Animal models of brain ageing and dementia.

Six animal models of human brain ageing or of age-related diseases, especially Alzheimer's disease, are evaluated. Special emphasis is laid on comparing the neuropathological symptoms found in aged human brains or in Alzheimer patients, with the neuronal symptoms induced experimentally by the different treatments described here. For each model, the experimental methods for induction of the neuropathological symptoms (lipofuscin accumulation, senile plaques, neurofibrillary degeneration, etc.) and for their identification are discussed. In addition, the behavioral significance of each animal mo

Animal models of inner ear vascular disturbances.

Animal models of vascular disorders are identified or developed for the evaluation of functional deficits and morphologic alterations. This information will serve a useful purpose for a better understanding of sudden deafness, Meniere's disease, and presbycusis. The study of microcirculation of the inner ear vessels reveals that their responses to various stimuli, such as anoxia, sympathetic nerve stimulation, hypothermia, and drugs, are different from those of the middle ear vessels. In sudden occlusion of the major vascular supply to the inner ear, the cochlea is found to be more vulnerable

Predictive experimental animal tumor models: a concept.

As an introduction to the session on Animal Tumor Models, a concept of animal model systems in presented that differentiates between models of organ-related diseases such as prostate or bladder cancer and models of a certain class of neoplasms found within a specific organ disease. Thus defined, no one animal tumor can completely represent and be predictive for a disease entity, no more so than can a single clinical experience with prostate cancer, e.g., be representative of all clinical cases. Rather, a block of animal tumors originating from a specific organ that reflects a spectrum of caref

Animal Models of Exercise and Cardiometabolic Disease.

Cardiometabolic diseases, encompassing cardiovascular and metabolic disorders, represent a significant global health challenge, driven in part by the rising prevalence of obesity and type 2 diabetes. Exercise offers profound benefits in mitigating the progression and impact of cardiometabolic diseases by improving glucose homeostasis, body composition, and cardiovascular function. However, gaps remain in our understanding of the molecular mechanisms underlying these benefits and how to optimize exercise prescriptions for cardiometabolic disease. Animal models are indispensable tools for unrave

[Research progress in animal models of pulpitis].

Pulpitis is a prevalent inflammatory disease in dentistry, and root canal therapy remains the primary clinical treatment for it. However, pulp removal leads to reduced tooth fracture resistance and complications such as secondary infection and tooth fracture. As a potential alternative, vital pulp therapy (VPT) relies on precise assessment of pulp status; yet current clinical diagnostic methods lack specificity. The establishment of appropriate animal models for pulpitis is crucial for investigating its pathogenesis, developing specific diagnostic biomarkers, and optimizing VPT strategies. Thi

Marburg Virus Aerosol Infection in Animal Models.

Aerosol challenge provides the ability to investigate in-depth mucosal virus exposure, including modeling of airborne transmission from high-consequence viral infection. For filoviruses, in particular Marburg and Ebola viruses, aerosol infection is considered in connection to their potential misuse. Aerosol exposure with filoviruses has been used to study disease progression, pathogenicity, and the efficacy of medical countermeasures. The aim of this protocol is to provide a general outline for aerosol exposure of animals with filoviruses, specifically Marburg virus.

Molecular Pathways and Animal Models of Cardiomyopathies.

Cardiomyopathies are a heterogeneous group of disorders of the heart muscle that ultimately result in congestive heart failure. Rapid progress in genetics, molecular and cellular biology with breakthrough innovative genetic-engineering techniques, such as next-generation sequencing and multiomics platforms, stem cell reprogramming, as well as novel groundbreaking gene-editing systems over the past 25 years has greatly improved the understanding of pathogenic signaling pathways in inherited cardiomyopathies. This chapter will focus on intracellular and intercellular molecular signaling pathways

Molecular Pathways and Animal Models of Arrhythmias.

Arrhythmias account for over 300,000 annual deaths in the United States, and approximately half of all deaths are associated with heart disease. Mechanisms underlying arrhythmia risk are complex; however, work in humans and animal models over the past 25 years has identified a host of molecular pathways linked with both arrhythmia substrates and triggers. This chapter will focus on select arrhythmia pathways solved by linking human clinical and genetic data with animal models.

Large animal models for Huntington's disease research.

