The unusual features of traumatic neurogenic muscular atrophy in the infant: an anatomic study.

A biopsy from the deltoid muscle of an 8-week old girl who had sustained a bilateral traumatic Erb's brachial plexus paresis at birth revealed the neurogenic atrophy to be different from that of adult muscle in that small, round fibers, similar to those observed in Werdnig-Hoffmann disease, were found. Detailed histochemistry of the fibers is submitted. On electron microscopy, there was focal degeneration of sarcomeres; a loss of myofibrils and myofilaments accompanied by increased autophagocytosis and increased amounts of glycogen; occasionally, preferential drop out of thick myofilaments and

Coordinated improvement of shrimp Litopenaeus vannamei preservation quality during super-chilling at -3 °C by combination of chitosan coating with vacuum packing: Efficiency and mechanism.

The quality deterioration of Litopenaeus vannamei during super-chilling exerts a critical challenge for shrimp industry. This study proposes a "antibacterial and oxygen removal-dependent coupling" mechanism to address this challenge. The efficacy of chitosan coating (CC) combined with vacuum packaging (VP) on quality indicators was investigated. Remarkably, the highest sensory score, desirable values of hardness and springiness, the lowest TVB-N contents and activities of protease and polyphenols oxidase (PPO), as well as the significant decrease in TVC and Pseudomonas counts were observed in

S-nitrosoproteomics profiling elucidates the regulatory mechanism of S-nitrosylation on beef quality.

This study aimed to quantitively profile the S-nitrosylation in beef semimembranosus (SM) with different treatments (nitric oxide donor or nitric oxide synthase inhibitor) by applying iodoTMT-based nitrosoproteomics. Results showed that 2096 S-nitrosylated cysteine sites in 368 proteins were detected in beef SM. Besides, differential SNO-modified proteins were screened, some of which were involved in crucial biochemical pathways, including calcium-releasing-related proteins, energy metabolic enzymes, myofibrils, and cytoskeletal proteins. GO analysis indicated that differential proteins were l

Internal structure and remodeling in dystrophin-deficient cardiomyocytes using second harmonic generation.

Duchenne muscular dystrophy (DMD) is a debilitating disorder related to dystrophin encoding gene mutations, often associated with dilated cardiomyopathy. However, it is still unclear how dystrophin deficiency affects cardiac sarcomere remodeling and contractile dysfunction. We employed second harmonic generation (SHG) microscopy, a nonlinear optical imaging technique that allows studying contractile apparatus organization without histologic fixation and immunostaining. Images were acquired on alive DMD (mdx) and wild type cardiomyocytes at different ages and at various external calcium concent

Functional aptitude of hake minces with added TMAO-demethylase inhibitors during frozen storage.

The ability of compounds of natural origin (black, white, red, and green tea extracts, phytic acid) to inhibit TMAO-demethylase enzyme was assayed. Black tea and phytic acid exerted the highest inhibiting activities, similar to the already known inhibitor sodium citrate. Hake minces incorporating these three compounds were prepared and stored frozen (150 days, -12 °C). TMAO-demethylase enzyme was partially inhibited (lower enzyme activity, reduction of formaldehyde accumulation). The study of physicochemical properties of the minces (salt-soluble proteins, water holding capacity, structural wa

Possible mechanism for disposal of degenerative cardiomyocytes in human failing hearts: phagocytosis by a neighbour.

The index case was a 51-year-old woman suffering from doxorubicin cardiomyopathy. In her endomyocardial biopsy specimen, we observed under electron microscopy six scenes in which degenerative cardiomyocytes were engulfed by neighbouring cardiomyocytes. The enclosed cardiomyocytes appeared more degenerative than the enclosing ones in every pair: the myofibrils were more severely damaged. At more degenerative stages, some desmosomes of the intercalated discs on the enclosed cardiomyocyte had disappeared. The membranes between the cardiomyocytes were occasionally disrupted, and there appeared to

In Vivo Maturation of Human Induced Pluripotent Stem Cell-Derived Cardiomyocytes in Neonatal and Adult Rat Hearts.

