Roles of amylin in satiation, adiposity and brain development.

Amylin plays an important role in the control of nutrient fluxes. It is cosecreted with insulin and reduces eating by promoting meal-ending satiation. This effect seems to depend on a stimulation of amylin receptors in the area postrema. Subsequent to area postrema activation, the neural signal is conveyed to the forebrain via distinct relays in the nucleus of the solitary tract and the lateral parabrachial nucleus to the lateral hypothalamic area and other hypothalamic nuclei; the functional roles of these relays in amylin's eating inhibitory effect have not been fully investigated. Amylin ma

Dopamine cell degeneration induced by intraventricular administration of 6-hydroxydopamine in the rat: similarities with cell loss in parkinson's disease.

In an attempt to find a convenient rat model to study cell vulnerability in Parkinson's disease, we have investigated the cell-loss profile in different midbrain dopaminergic nuclei and subnuclei of rats injected with 6-hydroxydopamine (6-OHDA) in the third ventricle. Following administration of different doses (5-1000 microgram) of 6-OHDA, motor behavior was evaluated and tyrosine hydroxylase-immunostained neurons were counted in the A8 group and different subdivisions of A9 and A10 groups. Animals developed hypokinesia, repetitive chewing movements, and catalepsia. Signs of cell degeneration

Aldosterone-sensitive HSD2 neurons in mice.

Sodium deficiency elevates aldosterone, which in addition to epithelial tissues acts on the brain to promote dysphoric symptoms and salt intake. Aldosterone boosts the activity of neurons that express 11-beta-hydroxysteroid dehydrogenase type 2 (HSD2), a hallmark of aldosterone-sensitive cells. To better characterize these neurons, we combine immunolabeling and in situ hybridization with fate mapping and Cre-conditional axon tracing in mice. Many cells throughout the brain have a developmental history of Hsd11b2 expression, but in the adult brain one small brainstem region with a leaky blood-b

Organization of projections from the ventromedial nucleus of the hypothalamus: a Phaseolus vulgaris-leucoagglutinin study in the rat.

The organization of projections from the four parts of the ventromedial nucleus (VMH) and a ventrolaterally adjacent region tentatively identified as the tuberal nucleus (TU) have been analyzed with small injections of the anterograde axonal tracer Phaseolus vulgaris-leucoagglutinin (PHA-L). Extrinsic and intranuclear projections of each part of the VMH display clear quantitative differences, whereas the overall patterns of outputs are qualitatively similar. Overall, the VMH establishes massive intrahypothalamic terminal fields in other parts of the medial zone, tending to avoid the periventri

Effect of sex chromosome complement on sodium appetite and Fos-immunoreactivity induced by sodium depletion.

Previous studies indicate a sex chromosome complement (SCC) effect on the angiotensin II-sexually dimorphic hypertensive and bradycardic baroreflex responses. We sought to evaluate whether SCC may differentially modulate sexually dimorphic-induced sodium appetite and specific brain activity due to physiological stimulation of the rennin angiotensin system. For this purpose, we used the "four core genotype" mouse model, in which the effect of gonadal sex and SCC is dissociated, allowing comparisons of sexually dimorphic traits between XX and XY females as well as in XX and XY males. Retrograde transport of brain-derived neurotrophic factor (BDNF) following infusion in neo- and limbic cortex in rat: relationship to BDNF mRNA expressing neurons.

Brain-derived neurotrophic factor (BDNF) was the second member of the nerve growth factor (NGF) family to be isolated. The ability of BDNF to be retrogradely transported following intraparenchymal infusion represents a unique neurobiological tool to determine the location of putative neuron-specific BDNF-responsive neuronal systems. In the present study, we infused recombinant human (rh) BDNF into the rodent neo- and limbic cortex and used a turkey anti-BDNF antibody to determine specific populations of neurons which retrogradely transport this neurotrophin. Frontal cortex infusion retrogradel

Neural Pathways of Craniofacial Muscle Pain: Implications for Novel Treatments.

Craniofacial muscle pain is highly prevalent in temporomandibular disorders but is difficult to treat. Enhanced understanding of neurobiology unique to craniofacial muscle pain should lead to the development of novel mechanism-based treatments. Herein, we review recent studies to summarize neural pathways of craniofacial muscle pain. Nociceptive afferents in craniofacial muscles are predominantly peptidergic afferents enriched with TRPV1. Signals from peripheral glutamate receptors converge onto TRPV1, leading to mechanical hyperalgesia. Further studies are needed to clarify whether hyperalges

Human brainstem activation underpinning offset analgesia changes in perceived pain intensity. An ultra-high field functional magnetic resonance imaging investigation.

