Cutaneous metastasis of extraskeletal osteosarcoma arising in the mediastinum.

Cutaneous metastases have been reported in 5%-10% of patients with internal malignancy. The most common clinical features are the sudden appearance of discrete, firm, and nontender nodules in a particular area of the body. The most frequent metastatic site of extraskeletal osteosarcoma (ESOS) is the lungs. Cutaneous metastasis of ESOS is rare. To our knowledge, only one case of ESOS has been reported with skin metastasis, histopathologically classified as the osteoblastic subtype. Here, we describe a case of chondroblastic ESOS arising in the mediastinum with metastasis to the skin. Our patien

Primary osteosarcoma of the breast: report of 2 cases.

Two distinct histologic variants of primary breast osteosarcoma in 2 elderly women are described. The first patient was an 88-year-old woman with a long-standing, slow-growing, 18-cm mass in her right breast. The second patient was a 96-year-old woman with a recently self-detected, painless, 7.5-cm lump in her left breast. Clinically, there was no evidence of metastasis, and both women underwent simple mastectomy. Histologic features of both specimens were those of high-grade primary breast osteosarcoma. The first patient's tumor was classified as a chondroblastic variant, and the second as an

A review of clinical and molecular prognostic factors in osteosarcoma.

Traditional prognostic determinants in osteosarcoma have included demographics (age, sex), tumour size, site, stage, and the response to chemotherapy. Many of these are determined using varying techniques and units of measurement, which can make comparison between studies difficult. The absence of survival difference between limb sparing surgery and amputation has been repeatedly demonstrated in primary disease, and even in the setting of pathological fracture. On the other hand, there is still some controversy over the existence of increased local recurrence for limb-sparing surgery, and the

Osteosarcoma presenting as a solitary focus of osteolysis in the calvarium.

Osteolysis of the skull is fairly common. The clinical presentation and results of laboratory or imaging tests may suggest a benign or a malignant disease. However, histology is the only means of obtaining a definitive diagnosis and should therefore be performed at the slightest doubt. We report the case of a 58-year-old man who presented with a recent painless lump in the right temporoparietal region. Laboratory tests were normal. Brain imaging studies and total body radionuclide scanning showed no other lesions. However, the heterogeneous and aggressive appearance of the skull lesion prompte

The clinical and diagnostic imaging findings of osteosarcoma of the jaw.

OBJECTIVE: To clarify the valuable clinical features and diagnostic imaging findings regarding the diagnosis of osteosarcoma of the jaw (OSJ). MATERIALS AND METHODS: The initial symptoms and diagnostic imaging findings of 10 patients with OSJ were analysed. The points analysed on the diagnostic images were as follows: any widening of the periodontal ligament space of the teeth on the periphery of the OSJ; the presence of radial spicules and Codman's triangle; any signs of bone destruction; and the patterns of osteogenesis. RESULTS: All patients had pain and/or swelling of the affected site,

Optimal management of methotrexate intoxication in a child with osteosarcoma.

OBJECTIVE: To describe the time course and management of methotrexate (MTX) toxicity in a 14-year-old Hispanic boy with osteosarcoma treated with high-dose MTX. CASE SUMMARY: During the sixth cycle of high-dose MTX, severe intoxication was observed with high MTX plasma concentrations, acute renal failure, and hepatitis, followed by mucositis and moderate myelosuppression. Intensification of urine alkalinization and increased leucovorin dosages did not decrease plasma concentrations of MTX or prevent systemic toxicities. Carboxypeptidase G2 and aminophylline were thus administered as a second-

Primary osteosarcoma arising in the colon: report of a case.

A 53-year-old female presented with abdominal pain, and computed tomography scan revealed a small, calcified lesion in the wall of the transverse colon. The symptoms later disappeared spontaneously, and she remained in good health. However, four years later, she developed lancinating abdominal pain and was admitted to our hospital. A large tumor with calcification was found in the left upper abdominal cavity. Curative resection of the tumor was performed, and the histology was compatible with extraskeletal osteosarcoma. We speculated that the tumor originated from the colonic wall, because a s

Osteosarcoma as a second malignancy after treatment for neuroblastoma.

A 4-month old girl was diagnosed as having stage IV neuroblastoma of the right adrenal gland. Preoperative chemotherapy was given, followed by local surgical excision. Postoperatively, irradiation of the tumor bed and adjuvant chemotherapy was given for 11 months. Nine years after cessation of chemotherapy, the patient developed left hip-joint pain. Biopsy of the ischium showed chondroblastic osteosarcoma. Limb salvage surgery was performed after preoperative chemotherapy. Postoperatively, adjuvant chemotherapy was given for 14 months. Twenty-two months after treatment for the secondary osteos

Atypical osteosarcomas in Werner Syndrome (adult progeria).

Werner syndrome (WS), adult progeria, is more common in Japan than elsewhere. It predisposes to osteosarcoma (OS) and five other rare tumors. To determine if and how OS is atypical in this genetic disorder, we studied the characteristics of ten Japanese cases with respect to clinical features, pathology, and radiographs, and compared them with a hospital series of 36 skeletal OS with the same atypical age-range, 35 - 57 years. The anatomic sites were also atypical: seven ankle / foot, two radius and one patella compared with only one at the ankle in the hospital series. The osteoblastic cell-t

Extraskeletal osteosarcoma of the mandibular salivary gland in a dog.