We hypothesized that the neonatal rat heart would bring transplanted human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) to maturity as it grows to adult size. In neonatal rat heart, engrafted hiPSC derivatives developed partially matured myofibrils after 3 months, with increasing cell size and sarcomere length. There was no difference between grafts from hiPSC-CMs or hiPSC-derived cardiac progenitors (hiPSC-CPs) at 3 months, nor was maturation influenced by infarction. Interestingly, the infarcted adult heart induced greater human cardiomyocyte hypertrophy and induction of

Neuromuscular regulation in zebrafish by a large AAA+ ATPase/ubiquitin ligase, mysterin/RNF213.

Mysterin (also known as RNF213) is a huge intracellular protein with two AAA+ ATPase modules and a RING finger ubiquitin ligase domain. Mysterin was originally isolated as a significant risk factor for the cryptogenic cerebrovascular disorder moyamoya disease, and was found to be involved in physiological angiogenesis in zebrafish. However, the function and the physiological significance of mysterin in other than blood vessels remain largely unknown, although mysterin is ubiquitously expressed in animal tissues. In this study, we performed antisense-mediated suppression of a mysterin orthologu

[Structural features of cardiomyocytes in the atrialized right ventricle in patients with Ebstein anomaly].

Intraoperative biopsy specimens were used to analyze the sizes and ultrastructure of cardiomyocytes (CMC) in the atrialized right ventricle (ARV) versus those in the right ventricle (RV) and right atrium (RA) in 32 patients aged 9 months to 57 years with Ebstein anomaly. The mean CMC diameter in ARV, RV, and RA did not differ significantly in patients less than 10 years; that was equal in ARV and RV, but higher in RA in patients over 10 years of age. No specific atrial granules were found in the CMC of ARV, RV, and RA; but T system channels and multiple intercalated disks were encountered sign

[Cardiac arrhythmia associated with malignant neuroleptic syndrome: description of 2 clinical cases].

Malignant neuroleptic syndrome (alteration of consciousness, muscle rigidity and hyperthermia) is a potentially lethal condition, due also to its life-threatening complications. In particular, hypokinetic and hyperkinetic arrhythmias can be rare and severe early manifestations of this illness, and they deserve a careful approach because of their drug-refractoriness. Arrhythmias associated with the malignant neuroleptic syndrome depend on various mechanisms: neurotransmitter receptor blockades typical of neuroleptic drugs, clustered lipid droplets among the cardiac myofibrils and possible elect

Congenital nystagmus: fine structure of human extraocular muscles.

The ultrastructure of the extraocular muscles of patients affected by congenital nystagmus is still to be defined. Specimens obtained from patients suffering from either oculomotor congenital nystagmus or nystagmus associated with strabismus were studied and compared with specimens obtained from patients enucleated for various pathologies but not affected by any oculomotor system disorder. The scleral myotendinous junction and the muscle body (venter) were examined. In both these areas, damaged muscle fibers were found. 'Mitochondrial cores' and concentric myofibrils characterized the extraocu

Murine acinar pancreatitis preceding necrotizing myocarditis after Coxsackievirus B3 inoculation.

Balb/C weanling mice were inoculated intraperitoneally with a myocarditic variant of coxsackie-virus B3, with the aim of characterizing more fully the cell damage induced in the heart as well as in other organs. During the first week postinfection (pi), all animals developed acinar pancreatitis, followed by focal myocarditis. In accordance with the increasing infectivity titers, such progressive histopathological changes correlated with local viral replication. From day 4 pi, acinar degeneration accompanied by diffuse inflammatory exudate was observed in the pancreas, followed by fatty tissue

Cellular mechanisms of alterations in myocardial contractile function in experimental cardiomyopathies.

The high-energy-phosphate content, myocardial ultrastructure, left ventricular (LV) pressure, and pump function of isolated rat hearts were determined in four kinds of chronic myocardial damage induced by either autoimmunization, treatment of rats with adriamycin or noradrenaline in increasing doses, or infection with smallpox virus. Mild fibrosis, swollen mitochondria, and hyper-contracted and overdistended sarcomeres were typical ultrastructural alterations. The ATP and phosphocreatine content as well as cardiac output at standard load conditions were substantially lower in all four experime

Ultrastructural features of diseased human atrial muscle cells.

