Safety and activity of selinexor in patients with myelodysplastic syndromes or oligoblastic acute myeloid leukaemia refractory to hypomethylating agents: a single-centre, single-arm, phase 2 trial.

BackgroundThe median overall survival of patients with high-risk myelodysplastic syndromes refractory to hypomethylating agents is less than 6 months. Currently, no standard therapy for such patients exists. Preclinical studies have shown that inhibition of the nuclear export protein exportin 1 (XPO1) causes nuclear accumulation of p53 and disruption of NF-κB signalling, both relevant targets for myelodysplastic syndromes. We therefore aimed to assess the safety and activity of selinexor in patients with myelodysplastic syndromes or oligoblastic acute myeloid leukaemia refractory to hypomethyl

ANXA2 suppresses antiviral immunity by impeding STING Golgi translocation and disrupting the TBK1/IKKε-IRF3 axis.

UNLABELLED: Upon viral infection, the cGAS-STING axis is pivotal for initiating type I interferon (IFN) production against DNA viruses. In this study, we identified Annexin A2 (ANXA2) as a novel and potent negative regulator of this antiviral pathway. Overexpression of ANXA2 inhibits herpes simplex virus-1 (HSV-1)-induced IFN-β production, thereby promoting viral replication, whereas ANXA2 deficiency enhances IFN responses and restricts HSV-1 replication. Mechanistically, ANXA2 operates through a dual mechanism. First, it interacts with the transmembrane domain of STING and inhibits its agonis

Extracellular proteins from Perkinsus olseni: LC-MS/MS-based proteomic profiling and functional insights into host-parasite interactions.

Perkinsus olseni is a protozoan parasite that causes significant economic losses in molluscan aquaculture due to perkinsosis in clams and oysters. However, the molecular mechanisms underlying its pathogenicity are still poorly understood. In this study, we performed high-resolution liquid chromatography-tandem mass spectrometry (LC-MS/MS) combined with integrative bioinformatics to generate the first comprehensive profile of the P. olseni extracellular proteome ("secretome"). A total of 619 proteins were identified, with functional annotations revealing a diverse secretome involved in multiple

Nucleolar Localization of HIV-1 Rev Is Required, Yet Insufficient for Production of Infectious Viral Particles.

Combination antiretroviral therapy fails in complete suppression of HIV-1 due to drug resistance and persistent latency. Novel therapeutic intervention requires knowledge of intracellular pathways responsible for viral replication, specifically those untargeted by antiretroviral drugs. An understudied phenomenon is the nucleolar localization of Rev phosphoprotein, which completes nucleocytoplasmic transport of unspliced/partially spliced HIV mRNA through multimerization with intronic cis-acting targets-the Rev-response element (RRE). Rev contains a nucleolar localization signal (NoLS) comprisi

XPO1 inhibition enhances the efficacy and durability of RAS-targeted therapy in preclinical models of KRASG12D mutant pancreatic ductal adenocarcinoma.

KRASG12D-selective and pan-RAS inhibitors have shown promise in pancreatic ductal adenocarcinoma (PDAC), yet adaptive resistance is anticipated to limit durability of response. Exportin 1 (XPO1), a nuclear export protein frequently overexpressed in PDAC, represents a potential vulnerability in KRAS-mutant cancers. We evaluated whether pharmacologic inhibition of XPO1 enhances therapeutic efficacy and durability of KRAS pathway inhibition. KRASG12D inhibitor- and pan-RAS inhibitor-resistant PDAC cellular models were generated and assessed for sensitivity to the second-generation XPO1 inhibitor

PET imaging of XPO1 engagement with [18F]selinexor: A pharmacodynamic theranostic strategy for multiple myeloma.

Exportin 1 (XPO1), a nuclear export protein frequently overexpressed in multiple myeloma (MM), represents a validated therapeutic target. However, noninvasive imaging approaches capable of assessing XPO1 engagement and pharmacodynamic modulation during therapy remain limited. Here, we present the radiosynthesis and preclinical evaluation of [18F]selinexor, an XPO1-targeted positron emission tomography (PET) radiotracer derived from the clinically used drug selinexor via an 18F/19F isotope exchange method. This labeling approach preserves the parent drug's molecular structure and pharmacologica

CD73 inhibitor enhances the antitumor activity of selinexor in multiple myeloma by restoring the activation of CD8+ T cells.

