[Effect of aminazine and triftazin on the synaptosome membranes of the rat cerebral cortex].

Fluorescent probes 1,6-diphenyl-1,3,5-hexatriene (DPH) and pyrene were employed in studying the effect of aminazine and triftazin versus that of imipramine on microviscosity of rat brain cortex synaptosomal membranes. Unlike imipramine, the neuroleptics decrease microviscosity of membrane's lipid bilayer. All drugs decrease fluorescence of endogenous tryptophan, but fail to change fluorescence of L-tryptophan in the solution. It is concluded that neuroleptics induce conformational perturbations in membrane-bound proteins modifying microviscosity of lipid bilayer whereas imipramine changes the

Probing Phospholipid Bilayer Structure and Interaction Using ATR-FTIR Spectroscopy.

The cell membrane plays a critical role in maintaining cellular integrity and regulating interactions between the cell and its environment. The understanding of many biological processes relies on obtaining molecular level information on the structure and dynamic behaviour of membranes, interactions between molecules within the membrane, as well as interactions between membrane and external compounds. Attenuated Total Reflection Fourier Transform Infrared (ATR-FTIR) spectroscopy has emerged as a powerful analytical technique for investigating biological membranes due to its surface sensitivity

Cellular uptake of nanoparticles by membrane penetration: a study combining confocal microscopy with FTIR spectroelectrochemistry.

It is well-known that nanomaterials are capable of entering living cells, often by utilizing the cells' endocytic mechanisms. Passive penetration of the lipid bilayer may, however, occur as an alternative process. Here we have focused on the passive transport of small nanoparticles across the plasma membranes of red blood cells, which are incapable of endocytosis. By using fluorescence microscopy, we have observed that zwitterionic quantum dots penetrate through the cell membranes so that they can be found inside the cells. The penetration-induced structural changes of the lipid bilayer were e

Familial mutations modulate α-synuclein interactions with key neuronal membrane lipids.

Progressive aggregation of α-synuclein (α-Syn) in the midbrain, hypothalamus and thalamus is linked to Parkinson's disease (PD), one of the fastest growing neurodegenerative diseases in the U.S. Studies of families with PD history revealed several mutations that are responsible for the early-onset (A30P, E46K, A53T) and late-onset (H50Q) forms of PD. A growing body of evidence indicates that phospho-/sphingolipids and cholesterol alter the aggregation properties of wild-type (WT) α-syn. However, the effects of these lipids on the rate of α-syn mutants remain unclear. In the current study, we d

Membrane binding and insertion of the predicted transmembrane domain of human scramblase 1.

Human phospholipid scramblase 1 (SCR) was originally described as an intrinsic membrane protein catalyzing transbilayer phospholipid transfer in the absence of ATP. More recently, a role as a nuclear transcription factor has been proposed for SCR, either in addition or alternatively to its capacity to facilitate phospholipid flip-flop. Uncertainties exist as well from the structural point of view. A predicted α-helix (aa residues 288-306) located near the C-terminus has been alternatively proposed as a transmembrane domain, or as a protein core structural element. This paper explores the possi

TMEM41B acts as an ER scramblase required for lipoprotein biogenesis and lipid homeostasis.

How amphipathic phospholipids are shuttled between the membrane bilayer remains an essential but elusive process, particularly at the endoplasmic reticulum (ER). One prominent phospholipid shuttling process concerns the biogenesis of APOB-containing lipoproteins within the ER lumen, which may require bulk trans-bilayer movement of phospholipids from the cytoplasmic leaflet of the ER bilayer. Here, we show that TMEM41B, present in the lipoprotein export machinery, encodes a previously conceptualized ER lipid scramblase mediating trans-bilayer shuttling of bulk phospholipids. Loss of hepatic TME

Membrane lipid alterations in hemoglobinopathies.

