Isolation of Mycobacterium avium subspecies paratuberculosis from muscle and peripheral lymph nodes using acid-pepsin digest prior to BACTEC culture.

Meat has received little attention regarding human exposure to Mycobacterium avium subsp. paratuberculosis, a possible infectious trigger of Crohn's disease. Meat has less contamination with other organisms than gut tissues, facilitating modifications to existing decontamination protocols prior to BACTEC culture that could increase analytical sensitivity. Using spiked meat samples we trialled enzymatic and chemical digestion techniques to concentrate larger starting samples, and modifications to existing clinical mycobacteriological decontamination protocols. An acid-pepsin digestion method us

Pathogens of zoonotic and biological importance in roe deer (Capreolus capreolus): Seroprevalence in an agro-system population in France.

Antibody prevalence for several infectious and parasitic diseases in a population of roe deer (Capreolus capreolus) inhabiting a mixed agricultural landscape (south of France) has been analyzed. Serological analyses with ELISA in 245 animals captured from 2008 to 2012 has been performed. We found a high prevalence of Toxoplasma gondii (46.4%), Chlamydophila abortus (17.27%) and Coxiella burnetii (11.26%) compared to other studies in Europe. Seroprevalence varied strongly among years for T. gondii (27-91%), C. abortus (0-42%) and C. burnetii (0-27%). T. gondii prevalence was lower in juvenile f

Paratuberculosis: The Hidden Killer of Small Ruminants.

Paratuberculosis (PTB) is a contagious and chronic enteric disease of ruminants and many non-ruminants caused by Mycobacterium avium subsp. paratuberculosis (MAP), and is characterised by diarrhoea and progressive emaciation with consequent serious economic losses due to death, early culling, and reduced productivity. In addition, indirect economic losses may arise from trade restrictions. Besides being a production limiting disease, PTB is a potential zoonosis; MAP has been isolated from Crohn's disease patients and was associated with other human diseases, such as rheumatoid arthritis, Hashi

Development and validation of a serum microRNA biomarker panel for bovine Johne's disease.

Johne's disease (JD) is a chronic wasting disease of ruminants caused by Mycobacterium avium subspecies paratuberculosis (MAP). For decades, JD management has been hampered by the lack of a sensitive diagnostic test. Here we have developed a machine learning (ML)-driven pipeline that identifies host-encoded microRNA (miRNA) biomarkers and deploys a classification model to provide a diagnostic output. This optimized model is integrated with a molecular assay to form the diagnostic test. Serum miRNA were profiled using RNA-sequencing from MAP-uninfected and MAP-infected cattle across Australia a

Genetic parameters for enzyme-linked immunosorbent antibody assay-based humoral response to Mycobacterium avium subspecies paratuberculosis in New Zealand dairy cattle.

Eradication of paratuberculosis is hindered by a lack of effective treatments and the difficulty of developing highly efficacious diagnostic tests. Breeding disease-resilient animals is a potential alternative and complementary strategy for controlling paratuberculosis. However, limited information is available on the estimates of genetic parameters for paratuberculosis in New Zealand dairy cattle. The present study used ELISA scores from 2 commercial kits (IDEXX and IDvet) using milk (n = 930 for both IDvet and IDEXX) and serum samples (n = 935 for IDvet and 1,121 for IDEXX) obtained from 4 d

Weighted gene co-expression network analysis reveals key genes and lncRNAs in Bos indicus chronically infected with Johne's disease.

BACKGROUND: Johne's disease (JD), caused by Mycobacterium avium subsp. paratuberculosis (MAP), is a chronic enteric disease affecting cattle worldwide, with considerable economic implications for dairy producers. MAP is a robust, intracellular pathogen highly resilient to environmental challenges. To elucidate the molecular mechanisms underlying MAP infection in desi cattle (Bos indicus), we performed a comprehensive transcriptomic analysis. METHODS: Diseased and healthy female desi cattle were selected for RNA sequencing based on phenotype. Raw data were processed, and differential expressio

Preliminary exploration of mRNA, lncRNA, and miRNA expressions in the bovine jejunum unveils novel aspects of Mycobacterium avium subspecies paratuberculosis infections.