Selinexor, a selective nuclear export inhibitor, blocks exportin-1 and reduces oncoprotein messenger RNA translation, and is approved for the treatment of relapsed/refractory multiple myeloma in combination with proteasome inhibitors, immunomodulators, anti-CD38 antibodies, etc. Nevertheless, selinexor presents limited monotherapy efficacy with an objective response rate of 29% and fails to completely overcome drug resistance. To improve the therapeutic outcome, this study investigated the combined treatment of selinexor and a CD73 inhibitor in a murine tumor model. The expression of CD73 in m

The x-ray crystal structure of the first RNA recognition motif and site-directed mutagenesis suggest a possible HuR redox sensing mechanism.

Hu-antigen R (HuR) is a ubiquitous RNA-binding protein that comprises three RNA recognition motifs (RRMs). The first two tandem RRMs are known to bind to AU-rich elements (AREs) in the 3'-untranslated region of many mRNAs. The third RRM is connected to the second RRM through a basic hinge region that contains a localization signal termed HuR nucleocytoplasmic shuttling. Binding of HuR to the ARE in the 3'-untranslated region of mRNA leads to nuclear export, stabilization, and/or translational de-repression of the mRNA, resulting in upregulation of the encoded protein. Among the various ARE bin

Reprimo (RPRM) mediates neuronal ferroptosis via CREB-Nrf2/SCD1 pathways in radiation-induced brain injury.

Neuronal ferroptosis has been found to contribute to degenerative brain disorders and traumatic and hemorrhagic brain injury, but whether radiation-induced brain injury (RIBI), a critical deleterious effect of cranial radiation therapy for primary and metastatic brain tumors, involves neuronal ferroptosis remains unclear. We have recently discovered that deletion of reprimo (RPRM), a tumor suppressor gene, ameliorates RIBI, in which its protective effect on neurons is one of the underlying mechanisms. In this study, we found that whole brain irradiation (WBI) induced ferroptosis in mouse brain

An external sensing system in Plasmodium falciparum-infected erythrocytes.

BACKGROUND: A number of experiments have previously indicated that Plasmodium falciparum-infected erythrocytes (pRBC) were able to sense host environment. The basis of this ability to detect external cues is not known but in screening signalling molecules from pRBC using commercial antibodies, a 34 kDa phosphorylated molecule that possesses such ability was identified. METHODS: The pRBC were exposed to different culture conditions and proteins were extracted for 1D or 2D gel electrophoresis followed by Western blot. The localization of 34 kDa protein was examined by biochemical fractionation

Molecular role of GATA binding protein 4 (GATA-4) in hyperglycemia-induced reduction of cardiac contractility.

BACKGROUND: Diabetic cardiomyopathy, a diabetes-specific complication, refers to a disorder that eventually leads to left ventricular hypertrophy in addition to diastolic and systolic dysfunction. In recent studies, hyperglycemia-induced reactive oxygen species (ROS) in cardiomyocytes have been linked to diabetic cardiomyopathy. GATA binding protein 4 (GATA-4) regulates the expression of many cardio-structural genes including cardiac troponin-I (cTnI). METHODS: Streptozotocin-induced diabetic rats and H9c2 embryonic rat cardiomyocytes treated with a high concentration of glucose (a D-glucose

Host Factor Nucleophosmin 1 (NPM1/B23) Exerts Antiviral Effects against Chikungunya Virus by Its Interaction with Viral Nonstructural Protein 3.

Chikungunya virus (CHIKV) hijacks host cell machinery to support its replication. Nucleophosmin 1 (NPM1/B23), a nucleolar phosphoprotein, is one of the host proteins known to restrict CHIKV infection; however, the mechanistic details of the antiviral role of NPM1 are not elucidated. It was seen in our experiments that the level of NPM1 expression affected the expression levels of interferon-stimulated genes (ISGs) that play antiviral roles in CHIKV infection, such as IRF1, IRF7, OAS3, and IFIT1, indicating that one of the antiviral mechanisms could be through modulation of interferon-mediated

Re-engineering segment 8 facilitates generation of a versatile live-attenuated influenza A virus vector platform for secretory protein delivery.

