When injected intravenously (1 mg/kg) and intrathecally (100 micrograms) to rats, serotonin produced an analgesic effect in thermal pain and failed to affect nociceptive responses under electric irritation. When given in the analgesic dose, it unchanged the baseline level of blood pressure, but enhanced its nociceptive pressor responses. Naloxone (0.1 mg/kg) reduced the sedative action of serotonin and prevented the increase of pressor responses of blood pressure induced by its intrathecal administration.

The antinociception of opiates is mediated through the activation of opioid receptors in several mid brain and brain stem areas. This paper reports that the forebrain area termed area tempestas (AT), first identified as a convulsant trigger area, is also a component of the endogenous pain suppression system. Unilateral AT application of DAMGO, morphine and U-50,488H in rats at doses in the nanogram range produced marked and dose-dependent increases in the latency to respond to nociceptive stimuli. A lower effect is found after application of DPDPE and DADLE. Antinociception is more evident in

The putative peptide neurotransmitter cholecystokinin (CCK) is co-localized with substance P (SP) in a dense cluster of neurons located in the rat Edinger-Westphal (EW) nucleus. In an attempt to record electrophysiologically from these CCK-containing neurons, we have identified a group of nociceptive neurons located within the confines of the EW nucleus. The firing pattern of these nociceptive neurons is erratic, sometimes with a bursting pattern and at other times with fairly regular rates which vary generally from 1 to 7 Hz. These neurons respond to noxious stimuli, such as toe pinch, with a

The present study describes the segmental variation in the distribution and morphology of nociceptive neurons (N cells) in the central nervous system of the leech. N cells of midbody ganglia can be segregated into lateral and medial types. We show that monoclonal antibodies specific for N cells can distinguish between the two populations. The monoclonal antibodies were used to map the complete distribution of the cells along the nervous cord. There are two pairs of the medial and lateral nociceptive neurons in the midbody ganglia, one pair of the medial type in the sex ganglia (5 and 6), and a

This study investigated the local inhibition of a reflexive cutaneous nociceptive response produced by nalbuphine and naloxone after subcutaneous administration and compared their pharmacological profiles with those of lidocaine. At the single equimolar dose tested (39.3 μmol/kg), nalbuphine produced significantly greater local cutaneous nociceptive blockade than naloxone. Dose-response analyses were subsequently performed for nalbuphine and lidocaine. Both nalbuphine and lidocaine produced dose-dependent local cutaneous nociceptive blockade, and no significant difference in ED50 (50% effectiv

INTRODUCTION: Naloxone blocks Na+ channels and produces cutaneous analgesia, but it is unknown whether naloxone or its analog, nalbuphine, produces spinal anesthesia. The purpose of this study was to assess the effects of intrathecal nalbuphine and naloxone on motor and nociceptive blockade.
METHODS: Sprague-Dawley rats received intrathecal injections of nalbuphine, naloxone, and the local anesthetic lidocaine (n = 6 for each group). Following intrathecal injection, motor function (extensor strength) and nociceptive responses (pinch test) were assessed. A combination of intrathecal nalbuphine

Direct oral anticoagulants (DOACs) are widely prescribed in patients at risk of thromboembolism. Although they are generally safer than warfarin, DOACs still carry a risk of bleeding and drug-drug interactions (DDIs). Nociceptive pain is common and may frequently occur in patients receiving DOAC therapy. In these cases, the European Society of Cardiology recommends avoiding non-steroidal anti-inflammatory drugs (NSAIDs) and preferring acetaminophen. However, acetaminophen is not effective when an inflammatory component is present. Consequently, non-pharmacological approaches or topical NSAIDs

Somatosensory synaptic transmission is an adapting phenomenon according to changes of synaptic environments composed of neurons and glial cells. Synaptic architecture in the spinal dorsal horn well recognizes and identifies sensory modalities, such as nociceptive and non-nociceptive, however, sensory neurons do not identify only single modality, therefore, synaptic transmission is dynamic and phasic events. More importantly, maladaptive synaptic plasticity developed chronically and critically drives the new setup of synaptic environments that results in enhanced or decreased synaptic transmiss

BACKGROUND: The trigeminal nerve innervates the face and the meninges via its three branches, with central relay neurons located in the brainstem's spinal trigeminal nucleus. We have shown a somatotopic organization on human brainstem level and aimed, using an independent new sample, to characterize the functional somatotopy with a focus on within-brainstem and brainstem-cortical connectivity.
METHODS: Electrical stimulation was applied to the three branches of the trigeminal nerve at two intensities (high and low) to examine the somatotopic organization in healthy participants during fMRI. N

Peripersonal space (PPS) is the area surrounding the body where we can reach, see, and interact with objects, acting as a dynamic interface between us and the environment. We expected that pain stimuli closer to the body center in the PPS would result in higher pain intensity, increment in heart rate (HR), and skin-conductance response (SCR). It was also assumed that proprioceptive systematic error toward the body center, and psychological factors such as negative affect, pain catastrophizing, and somatosensory amplification would show a positive association with pain. Sixty individuals partic

