Antibody localization of extensin in cell walls of carrot storage roots.

The accumulation and cross-linking of hydroxyproline-rich glycoproteins (HRGPs) in cell walls of dicotyledonous plants has been correlated with a number of wall-strengthening phenomena. Polyclonal antibodies raised against glycosylated extensin-1, the most abundant HRGP in carrot (Daucus carota L.) cell walls, recognize this antigen on gel and dot blots and on thin sections of epoxy-embedded carrot-root cell walls. Since wall labeling can be largely reduced by preincubating the antibodies with purified extensin-1, most labeling can be attributed to recognition of this antigen. The remaining la

Single-Cell Insights into Medicinal Plant Development and Metabolism.

Medicinal plants are major sources of therapeutic natural products, yet the cell-type-specific organization that governs metabolite biosynthesis, transport, and storage remains imperfectly resolved by organ-level omics. This review synthesizes studies published up to June 2026 that used single-cell, single-nucleus, spatial, metabolomic, and epigenomic approaches to medicinal plant systems, following a PRISMA-guided literature search across PubMed, Web of Science, Scopus, and CNKI. Emerging evidence shows that specialized metabolism is organized through discrete and often rare cell populations,

The insect effector CSPs activate an atypical plant TNL immune receptor.

CSP-mediated activation of RCSP involves noncanonical structural and catalytic mechanisms-including BB-loop deletion, α5α6-loop insertion, and atypical catalytic residues within the RCSP-TIR domain. Chemosensory proteins (CSPs), a conserved effector family in phloem-feeding insects such as aphids and the brown planthopper (Nilaparvata lugens), induce dwarfism in Nicotiana benthamiana through recognition by the Toll-interleukin-1 receptor (TIR) domain-containing nucleotide-binding leucine-rich repeat receptor (TNL) RCSP. However, CSP-mediated RCSP activation and enzymatic mechanism remain unkno

Jasmonic and salicylic acid pathways shape the rhizosphere microbiome, affecting aphid herbivory and soil-mediated insect-plant interactions in a density-dependent manner.

Aboveground induction of plant defense pathways can shape root-associated microbial communities. However, whether these changes are pathway-specific and how they affect plant growth and resistance remain unclear. We evaluated how induction of the jasmonic acid (JA) and salicylic acid (SA) defense pathways shapes the root microbiome of Brassica oleracea and whether these soil-mediated shifts affect plant growth and resistance to herbivory in a subsequent generation using a plant-soil feedback approach. In the conditioning phase, defense pathways were induced either through foliar application of

Multilevel Mechanisms by Which Membrane Transport Proteins Regulate Plant Responses to Heavy Metal Stress: From Ion Perception and Uptake Control to Systemic Signaling and Translational Perspectives.

Soil heavy metal contamination-including cadmium (Cd), arsenic (As), lead (Pb), mercury (Hg), zinc (Zn) excess, and copper (Cu) excess-poses a critical threat to sustainable agriculture and food safety. Membrane transport proteins serve as primary interfaces governing ion entry, subcellular compartmentalization, and long-distance redistribution. This review integrates recent advances across multiple scales to construct a unified regulatory framework: (i) at the cell periphery, plasma membrane transporters of the NRAMP, ZIP, and COPT families mediate metal influx, whereas efflux transporters an

CsNIP2 and CsNIP16 facilitated Al-F transport and hyperaccumulation in tea plants (Camellia sinensis).

Although Al-F complexes (e.g., AlF2+, AlF2+) exist in the tea apoplast and symplast, their root uptake mechanisms and translocation to mature leaves remain elusive. Here, we identified two plasma membrane-localized Nodulin 26-like Intrinsic Proteins (NIPs), CsNIP2 and CsNIP16, as crucial mediators of Al-F complex transport. CsNIP2 is predominantly expressed in roots and enriched in the phloem-epidermic part tissues, whereas CsNIP16 is predominantly expressed in stems and abundantly in xylem tissues. Heterologous expression in yeast revealed that both transporters facilitate Al-F uptake, signif

Unveiling the Pharmacognosy of an Aerial Parasite: Scurrula cordifolia and the Transfer of Medicinal Potential from Host to Mistletoe.

