Trichomonas vaginalis: metronidazole and other nitroimidazole drugs are reduced by the flavin enzyme thioredoxin reductase and disrupt the cellular redox system. Implications for nitroimidazole toxicity and resistance.

Infections with the microaerophilic parasite Trichomonas vaginalis are treated with the 5-nitroimidazole drug metronidazole, which is also in use against Entamoeba histolytica, Giardia intestinalis and microaerophilic/anaerobic bacteria. Here we report that in T. vaginalis the flavin enzyme thioredoxin reductase displays nitroreductase activity with nitroimidazoles, including metronidazole, and with the nitrofuran drug furazolidone. Reactive metabolites of metronidazole and other nitroimidazoles form covalent adducts with several proteins that are known or assumed to be associated with thiored

Effect of chloridazone on the animal organism.

The acute toxic effect of the herbicide chloridazone and mitochondrial respiration were investigated and typical clinical signs of intoxication were described in rats (Wistar), pheasants (Phasianus colchicus) and sheep (Slovak Merino). The LD50 of chloridazone was calculated to be for rats 800 mg/kg bw (range 552 to 1160 mg/kg bw) and for pheasants 3684 mg/kg bw (range 1768 to 7677 mg/kg bw). According to WHO chloridazone is moderately toxic for rats and slightly toxic for pheasants. The LD50 for sheep is 161 mg/kg bw (range 76 to 340 mg/kg bw). Chloridazone thus presents an acute risk for rum

Parkinson-like disease by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) toxicity in Macaca fascicularis: synaptosomal metabolism and action of dihydroergocriptine.

The maximal rates (Vmax) of some enzyme activities related to synaptosomal energy metabolism were studied in different types of synaptosomes from cerebellar cortex of Macaca Fascicularis (Cynomolgus monkey). Different synaptosomal populations, namely "large" and "small" synaptosomes, were isolated from the anterior lobule of the cerebellar cortex of monkeys treated p.o. with dihydroergocriptine at the dose of 12 mg/kg/day before and during the induction of a Parkinson's-like syndrome by MPTP administration (i.v., 0.3 mg/kg/day for 5 days). The enzymes were chosen according to their regulatory

The relationship between the cardiac contractile function, adenine nucleotides and amino acids of cardiac tissue and mitochondria at acute respiratory hypoxia.

The effect of asphyxia and subsequent resumption of respiration on the content of adenine nucleotides and some amino acids in heart tissue and mitochondria, as well as respiration of heart mitochondria was studied in rats. The depression of cardiac contractile function during asphyxia showed a better correlation with losses in mitochondrial adenine nucleotides (ATP + ADP + AMP) than those in cardiac tissue. The decrease in the heart work index was accompanied by a decrease in state 3 respiration with glutamate and malate as well as uncoupled respiration with these substrates. This did not occu

Changes in enzyme pattern of Ehrlich ascites tumor cells following serial cultivation in media with increased (hypertonic) NaCl content.

Adaptation of Ehrlich ascites tumor cells to serial cultivation in media with progressively elevated (hypertonic) NaCl content ("high NaCl"-tolerant cells) has resulted in progressive increases of the cellular activities of NAD-dependent glycerol-3-phosphate dehydrogenase (EC 1.1.1.8), NAD-dependent malate dehydrogenase (EC 1.1.1.37), glutamate--oxalacetate transaminase (EC 2.6.1.1), NAD (P)-dependent glutamate dehydrogenase (EC 1.4.1.3), NADP-dependent isocitrate dehydrogenase (EC 1.1.1.42). Hanseniaspora species can create a privileged niche for Oenococcus oeni during triple-strain simultaneous alcoholic-malolactic fermentation based on metabolomics analysis.

Previous studies have demonstrated that non-Saccharomyces yeasts can enhance malolactic fermentation (MLF) efficiency during co-fermentation. However, the underlying mechanisms remain unclear. Here, we demonstrate how specific strains create favorable metabolic niches through tripartite fermentation assays involving 16 strains from 8 species co-cultured with Saccharomyces cerevisiae F5 and Oenococcus oeni SD-2a, Hanseniaspora spp. emerged as optimal partners, reducing MLF duration by 50 % (≤48 h vs. 96 h in controls) while increasing O. oeni biomass by > 10-fold. Effects of the addition of fumaric acid on the spontaneous alcoholic fermentation of white grape must.

Consumer demand is driving the production of wines without added sulfur dioxide or yeast inoculation. However, this approach can lead to organoleptic defects or health risks due to unwanted microbial activity during alcoholic fermentation (AF). Lactic acid bacteria (LAB), responsible for malolactic fermentation (MLF), may also affect wine quality. Fumaric acid (FUM) inhibits LAB growth and could serve as an alternative to SO₂ in controlling MLF. The objective of this work was to study the effects of adding FUM (0.6 g/L) to natural white grape must without SO2 on the growth of LAB during sponta

Novel calcium carboxyphosphonate/polycarboxylate inorganic-organic hybrid materials from demineralization of calcitic biomineral surfaces.

Dissolution of biologically important sparingly soluble salts, such as calcium carbonate and calcium oxalate, is possible by use of carboxyl- and carboxyl/phosphonate-bearing, anionic additives, citrate, malate, carboxyphosphonate, and butane tetracarboxylate. Calcium-containing dissolution products have been identified, characterized, and independently synthesized. These are polymeric materials composed of calcium and the additive as the ligand. Their full characterization was carried out by single-crystal X-ray crystallography and other techniques.

Detection of NADH/NAD+ dysregulation in MELAS via diazo-carboxyl click derivatization mass spectrometry.

