Phloroglucinol inhibits oxytocin-induced contraction in rat gastric circular muscle and uterine smooth muscle.

Phloroglucinol is commonly used to alleviate dysmenorrhoea and stomach cramps. However, there is little evidence of phloroglucinol in the mechanism of primary dysmenorrhoea (PD) development. In this study, a PD rat model was established. The effects of phloroglucinol on the contraction of rat gastric circular muscle and uterine smooth muscle induced by oxytocin (OT) were investigated. The writhing response, and levels of oestradiol (E2), prostaglandin e2 (PGE2), and prostaglandin f2α (PGF2α) were determined. The protein and mRNA levels of OT receptor (OTR) were detected. OT showed a significan

The airway smooth muscle and the pipe dream of better bronchodilators.

Research on airway smooth muscle has traditionally focused on its putative detrimental role in asthma, emphasizing on how its shortening narrows the airway lumen, without much consideration about its potential role in subserving the function of the entire respiratory system. New experimental evidence on mice suggests that not only the smooth muscle is required to sustain life postnatally, but its stiffening effect on the lung tissue also protects against excessive airway narrowing and, most importantly, against small airway narrowing heterogeneity and closure. These results suggest that the sm

Nasal Smooth Muscle Hamartoma in a Dog: a Novel Subtype.

A 3-year-old, female spayed, Pug-cross dog presented with a 2-year history of nasal congestion and focal facial deformity. Complete surgical excision was elected following nasopharyngoscopy, endoscopic biopsy and subsequent computed tomography (CT) scan. Nasopharyngoscopy revealed a friable, polypoid mass of the right nasal conchae occluding the right nasal passage. Biopsies of the mass showed expansion of the submucosa by dense bundles of well-differentiated smooth muscle cells that were strongly immunoreactive for ⍺-smooth muscle actin and desmin. Post-procedure CT revealed a large space-occ

Fate and State of Vascular Smooth Muscle Cells in Atherosclerosis.

Vascular smooth muscle cells (VSMCs) have long been associated with phenotypic modulation/plasticity or dedifferentiation. Innovative technologies in cell lineage tracing, single-cell RNA sequencing, and human genomics have been integrated to gain unprecedented insights into the molecular reprogramming of VSMCs to other cell phenotypes in experimental and clinical atherosclerosis. The current thinking is that an apparently small subset of contractile VSMCs undergoes a fate switch to transitional, multipotential cells that can adopt plaque-destabilizing (inflammation, ossification) or plaque-st

Development of the smooth muscle layer in the ileum of mouse embryos.

The smooth muscle layer (SML) comprises a significant portion of the intestines and other tubular organs. Whereas epithelial development has recently been extensively studied, SML development has drawn relatively less attention. Previous morphological reports revealed that the inner circular layer (IC) differentiates earlier than the outer longitudinal layer (OL), but detailed development of the SML, including chronological changes in the cell layer number, precise cell orientation, and regional differences in relation to the mesentery, has not been reported. We here observed the development o

Regulation of Airway Smooth Muscle Contraction in Health and Disease.

Airway smooth muscle (ASM) extends from the trachea throughout the bronchial tree to the terminal bronchioles. In utero, spontaneous phasic contraction of fetal ASM is critical for normal lung development by regulating intraluminal fluid movement, ASM differentiation, and release of key growth factors. In contrast, phasic contraction appears to be absent in the adult lung, and regulation of tonic contraction and airflow is under neuronal and humoral control. Accumulating evidence suggests that changes in ASM responsiveness contribute to the pathophysiology of lung diseases with lifelong health

Smooth muscle of the male pelvic floor: An anatomic study.

Knowledge of the anatomy of the male pelvic floor is important to avoid damaging the pelvic floor muscles during surgery. We set out to explore the structure and innervation of the smooth muscle (SM) of the whole pelvic floor using male fetuses. We removed en-bloc the entire pelvis of three male fetuses. The specimens were serially sectioned before being stained with Masson's trichrome and hematoxylin and eosin, and immunostained for SMs, and somatic, adrenergic, sensory and nitrergic nerve fibers. Slides were digitized for three-dimensional reconstruction. We individualized a middle compartme

Airway smooth muscle as an underutilised biomarker: a case report.

BACKGROUND: Severe asthma and chronic obstructive pulmonary disease (COPD) can be challenging to manage, particularly when the clinical features may be similar. With the increased availability of advanced therapies for both entities, it is more important than ever to diagnose and phenotype accurately to inform appropriate treatment decisions. This case highlights the use of endobronchial biopsies to allow for histological evaluation of airways disease, and in particular the role of airway smooth muscle mass as an additional biomarker that could facilitate the diagnostic process. KV channels and the regulation of vascular smooth muscle tone.

VSMCs in resistance arteries and arterioles express a diverse array of KV channels with members of the KV 1, KV 2 and KV 7 families being particularly important. Members of the KV channel family: (i) are highly expressed in VSMCs; (ii) are active at the resting membrane potential of VSMCs in vivo (-45 to -30 mV); (iii) contribute to the negative feedback regulation of VSMC membrane potential and myogenic tone; (iv) are activated by cAMP-related vasodilators, hydrogen sulfide and hydrogen peroxide; (v) are inhibited by increases in intracellular Ca2+ and vasoconstrictors that signal through Gq

A short review of adipokines, smooth muscle and uterine contractility.

