Alzheimer's disease (AD) is a prevalent neurodegenerative disorder linked to cognitive decline. To understand how specific neuronal circuits are impaired in AD, we have used optogenetic and electrophysiological approaches to reveal the functional changes between prefrontal cortex (PFC) and basal forebrain (BF), 2 key regions controlling cognitive processes, in a tauopathy mouse model. We found that the glutamatergic synaptic responses in BF cholinergic neurons from P301S Tau mice (6-8 months old) were markedly diminished. The attenuated long-range PFC to BF pathway in the AD model significantl

Toxoplasma gondii is an intracellular protozoan parasite, with approximately one-third of the worlds' population chronically infected. In chronically infected individuals, the parasite resides in tissue cysts in neurons in the brain. The chronic infection in immunocompetant individuals has traditionally been considered to be asymptomatic, but increasing evidence indicates that chronic infection is associated with diverse neurological disorders such as schizophrenia, cryptogenic epilepsy, and Parkinson's Disease. The mechanisms by which the parasite exerts affects on behavior and other neuronal

Understanding how animals perceive environmental stimuli is essential for reconstructing the evolution of their sensory systems. Nematodes provide a useful model for studying sensory adaptation due to their relatively simple nervous systems and broad ecological diversity. The amphid, the primary sensory organ in nematodes, has been well characterized in Caenorhabditis elegans and other bacterivorous species. However, comparatively little is known about amphid structures in nematodes with different ecological niches. In this study, we performed serial section transmission electron microscopy a

BACKGROUND: Omnipause neurons (OPN) are glycinergic neurons that tonically inhibit burst neurons between saccades. In primates, OPNs are located bilaterally around the midline at the level of the traversing rootlets of the abducens nerve in the pontine brainstem forming the nucleus raphe interpositus (RIP). Healthy OPNs are previously characterized by dense perineuronal net (PN) ensheathment, parvalbumin (PAV) and voltage-gated potassium channel Kv1.1 and Kv3.1 expression.
MOTIVATION: The ion channel and transmitter profile of OPNs in human has not been established. The further characterizati

CDKL5 deficiency disorder (CDD) is a rare developmental and epileptic encephalopathy resulting from variants in cyclin-dependent kinase-like 5 (CDKL5) that lead to impaired kinase activity or loss of function. CDD is one of the most common genetic etiologies identified in epilepsy cohorts. To study how CDKL5 variants impact human neuronal activity, gene expression and morphology, CDD patient-derived induced pluripotent stem cells and their isogenic controls were differentiated into excitatory neurons using either an NGN2 induction protocol or a guided cortical organoid differentiation. Patient

Successful execution of behavior requires coordinated activity and communication between multiple cell types. Studies using the relatively simple neural circuits of invertebrates have helped to uncover how conserved molecular and cellular signaling events shape animal behavior. To understand the mechanisms underlying neural circuit activity and behavior, we have been studying a simple circuit that drives egg-laying behavior in the nematode worm Caenorhabditis elegans Here we show that the sex-specific, ventral C (VC) motor neurons are important for vulval muscle contractility and egg laying in

Sensory cortices process stimuli in manners essential for perception. Very little is known regarding interactions between olfactory cortices. The piriform "primary" olfactory cortex, especially its anterior division (aPCX), extends dense association fibers into the ventral striatum's olfactory tubercle (OT), yet whether this corticostriatal pathway is capable of shaping OT activity, including odor-evoked activity, is unknown. Further unresolved is the synaptic circuitry and the spatial localization of OT-innervating PCX neurons. Here we build upon standing literature to provide some answers to

Motor neuron circuitry is one of the most elaborate circuitries in our body, which ensures voluntary and skilled movement that requires cognitive input. Therefore, both the cortex and the spinal cord are involved. The cortex has special importance for motor neuron diseases, in which initiation and modulation of voluntary movement is affected. Amyotrophic lateral sclerosis (ALS) is defined by the progressive degeneration of both the upper and lower motor neurons, whereas hereditary spastic paraplegia (HSP) and primary lateral sclerosis (PLS) are characterized mainly by the loss of upper motor n

We investigated the immunohistochemical localization of neuropeptide Y (NPY) and galanin (GAL) in the brain of the masu salmon Oncorhynchus masou in order to clarify the interaction between these neuropeptide hormones in the brain. NPY-immunoreactive (ir) cell bodies were observed in the ventral and lateral regions of the ventral telencephalon (Vv and Vl, respectively), and in the dorsolateral midbrain tegmentum. NPY-ir fibers were observed throughout the brain, mainly in the ventral telencephalon, hypothalamus, optic tectum, and midbrain. GAL-ir cell bodies were observed in the nucleus preopt

Neuropeptide Y (NPY) is found in neurons of the brain and in the neurons that innervate abdominal organs including liver. Major biological function of hypothalamic NPY is regulation of appetite and body weight homeostasis. In the periphery, biological function of NPY varies, depending on the organ/tissue. Increased hypothalamic NPY mRNA level in response to chronic caloric restriction is a well documented phenomenon. The effect of food restriction on NPY mRNA level in neurons that innervate liver has not been published so far. To evaluate how chronic food restriction affects liver (and other a

