produces a racemic mixture (R and S forms) of methionine sulfoxide (MetSO). Reduction of L-MetSO (free or protein-bound) to L-Met is catalyzed by methionine sulfoxide reductases (MSRs). Bioinformatics analyses identified the coding sequence for a free-R-MSR (fRMSR) enzyme in the

Cysteine Supplementation May be Beneficial in a Subgroup of Mitochondrial Translation Deficiencies.

BACKGROUND: Mitochondrial encephalomyopathies are severe, relentlessly progressive conditions and there are very few effective therapies available to date. We have previously suggested that in two rare forms of reversible mitochondrial disease (reversible infantile respiratory chain deficiency and reversible infantile hepatopathy) supplementation with L-cysteine can improve mitochondrial protein synthesis, since cysteine is required for the 2-thiomodification of mitochondrial tRNAs. OBJECTIVES: We studied whether supplementation with L-cysteine or N-acetyl-cysteine (NAC) results in any improv

Long-term survival of Legionella pneumophila associated with Acanthamoeba castellanii vesicles.

Legionella pneumophila, the causative agent of Legionnaires' disease, is ubiquitously found in aquatic environments, associated with free living amoebae. Trophozoite forms of the genus Acanthamoeba have been shown to support the intracellular growth of Legionella while it has been proposed that cyst forms are related to survival in harsh environments. This underlines that amoebae are of primary importance in Legionella spreading. In this study, we followed the survival of L. pneumophila Lens over 6 months in a poor medium, with or without Acanthamoeba castellanii. The results demonstrated that

The role of the spleen in a lactate dehydrogenase mutant mouse (Ldh-1c/Ldh-1c) with hemolytic anemia.

The lactate dehydrogenase mouse mutant Ldh-1c/Ldh-1c is afflicted with a severe hemolytic anemia associated with extreme reticulocytosis (95%) and splenomegaly. Ninety-one percent of the total body colony-forming units--erythroid (CFU-E) have been quantified in the seven- to ten-times enlarged spleens of the mutant mice. Moreover, the splenic fraction of morphologically recognizable erythroid precursors was 134 times normal. From these data it was apparent that the spleen crucially contributes to the maintenance of steady state erythropoiesis in the mutants. On the other hand, an enhanced sequ
