Trace-Cu(II)/CaO2 pre-oxidation coupled with coagulation for algal elimination: A triple-mechanism cascade involving functional inactivation driven by Cu(III).

Harmful algal blooms release microcystins and other metabolites that threaten aquatic ecosystems and human health. Conventional removal methods often induce cell lysis and are affected by matrix interference in natural waters, limiting their practical applicability. A sequential process coupling trace-Cu(II)/CaO2 pre-oxidation with PAC coagulation was developed for efficient algal removal. The system removed 98.5% of M. aeruginosa from lake water. During the 10-min pre-oxidation stage, trace-Cu(II)/CaO2 effectively degraded extracellular microcystin-LR (MC-LR) and the dissolved organic matter

B-PER tandem assembly CRISPR/Cas12a cascade amplification strategy based fluorescence/colorimetric dual-mode for detection of MC-LR.

Microcystins are the most common, most powerful and most toxic types of cyanobacteria, which seriously threaten the public health and ecological environment. Biosensor has been widely used in Microcystin-Leucine-Arginine (MC-LR) detection. In this study, we report an efficient single-hairpin with double primers (the trigger chain T hybridizes with the left end a, and the amplified product ab hybridizes with the right end a) for bidirectional PER (B-PER) amplification strategy for uniform visual and fluorescence detection of trace amounts of MC-LR in the environment using quantum dots (QDs) as

Adverse outcome pathway of Alzheimer's disease-like changes resulting from autophagy flux blockade after MC-LR exposure.

Microcystins (MCs) pollution is a worldwide environmental issue concerning about human health. Microcystin-leucine-arginine (MC-LR), the most common type of MCs produced by cyanobacteria, could enter the brain and bring about damage to the nervous system. Up to date, it is not clear about the mechanism of MC-LR-induced neurotoxicity. Amyloid-β (Aβ) deposits are hallmark of Alzheimer's disease (AD). In this study, we revealed that MC-LR exposure at environment-related doses (1, 7.5, 15 μg/L) could promote Aβ accumulation in mouse brain. Mechanically, we firstly found that Aβ accumulation is clo

Sperm PP1gamma2 is regulated by a homologue of the yeast protein phosphatase binding protein sds22.

Serine/threonine phosphatase PP1gamma2 is a testis-specific protein phosphatase isoform in spermatozoa. This enzyme appears to play a key role in motility initiation and stimulation. Catalytic activity of PP1gamma2 is higher in immotile compared with motile spermatozoa. Inhibition of PP1gamma2 activity causes both motility initiation and motility stimulation. Protein phosphatases, in general, are regulated by their binding proteins. The objective of this article is to understand the mechanisms by which PP1gamma2 is regulated, first by identifying its regulatory proteins. We had previously show

On the regulation of the expressed L-type calcium channel by cAMP-dependent phosphorylation.

The Ca2+ channel subunits alpha 1C-a and alpha 1C-b were stably expressed in Chinese hamster ovary (CHO) and human embryonic kidney (HEK) 293 cells. The peak Ba2+ current (IBa) of these cells was not affected significantly by internal dialysis with 0.1 mM cAMP-dependent protein kinase inhibitor peptide (mPKI), 25 microM cAMP-dependent protein kinase catalytic subunit (PKA), or a combination of 25 microM PKA and 1 microM okadaic acid. The activity of the alpha 1C-b channel subunit expressed stably in HEK 293 cells was depressed by 1 microM H 89 and was not increased by superfusion with 5 microM

Reduced Zebrafish Transcriptome Atlas toward Understanding Environmental Neurotoxicants.

Transcriptomic approaches monitoring gene responses at genome-scale are increasingly used in toxicological research and help to clarify the molecular mechanisms of adverse effects caused by environmental toxicants. However, their applications for chemical assessment are hampered due to high expenses required and more importantly the lack of in-depth data mining and mechanistic perspectives. Here, we described a reduced transcriptome atlas (RTA) approach which integrates transcriptomic data sets and a comprehensive panel of genes generated to represent neurogenesis and the early neuronal develo

Enhanced coagulation of Microcystis aeruginosa using titanium xerogel coagulant.

Cyanobacterial blooms are prevalent globally and present a significant threat to water security. Titanium salt coagulants have garnered considerable attention due to their superior coagulation properties and the absence of metal residue risks. This paper explored the influencing factors in the coagulation process of titanium xerogel coagulant (TXC), the alterations in cell activity during floc storage, and the release of cyanobacterial organic matters, thereby determining the application scope of TXC for cyanobacterial water treatment. The findings indicated that at a TXC dosage of 8 mg Ti/L,

Microcystis genotypes in a tropical freshwater lake: Discovery of novel MIB-producing Microcystis with potentially unique synthesis pathway.

Harmful algal blooms (HABs) are a threat to freshwater systems over the world due to the production of hepatotoxins like microcystin (MC), and nuisance taste and odour (T&O) compounds like 2-methylisoborneol (MIB). While MCs are known to cause detrimental effects to both water quality and human health, MIB is only reported to cause aesthetical problems. In this study, we investigated a tropical, urban lake that was experiencing persistent MC and MIB events. Although it was dominated by Microcystis blooms, analysis revealed that the toxigenic Microcystis were not the only species driving the MC

Inhibition of PP2A ameliorates intervertebral disc degeneration by reducing annulus fibrosus cells apoptosis via p38/MAPK signal pathway.

