Diabetes through a 3D lens: organoid models.

Diabetes is one of the most challenging health concerns facing society. Available drugs treat the symptoms but there is no cure. This presents an urgent need to better understand human diabetes in order to develop improved treatments or target remission. New disease models need to be developed that more accurately describe the pathology of diabetes. Organoid technology provides an opportunity to fill this knowledge gap. Organoids are 3D structures, established from pluripotent stem cells or adult stem/progenitor cells, that recapitulate key aspects of the in vivo tissues they mimic. In this re

Human Organoids for the Study of Retinal Development and Disease.

Recent advances in stem cell engineering have led to an explosion in the use of organoids as model systems for studies in multiple biological disciplines. Together with breakthroughs in genome engineering and the various omics, organoid technology is making possible studies of human biology that were not previously feasible. For vision science, retinal organoids derived from human stem cells allow differentiating and mature human retinal cells to be studied in unprecedented detail. In this review, we examine the technologies employed to generate retinal organoids and how organoids are revoluti

Organoid as a culture system for viral vaccine strains.

Organoid is an in vitro multicellular form mimicking in vivo organ. Its similarity to human organ including cellular organization, molecular expression patterns, as well as genetic signatures enables to study the characteristics of infectious agents and host-pathogen interaction. For the features of organoid, this system also can be potentially used to cultivate currently uncultivable viruses of vaccine candidates. This paper will briefly describe problems in the current culture system for virus production and the possibility of organoid as culture system for viral vaccine and their current li

Patient-derived organoids from pancreatic cancer after pancreatectomy: Feasibility and organoid take rate in treatment-naïve periampullary tumors.

BACKGROUND/OBJECTIVE: Patient-derived organoids (PDOs) have emerged as essential for ex vivo modelling for pancreatic cancer (PDAC) but reports on efficacy and organoid take rate are scarce. This study aimed to assess the feasibility of establishing PDOs from resected specimens in periampullary tumors. METHODS: Patients undergoing surgery for suspected periampullary cancer were included. PDO protocol amendments were tested, with organoid take rate as outcome measure. Samples from resected specimens were processed and expanded per protocol. Pooled estimate of take rates of PDOs in PDAC was der

Developing liver organoids from induced pluripotent stem cells (iPSCs): An alternative source of organoid generation for liver cancer research.

Primary liver cancer (PLC) represents a significant proportion of all human cancers and constitutes a substantial health and economic burden to society. Traditional therapeutic approaches such as surgical resection and chemotherapy often fail due to tumour relapse or innate tumour chemoresistance. There is a dearth of efficient treatments for PLC in part due to the poor capacity of current laboratory models to reflect critical features of the native tumour in vivo. The increasing incorporation of organoid systems has led to a resurgence of interest in liver cancer research. Organoid systems sh

CODNet models enable real-time assessment of colorectal cancer organoids and uncover the integral role of LTBP2 in the diffuse organoid subtype.

To achieve precise classification of colorectal cancer (CRC) at the patient-derived organoids (PDOs) level, we constructed a real-time CRC PDO assessment system through artificial intelligence (AI) assisted model using an optimized object detection algorithm. We constructed the CODNetV1 model for the identification, feature extraction and drug sensitivity detection of CRC PDOs (mAP of 88.05%). And we optimized CODNetV2 to classify four morphological subtypes of CRC PDOs (mAP of 85.65%). CODNetV1 and CODNetV2 achieved real-time, automated, multi-parameter detection of CRC PDOs, directly facilit

The Organoid Decade: Leveraging 3D Patient-Derived Organoids to Bridge the Translational Gap in Triple-Negative Breast Cancer: A Systematic Review.

Triple-negative breast cancer (TNBC) is an aggressive subtype of breast cancer with limited effective therapies. Two-dimensional (2D) in vitro models poorly recapitulate tumor microenvironment (TME) interactions, impeding the translational relevance of TNBC immunotherapy research. Three-dimensional patient-derived tumor organoids (3D PDTOs) have emerged as advanced preclinical models that better mimic tumor-immune interactions. The objective of this systematic review was to assess the landscape of 3D PDTO adoption in TNBC research and evaluate their application in addressing key bottlenecks in

Adrenomedullary Organoids Derived from Neural Crest and Schwann Cell Precursors.

The adrenal gland, essential for survival, contains the medulla and cortex. Chromaffin cells (ChrCs) in the adrenal medulla control the fight-or-flight response by releasing catecholamines like epinephrine and norepinephrine. Recent findings reveal that ChrCs originate from two progenitors: 20% from developmentally early, neural crest-derived sympathoadrenal progenitors (SAPs) and 80% from the later Schwann cell precursors (SCPs). We generated functional adrenomedullary organoids from human pluripotent stem cells (hPSCs) via both pathways and in mixed (20% SAP, 80% SCP-derived) cultures. When

Modeling Respiratory Infections Using Patient-Derived Apical-Out Airway Organoids.

Airway organoids bridge the gap between conventional epithelial cultures and animal models by combining human tissue architecture, cellular diversity, and experimental accessibility. However, because these systems are dynamic, reproducible infection modeling requires careful definition of parameters influencing infection readouts, including epithelial polarity, maturation, cellular composition, and access to a physiologically relevant apical surface. We established patient-derived apical-out airway organoids and characterized them longitudinally using multiple modalities throughout differentia

Organoids in polycystic ovary syndrome: Emerging platforms and future directions.