Left atrial myocardial tissues, excised at the time of surgery from 30 Japanese patients, were studied using light and electron microscopy and the freeze-fracture technique. All had mitral valvular disease, and, in 22 (74%) patients, atrial fibrillation also was present. In all patients, most of the left atrial cardiocytes were hypertrophied and surrounded by various amounts of fibrous tissue. Degenerative alterations included large masses of lipofuscin granules and lamellar bodies, disorganized myofibrils, widened Z-line material, selective loss of thick filaments, and prominent tubules of sa

Changes in creatine kinase and its isoenzymes in human fetal muscle during development.

The total creatine kinase activity and its isoenzyme patterns were investigated in the skeletal muscle of 87 fetuses, of which 80 were presumed normal, 5 were anencephalic and 2 were "at risk" for Duchenne muscular dystrophy (DMD). No differences, either in total enzyme activity or in the isoenzyme distributions, were found between the anencephalic fetuses or those at risk for DMD, when compared to normal fetuses of similar gestation. Creatine kinase activity was found to rise steadily throughout embryonic life. During fetal development, the isoenzyme pattern in skeletal muscle was observed to

Mechanisms for the transition from physiological to pathological cardiac hypertrophy.

The heart is capable of responding to stressful situations by increasing muscle mass, which is broadly defined as cardiac hypertrophy. This phenomenon minimizes ventricular wall stress for the heart undergoing a greater than normal workload. At initial stages, cardiac hypertrophy is associated with normal or enhanced cardiac function and is considered to be adaptive or physiological; however, at later stages, if the stimulus is not removed, it is associated with contractile dysfunction and is termed as pathological cardiac hypertrophy. It is during physiological cardiac hypertrophy where the f

Cardiac cytoarchitecture - why the "hardware" is important for heart function!

Cells that constitute fully differentiated tissues are characterised by an architecture that makes them perfectly suited for the job they have to do. This is especially obvious for cardiomyocytes, which have an extremely regular shape and display a paracrystalline arrangement of their cytoplasmic components. This article will focus on the two major cytoskeletal multiprotein complexes that are found in cardiomyocytes, the myofibrils, which are responsible for contraction and the intercalated disc, which mediates mechanical and electrochemical contact between individual cardiomyocytes. Recent st

Ultrastructural changes in the advanced third larval stage of Gnathostoma binucleatum Almeyda-Artigas, 1991 (Nematoda: Spirurida) following albendazole and albendazole sulfoxide in vitro exposure.

We report here the ultrastructural changes in the third advanced larval stage (AL3) of Gnathostoma binucleatum Almeyda-Artigas, 1991, induced by in vitro treatment with albendazole and the metabolite albendazole sulfoxide. During the whole period of the experiment, the controls remained active and vital throughout the 108-h exposure period. The primary site of action of both drugs appears to be the mitochondria and myofibrils of the polymyarian musculature. Degenerative changes in the secretory portion of the esophagus and somatic musculature were clearly observed. These degenerative changes,

An AI-informed framework for flavor design: How κ-carrageenan sculpts low-sodium myofibrillar protein architecture for aldehyde capture and bitterness masking.

Designing healthy, palatable foods is a grand challenge in materials science, particularly in quest for low-sodium products where flavor is often compromised. The central problem is not merely replacing NaCl, but re-engineering the food matrix itself to manage off-flavors and retain desirable aromas. Here, an AI-informed framework demonstrates how the biopolymer κ-Carrageenan (KC) can sculpt the myofibrillar protein (MP) architecture for enhanced aldehyde capture and bitterness masking in low-sodium systems. Results showed KC acts as a molecular scaffold, transforming the protein from a rigid

Thermo-reversible gelation and enhanced umami perception of myofibrillar proteins induced by protein-glutaminase-mediated deamidation.

In this study, the absolute electrostatic charge of myofibrillar protein (MP) was substantially increased by protein-glutaminase (PG) treatment, which was a critical step for achieving the dissociation and solubility of MP under low salt condition. The PG-treated MP exhibited the capacity to form thermo-reversible gels that could be melted through heating and subsequently reformed into a stable gel structure upon refrigeration. The results of SDS-PAGE further revealed that the levels of soluble monomeric myosin and actin in the supernatant of deamidated MP (DMP) gels were markedly elevated, an