Huntington's disease (HD) is a hereditary neurodegenerative disorder for which there is currently no effective treatment available. Consequently, the development of appropriate disease models is critical to thoroughly investigate disease progression. The genetic basis of HD involves the abnormal expansion of CAG repeats in the huntingtin ( HTT) gene, leading to the expansion of a polyglutamine repeat in the HTT protein. Mutant HTT carrying the expanded polyglutamine repeat undergoes misfolding and forms aggregates in the brain, which precipitate selective neuronal loss in specific brain region

Animal Models for the Study of Keratoconus.

Keratoconus (KC) is characterized by localized, central thinning and cone-like protrusion of the cornea. Its precise etiology remains undetermined, although both genetic and environmental factors are known to contribute to disease susceptibility. Due to KC's complex nature, there is currently no ideal animal model to represent both the corneal phenotype and underlying pathophysiology. Attempts to establish a KC model have involved mice, rats, and rabbits, with some additional novel animals suggested. Genetic animal models have only been attempted in mice. Similarly, spontaneously occurring ani

Animal models in systemic sclerosis: an update.

PURPOSE OF REVIEW: Systemic sclerosis (SSc) is a multisystem autoimmune connective tissue disease characterized by early inflammation followed by excessive fibrosis in the skin and internal organs. Enhancing our comprehension of SSc pathogenesis is essential to develop effective therapeutic strategies. Animal models that mimic one or more aspects of SSc have been proven to be a valuable resource for investigating disease mechanisms. This review aims to provide an updated overview of the existing SSc animal models and the potentially relevant pathways to SSc pathogenesis. RECENT FINDINGS: This

Animal models of pediatric abusive head trauma.

BACKGROUND: Abusive head trauma (AHT), previously known as the shaken baby syndrome, is a severe and potentially fatal form of traumatic brain injury in infant children who have been shaken, and sometimes also sustained an additional head impact. The clinical and autopsy findings in AHT are not pathognomonic and, due to frequent obfuscation by perpetrators, the circumstances surrounding the alleged abuse are often unclear. The concept has evolved that the finding of the combination of subdural hemorrhage, brain injury, and retinal hemorrhages ("the triad") is the result of shaking of an infant

Animal Models of Cardiovascular Complications of Pregnancy.

Cardiovascular complications of pregnancy have risen substantially over the past decades, and now account for the majority of pregnancy-induced maternal deaths, as well as having substantial long-term consequences on maternal cardiovascular health. The causes and pathophysiology of these complications remain poorly understood, and therapeutic options are limited. Preclinical models represent a crucial tool for understanding human disease. We review here advances made in preclinical models of cardiovascular complications of pregnancy, including preeclampsia and peripartum cardiomyopathy, with a

Understanding main pregnancy complications through animal models.

Since human pregnancy is an inefficient process, achieving desired and pleasant outcome of pregnancy - the birth of a healthy and fit baby - is the main goal in any pregnancy. Spontaneous pregnancy failure is actually the most common complication of pregnancy and Most of these pregnancy losses are not known. Animal models have been utilized widely to investigate the system of natural biological adaptation to pregnancy along with increasing our comprehension of the most important hereditary and non-hereditary factors that contribute to pregnancy disorders. We use model organisms because their c

Application of Animal Models in Diabetic Cardiomyopathy.

Diabetic heart disease is a growing and important public health risk. Apart from the risk of coronary artery disease or hypertension, diabetes mellitus (DM) is a well-known risk factor for heart failure in the form of diabetic cardiomyopathy (DiaCM). Currently, DiaCM is defined as myocardial dysfunction in patients with DM in the absence of coronary artery disease and hypertension. The underlying pathomechanism of DiaCM is partially understood, but accumulating evidence suggests that metabolic derangements, oxidative stress, increased myocardial fibrosis and hypertrophy, inflammation, enhanced

[Radiation Carcinogenesis in Animal Models: Part 1].

Exposure to ionizing radiation (IR) increases the risk of cancers, as epidemiology studies of atomic bomb survivors and patients who have received radiotherapy show. The carcinogenic effects of IR are well-documented, although the effects of radiation carcinogenesis change in each organ. The mammary gland is known to be highly susceptible to radiation-induced cancer. We have previously reported that (i) differential DNA methylation patterns in rat mammary carcinomas induced by pre-and post-pubertal IR; (ii) the effect of parity on rat mammary carcinogenesis varies between pre-and post-pubertal