BACKGROUND: Intervertebral disc degeneration (IVDD) is considered one of the main reasons for low back pain (LBP). To date, the specific pathology of IVDD remains unclear. The annulus fibrosus (AF) is an important part of the intervertebral disc, and AF cell oxidative stress, apoptosis plays a vital role in disc degeneration. Protein phosphatase 2 A (PP2A), a serine/threonine phosphatase, has regulatory functions in various processes, including apoptosis and autophagy. However, thus far, the effect of PP2A on IVDD is not clear. METHODS: AF cells derived from caudal intervertebral discs in SD

Multilocus sequence typing (MLST) reveals high genetic diversity and clonal population structure of the toxic cyanobacterium Microcystis aeruginosa.

Microcystis aeruginosa is one of the most prevalent bloom-forming cyanobacteria and has been the cause of increasing public health concern due to the production of hepatotoxins (microcystins). To investigate the genetic diversity, clonality and evolutionary genetic background with regard to the toxicity of M. aeruginosa, a multilocus sequence typing (MLST) scheme was developed, based on seven selected housekeeping loci (ftsZ, glnA, gltX, gyrB, pgi, recA and tpi). Analysis of a collection of 164 isolates from Japan and other countries identified 79 unique sequence types (STs), revealing a high

Evaluation of methods for the isolation, detection and quantification of cyanobacterial hepatotoxins.

Cyanobacterial hepatotoxins such as microcystins and nodularins have been responsible for the poisoning of both animals and humans who ingest or come into contact with toxic blooms. They are extremely stable in water due to their stable chemical structure and can tolerate radical changes in water chemistry, including pH and salinity. Different methods for the extraction and detection of these compounds have been reported. Extraction methods utilizing both aqueous and organic solvent systems have been reported. The detection methods ranging from immunological or biochemical assays such as enzym

Metabolomic analysis reveals the toxicity mechanisms of bisphenol A on the Microcystis aeruginosa under different phosphorus levels.

Harmful cyanobacterial blooms have been a global environmental problem. Discharge of anthropogenic pollutants and excess nutrient import into the freshwater bodies may be the biggest drivers of bloom. Bisphenol A (BPA), a typical endocrine-disrupting compound, is frequently detected in different natural waters, which was a threat to the balance of aquatic ecosystem. Yet mechanistic understanding of the bloom and microcystin generation under combined pollution conditions is still a mystery. Herein, the cellular and metabolomic responses to BPA exposure and phosphorus (P) levels in Microcystis a

Solar-light-driven inactivation of toxigenic Microcystis by floating photocatalytic mesh system: Insights on physiological mechanism and application.

Harmful algal blooms (HABs) pose serious ecological and health risks with releasing waterborne toxins. In this study, a novel floating Bi2WO6-based photocatalytic mesh (BPM) system was developed for efficient water purification by targeting the inactivation of HABs-dominated Microcystis and detoxification of associated microcystins (MCs) under solar light irradiation. The superior flotation, durability and sunlight utilization of the BPM system were achieved by its super-hydrophobic surface, robust stainless-steel mesh carrier, and efficient light transmission, respectively. Harmful and beneficial properties of cyanotoxins: Two sides of the same coin.

Cyanotoxins are by definition "harmful agents" produced by cyanobacteria. Their toxicity has been extensively studied and reviewed over the years. Cyanotoxins have been commonly classified, based on their poisonous effects on mammals, into three main classes, neurotoxins, hepatotoxins and dermatotoxins, and, considering their chemical features, mainly identified as peptides, alkaloids and lipopolysaccharides. Here we propose a broader subdivision of cyanotoxins into eight distinct classes, taking into account their molecular structures, biosynthesis and modes of action: alkaloids, non-ribosoma

The coexistence of Bacillus haynesii and Bacillus tequilensis achieves synchronous algicidal activity and denitrification and enhances the MC-LR degradation of phycosphere microbiota.

The key challenge in current biological technologies for the control of harmful algal blooms (HABs) is that algicidal bacteria's lytic mechanisms during bloom treatment results in structural rupture of algal cells. This results in the massive release of intracellularly stored nutrients (such as nitrogen/phosphorus compounds) and secondary metabolites such as microcystins into aquatic environments, causing secondary pollution. This study isolated two Bacillus strains (Bacillus haynesii and Bacillus tequilensis) from phycosphere microbiota, applying them synergistically against Microcystis aerug

Contaminant Exposure and Liver and Kidney Lesions in North American River Otters in the Indian River Lagoon, Florida.

The harmful algal bloom (HAB) liver toxin microcystin (MC) and trace element biomagnification were previously detected in organisms in the Indian River Lagoon (IRL), Florida. Since there are no routine screening programs for these contaminants, liver tissue from North American river otters (Lontra canadensis), an important sentinel species in the IRL, was screened for MC via enzyme-linked immunoassay (ELISA), followed by confirmatory analyses via liquid-chromatography/mass spectrometry methods (LC-MS/MS). Liver and kidney samples were evaluated for trace element (As, Cd, Co, Cu, Fe, Hg, Mn, Mo
