Potentially pathogenic free-living amoebae at very high altitude: Detection by multiplex qPCR in the Northern Altiplano fascioliasis hyperendemic area in Bolivia.

Free-living amoebae (FLA), which are frequently found in the environment, include opportunistic pathogenic genera/species such as Acanthamoeba spp., Balamuthia mandrillaris, Naegleria fowleri and Vermamoeba vermiformis. These pathogenic FLA are causative agents of amoebic encephalitis and keratitis in the case of Acanthamoeba genus and V. vermiformis. In addition, amoebic infections are often related to contamination of domestic and recreational water sources. This study aimed to identify potentially pathogenic FLA in the hyperendemic area of human fascioliasis in a very-high-altitude area (38

Fatal Primary Amebic Meningoencephalitis in Nebraska: Case Report and Environmental Investigation, August 2022.

Primary amebic meningoencephalitis (PAM) is a rare and lethal infection caused by Naegleria fowleri. We report an epidemiological and environmental investigation relating to a case of PAM in a previously healthy boy age 8 years. An interview of the patient's family was conducted to determine the likely exposure site and to assess risk factors. Data from the United States Geological Survey site at Waterloo, NE, on the Elkhorn River were used to estimate water temperature and streamflow at the time and site of exposure. Data from the National Weather Service were used to estimate precipitation a

Characterization of brain inflammation during primary amoebic meningoencephalitis.

Naegleria fowleri is a free-living amoeba and the etiologic agent of primary amoebic meningoencephalitis (PAM). Trophozoites reach the brain by penetrating the olfactory epithelium, and invasion of the olfactory bulbs results in an intense inflammatory reaction. The contribution of the inflammatory response to brain damage in experimental PAM has not been delineated. Using both optical and electron microscopy, we analyzed the morphologic changes in the brain parenchyma due to inflammation during experimental PAM. Several N. fowleri trophozoites were observed in the olfactory bulbs 72 h post-in

Drug Target Discovery Methods In Targeting Neurotropic Parasitic Amoebae.

Neurotropic parasitic amoebal infections have imposed an enormous challenge to chemotherapy in patients who fall victims to the infections caused by them. Conventional antibiotics that are given to treat these infections have a low patient compliance because of the serious adverse effects that are associated with their use. Additionally, the growing incidence of the development of drug resistance by the neurotropic parasites like Naegleria fowleri, Balamuthia mandrillaris, and Acanthamoeba spp has made the drug therapy more challenging. Recent studies have reported some cellular targets in the

Exploitation of Mangrove Endophytic Fungi for Infectious Disease Drug Discovery.

There is an acute need for new and effective agents to treat infectious diseases. We conducted a screening program to assess the potential of mangrove-derived endophytic fungi as a source of new antibiotics. Fungi cultured in the presence and absence of small molecule epigenetic modulators were screened against Mycobacterium tuberculosis and the ESKAPE panel of bacterial pathogens, as well as two eukaryotic infective agents, Leishmania donovani and Naegleria fowleri. By comparison of bioactivity data among treatments and targets, trends became evident, such as the result that more than 60% of

Primary amoebic meningoencephalitis: fifteen years later.

Primary amoebic meningoencephalitis is a fulminant and rapidly fatal diseases which principally affects children and young adults. The causative organism is Naegleria fowleri, an amoebo-flagellate found in most soil and freshwater habitats. The portal of entry is the nasopharynx from which the amoeba makes its way into the brain by penetration of the olfactory mucosa and cribriform plate. Diagnosis should be suspected in all cases of purulent meningitis and meningoencephalitis in which bacteria are not evident in the cerebrospinal fluid. Diagnosis can be made by microscopic examination of a fr

Ultrastructural observations of experimental Naegleria meningoencephalitis in mice: intranuclear inclusions in amebae and host cells.

Primary amebic meningoencephalitis was experimentallly produced in mice through intranasal instillation of pathogenic Naegleria fowleri. Experimental animals had a 64% mortality with average time of onset of symtoms of death occurring on the 7-8th day following inoculation. Ultrastructural studies of the olfactory lobes from brains of dead (or sacrificed) animals revealed major concentrations of amebae in the perivascular regions; amebae were also seen to be under attack by host polymorphonuclear leukocytes, and in the lumina of blood vessels. Amebae in brain tissue contained 30 nm intranuclea

Amebic meningoencephalitides and keratitis: challenges in diagnosis and treatment.

PURPOSE OF REVIEW: Acanthamoeba spp., Balamuthia mandrillaris, and Naegleria fowleri, although free-living amebae, also cause devastating diseases in humans leading to death. Acanthamoeba spp. and B. mandrillaris cause granulomatous amebic encephalitis, cutaneous and nasopharyngeal as well as disseminated infection. Acanthamoeba also causes a vision-threatening infection of the cornea, Acanthamoeba keratitis, principally in contact lens wearers. N. fowleri causes an acute, fulminating infection of the central nervous system, primary amebic meningoencephalitis, in healthy children and young adu

Primary Amebic Meningoencephalitis Mimicking Acute Bacterial Meningitis: Survival Following Early Empirical Therapy.