OBJECTIVES: Mitochondrial encephalomyopathy, lactic acidosis, and stroke-like episodes (MELAS) is a mitochondrial disorder driven by mutations in mitochondrial or nuclear DNA, involving an altered NADH/NAD+-associated redox metabolism as a key pathological mechanism. The traditional metabolomic analyses in MELAS face sensitivity and sample volume limitations, particularly for carboxylic acid metabolites. This study employed a recently established diazo-carboxyl/hydroxylamine-ketone double-click derivatization (DQmB-HA) mass spectrometry method to overcome these barriers, enabling highly sensit

In Situ Imaging for Monitoring Biomass Content and Pellet Size in Submerged Filamentous Stirred-Tank Cultivation.

Filamentous microorganisms play a central role in industrial biotechnology, where pellet morphology affects mass transfer, rheology, and process performance. However, time-resolved monitoring of filamentous biomass and pellet size dynamics in stirred-tank reactors remains challenging due to offline sampling limitations. We present a noninvasive in situ imaging approach to observe biomass-related light scattering and projected pellet size. A custom-built camera and ring-light setup with MATLAB-based image acquisition and processing was used to quantify image luminosity as a proxy for biomass co

Tunable and Efficient CO2 Delivery Supports Robust Malic Acid Production by Escherichia coli in a Hollow Fiber Membrane Bioreactor.

Malic acid, a four-carbon dicarboxylic acid, represents a top value-added chemical from biomass and important building-block molecule for various industrial chemical processes. Currently, however, it is primarily produced from petrochemicals via an unsustainable process with high greenhouse gas intensity. Alternatively, malic acid can be produced from renewable biomass sugars via anaerobic fermentation by engineered Escherichia coli. While inorganic carbon (Ci) serves as an essential co-substrate in this carbon-negative process, its typical delivery methods suffer from several key limitations.

Assessing Lanthanide-Dependent Methanol Dehydrogenase Activity: The Assay Matters.

Artificial dye-coupled assays have been widely adopted as a rapid and convenient method to assess the activity of methanol dehydrogenases (MDH). Lanthanide(Ln)-dependent XoxF-MDHs are able to incorporate different lanthanides (Lns) in their active site. Dye-coupled assays showed that the earlier Lns exhibit a higher enzyme activity than the late Lns. Despite widespread use, there are limitations: oftentimes a pH of 9 and activators are required for the assay. Moreover, Ln-MDH variants are not obtained by isolation from the cells grown with the respective Ln, but by incubation of an apo-MDH wit

Preparation and characterization of the NAD vinylogue, 3-pyridylacryloamide adenine dinucleotide.

The vinylogue of NAD, 3-pyridylacryloamide adenine dinucleotide, was prepared from NAD and 3-pyridylacryloamide through the snake venom NADase-catalyzed transglycosidation reaction. The analog, purified by ion-exchange chromatography, was obtained in a 55% yield. The cyanide adduct and reduced form of the analog exhibited absorbance maxima at 358 nm and 378 nm, respectively, with extinction coefficients in each case being 2.3-times higher than those reported for the corresponding NAD derivatives. 3-Pyridylacryloamide adenine dinucleotide served as a coenzyme with bovine liver glutamic dehydrog

Independent regulation of synthesis of form I and form II ribulose bisphosphate carboxylase-oxygenase in Rhodopseudomonas sphaeroides.

Ribulose 1,5-bisphosphate carboxylase-oxygenase (RuBPC-O) activity was greatly enhanced when Rhodopseudomonas sphaeroides was grown in a mineral salts medium supplied with 1.5% CO2 in hydrogen. Analysis of cell extracts by sodium dodecyl sulfate-polyacrylamide gel electrophoresis indicated that cells growing on 1.5% CO2 in H2 specifically accumulated RuBPC-O polypeptides. Quantitative immunological determinations revealed that accumulation of form I and form II RuBPC-O closely correlates with the increase of specific activity. However, the two enzymes appeared to be derepressed at different le

Vitamin A deficiency and tri-iodothyronine action at the cellular level in the rat.

Young rats were given a diet with or without vitamin A for a period of 7 weeks. In rats on the vitamin A-deficient diet, the formation of tri-iodothyronine (T3) complexes with nuclear proteins was reduced, and Scatchard analysis showed a decreased capacity of nuclear T3 receptors. In these rats, a decrease in growth and a decrease in the activity of lipogenic enzymes (glucose-6-phosphate dehydrogenase, phosphogluconate dehydrogenase and L-malate dehydrogenase) occurred, which could be related to the decreased formation of T3-nuclear protein complexes.

The possible metabolic diversions adapted by the cockroach, Periplaneta americana to counteract the toxicity of fenvalerate.

Effects of sublethal doses of fenvalerate through topical application were monitored in the central nervous system (CNS) of P. americana. A decrease in total and soluble proteins with an increase in free amino acids, alanine aminotransferase (AlAT) and aspartate aminotransferase (AAT) was observed during fenvalerate toxicity. Further the levels of glycogen, pyruvate and activities of succinate dehydrogenase (SDH) and malate dehydrogenase (MDH) dropped significantly. Lactate content and lactate dehydrogenase (LDH) activity also showed an elevation following fenvalerate toxicity.

Inactivation of thioredoxin by sulfite ions.

Oxidized thioredoxin undergoes sulfitolysis of its single disulfide bond at low concentrations of sulfite ions and protein and in the absence of denaturing agents. The reaction, which has an optimum at pH 8, was studied using [35S]sulfite and E. coli thioredoxin as model. The product, thioredoxin-S-sulfonate, has a half-life of several hours in solution. It is unable to activate chloroplast NADP malate dehydrogenase. Thioredoxin sulfitolysis may therefore be a physiologically important factor in mediating the phytotoxic effects of sulfur dioxide in plants.