Mistletoes are obligate hemi-parasitic angiosperms that establish complex ecological and physiological relationships with their forest hosts through specialized haustorial systems. To maintain nutrient flux, these parasites withstand more negative water potentials than their hosts, utilizing succulent leaves for strategic rehydration. Although root parasites have been extensively studied, the absorptive structures and host-parasite dynamics of the Loranthaceae family, the largest group of mistletoes, remain under-investigated. This study explores the genus Scurrula, specifically the less-studi

Temperature-Directed Reprogramming of Volatile and Semi-Volatile Metabolism in Ginkgo biloba Microclones Under Cold and Heat Stress.

Temperature is a major determinant of plant metabolic plasticity, yet its role in directing volatile and semi-volatile specialized metabolism in Ginkgo biloba remains poorly understood. In this study, we investigated how contrasting low- and high-temperature treatments reshape secondary metabolite contents in G. biloba microclones cultivated in vitro. Plants were exposed to cold (+3 °C) and heat (+30 °C) conditions, and their responses were analyzed using GC-MS profiling, anatomical measurements, chlorophyll fluorescence, and multivariate statistics. Cold treatment selectively increased the ab

A comprehensive characteristic of Anastatica hierochuntica.

Anastatica hierochuntica (Brassicaceae) is a desert plant used globally to treat various diseases. To advance research on A. hierochuntica, we conducted studies on its morphology, anatomy, physicochemical parameters, and antioxidant properties. Morphological analysis revealed that the whole plant was light yellow with an irregular umbrella shape; the stem, roots, leaves, and fruits each have distinct structural features. Light microscopy and scanning electron microscope (SEM) identified numerous vessels, including reticulate and spiral vessels, along with non-glandular hairs, phloem fibers, ep

Host-driven structuring shapes the landscape of phytoplasma diversity across Indian agroecosystems revealed by multilocus phylogenomics.

Phytoplasmas are phloem-limited, wall-less plant pathogens that impose significant biotic stress on host plants, resulting in substantial yield losses worldwide. However, their genetic diversity, host associations, and population structure in tropical agroecosystems remain poorly understood. This study presents a nationwide multilocus assessment of 'Ca. Phytoplasma' diversity, distribution, and population structure across Indian agroecosystems. A survey (2020-2023) across ten states identified 124 phytoplasma-positive samples from 42 host species and associated leafhopper insect vectors. Multi

Foliar-Applied EDTA-Zn Reduces Grain Cd Content in Triticum aestivum L. Grown in Alkaline Fields with Cd Contamination by Regulating Vascular Bundle-Mediated Transport and Sequestration of Cd in Nodes and Rachises.

Foliar application of zinc (Zn) compounds is a promising strategy for mitigating cadmium (Cd) accumulation in wheat grains in pot experiments. However, its inconsistent field efficacy, especially in alkaline soils with long-term Cd contamination, poses a unique challenge. Herein, field experiments and demonstrations were conducted at three sites across four wheat-growing winter seasons to evaluate the efficacy of foliar Zn application in reducing grain Cd accumulation in typical Cd-contaminated alkaline soils. Foliar application of EDTA-Zn at the booting and milk stages increased grain yield b

Foliar-applied honokiol exhibits basipetal translocation, offering a strategy for root disease management in precision agriculture systems.

INTRODUCTION: Root diseases in crops are increasingly becoming the most difficult to control due to the challenge of fungicides reaching infection sites. Honokiol, a natural fungicidal product derived from Magnolia officinalis, is primarily biosynthesized in leaves and subsequently translocated to roots to resist pathogen infection. METHODS: In this study, honokiol applied to the leaves of Polygonatum cyrtonema Hua (PCH) could be transported downward to the roots and exhibited a control effect against root rot disease. RESULTS: Subsequent research showed that concentration, temperature, pH,

Spider venom neurotoxin based bioinsecticides: A novel bioactive for the control of the Asian citrus psyllid Diaphorina citri (Hemiptera).

The Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Psyllidae), is a key vector of the phloem-limited bacteria Candidatus Liberibacter asiaticus (CLas) associated with huanglongbing (HLB), the most serious and currently incurable disease of citrus worldwide. Here we report the first investigation into the potential use of a spider venom-derived recombinant neurotoxin, ω/κ-HxTx-Hv1h (hereafter HxTx-Hv1h) when delivered alone or when fused to snowdrop lectin (Galanthus nivalis agglutinin; GNA) to control D. citri. Proteins, including GNA alone, were purified from fermented transforme

Mutations in CmVPS41 controlling resistance to cucumber mosaic virus display specific subcellular localization.

Resistance to cucumber mosaic virus (CMV) in melon (Cucumis melo L.) has been described in several exotic accessions and is controlled by a recessive resistance gene, cmv1, that encodes a vacuolar protein sorting 41 (CmVPS41). cmv1 prevents systemic infection by restricting the virus to the bundle sheath cells, preventing viral phloem entry. CmVPS41 from different resistant accessions carries two causal mutations, either a G85E change, found in Pat-81 and Freeman's cucumber, or L348R, found in PI161375, cultivar Songwhan Charmi (SC). Here, we analyzed the subcellular localization of CmVPS41 in

Iron Availability within the Leaf Vasculature Determines the Magnitude of Iron Deficiency Responses in Source and Sink Tissues in Arabidopsis.

Iron (Fe) uptake and translocation in plants are fine-tuned by complex mechanisms that are not yet fully understood. In Arabidopsis thaliana, local regulation of Fe homeostasis at the root level has been extensively studied and is better understood than the systemic shoot-to-root regulation. While the root system is solely a sink tissue that depends on photosynthates translocated from source tissues, the shoot system is a more complex tissue, where sink and source tissues occur synchronously. In this study, and to gain better insight into the Fe deficiency responses in leaves, we overexpressed

The salivary effector protein Sg2204 in the greenbug Schizaphis graminum suppresses wheat defence and is essential for enabling aphid feeding on host plants.

Aphids secrete diverse repertoires of salivary effectors into host plant cells to promote infestation by modulating plant defence. The greenbug Schizaphis graminum is an important cereal aphid worldwide. However, the secreted effectors of S. graminum are still uncharacterized. Here, 76 salivary proteins were identified from the watery saliva of S. graminum using transcriptome and proteome analyses. Among them, a putative salivary effector Sg2204 was significantly up-regulated during aphid feeding stages, and transient overexpression of Sg2204 in Nicotiana benthamiana inhibited cell death induc

The complete genome sequence of 'Candidatus Liberibacter solanacearum', the bacterium associated with potato zebra chip disease.

Zebra Chip (ZC) is an emerging plant disease that causes aboveground decline of potato shoots and generally results in unusable tubers. This disease has led to multi-million dollar losses for growers in the central and western United States over the past decade and impacts the livelihood of potato farmers in Mexico and New Zealand. ZC is associated with 'Candidatus Liberibacter solanacearum', a fastidious alpha-proteobacterium that is transmitted by a phloem-feeding psyllid vector, Bactericera cockerelli Sulc. Research on this disease has been hampered by a lack of robust culture methods and p

The determination of ferric iron in plants by HPLC using the microbial iron chelator desferrioxamine E.

Common methods for plant iron determination are based on atomic absorption spectroscopy, radioactive measurements or extraction with subsequent spectrophotometry. However, accuracy is often a problem due to background, contamination and interfering compounds. We here describe a novel method for the easy determination of ferric iron in plants by chelation with a highly effective microbial siderophore and separation by high performance liquid chromatography (HPLC). After addition of colourless desferrioxamine E (DFE) to plant fluids, the soluble iron is trapped as a brown-red ferrioxamine E (Fox