A 14-year-old spayed female shepherd/collie crossbred dog had a 6 x 4-cm mass excised from below the right ear. The partially encapsulated, neoplastic mass had a necrotic core and was embedded in the mandibular salivary gland. Histologically, the mass was composed of numerous multinucleate giant cells and large, oval, pleomorphic cells that produced osteoid. Neoplastic cells were immunoreactive for vimentin and actin but not for keratin, desmin, or S-100 protein. At postmortem examination 1 month later, an 11-cm spherical mass had recurred at the surgical site, and there were metastatic nodule

Osteosarcoma of the pelvis. Oncologic results of 40 patients.

The cases of 40 patients with osteosarcoma of the pelvis treated between 1977 and 1994 were reviewed. The location of the tumor was ilium in 30 patients, ischium in four, pubis in one, and sacrum in five. Most (58%) of the tumors were of the chondroblastic subtype. Thirty patients had surgical excision of the tumors: 10 with hemipelvectomies and 20 with limb sparing procedures. A wide margin was achieved in 16 of 30 (53%) patients, including 12 of 14 who had no sacral tumor involvement. Positive margins occurred at the sacrum in 11 patients, lumbar vertebra in one, perirectal space in one, and

Osteosarcoma of the scaphoid. A case report and review of the literature.

We describe a case of osteosarcoma of the scaphoid bone, which to our knowledge is only the second reported case of osteosarcoma in the carpus. A 38-year-old man complained of intense pain in the right wrist and had curettage and a bone graft for a lesion in the scaphoid. Histological examination showed this to be an osteosarcoma. Below-elbow amputation was performed and adjuvant chemotherapy given. There has been no evidence of recurrence or metastases at 33 months after amputation.

The surgical treatment of parosteal osteosarcoma in long bones.

The cases of twenty-five patients with Stage-I parosteal osteosarcoma were analyzed to ascertain the incidence of recurrence after forty-two various surgical procedures that produced an intracapsular, marginal, wide, or radical excisional margin. Ten intracapsular resections were followed by local recurrence in each patient. This procedure usually was done to shell-out a presumably benign lesion. Marginal excision was successful in nine of seventeen patients. When a marginal excision was knowingly done to preserve a major neurovascular bundle, the risk of recurrence was less (three of eleven)

[Primary osteosarcoma of the base of the skull in Garcin syndrome].

This contribution presents a case study of a 17-year-old female patient with unilateral failure of several brain nerves within a period of five months. Further findings included fleeting homolateral signs of the small brain and, during the terminal phase, discrete hemiparesis. Diagnosis was based on the tomograms of the skull base and computerized tomography, which revealed a tumour at the base of the skull. During the obduction this was found to be a primary osteosarcoma. According to the literature it is rare for this kind of tumour to lead to Garcin 's syndrome.

Molecular characterization of cell dynamics during osteosarcoma progression.

BACKGROUND: Osteosarcoma is the most common malignant bone disease. The current clinical treatment is difficult to deal with the high heterogeneity within tumors and the cellular dynamic remodeling during the metastasis process of osteosarcoma. METHODS: Cell annotation was performed using the SingleR algorithm in combination with specific marker genes. Further subcluster analysis was conducted on osteoblasts, myeloid cells, fibroblasts, malignant cells and T/NK cells. Cell differentiation trajectories were constructed using Monocle2 and scTour for pseudotime analysis. The spatial transcriptom

Ldha Regulates Osteosarcoma Lung Metastasis through Hedgehog Signaling.

UNLABELLED: Osteosarcoma is the most common malignant bone tumor in children and adolescents, with a high propensity for lung metastasis, leading to poor prognosis. Lactate dehydrogenase A (LDHA), a key glycolytic enzyme, has been implicated in the progression and metastasis of various cancers. In this study, we demonstrated that Ldha knockdown reduced cell migration and invasion in murine osteosarcoma cells and decreased lung metastasis. RNA sequencing analysis revealed that Ldha knockdown was associated with the downregulation of Hedgehog signaling. Consistently, pharmacologic inhibition of

Case Report: Feline spinous process giant-cell osteosarcoma.

Two adult domestic shorthair cats were evaluated for progressive neurologic deficits caused by spinal cord compression secondary to vertebral osteosarcoma, giant-cell subtype. Feline giant cell osteosarcoma represents a rare tumor type, particularly uncommon in the vertebral column. The cats showed progressive paraparesis or hemiparesis, with neuroanatomic localization to C1-C5 and T3-L3 respectively. Magnetic resonance imaging revealed a single, well-demarcated osseous mass arising from the spinous process for and lamina for each patient (case 1 at C6 and case 2 at T4). Both lesions produced

Systemic strategies for osteosarcoma: advances and future directions.

Osteosarcoma is a common and aggressive bone cancer in children and adolescents, typically affecting the long bones during growth spurts. Treatment involves a combination of surgery, chemotherapy, and radiotherapy. Although surgery has become more limb-preserving, the overall prognosis remains challenging, with a 5-year survival rate of 60-70% for localized disease and much lower for advanced stages. Chemotherapy is the mainstay treatment; however, it can cause severe side effects. Targeted therapies (e.g., IGF-1R inhibitors, TKIs) and immunotherapy (e.g., checkpoint inhibitors, CAR-T therapie