The influence of central vagal stimulation induced by 2h cold exposure or intracisternal injection of thyrotropin-releasing hormone (TRH) analog, RX-77368, on gastro-duodenal enteric cholinergic neuronal activity was assessed in conscious rats with Fos and peripheral choline acetyltransferase (pChAT) immunoreactivity (IR). pChAT-IR was detected in 68%, 70% and 73% of corpus, antrum and duodenum submucosal neurons, respectively, and in 65% of gastric and 46% of duodenal myenteric neurons. Cold and RX-77368 induced Fos-IR in over 90% of gastric submucosal and myenteric neurons, while in duodenum

Both alpha- and beta-adrenergic receptors (ARs) are involved in the facilitation of the monosynaptic jaw-closing reflex in the trigeminal motor nucleus (MoV) caused by norepinephrine (NE). The amplitude of muscle spindle afferent-evoked EPSPs in masseter motor neurons is 65% greater when noradrenergic axons to the motor nucleus are concomitantly activated and seems to be due to a presynaptic mechanism (Vornov, J. J., and J. Sutin. 1986. J. Neurosci. 6: 30-37). To determine the subtypes of ARs located on motor neurons and other cells, the cytotoxic lectin Ricin communis was injected into the ma

Evidence is given of the location in man of the fibres going to the sympathetic neurons of the lateral horn that supply the intrinsic and extrinsic muscles of the eye and the sweat glands of the head and neck. For the region of the pons and medulla, the evidence is abstracted from the literature. For the cervical spinal cord, the evidence is from our cases of anterolateral cordotomy. In the medulla, thrombosis of the artery of the fossette latérale destroys the fibres; this locates the fibres in the posterolateral retro-olivary area. But not all fibres to the sudomotor neurons lie there: some

Spinal muscular atrophy (SMA), a progressive neurodegenerative disease affecting motor neurons, is caused by mutations or deletions of the SMN1 gene encoding the survival of motor neuron (SMN) protein. In immortalized non-neuronal cell lines, SMN has been shown to form a ribonucleoprotein (RNP) complex with Gemin proteins, which is essential for the assembly of small nuclear RNPs (snRNPs). An additional function of SMN in neurons has been hypothesized to facilitate assembly of localized messenger RNP complexes. We have shown that SMN is localized in granules that are actively transported into

Spinal muscular atrophy (SMA) is a recessive disorder involving the loss of motor neurons from the spinal cord. Homozygous absence of the survival of motor neuron 1 gene (SMN1) is the main cause of SMA, but disease severity depends primarily on the number of SMN2 gene copies. SMN protein levels are high in normal spinal cord and much lower in the spinal cord of SMA patients, suggesting neuron-specific regulation for this ubiquitously expressed gene. We isolated genomic DNA from individuals with SMN1 or SMN2 deletions and sequenced 4.6 kb of the 5' upstream regions of the these. We found that t

Cerebrospinal Fluid-contacting neurons (CSF-cNs) are GABAergic bipolar neurons found, in contact with the cerebrospinal fluid, along the vertebrate medullo-spinal central canal. They express Polycystin Kidney Disease 2-Like 1 channels (PKD2L1), members of the Transient Receptor Potential superfamily, and were shown to modulate motor activity and therefore suggested to act as a novel sensory system. However, in mice, they remain largely uncharacterized and it is crucial to comprehensively characterize their morphological and electrophysiological properties to determine whether they form a homog

Unconsciousness can be induced by sleep and general anaesthesia alike; however, the extent to which these states differ is not well understood. In this study, we aimed to determine the similarities and differences between the unconscious states caused by sleep and those caused by anaesthesia, as well as investigate gene expression profiles by applying tensor decomposition-based unsupervised feature extraction to the two gene expression profiles. One of the two expression profiles was obtained from mice treated with sevoflurane, a type of inhaled anaesthesia, whereas the other two expression pr

The caudal nucleus of the solitary tract (NTS) serves as the site of the first synapse for visceral sensory inputs to the central nervous system. The NTS sends functional projections to multiple brain nuclei, with gastric-related projections primarily targeting the dorsal motor nucleus of the vagus (DMV). Previous studies have demonstrated that the majority of caudal NTS neurons that project to the DMV respond robustly to nicotine and express nicotinic acetylcholine receptors (nAChRs). However, the cytochemical identity and relationship with specific viscera of DMV-projecting, nicotine-respons

Kv2 family "delayed-rectifier" potassium channels are widely expressed in mammalian neurons. Kv2 channels activate relatively slowly and their contribution to action potential repolarization under physiological conditions has been unclear. We explored the function of Kv2 channels using a Kv2-selective blocker, Guangxitoxin-1E (GxTX-1E). Using acutely isolated neurons, mixed voltage-clamp and current-clamp experiments were done at 37°C to study the physiological kinetics of channel gating and action potentials. In both rat superior cervical ganglion (SCG) neurons and mouse hippocampal CA1 pyram

BACKGROUND: Ileal transposition (IT) can effectively resolve obesity and improve type 2 diabetes. IT is associated with increased glucagon-like peptide 1 secretion. The mechanisms mediating the effects of IT on obesity and diabetes remain undefined. Given the role of pro-opiomelanocortin neurons in energy balance, we sought to determine its potential role in these processes.
METHODS: Twenty non-obese diabetic Goto-Kakizaki rats underwent either IT or sham operation. Various measures including food intake, body weight, fasting plasma glucose, glucagon-like peptide 1 level, activated pro-opiome