Obesity is a major health problem worldwide. The prevalence of obesity is increasing in both developed and developing countries. In the UK, for example, 60% of adults are overweight and 25% are obese. Obesity is associated with many pathological complications including respiratory, cardiovascular and endocrine, but it also affects fertility and is associated with many reproductive complications. This has led us and others to investigate links between women with high BMI, pregnancy outcome and uterine function. These studies in turn have led investigators to ask how obesity can have such an imp

Is there a need for smooth muscle cell transplantation in urethral reconstruction?

BACKGROUND: Hypospadias and urethral strictures are conditions requiring additional tissue for reconstruction. Due to a limited source of tissue, autologous skin and oral mucosa are frequently used. However, long-term follow-up studies demonstrated significant complications and diminished quality of life. Recently, a variety of tubular biodegradable biomaterials have been used. Cell seeding seems to be important to improve the host acceptance and neovascularization. OBJECTIVE: To compare in vivo performance of smooth muscle cell (SMC)-seeded and unseeded tubular collagen-based scaffolds in a

Congenital smooth muscle hamartoma of the nipple: a case report.

BACKGROUND: Hamartoma is a benign developmental disorder. This focal malformation is composed of mature tissues normally observed in a given organ but randomly distributed, often in disturbed quantitative ratio, with growth rate similar to that of the surrounding tissues. Congenital smooth muscle hamartoma presents as flesh-colored or variably pigmented plaque or macule. To our knowledge this is the first case of congenital smooth muscle hamartoma of the nipple reported in the available literature. CASE: A 44-year-old woman with a history of left breast cancer, for which she had mastectomy an

Characteristics of emodin on modulating the contractility of jejunal smooth muscle.

Emodin is traditionally used as a laxative and is found to increase or decrease the contractility of intestinal smooth muscle in low doses and high doses, respectively. In this study, we propose that bidirectional regulation (BR) on the contractility of jejunal smooth muscle (CJSM) is inducible by emodin in the absence of control by the central nervous system. The results indicated that emodin-induced BR had the following characteristics. A stimulatory effect on CJSM was induced by emodin at 7 low contractile states, and an inhibitory effect was induced on CJSM at 7 high contractile states. Em

Analysis of the polycystins in aortic vascular smooth muscle cells.

The leading cause of death in autosomal dominant polycystic kidney disease (ADPKD) is cardiovascular. However, little is known about the pathogenesis of these manifestations. The present study was undertaken to characterize the ADPKD proteins, the polycystins, in vascular smooth muscle cells. It was demonstrated that the expression of polycystin-1 is developmentally regulated, whereas polycystin-2 has a more constant level of expression. A polycystin-1 subpopulation was immunoprecipitated by polycystin-2, indicating an in vivo interaction of these two proteins. Analysis with glycosidase and ce

Expression of smooth muscle actin in osteoblasts in human bone.

It is well known that certain connective tissue cells (viz., dermal fibroblasts) can express the gene for a muscle actin--alpha-smooth muscle actin--and can contract. This process contributes to skin wound closure and is responsible for Dupuytren's contracture. The objective of this study was to determine if human osteoblasts can also express the gene for alpha-smooth muscle actin. Immunohistochemistry using a monoclonal antibody for alpha-smooth muscle actin was performed on human cancellous bone samples obtained from 20 individuals at the time of total joint arthroplasty. The percentages of

Histaminergic regulation of smooth muscles in rabbit pulmonary arteries.

Histaminergic contractile reactions of smooth muscles in rabbit lobar pulmonary arteries were studied using in vitro mechanographic technique. Histamine via activation of H-receptors induced a dose-dependent constriction of smooth muscle segments, while endothelium inhibited histamine-induced contractile reactions. Histamine increased sensitivity of smooth muscle cells in pulmonary artery to sodium nitroprusside.

Properties of junction potentials in gastric smooth muscle of the rat.

In isolated smooth muscles of the rat stomach, the properties of electrical responses of the membrane elicited by transmural nerve stimulation were studied. The smooth muscle membranes were quiescent in the fundus and spontaneously active with slow waves and often action potentials superimposed on top of the slow wave in the antrum and pylorus. The maximum membrane potentials were larger in the antrum (-50 to -55 mV) than in the fundus (-40 to -45 mV). Transmural nerve stimulation elicited an excitatory junction potential (e.j.p.) which was followed by an inhibitory junction potential (i.j.p.)

[Effects of puerarin on cat vascular smooth muscle in vitro].

Puerarin is an isoflavone compound isolated from Pueraria lobata (Wild). This study characterized the vascular action of puerarin in order to more clearly elucidate its pharmacological activities. Helically cut strips of cat femoral veins and renal arteries were suspended in an organ chamber filled with Krebs bicarbonate solution for measurement of isometric force development. In the cases of femoral veins and renal arteries isoproterenol (0.01 mumol.L-1 -0.1 mmol.L-1) caused relaxation of methoxamine-induced contraction (0.01 mmol.L-1) in a concentration-related manner. In-vitro motility of normal and pathological smooth muscle of colon.

This study is based on the in-vitro analysis of the normal and pathological colon smooth muscle contraction. Two different methods, supported by computer analysis, are presented. This approach will probably help the physician and the surgeon to better understand the physiopathology of some characteristical diseases, such as Hirschsprung's disease.
