Neuronal inhibitory mechanisms in overweight/obesity and food/eating addiction: The role of frontal brain coherence, dysregulated eating, and depressive symptoms.

BACKGROUND AND AIMS: Obesity and food/eating addiction are associated with deficits in inhibitory control mechanisms manifested in the lateral prefrontal cortex (LPFC) bilaterally. We explored abnormalities in the functional coherence between the hemispheres and within each of them. METHODS: Resting-state electroencephalography (EEG) was utilized to examine intra- and inter-hemispheric frontal brain coherence in individuals with obesity and food/eating addiction (FAOB, n = 27), those with obesity without addiction (Non-FAOB, n = 11), and healthy controls (HC, n = 15). Participants were includ

Perceptual Competition Promotes Suppression of Reward Salience in Behavioral Selection and Neural Representation.

Visual attentional selection is influenced by the value of objects. Previous studies have demonstrated that reward-associated items lead to rapid distraction and associated behavioral costs, which are difficult to override with top-down control. However, it has not been determined whether a perceptually competitive environment could render the reward-driven distraction more susceptible to top-down suppression. Here, we trained both genders of human subjects to associate two orientations with high and low magnitudes of reward. After training, we collected fMRI data while the subjects performed

Anterior Thalamic Excitation and Feedforward Inhibition of Presubicular Neurons Projecting to Medial Entorhinal Cortex.

The presubiculum contains head direction cells that are crucial for spatial orientation. Here, we examined the connectivity and strengths of thalamic inputs to presubicular layer 3 neurons projecting to the medial entorhinal cortex in the mouse. We recorded pairs of projection neurons and interneurons while optogenetically stimulating afferent fibers from the anterior thalamic nuclei. Thalamic input differentially affects presubicular neurons: layer 3 pyramidal neurons and fast-spiking parvalbumin-expressing interneurons are directly and monosynaptically activated, with depressing dynamics, wh

Diversity of vestibular nuclei neurons targeted by cerebellar nodulus inhibition.

A functional role of the cerebellar nodulus and ventral uvula (lobules X and IXc,d of the vermis) for vestibular processing has been strongly suggested by direct reciprocal connections with the vestibular nuclei, as well as direct vestibular afferent inputs as mossy fibres. Here we have explored the types of neurons in the macaque vestibular nuclei targeted by nodulus/ventral uvula inhibition using orthodromic identification from the caudal vermis. We found that all nodulus-target neurons are tuned to vestibular stimuli, and most are insensitive to eye movements. Such non-eye-movement neurons

Tonic and phasic drive to medullary respiratory neurons during periodic breathing.

It is unknown how central neural activity produces the repetitive termination and restart of periodic breathing (PB). We hypothesized that inspiratory and expiratory neural activities would be greatest during the waxing phase and least during the waning phase. We analyzed diaphragmatic and medullary respiratory neural activities during PB in intact unanesthetized adult cats. Diaphragmatic activity was increased and phasic during the waxing phase and was decreased and tonic during the waning phase. Activity of expiratory (n=21) and inspiratory (n=40) neurons was generally increased and phasic d

Decreased spinal inhibition leads to undiversified locomotor patterns.

During walking and running, animals display rich and coordinated motor patterns that are generated and controlled within the central nervous system. Previous computational and experimental results suggest that the balance between excitation and inhibition in neural circuits may be critical for generating such structured motor patterns. In this paper, we explore the influence of this balance on the ability of a reservoir computing artificial neural network to learn human locomotor patterns, using mean-field theory and simulations. We created networks with varying neuron numbers, connection perc

Reduced interhemispheric inhibition in mild cognitive impairment.

In mild cognitive impairment (MCI), the corpus callosum is known to be affected structurally. We evaluated callosal function by interhemispheric inhibition (IHI) using transcranial magnetic stimulation (TMS) in MCI patients. We investigated 12 amnestic MCI patients and 16 healthy age-matched control subjects. The IHI was studied with a paired-pulse TMS technique. The conditioning TMS was given over the right primary motor cortex (M1) and the test TMS over the left M1. Motor evoked potentials were recorded from the relaxed first dorsal interosseous muscle. We also studied other motor cortical c

Facilitation and antagonism of kindled seizure development in the limbic system of the rat.

To study the relative contribution of positive and negative transfer during the kindling of motor seizures, we electrically stimulated the septal area and entorhinal cortex in rats according to two different stimulation paradigms. The first consisted of the usual transfer paradigm in which one site was consecutively stimulated until generalized convulsion were elicited and then the other, secondary site was consecutively stimulated ('transfer kindling'). The second paradigm differed only in that both sites were kindled concurrently by alternating stimulation--one site on one trial, the other o

Wilms Tumor 1 Associated Protein (WTAP) Inhibits Inflammation Provoked by Mycobacterium tuberculosis in Microglial BV-2 Cells and Promotes Differentiation of Neural Stem/Progenitor Cells into Neurons by Elevating Expression of HMGN3 Protein Resulted from Modulation of m6A Metilation of Its RNA.