UNLABELLED: Influenza A viruses (IAVs) lacking the nonstructural protein 1 (NS1) gene (IAV-ΔNS1) are attenuated both in vitro and in vivo, yet retain replicative capacity in interferon signaling-deficient systems. To date, several efforts have been pursued to expand its functionality by incorporating transgenes in lieu of the NS1 gene. However, such vectors require complex segment designs, retain residual NS1 peptide stretches, or display suboptimal cytokine secretion, largely due to the constraints related to the partial nuclear export protein (NEP) open reading frame (ORF) upstream of the tr

Anti-Influenza Activity of the Ribonuclease Binase: Cellular Targets Detected by Quantitative Proteomics.

Unpredictable influenza pandemics, annual epidemics, and sporadic poultry-to-human avian influenza virus infections with high morbidity and mortality rates dictate a need to develop new antiviral approaches. Targeting cellular pathways and processes is a promising antiviral strategy shown to be effective regardless of viral subtypes or viral evolution of drug-resistant variants. Proteomics-based searches provide a tool to reveal the druggable stages of the virus life cycle and to understand the putative antiviral mode of action of the drug(s). Ribonucleases (RNases) of different origins not on

The Toxoplasma gondii effector GRA83 modulates the host's innate immune response to regulate parasite infection.

UNLABELLED: Toxoplasma gondii 's propensity to infect its host and cause disease is highly dependent on its ability to modulate host cell functions. One of the strategies the parasite uses to accomplish this is via the export of effector proteins from the secretory dense granules. Dense granule (GRA) proteins are known to play roles in nutrient acquisition, host cell cycle manipulation, and immune regulation. Here, we characterize a novel dense granule protein named GRA83, which localizes to the parasitophorous vacuole in tachyzoites and bradyzoites. Disruption of GRA83 results in increased vi

Marine-derived oral nanovesicles from Sargassum fusiforme ameliorate steatohepatitis by activating the HO-1 pathway to inhibit NF-κB signaling and restore small intestinal homeostasis.

BACKGROUND: Marine algae, notably the edible brown seaweed Sargassum fusiforme (Hijiki), harbor a vast repertoire of bioactive molecules with potent anti-inflammatory and metabolic regulatory properties. However, their clinical translation is severely hampered by poor systemic bioavailability and susceptibility to gastrointestinal degradation. To circumvent these bottlenecks, we engineered Sargassum fusiforme-derived nanovesicles (SF-NVs) as a robust, intrinsic oral delivery platform. These nanovesicles effectively encapsulate and protect bioactive cargos, significantly augmenting their stabil

Subcellular Localization of HIV-1 gag-pol mRNAs Regulates Sites of Virion Assembly.

Full-length unspliced human immunodeficiency virus type 1 (HIV-1) RNAs serve dual roles in the cytoplasm as mRNAs encoding the Gag and Gag-Pol capsid proteins as well as genomic RNAs (gRNAs) packaged by Gag into virions undergoing assembly at the plasma membrane (PM). Because Gag is sufficient to drive the assembly of virus-like particles even in the absence of gRNA binding, whether viral RNA trafficking plays an active role in the native assembly pathway is unknown. In this study, we tested the effects of modulating the cytoplasmic abundance or distribution of full-length viral RNAs on Gag tr

Thrombosis-related circulating miR-16-5p is associated with disease severity in patients hospitalised for COVID-19.

SARS-CoV-2 tropism for the ACE2 receptor, along with the multifaceted inflammatory reaction, is likely to drive the generalized hypercoagulable and thrombotic state seen in patients with COVID-19. Using the original bioinformatic workflow and network medicine approaches we reanalysed four coronavirus-related expression datasets and performed co-expression analysis focused on thrombosis and ACE2 related genes. We identified microRNAs (miRNAs) which play role in ACE2-related thrombosis in coronavirus infection and further, we validated the expressions of precisely selected miRNAs-related to thro
