Cationic polystyrene nanosphere toxicity depends on cell-specific endocytic and mitochondrial injury pathways.

The exponential increase in the number of new nanomaterials that are being produced increases the likelihood of adverse biological effects in humans and the environment. In this study we compared the effects of cationic nanoparticles in five different cell lines that represent portal-of-entry or systemic cellular targets for engineered nanoparticles. Although 60 nm NH(2)-labeled polystyrene (PS) nanospheres were highly toxic in macrophage (RAW 264.7) and epithelial (BEAS-2B) cells, human microvascular endothelial (HMEC), hepatoma (HEPA-1), and pheochromocytoma (PC-12) cells were relatively res

Extracorporeal adsorption of anti-factor VIII allo-antibodies on randomly functionalized polystyrene resins.

The occurrence of anti-factor VIII (FVIII) allo-antibodies is a severe complication of the treatment of haemophilia A patients, leading to the inhibition of transfused FVIII activity. The effective elimination of these inhibitory antibodies plays a decisive role in the management of affected patients. To achieve this, immunoadsorption devices employing synthetic adsorbers, which selectively eliminate inhibitors, are of interest in the treatment strategy of haemophilia A patients with inhibitors. Adsorbers consisting of polystyrene-based beads substituted with sulphonate and L-tyrosyl methylest

Use of floating layers of polystyrene beads to control populations of the filaria vector Culex quinquefasciatus.

Floating layers of polystyrene beads suffocate mosquito larvae and pupae and inhibit egg laying. The layers are very durable in breeding sites with water contained within walls, as in wet pit latrines and soakage pits. In some areas such pits constitute an important breeding site for Culex quinquefasciatus. Trials have been conducted in communities in Zanzibar, Tanzania, and in Tamil Nadu, India, where such mosquito populations were the vectors of Wuchereria bancrofti. In each case, treatment of all the pits with polystyrene beads was integrated with mass treatment of the people with antifilar

Comparative efficacy of thallium adsorption by activated charcoal, prussian blue, and sodium polystyrene sulfonate.

BACKGROUND: Although Prussian blue is considered the antidote of choice for thallium poisoning, the lack of a Food and Drug Administration-approved pharmaceutical formulation has led to the search for other adsorbents. Activated charcoal has been demonstrated to adsorb thallium in vitro, and the similarity between thallium and potassium has led some authors to consider the use of sodium polystyrene sulfonate as a potential adsorbent. This experiment was designed to compare the relative thallium binding efficacy of these agents in a standard isotherm model. METHODS: A standard aqueous solution

Polystyrene Microplastics Impair Cellular Immune Responses in Galleria mellonella Following Controlled Internal Exposure.

Microplastics (MPs) are emerging environmental contaminants with increasing evidence of immunotoxic effects in invertebrates; however, their underlying cellular mechanisms remain insufficiently understood. This study investigated whether short-term internal exposure to 1 μm polystyrene microplastics affects immune and oxidative responses in Galleria mellonella. Larvae were injected with 1, 5, 20, 40, and 80 mg/mL PS-MPs (n = 16 per parameter), and total hemocyte count, encapsulation and melanization responses, phenoloxidase (PO) activity, antioxidant enzymes (SOD and CAT), malondialdehyde (MDA

Polystyrene nanoplastics impair mitochondrial function, quality control, and ER-mitochondria crosstalk in HeLa cells.

INTRODUCTION: Polystyrene nanoplastics (PS-NPs) represent persistent environmental contaminants derived from the degradation of plastic debris and are increasingly detected in biological systems, raising concerns regarding their potential effects on human health. Emerging evidence suggests that, following cellular internalization, PS-NPs can induce intracellular stress responses and interfere with mitochondrial homeostasis, highlighting mitochondria as key targets of nanoplastic-induced cellular dysfunction. This study aimed to investigate the effects of PS-NPs on mitochondrial function in HeL

The impacts of polystyrene nanoplastics on colorectal cancer: Evidence from integrated network toxicity and in vitro approaches.

Environmental ingestion of polystyrene nanoplastics (PNPLs) raises critical public health concerns, yet their impacts on colorectal cancer (CRC) remain obscure. In this study, network toxicology, and in vitro validation were integrated to evaluate the molecular mechanisms linking PNPLs to CRC. We identified 154 overlapping targets by integrating GEO-derived differentially expressed genes with PNPLs-predicted targets and CRC-associated genes, which are enriched in pathways associated with cancer signaling, apoptosis, oxidative stress, and inflammation. Using protein-protein interaction network,

Factors Influencing the Clearance of Polystyrene Microplastics by Oysters: Roles of Environmental Conditions, Food Availability and Selection.