Primary amebic meningoencephalitis (PAM) is a rare but rapidly fatal infection of the central nervous system caused by the thermophilic free-living amoeba Naegleria fowleri. The disease often presents with clinical and cerebrospinal fluid (CSF) findings that closely mimic acute bacterial meningitis, making early diagnosis particularly challenging. We report the case of a 29-year-old woman with poorly controlled type 1 diabetes who presented with acute fever, severe headache, vomiting, and meningeal signs. Within 24 hours, her condition progressed to visual and auditory hallucinations, generali

A rare but fatal case of granulomatous amebic encephalitis with brain abscess: the first case reported from Turkey.

The incidence of protozoal and helminthic infestations of the central nervous system (CNS) is less than 1%, but these infestations tend to follow a fatal course. They are more common among children, the elderly and immunocompromised individuals. CNS infections due to Entamoeba histolytica have been known for a long time. In recent years, especially in developing countries, there has been an increase in CNS infections due to free-living amebas (FLAs). Acute CNS infection due to Naegleria fowleri, which ends in death within 2-7 days, is termed primary amebic meningoencephalitis (PAM); subacute o

CNS infections with free-living amebas: neuroimaging findings.

OBJECTIVE: The purpose of this report is to describe the clinical history, treatment, pathology, and imaging in two cases of rare CNS infection caused by free-living amebas. The Naegleria fowleri and Acanthamoeba species cause primary amebic meningoencephalitis and granulomatous amebic encephalitis, respectively. We describe the neuroimaging findings of a case involving a nonspecific cerebral edema pattern in primary amebic meningoencephalitis and a case involving focal enhancing lesions in granulomatous amebic encephalitis. CONCLUSION: Primary amebic meningoencephalitis and granulomatous ame

Pathogenic and free-living protozoa cultured from the nasopharyngeal and oral regions of dental patients: II.

Protozoa of the nose, mouth, and pharynx of 30 randomly chosen male caries patients at an odontological clinic of the National Autonomous University of Mexico, in Mexico City, were surveyed by culture from swabs. Culture tubes from swabs were observed every other day for 5 weeks. Pathogens found included Entamoeba histolytica, Naegleria fowleri, Acanthamoeba castellanii, A. culbertsoni, A. polyphaga, and Giardia lamblia. Such isolations of pathogens suggest that patients may be healthy carriers of cysts and trophic forms of protozoa responsible for several human diseases. Amoebae responsible f

Non-granulomatous meningoencephalitis with Balamuthia mandrillaris mimicking a tumor: First confirmed case from Pakistan.

BACKGROUND: Free-living amoebae rarely instigate intracranial infections that may resemble neoplastic conditions on imaging. Naegleria fowleri precipitates an acute, swiftly fatal meningoencephalitis, whereas Acanthamoeba and Balamuthia species typically manifest with a less aggressive onset but carry equally dire consequences. CASE DESCRIPTION: The case describes a 33-year-old woman with subacute encephalitis caused by Balamuthia mandrillaris. She experienced 2 months of back pain, 1 month of headaches, and 2 weeks of vomiting without fever, recent travel, aquatic activities, or animal expos

Notes from the field: primary amebic meningoencephalitis associated with ritual nasal rinsing--St. Thomas, U.S. Virgin islands, 2012.

On November 21, 2012, the U.S. Virgin Islands (USVI) Department of Health documented the first case and death from primary amebic meningoencephalitis (PAM) in the territory. PAM, a rare and almost universally fatal condition, results when Naegleria fowleri, a free-living thermophilic ameba found in warm freshwater, enters the nose and migrates to the brain. The patient was a man aged 47 years whose only reported freshwater exposures were the use of tap water for daily household activities and for ablution, a ritual cleansing that he practiced several times a day in preparation for Islamic pray

Meningoencephalitis due to Acanthamoeba SP. Pathogenesis and clinico-pathological study.

Amebic Meningoencephalitis (AM) and Primary Amebic Meningoencephalitis (PAM) are infectious diseases essentially confined to the Central Nervous System (CNS) and caused by free-living amebas of the genus Acanthamoeba (A.) and Naegleria (N.) respectively. AM due to A. sp. (Acanthamoeba castellanii and Acanthamoeba culbertsoni) have been reported in chronically ill debilitated individuals, some of them under immunosuppressive therapy, or in immunologically impaired patients without a history of recent swimming in contrast to cases due to N. sp. which usually occurs in healthy, young individuals