BACKGROUND: Paratuberculosis (Johne's disease, JD) is a chronic and enteric disease in a range of ruminants, often caused by Mycobacterium avium subspecies paratuberculosis (MAP) infection, leading to substantial economic losses worldwide. Yet, the molecular underpinning of paratuberculosis remains elusive. Here we performed RNA sequencing (RNA-seq) and small RNA sequencing (sRNA-seq) of the jejunum tissues from the Holstein cows with three distinct statuses of paratuberculosis, i.e., healthy, subclinical, and clinical to screen potential genes, lncRNAs, and miRNAs associated with the resistan

Paratuberculosis in small ruminants in the Sudan: prevalence and risk factors.

BACKGROUND: Paratuberculosis (PTB), caused by Mycobacterium avium subsp. paratuberculosis (MAP), is a contagious and chronic enteric disease of ruminants and many non-ruminants leading to emaciation and death of the animal. PTB is poorly investigated in sheep and goats in Sudan, where these animals contribute significantly to food security and poverty alleviation as sources of income. They also play an important role in the national economy through animal exports. The objectives of this study were to investigate the prevalence of PTB and associated risk factors in small ruminants. Mycobacterium avium subspecies induce differential expression of pro-inflammatory mediators in a murine macrophage model: evidence for enhanced pathogenicity of Mycobacterium avium subspecies paratuberculosis.

Mycobacterium avium subspecies (ssp.) paratuberculosis (MAP) is the etiological agent of paratuberculosis, a chronic, non-treatable granulomatous enteritis of ruminants. MAP is the only mycobacterium affecting the intestinal tract, which is of interest since it is presently the most favoured pathogen linked to Crohn's disease (CD) in humans due to its frequent detection in CD tissues. MAP is genetically closely related to other M. avium ssp. such as M. avium ssp. avium (MAA) and M. avium ssp. hominissuis (MAH) which can cause mycobacteriosis in animals and immunocompromised humans. We have rec

Prevalence and diversity of non-tuberculous mycobacteria isolated from slaughtered tuberculin-positive bovine samples: Epidemiological and Diagnostic implications.

The isolation of non-tuberculous mycobacteria (NTM) is common in cattle positive for tuberculosis (TB) in official diagnostic tests, whereas data on specific NTM species in Spanish cattle remain limited. This study identifies the most frequently isolated NTM species from Single Intradermal Tuberculin Test (SITT)-positive cattle in Extremadura, western Spain. Among 1669 Mycobacterium Growth Indicator Tube (MGIT) positive cultures collected in 2018, 493 (29.54 %) were identified as NTM, and 194 were randomly selected for further analysis. Genome-wide association analysis for susceptibility to infection by Mycobacterium avium ssp. paratuberculosis in US Holsteins.

Paratuberculosis, or Johne's disease, is a chronic, granulomatous, gastrointestinal tract disease of cattle and other ruminants caused by the bacterium Mycobacterium avium subspecies paratuberculosis (MAP). Control of Johne's disease is based on programs of testing and culling animals positive for infection with MAP and concurrently modifying management to reduce the likelihood of infection. The current study was motivated by the hypothesis that genetic variation in host susceptibility to MAP infection can be dissected and quantifiable associations with genetic markers identified. WC1+ and WC1neg γδ T lymphocytes in intestinal mucosa of healthy and Mycobacterium avium subspecies paratuberculosis-infected calves.

Mucosal surfaces such as the gastrointestinal tract, and skin are the front line of host defence and immunity against many pathogens. Gamma delta (γδ) T lymphocytes preferentially localize to the mucosal surfaces in several species including cattle, and are thought to play crucial roles in immunosurveillance and host defence, particularly against mycobacteria. Many γδ T cells are present in young calves, which is the period when calves are thought to be initially exposed to Mycobacterium avium subspecies paratuberculosis (Map). The role of mucosal γδ T cells in cattle, especially during host-p

Application of multiple laboratory tests for Mycobacterium avium ssp. paratuberculosis detection in Crohn's disease patient specimens.

The difficulties involved in detecting and enumerating Mycobacterium avium subsp. paratuberculosis (MAP) as a pathogen potentially involved in Crohn's disease (CD) are well known. This study aimed to improve this situation through the application of multiple laboratory diagnostic tests to detect and isolate this bacterium from different specimens collected from CD-patients and non-CD subjects as controls. A total of 120 samples (terminal ileum and colon biopsies, blood and stool) were obtained from 19 CD-patients and from 11 individuals who did not have a clinicopathological diagnosis of CD (n

Assessment of Dietzia subsp. C79793-74 for treatment of cattle with evidence of paratuberculosis.