This study investigated the abiotic and biotic factors regulating the removal of polystyrene microplastics (PS-MPs) from the water column by the oyster Crassostrea hongkongensis. Our data revealed that MP sequestration is highly dependent on environmental conditions and food characteristics. Optimal clearance occurred at 25 °C and medium estuarine salinity (20 psu), while extreme thermal stress (30 °C) and hyposaline conditions (5 psu) significantly depressed filtration. Light condition had little effect on the removal of PS-MPs. Increased food density remarkably stimulated oyster's clearance

Aged polystyrene nanoplastics induce cardiac developmental toxicity associated with mitochondrial dysfunction in zebrafish larvae.

Environmental aging alters the physicochemical properties of micro/nanoplastics (MNPs) and may modify their biological effects; however, the cardiac developmental toxicity of aged MNPs remains poorly understood. In this study, ultraviolet-aged polystyrene nanoplastics (aged PSNPs) were prepared as a laboratory model of photoaged nanoplastics, and zebrafish embryos were exposed to pristine or aged PSNPs for 7 days to evaluate developmental cardiotoxicity and mitochondrial mechanisms. UV aging increased surface roughness and cracks, reduced particle diameter from 93.43 nm to 87.5 nm, shifted zet

Polystyrene nanoplastics induce pyroptosis in HepG2 cells via the YAP1-cGAS-STING signaling axis.

BACKGROUND: Polystyrene nanoplastics (PS-NPs) are emerging environmental contaminants with documented hepatotoxic potential, yet the mechanisms underlying their liver toxicity remain incompletely understood. OBJECTIVE: This study aimed to determine whether PS-NPs induce pyroptosis in human hepatocellular carcinoma (HepG2) cells and to elucidate the involvement of the Yes-associated protein 1 (YAP1)-cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) signaling axis. METHODS: HepG2 cells were exposed to PS-NPs, followed by evaluation of cytotoxicity, reactive oxygen species (

Dual-Functional Polystyrene-Based Borate Complexes: Recyclable Alkyl Radical Sources and Photodegradable Absorbents.

Recyclable polymeric materials capable of generating radicals under mild conditions are valuable materials in synthetic chemistry. Herein, we report polystyrene-based copolymers bearing 2,2'-(pyridine-2,6-diyl)diphenol (PDP) moieties that form alkyl borate complexes and release alkyl radicals upon blue-light irradiation. The generated primary and secondary alkyl radicals efficiently participate in Giese-type addition reactions, affording products in good to excellent yields. The polymer-supported system enables straightforward purification by simple reprecipitation at both the borate complex f

Maternal co-exposure to polystyrene microplastics and DEHP impairs thyroid function in adult rat offspring.

Maternal combined exposure to polystyrene microplastics (PS-MPs) and di-(2-ethylhexyl) phthalate (DEHP) poses potential risks to offspring endocrine development; however, the combined effects of these two contaminants remain poorly characterized. Pregnant rats were exposed to PS-MPs, DEHP, or a combination of both throughout pregnancy and lactation. The hypothalamic-pituitary-thyroid (HPT) axis function and gut microbiota composition were then assessed in adult offspring. Single-contaminant exposure altered thyroid hormone levels and HPT axis gene expression. Co-exposure exacerbated these disr

Co-exposure to polystyrene microplastics and glyphosate induces gut microbiota dysbiosis and cognitive impairment in honeybees.

Microplastics, as emerging persistent environmental contaminants, can act as vectors for the absorption of other pollutants in ecosystems, such as pesticide residues. However, the combined toxicological effects of microplastics and pesticides on pollinators remain poorly understood. Here, we evaluated the toxicity of polystyrene microplastics (PS) and glyphosate (GLY), both individually and in combination, in honeybees (Apis mellifera). While no significant changes in body weight gain or food consumption were observed after 20 days of exposure, co-exposure to PS and GLY significantly increased

How temperature modulates the combined toxicity of aged polystyrene microplastics and copper in early zebrafish development.