The red blood cell (RBC) membrane is a complex mixture of lipids and proteins. Hundreds of phospholipid molecular species spontaneously arrange themselves in a lipid bilayer and move rapidly in the plane as well as across the bilayer in a dynamic but highly organized fashion. Areas enriched in certain lipids determine proper protein function. Phospholipids are asymmetrically distributed across the lipid bilayer with phosphatidylserine (PS) exclusively on the inside. Both the composition and organization of the RBC membrane is well maintained. Alterations lead to apoptosis during erythropoiesis

Confocal Raman Microscopy of Hybrid-Supported Phospholipid Bilayers within Individual C18-Functionalized Chromatographic Particles.

Measuring lipid-membrane partitioning of small molecules is critical to predicting bioavailability and investigating molecule-membrane interactions. A stable model membrane for such studies has been developed through assembly of a phospholipid monolayer on n-alkane-modified surfaces. These hybrid bilayers have recently been generated within n-alkyl-chain (C18)-modified porous silica and used in chromatographic retention studies of small molecules. Despite their successful application, determining the structure of hybrid bilayers within chromatographic silica is challenging because they reside

Groove architecture controls lipid scrambling in simulations of protein and model systems.

Lipid bilayers are essential to life as they surround most cells and membrane-bound organelles. The integrity and fate of cells depend on the asymmetric makeup of lipid bilayers with various membrane proteins regulating the lipid composition of a bilayer's two leaflets. Lipids scramblases are one of the primary regulators of lipid asymmetry in bilayers, spontaneously transferring lipids between membrane leaflets. Members of the TMEM16, OSCA/TMEM63, and TMC families have been suggested to be lipid scramblases. Despite significant differences, these proteins share a common structural architectur

Functionalization of cobalt porphyrin-phospholipid bilayers with his-tagged ligands and antigens.

Methods to attach polypeptides to lipid bilayers are often indirect and ineffective, and can represent a substantial bottleneck in the formation of functionalized lipid-based materials. Although the polyhistidine tag (his-tag) has been transformative in its simplicity and efficacy in binding to immobilized metals, the successful application of this approach has been challenging in physiological settings. Here we show that lipid bilayers containing porphyrin-phospholipid conjugates that are chelated with cobalt, but not with other metals, can effectively capture his-tagged proteins and peptides

Exploring photosensitization as an efficient antifungal method.

Lipid bilayers containing ergosterol show signs of destruction when they are treated with singlet oxygen, due to the conversion of ergosterol into its peroxy derivative. Applying this previous knowledge, an antifungal method was explored using Candida tropicalis as model, and membrane permeation under photosensitization conditions became evident. These data were complemented through AFM images of artificial lipid bilayers, using cholesterol or ergosterol as structural sterols, showing their corresponding morphologies at the nanoscale. Based on these results, an antifungal method was developed,

Curvature-Induced Membrane Remodeling by the Cell-Penetrating Peptide Pep-1.

The cell-penetrating peptide Pep-1 interacts with lipid membranes through combined electrostatic and hydrophobic forces, yet the structural details of its membrane remodeling activity remain unclear. Using atomic force microscopy (AFM), we examined how Pep-1 perturbs supported lipid bilayers of varying composition and geometry. In zwitterionic POPC bilayer patches, Pep-1 preferentially targeted patch boundaries, where lipid packing is less constrained, leading to edge erosion and detergent-like disintegration. Incorporation of anionic POPS enhanced peptide binding and localized disruption, giv

Factors Affecting Enhanced Permeation of Amphotericin B Across Cell Membranes and Safety of Formulation.

The aim of this study was to determine amphotericin B (AmB) permeation across lipid bilayer membranes mounted on Transwell® and to observe the phagocytosis of the AmB and the AmB-lipid formulations by alveolar macrophage (AM) cell lines using a fluorescence microscope. The lipid bilayer membranes were prepared from phospholipid and ergosterol as well as phospholipid and cholesterol in a ratio (67:33 mol%). AmB-lipid formulations were prepared from AmB incorporated with four lipid derivatives during a lyophilization process. In vitro cytotoxicity studies were carried out on kidney cells by the

Reaction rate calculations indicate that α-tocopherol primarily acts as a membrane protein antioxidant in vivo.