The objective of the present investigation was to determine whether the bacterium Dietzia subsp. C79793-74, previously shown to inhibit growth of Mycobacterium subsp. paratuberculosis under in vitro culture conditions, has therapeutic value as a probiotic for adult cattle with paratuberculosis.  Animals were obtained from several herds with evidence of disease based on seropositivity and/or fecal shedding.  Sixty-eight cows with initial evidence of Stage II or III paratuberculosis and 2 with an initial Stage IV disease were evaluated longitudinally.  Animals were either treated daily with vari

Ovine small intestinal adenocarcinomas are not associated with infection by herpesviruses, Helicobacter species or Mycobacterium avium subspecies paratuberculosis.

Sheep in New Zealand more frequently develop small intestinal adenocarcinoma (SIA) than sheep in other countries. The reasons for this high rate of intestinal neoplasia are not known. In man, differences between countries in the incidence of neoplasia are often due to differences in the rate of infection by carcinogenic viruses or bacteria. Therefore, it was hypothesized that New Zealand sheep more frequently develop SIA as they are more frequently exposed to an infectious agent. This study compared rates of detection of herpesviruses, Helicobacter species, and Mycobacterium avium subspecies p

Dietary calcium modulates Mycobacterium paratuberculosis infection in beige mice.

A 6-month study was conducted to evaluate the effects of feeding different levels of dietary calcium (Ca) on the persistence of Mycobacterium paratuberculosis infection using a mouse model. Beige mice, averaging 8 weeks of age, were randomly assigned to one of the following dietary treatments: 1) 0.02% Ca, 2) 0.15% Ca, 3) 0.45% Ca, and 4) 1.0% Ca. Mice were infected intraperitoneally with 10(8) CFU viable M. paratuberculosis for 1, 3, and 6 month periods. Plasma Ca levels was unaffected by dietary Ca (x = 7.3 mg/dl). Plasma levels of 1,25(OH)2D3 was elevated significantly in 0.02% and 0.15% Ca

Cell wall peptidolipids of Mycobacterium avium: from genetic prediction to exact structure of a nonribosomal peptide.

Mycobacteria have a complex cell wall structure that includes many lipids; however, even within a single subspecies of Mycobacterium avium these lipids can differ. Total lipids from an M. avium subsp. paratuberculosis (Map) ovine strain (S-type) contained no identifiable glycopeptidolipids or lipopentapeptide (L5P), yet both lipids are present in other M. avium subspecies. We determined the genetic and phenotypic basis for this difference using sequence analysis as well as biochemical and physico-chemical approaches. This strategy showed that a nonribosomal peptide synthase, encoded by mps1, c

Targeting the Mincle and TLR3 receptor using the dual agonist cationic adjuvant formulation 9 (CAF09) induces humoral and polyfunctional memory T cell responses in calves.

There is a need for the rational design of safe and effective vaccines to protect against chronic bacterial pathogens such as Mycobacterium tuberculosis and Mycobacterium avium subsp. paratuberculosis in a number of species. One of the main challenges for vaccine development is the lack of safe adjuvants that induce protective immune responses. Cationic Adjuvant Formulation 01 (CAF01)-an adjuvant based on trehalose dibehenate (TDB) and targeting the Mincle receptor-has entered human trials based on promising pre-clinical results in a number of species. However, in cattle CAF01 only induces wea

Retrospective Single Nucleotide Polymorphism Analysis of Host Resistance and Susceptibility to Ovine Johne's Disease Using Restored FFPE DNA.

Johne's disease (JD), also known as paratuberculosis, is a chronic, untreatable gastroenteritis of ruminants caused by Mycobacterium avium subsp. paratuberculosis (MAP) infection. Evidence for host genetic resistance to disease progression exists, although it is limited due to the extended incubation period (years) and diagnostic challenges. To overcome this, previously restored formalin-fixed paraffin embedded tissue (FFPE) DNA from archived FFPE tissue cassettes was utilized for a novel retrospective case-control genome-wide association study (GWAS) on ovine JD.