Exposing the skin to high levels of ultraviolet B (UVB) radiation induces an inflammatory response that upregulates local nociceptive processing; this, in turn, facilitates protective responses to limit further injury. In this study, the UVB model was used to explore additional effects of inflammation on supraspinal nociceptive processing. Thirty-one healthy participants attended two sessions approximately 24 h apart. In each session, pressure-pain thresholds and sensitivity to sharp stimulation and heat were assessed in both forearms, and pressure-pain thresholds and sensitivity to sharp stim

The periaqueductal gray (PAG) is located in the mesencephalon in the upper brainstem and, as part of the descending pain modulation, is considered a crucial structure for pain control. Its modulatory effect on painful sensation is often seen as a systemic function affecting the whole body similarly. However, recent animal data suggest some kind of somatotopy in the PAG. This would make the PAG capable of dermatome-specific analgesic function. We electrically stimulated the three peripheral dermatomes of the trigemino-cervical complex and the greater occipital nerve in 61 humans during optimize

BACKGROUND: The nociceptive flexion reflex (NFR) is a polysynaptic and multisegmental spinal reflex that develops in response to a noxious stimulus and is characterized by the withdrawal of the affected body part. The NFR possesses two excitatory components: early RII and late RIII. Late RIII is derived from high-threshold cutaneous afferent A-delta fibers, which are prone to injury early in the course of diabetes mellitus (DM) and may lead to neuropathic pain. We investigated NFR in patients with DM with different types of polyneuropathies to analyze the role of NFR in small fiber neuropathy

Temporomandibular disorders (TMDs) are characterized by numerous pain manifestations. Their treatment often involves the use of an oral splint. Recent research has found a relationship between migraines, nociceptive pain and TMDs. The aim of the present study was to perform a scoping review of studies in order to evaluate the effectiveness of the various types of oral splint in the treatment of migraine or nociceptive pain. Publications were retrieved from seven databases (PubMed, Web of Science, EMBASE, Scopus, ProQuest, SpringerLink and Ovid). Out of the 15 included publications, three studi

OBJECTIVE: Neurophysiological investigation of nociceptive pathway has so far been limited to late cortical responses. We sought to detect early components of the cortical evoked potentials possibly reflecting primary sensory activity.
METHODS: The 150 IDE micropatterned electrode was used to selectively activate Aδ intraepidermic fibres of the right hand dorsum in 25 healthy subjects and 3 patients suffering from trigeminal neuralgia. Neurographic recordings were performed to assess type of stimulated fibres and check selectivity. Cortical evoked potentials were recorded from C3'-Fz and Cz-A

Dorsal root ganglia (DRG) sensory neurons can transmit information about noxious stimulus to cerebral cortex via spinal cord, and play an important role in the pain pathway. Alterations of the pain pathway lead to CIPA (congenital insensitivity to pain with anhidrosis) or chronic pain. Accumulating evidence demonstrates that nerve damage leads to the regeneration of neurons in DRG, which may contribute to pain modulation in feedback. Therefore, exploring the regeneration process of DRG neurons would provide a new understanding to the persistent pathological stimulation and contribute to reshap

Orofacial pain or tenderness is a primary symptom associated with temporomandibular joint (TMJ) disorders (TMDs). To understand the pathological mechanisms underlying TMDs, several mouse models have been developed, including mechanical stimulus-induced TMD and genetic mouse models. However, a lack of feasible approaches for assessing TMD-related nociceptive behaviours in the orofacial region of mice has hindered the in-depth study of TMD-associated mechanisms. This study aimed to explore modifications of three existing methods to analyse nociceptive behaviours using two TMD mouse models: (1) m

KEY POINTS: We investigated the cardiovascular and respiratory responses of the normotensive Wistar-Kyoto (WKY) rat and the spontaneously hypertensive (SH) rat to inhalation and intravenous injection of the noxious stimuli allyl isothiocyanate (AITC). AITC inhalation evoked atropine-sensitive bradycardia in conscious WKY rats, and evoked atropine-sensitive bradycardia and atenolol-sensitive tachycardia with premature ventricular contractions (PVCs) in conscious SH rats. Intravenous injection of AITC evoked bradycardia but no tachycardia/PVCs in conscious SHs, while inhalation and injection of

Pain is a common nonmotor symptom of Parkinson's disease (PD) that remains neglected and misunderstood. Elucidating the nondopaminergic circuitry may be key to better understanding PD and improving current treatments. We investigated the role of monoamines in nociceptive behavior and descending analgesic circuitry in a rat 6-hydroxydopamine (6-OHDA)-induced PD model and explored the resulting motor dysfunctions and inflammatory responses. Rats pretreated with noradrenaline and serotonin reuptake inhibitors were given unilateral striatal 6-OHDA injections and evaluated for mechanical hyperalges