Offset analgesia is a pain modulatory phenomenon whereby a slight decrease in the intensity of a painful stimulus results in a large, disproportionate reduction in perceived pain. While human brain imaging studies have begun to map the brain circuitry underpinning offset analgesia, the brainstem circuits responsible for this phenomenon remain unknown. Furthermore, it is now recognised that previously described analgesic paradigms can evoke pain increases in some individuals, but whether the same brainstem circuitry is responsible for inhibitory and paradoxical facilitatory responses remains to

Presynaptic Inputs to Any CNS Projection Neuron Identified by Dual Recombinant Virus Infection.

Although neuroanatomical tracing studies have defined the origin and targets of major projection neurons (PN) of the central nervous system (CNS), there is much less information about the circuits that influence these neurons. Recently, genetic approaches that use Cre recombinase-dependent viral vectors have greatly facilitated such circuit analysis, but these tracing approaches are limited by the availability of Cre-expressing mouse lines and the difficulty in restricting Cre expression to discrete regions of the CNS. Here, we illustrate an alternative approach to drive Cre expression specifi

Postnatal expression of the transcription factor Ebf2 in motivation, reward, and pain-related circuits of the mouse brain.

INTRODUCTION: Early B-cell factor 2 (Ebf2) is a transcription factor required for neuronal differentiation. However, its postnatal expression pattern and functional roles in the brain are not well characterized. This study examined the spatial distribution of Ebf2 in postnatal day 10 (P10) mouse brains and investigated its association with neural circuits mediating motivation, reward, and nociception. MATERIALS AND METHODS: Ebf2-TGFP transgenic mice, which express green fluorescent protein (GFP) as a reporter for Ebf2, were utilized. Immunofluorescence labeling and high-resolution microscopy

Opioid receptors mRNAs expression and opioids agonist-dependent G-protein activation in the rat brain following neuropathy.

Potent opioid-based therapies are often unsuccessful in promoting satisfactory analgesia in neuropathic pain. Moreover, the side effects associated with opioid therapy are still manifested in neuropathy-like diseases, including tolerance, abuse, addiction and hyperalgesia, although the mechanisms underlying these effects remain unclear. Studies in the spinal cord and periphery indicate that neuropathy alters the expression of mu-[MOP], delta-[DOP] or kappa-[KOP] opioid receptors, interfering with their activity. However, there is no consensus as to the supraspinal opioidergic modulation provok

Effects of albumin-conjugated PYY on food intake: the respective roles of the circumventricular organs and vagus nerve.

The mechanism and routes through which peptide tyrosine-tyrosine (PYY) exerts its anorectic effects are still largely unknown. In the present study, we investigated the roles of the area postrema (AP), subfornical organ (SFO) and vagus nerve in mediating the anorectic effect of PYY using PYY(3-36) conjugated to human serum albumin (PYY(3-36)-HSA) in rats. PYY(3-36)-HSA is a large molecule that does not penetrate the blood-brain barrier, and thus provides a useful tool to discriminate between the central (brain) and peripheral actions of this peptide. PYY(3-36)-HSA induced significant reduction

The anorectic hormone amylin contributes to feeding-related changes of neuronal activity in key structures of the gut-brain axis.

Amylin is a peptide hormone that is cosecreted with insulin from the pancreas during and after food intake. Peripherally injected amylin potently inhibits feeding by acting on the area postrema (AP), a circumventricular organ lacking a functional blood-brain barrier. We recently demonstrated that AP neurons are excited by a near physiological concentration of amylin. However, the subsequent neuronal mechanisms and the relevance of endogenously released amylin for the regulation of food intake are poorly understood. Therefore, we investigated 1) amylin's contribution to feeding-induced c-Fos ex

Detailed mapping of CGRP mRNA expression in the rat central nervous system: comparison with previous immunocytochemical findings.