Tuberculous meningitis (TBM) is the most prevalent and severe manifestation of tuberculosis in CNS. The mechanisms of neurological injury caused by TBM are not well understood. Our study showed that overexpression of WTAP (Wilms tumor 1 associated protein) reduced inflammatory factors and Iba-1 expression induced in BV-2 by H37Rv. It also increased proliferation of neural stem/progenitor cells (NSPCs) and expression of the neuronal marker DCX in these cells. WTAP enhanced expression of high mobility group nucleosome-binding domain-containing protein 3 (HMGN3) by promoting m6A methylation of it

The role of inhibitory neuronal variability in modulating phase diversity between coupled networks.

Neuronal heterogeneity, characterized by a multitude of spiking neuronal patterns, is a widespread phenomenon throughout the nervous system. In particular, the brain exhibits strong variability among inhibitory neurons. Despite the huge neuronal heterogeneity across brain regions, which in principle could decrease synchronization due to differences in intrinsic neuronal properties, cortical areas coherently oscillate during various cognitive tasks. Therefore, the functional significance of neuronal heterogeneity remains a subject of active investigation. Previous studies typically focus on the

Short interval intracortical inhibition: Variability of amplitude and threshold-tracking measurements with 6 or 10 stimuli per point.

Reduced short-interval intracortical inhibition (SICI) in motor neuron disease has been demonstrated by amplitude changes (A-SICI) and threshold-tracking (T-SICI) using 10 stimuli per inter-stimulus interval (ISI). To test whether fewer stimuli would suffice, A-SICI and T-SICI were recorded twice from 30 healthy subjects using 6 and 10 stimuli per ISI. Using fewer stimuli increased mean A-SICI variances by 23.8% but the 7.3% increase in T-SICI variance was not significant. We conclude that our new parallel threshold-tracking SICI protocol, with 6 stimuli per ISI, can reduce time and stimulus n

Response inhibition alterations in migraine: evidence from event-related potentials and evoked oscillations.

BACKGROUND: Migraine is characterized by a hypersensitivity to environmental stimulation which climaxes during headache attacks but persists during attack-free period. Despite ongoing debates about the nature of the mechanisms giving rise to this abnormality, the presence of deficient inhibitory cortical processes has been proposed to be one possible mechanism underlying its pathogenesis. Empirical evidence supporting this claim is mainly based on previous accounts showing functional cortical disexcitability in the sensory domain. Considering that a general inhibitory control process can play

A fronto-striato-subthalamic-pallidal network for goal-directed and habitual inhibition.

Classically, the basal ganglia have been considered to have a role in producing habitual and goal-directed behaviours. In this article, we review recent evidence that expands this role, indicating that the basal ganglia are also involved in neural and behavioural inhibition in the motor and non-motor domains. We then distinguish between goal-directed and habitual (also known as automatic) inhibition mediated by fronto-striato-subthalamic-pallido-thalamo-cortical networks. We also suggest that imbalance between goal-directed and habitual action and inhibition contributes to some manifestations

The role of the subiculum within the behavioural inhibition system.

This paper summarises the picture of the subiculum presented by Gray and McNaughton [Gray JA, McNaughton N. The neuropsychology of anxiety: an enquiry into the functions of the septo-hippocampal system. Oxford: Oxford University Press; 2000]. It is a key node in their "Behavioural Inhibition System" and, as such, is embedded within a hierarchy of structures controlling anxiety that is in parallel with a separate hierarchy controlling fear. It can nonetheless be viewed as operating in a fairly simple manner. It receives information about available goals from areas that plan motor action. This i

Relative distribution of Ca2+ channels at the crayfish inhibitory neuromuscular junction.

The use of LiCl in clinical psychiatry is routinely complicated by overt nephrogenic diabetes insipidus (NDI), the mechanism of which is incompletely understood. In vitro studies indicate that lithium can induce renal medullary interstitial cell cyclooxygenase 2 (COX2) protein expression via inhibition of glycogen synthase kinase-3beta (GSK-3beta). Both COX1 and COX2 are expressed in the kidney. Renal prostaglandins have been suggested to play an important role in lithium-induced polyuria. The present studies examined whether induction of the COX2 isoform contributes to LiCl-induced polyuria.

Reciprocal inhibition in forearm muscles in patients with essential tremor.

Reciprocal inhibition of the H-reflex in the forearm flexor muscles was studied in 11 patients with essential tremor and in 10 normal controls. Whereas patients and controls had a similar first, disynaptic phase of reciprocal inhibition, patients had a significantly reduced second phase. Patients with more severe functional impairment had more pronounced abnormalities of reciprocal inhibition. Abnormalities of reciprocal inhibition may play a role in the pathophysiology of essential tremor and probably arise from defective suprasegmental control.

The physiology of idiopathic dystonia.

Dystonia is mysterious and its pathophysiology is uncertain. The fundamental motor abnormality is an abnormality of muscle command signals, such that the wrong agonists may be activated for too long, there is abnormal co-contraction of agonist and antagonists, and there is excessive and misdirected action of synergists and postural fixators. The result is action dystonia. In addition, muscle spasms may occur spontaneously. The peripheral motor apparatus, the corticomotoneurone pathway, and (as far as is known) the proprioceptive feedback machinery, are all intact in primary dystonia. However,