Environmental factors, such as temperature, exert a critical influence on aquatic ecosystems. It is important to assess how climate change and pollutants affect contaminants released into water. In this study, the combined effects of environmentally relevant temperature variation, polystyrene microplastics (PSMPs), and copper were evaluated in zebrafish. Toxicity tests were conducted at 23, 26, and 29°C. The PSMPs were aged via the Fenton reaction, to simulate environmental oxidative conditions. Temperature influenced embryonic development, with hatching strongly delayed at 23 °C, while contam

Polystyrene microplastic uptake drives Inflammatory, Epitranscriptomic, and Metabolic Reprogramming in Human Aortic Endothelial cells.

Microplastics (MPLs) are pervasive environmental pollutants increasingly linked to adverse human health outcomes, including atherosclerosis. However, the underlying mechanisms remain poorly understood. Human aortic endothelial cells (HAECs), which line the inner surface of blood vessels, play a critical role in maintaining vascular homeostasis and in the development of atherosclerosis. This study demonstrates that polystyrene microplastics enter HAECs through clathrin-mediated endocytosis and macropinocytosis and subsequently co-localize with mitochondria and lysosomes. Exposure to MPLs induce

Polystyrene nanoplastics elicit early mitochondria-associated phenotypic, metabolic, and functional responses in human hepatocytes.

Nanoplastics have recently been detected in human liver tissue, raising concerns about their potential impact on liver function. However, early hepatocyte responses associated with nanoplastics exposure remain poorly understood. Here, we combined high-throughput Cell Painting-based phenomics, untargeted metabolomics, and Seahorse mitochondrial functional assay to investigate the effects of 100 nm polystyrene nanoplastics on human HepaRG hepatocytes, a surrogate for primary human hepatocytes. At the tested concentrations (6.25-100 µg/mL), exposure did not induce overt cytotoxicity, enabling ass

Simultaneous challenge with polystyrene nanoplastics and BDE-47 elicits cooperative cytotoxic impairment in human spermatozoa.

The progressive deterioration of human male reproductive capacity has paralleled the mounting global dispersal of persistent organic pollutants (POPs) and plastic-associated contaminants. Nevertheless, the collective consequences of simultaneous exposure to these ubiquitous environmental stressors on spermatozoal function remain inadequately characterized. This study undertook a comprehensive evaluation of the interactive reproductive harm posed by 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) and 25 nm polystyrene nanoplastics (PS-NPs) on human spermatozoa. Whereas isolated challenges with ecol

Polystyrene microplastics impair trophoblast invasion in vitro and alter microRNA expression targeting the angiogenesis pathway.

The presence of microplastics (MPs) in human blood and placenta provides evidence of human exposure to MPs and their accumulation in biological tissues. Adequate trophoblast invasion is essential for normal placentation. However, the effects of MPs exposure on microRNA (miRNA) expression regulating angiogenesis during early placental development on trophoblast function remain poorly understood. In this study, the interaction of polystyrene microplastics (PS-MPs) with the HTR8/SVneo trophoblast cell line was investigated, with a particular focus on miRNA, gene, and protein expression associated

Changes in dysbiosis and gene expression in the gut of wharf roach (Ligia spp.) fed with expanded polystyrene.

The East Asian region, with its intensive human and fishery activity, suffers from serious marine plastic pollution along its coastlines. Wharf roaches (Ligia spp.) on the coast of western Japan frequently ingest expanded polystyrene (EPS) and excrete it as microplastic in their feces, yet the effects on their gut remain unclear. Under controlled laboratory exposure, we examined both the host gut transcriptome, and the gut microbiome of wharf roaches fed EPS versus a fasted control group. DESeq2 and PyDESeq2 yielded a high-confidence set of 25 differentially expressed genes, among which xenobi

Polystyrene microplastic stress induces adaptive responses and functional remodeling of gastrointestinal microbiota in fattening lambs.

INTRODUCTION: This study aimed to investigate the effects of polystyrene microplastics (PS MPs) on microbial communities in the rumen, jejunum, ileum, cecum and colon of fattening Hu sheep lambs. METHODS: The 16S ribosomal RNA gene amplicon sequencing was employed to analyze the gastrointestinal microbial diversity and composition between the control (CON) and microplastic-exposed (MP, 100 μm PS MPs, 150 mg/d per lamb for 40 d) groups, each with 10 lambs. RESULTS: Alpha diversity analysis revealed that microplastics exposure slightly reduced the ruminal Shannon index (p = 0.087), but signifi