The localization of CGRP mRNA in neurons of the rat brain and spinal cord was assessed by in situ hybridization histochemistry (ISH) using a radiolabeled synthetic 57-mer oligodeoxynucleotide probe complementary to the rat prepro CGRP mRNA. Results were compared with previously published findings of CGRP-immunoreactive (CGRP-IR) cell bodies revealed by an indirect immunofluorescence technique. The highest numbers of CGRP mRNA expressing neurons as well as the greatest intensity of staining were found in the lateral hypothalamic area, the parabrachial nuclei, and among the cranial motor nuclei,

Inhibition of cardiopulmonary input to thoracic spinothalamic tract cells by stimulation of the subcoeruleus-parabrachial region in the primate.

Effects of electrically stimulating the subcoeruleus-parabrachial (SC-PB) region on 31 spinothalamic tract neurons which receive excitatory input from cardiopulmonary sympathetic afferent fibers were studied in 21 monkeys anesthetized with chloralose. A conditioning stimulus to the SC-PB region inhibited the activity of 5 cells responding to the test stimulus applied to sympathetic afferent fibers. At a conditioning-test interval of 10 ms, test responses were maximally reduced to 47 +/- 6% of the control. Inhibitory effects were present at conditioning-test intervals up to 150 ms. Excitatory e

Parabrachial Calca Neurons Influence Aversive and Appetitive Taste Function.

The parabrachial (PB) nucleus participates in taste processing and integration with other senses. PB neurons that express the Calca gene support sensory-integrative responses, albeit only limited data have addressed their influence on taste. Here we investigated how chemogenetic dampening of PB-Calca neurons affected mouse orosensory preferences for diverse taste stimuli in brief-access fluid exposure tests, which capture oral sensory/tongue control of licking behavior. Intracranial delivery of Cre-dependent viruses in female and male Calca Cre/+ mice induced expression of the inhibitory desig

METTL14 integrates tumor-derived SAM to drive parabrachial epigenetic rewiring in pancreatic cancer.

Tumor-neural crosstalk drives cancer progression and neurological symptoms, yet how tumors rewire brain circuits remains unclear. Here, we identified upregulated methyltransferase-like 14 (METTL14) in lateral parabrachial nucleus glutamatergic neurons (LPBNGlu) as a key regulator of pain-depression comorbidity in pancreatic ductal adenocarcinoma (PDAC) mice. METTL14 inhibition reversed PDAC-induced neuronal hyperexcitability and alleviated behavioral deficits. Mechanistically, METTL14 coordinated the tumor-derived S-adenosylmethionine (SAM) to promote N6-methyladenosine (m6A) modification of a

Satiation and re-intake after partial withdrawal of gastric food contents: A dissociation effect in external lateral parabrachial lesioned rats.

Sensory information from the gastrointestinal system can be transmitted to the brain through the vagus nerve, the intermediate-caudal region of the nucleus of the solitary tract (NST), and various subnuclei of the parabrachial complex, notably the external lateral subnucleus (LPBe). The objective of the present study was to examine the relevance of this subnucleus in satiation and food reintake after gastrointestinal food removal. LPBe-lesioned animals were subjected to a re-intake task following the partial withdrawal of gastric food contents shortly after satiation. Lesioned and control anim

A spatial and projection-based transcriptomic atlas of paraventricular hypothalamic cell types.

The paraventricular hypothalamus (PVH) controls behavioral and physiologic processes, including appetite, social behavior, autonomic outflow, and pituitary hormone secretion. However, molecular markers for centrally projecting PVH neuron populations remain largely undefined, and a complete census of PVH cell types has not been established. Therefore, we performed extensive single-cell/nucleus RNA sequencing to catalog PVH neuron subtypes and multiplexed error-robust fluorescence in situ hybridization (MERFISH) to map them spatially. Our spatial transcriptomic atlas resolves 26 Sim1+ and 29 GAB

Parabrachial oxytocin receptor-expressing neurons link social observation of distress to defensive behavior.

The ability to detect and respond to threat signals in the environment, including those conveyed by the distress of a familiar social partner, is fundamental to survival and disrupted in a range of neuropsychiatric conditions. This study identifies oxytocin receptor (Oxtr)-expressing neurons within the lateral parabrachial nucleus (lPBN) of mice as a key node in the neural circuitry underlying threat-related and social behaviors. These Oxtr neurons are activated by aversive stimuli and by observing demonstrator mice in stressful situations, including foot shock or inflammatory pain. Chemogenet