Lipid peroxidation is a complex free radical chain reaction in which lipid peroxyl radicals serially attack other components of the lipid bilayer. As a condensed phase, the lipid bilayer contains numerous potentially reactive substrates beyond unsaturated fatty acids, including membrane proteins and dissolved small molecules like α-tocopherol, the major lipid-soluble antioxidant. Despite a long history of investigation, the reaction cascades that do succeed in vivo to produce tocopherol's pronounced antioxidant effect are still arguable. Here, we have revisited the tocopherol reaction cascade

Examining the thermotropic properties of large circularized nanodiscs.

Nanodiscs, soluble membrane mimetics composed of an amphipathic membrane scaffold protein encircling a lipid bilayer, are widely used in biophysical and structural studies of membrane proteins. Because many membrane proteins are responsive to their membrane environment, through specific protein-lipid interactions and bulk membrane shape and structure, it is important to understand the properties of lipid bilayers contained within nanodiscs in order to interpret studies using this technology. Nanodiscs are known to alter lipid properties, such as membrane thickness and melting temperature, and

Deciphering Melatonin-Stabilized Phase Separation in Phospholipid Bilayers.

Lipid bilayers are fundamental building blocks of cell membranes, which contain the machinery needed to perform a range of biological functions, including cell-cell recognition, signal transduction, receptor trafficking, viral budding, and cell fusion. Importantly, many of these functions are thought to take place in the laterally phase-separated regions of the membrane, commonly known as lipid rafts. Here, we provide experimental evidence for the "stabilizing" effect of melatonin, a naturally occurring hormone produced by the brain's pineal gland, on phase-separated model membranes mimicking

Spherical Nanovesicles Transform into a Multitude of Nonspherical Shapes.

Nanovesicles are closed, bubblelike surfaces with a diameter between 20 and 200 nm, formed by lipid bilayers and biomembranes. Electron microscopy (EM) studies have shown that these vesicles can attain both spherical and nonspherical shapes. One disadvantage of EM methods is that they provide only a single snapshot of each vesicle. Here, we use molecular dynamics simulations to monitor the morphological transformations of individual nanovesicles. We start with the assembly of spherical vesicles that enclose a certain volume of water and contain a certain total number of lipids. When we reduce

When barriers ignore the "rule-of-five".

Why are a few drugs with properties beyond the rule of 5 (bRo5) absorbed across the intestinal mucosa while most other bRo5 compounds are not? Are such exceptional bRo5 compounds exclusively taken up by carrier-mediated transport or are they able to permeate the lipid bilayer (passive lipoidal diffusion)? Our experimental data with liposomes indicate that tetracycline, which violates one rule of the Ro5, and rifampicin, violating three of the rules, significantly permeate a phospholipid bilayer with kinetics similar to labetalol and metoprolol, respectively. Published data from experimental wo

Sphingomyelin and GM1 Influence Huntingtin Binding to, Disruption of, and Aggregation on Lipid Membranes.

Huntington disease (HD) is an inherited neurodegenerative disease caused by the expansion beyond a critical threshold of a polyglutamine (polyQ) tract near the N-terminus of the huntingtin (htt) protein. Expanded polyQ promotes the formation of a variety of oligomeric and fibrillar aggregates of htt that accumulate into the hallmark proteinaceous inclusion bodies associated with HD. htt is also highly associated with numerous cellular and subcellular membranes that contain a variety of lipids. As lipid homeostasis and metabolism abnormalities are observed in HD patients, we investigated how va

A coarse-grained molecular dynamics study of nanoscale phospholipid-cholesterol vesicles: curvature effects on cholesterol distribution and vesicle structure.

Nanoscale lipid bilayer vesicles are widely used as model systems for exosomes and synaptic vesicles, released by cells for intercellular communication, in which strong membrane curvature couples molecular packing to asymmetry between the two lipid-bilayer monolayers. Here, we investigate DPPC, POPC or DLiPC phospholipid (PC)/cholesterol bilayer vesicles using coarse-grained molecular dynamics simulations, systematically varying phospholipid chain unsaturation, cholesterol content (0-50 mol%), and vesicle diameter (10-30 nm for the DPPC system). Cholesterol undergoes rapid redistribution on su