Polycystic ovary syndrome (PCOS) is a prevalent and heterogeneous endocrine disorder affecting women of reproductive age, characterized by hyperandrogenism, ovulatory dysfunction, polycystic ovarian morphology, and metabolic disturbances, including insulin resistance. Despite its high global burden, mechanistic understanding remains incomplete due to limitations of conventional animal models and two-dimensional cultures, which fail to recapitulate the structural organization, endocrine dynamics, and patient-specific variability of human reproductive tissues. Organoid technology has emerged as

LNA-Probe-Based MicroRNA In Situ Hybridization in Brain Organoids.

MicroRNAs (miRNAs) are short regulatory RNAs that take part in posttranscriptional regulation. Their mature and active form is limited to 18-24 nucleotides and therefore sets demands on probe design for specific detection by in situ hybridization (ISH) analysis. Locked nucleic acids (LNA) are synthetic RNA analogs that augment binding affinity when incorporated into antisense oligos. In this ISH protocol, we applied LNA-enhanced probes in an automated set-up using a Ventana Discovery Ultra instrument for detection of miRNAs in formalin-fixed and paraffin embedded (FFPE) iPSC-derived brain orga

Intermittent Propofol Exposure Induces Neurodevelopmental Alterations in Human Brain Organoids.

The administration of anaesthesia during pregnancy may have implications for fetal brain development. This study used H1 embryonic stem cell-derived human brain organoids (HBOs) to investigate effects of intermittent propofol exposure (IPE). HBOs were subjected to early IPE from 47 to 50 days in vitro (div), or late IPE from 77 to 80 div, using a clinically supra-anaesthetic concentration of 50 µM propofol. This was followed by cultivation without propofol for an additional 10 div, and HBOs were subsequently analysed at 60 or 90 div. Determination of HBO growth and lactate release did not prov

Modeling cardiac electrical activity using cardiac organoids for drug screening.

Arrhythmia is one of the leading causes of mortality and lacks diagnostic and therapeutic options. Electrical activity at the cellular level is difficult to use as a basis for evaluating drug effects on cardiac rhythm. Cardiac organoids, as organized and functional cell clusters, offer significant advantages as models for the pathophysiological mechanisms of arrhythmia or as drug screening platforms. Here, we applied a standardized set of electrophysiological techniques to integrate electrophysiological activity of cardiac organoids, including multielectrode array (MEA), calcium signal optical

Human thyroid organoids: tools for understanding thyroid diseases and development.

The thyroid gland develops in five main stages, each characterized by specific molecular and cellular processes that are highly conserved across invertebrates and vertebrate species. In vivo studies have elucidated the key molecular players orchestrating each stage of thyroid organogenesis and thyroid hormone biosynthesis, laying the groundwork for developing thyroid organoids in vitro. Thyroid organoids have been successfully derived from mouse and human pluripotent stem cells, including embryonic stem cells and induced pluripotent stem cells, as well as from fetal and adult thyroid tissues.

Development and characterization of functional sheep endometrial luminal epithelial organoids.

The luminal epithelium (LE) serves as the primary interface for embryo-maternal communication during implantation. In ruminants such as sheep, the LE orchestrates critical reproductive events, including noninvasive adhesion, conceptus elongation, and pregnancy recognition. However, existing models face significant limitations: conventional two-dimensional (2D) cultures fail to recapitulate endometrial complexity, while current organoid systems are predominantly derived from glandular epithelium. Herein, we report the establishment of a robust three-dimensional (3D) culture system for generatin

Human hair regeneration using organoids and hair-on-chip technologies.

The hair follicle is a complex, dynamic mini organ that plays various important roles in facilitating the function of human skin. As human hair follicles do not naturally form after birth, there is a demand for methods to restore hair follicles in conditions involving permanent hair loss, such as scarring alopecia. However, robust therapeutics to regenerate hair follicles in humans have yet to be identified. Recent advances in the field of regenerative medicine show promise for human hair restoration. Regenerative medicine aims to restore lost tissues, such as hair follicles, by generating bio

Accessory Inner Segment-Like Structures Develop in Human Retinal Organoids.

PURPOSE: A novel photoreceptor structure, called the accessory inner segment (aIS), was recently identified in human rod photoreceptors. It is described as a microtubule-based, mitochondria-rich extension of the conventional inner segment attached externally to the outer segment. Human retinal organoids recapitulate many developmental and mature cellular features of the human retina. Thus, we sought to determine if retinal organoids exhibit an aIS-like structure, to support future in vitro studies of its role in photoreceptor structure and disease. METHODS: Mature retinal organoids were deriv

Cardiac organoids bridge translational gaps in 3R-compliant safety testing.

Cardiotoxicity remains a leading cause of late-stage drug attrition and post-marketing withdrawal, underscoring persistent limitations in current preclinical safety assessment paradigms. Traditional animal models are constrained by interspecies differences in electrophysiology, ion channel composition, myocardial remodeling, and pharmacokinetics, while two-dimensional human induced pluripotent stem cel-derived cardiomyocytes (hiPSC-CMs) lack tissue-level, functional maturity, organization and multicellular complexity. These limitations create a translational gap between cellular assays and who

Patient-derived organoids to study glycosylation dynamics during gastric disease.

Aberrant cellular glycosylation is a key event that accompanies and actively sustains gastric neoplastic transformation. Patient-derived organoids (PDOs) have recently emerged as promising ex vivo models to study human gastric diseases; however, their glycosylation landscape remains unknown. To evaluate gastric PDOs as avatars of in vivo tissue glycosylation, a biobank of gastric PDOs (n = 56) was generated from gastric mucosa samples of non-tumoral obese patients (n = 11), adjacent tumor mucosa (n = 26), and gastric tumor tissue (n = 19). PDOs reproduce distinct stages of gastric carcinogenes
