Effects of some drugs on the responses of the rat isolated, innervated urinary bladder to indirect electrical stimulation.

1. The effects of some drugs known to inhibit transmission in the superior cervical ganglion and at the neuromuscular junction were investigated on the cholinergic nerve-smooth muscle junction, using the rat isolated innervated urinary bladder preparation.2. HC-3 and Win 4981 inhibited the indirectly evoked contractions; the block was typically slow in onset, depended on the rate of stimulation and was partially reversed by choline. The moderate coincident inhibition of acetylcholine-responses disappeared after washing the tissue, while the block of the neuronally evoked contractions persisted

Corticofugal influence on taste responses in the parabrachial pons of the rat.

Although the anatomy of centrifugal input to gustatory neural structures has been described, little is known of the physiological mechanisms that convey this influence or of their functional significance. As a first step in the investigation of these issues, the effect of a reversible lesion in the gustatory neocortex (GN) on the neural code for taste in the parabrachial nucleus of the pons (PbN) was studied in rats. Electrophysiological responses to taste stimuli bathed over the tongue were recorded from single units in the PbN before, after and following recovery from an infusion of procaine

Pharmacology of local anaesthetic agents.

The most important clinical properties of local anaesthetic agents are potency, onset, duration of action and relative blockade of sensory and motor fibres. These qualities are related primarily to the physicochemical properties of the various compounds. In general, lipid solubility determines the relative intrinsic potency of the various agents, while protein binding influences the duration of anaesthesia and pKa is correlated with the onset of action. In general, the local anaesthetics for infiltration, peripheral nerve blockade, and extradural anaesthesia can be classified into three groups

Further studies of porcine malignant hyperthermia.

A non-lethal procedure for identifying pigs apt to develop malignant hyperthermia is described. Susceptible animals were exposed to a variety of anaesthetic and other agents and it was shown that thiopentone sodium and CT 1341 (Glaxo) afforded a measure of protection against the development of the syndrome. Pretreatment with procaine did not prevent the onset of the condition and the administration of procaine when muscle rigidity was present failed to prevent a fatal outcome. The syndrome was induced in susceptible animals by halothane, chloroform, and a combination of halothane with suxameth

Local anaesthetic induced turbidity increases: implications of interactions with intact bacterial cells and with subcellular fractions.

Four local anaesthetics (LAs), amethocaine and procaine (esters) and cinchocaine and lignocaine (amides) have been assessed in terms of their ability to induce turbidity increases in non-growing cells of Escherichia coli suspended in a carbohydrate free defined medium (CFM) or distilled water or in 0.2 M phosphate buffer. Procaine and lignocaine did not induce turbidity increases. The turbidity increase-LA concentration profiles for both amethocaine and cinchocaine are similar in all three systems. Slight variation are discussed in terms of the ionic content of the suspending media. Interactio

The effect of local anaesthetics and antiarrhythmic agents on the responses of rabbit platelete to ADP and thrombin.

Local anaesthetics and antiarrhythmic agents produced changes in responses of rabbit platelets to ADP and thrombin that varied with the agent and its concentration. In high concentrations all local anaesthetics decreased aggregation rates. At lower concentrations several local anaesthetics increased aggregation rates. Depending on the agonist and local anaesthetic, increases were produced at all or only at low or high agonist concentrations. Local anaesthetics failed to influence shape change except at concentrations much greater than those which inhibited aggregation. Inhibition of aggregatio

Local anesthetic activity of 1-(morpholino or piperidino)-2-propanol derivatives.

The local anesthetic activities of 1-(morpholino or piperidino)-2-propanol derivatives were investigated. These activities were observed with nerve trunk anesthesia, rabbit's corneal anesthesia and intradermal injection anesthesia. Among these propanol derivatives, No. 24 and No. 25 which possess thioether-type sulfur were the most active and the effective potency was enhanced by increasing the dissociation constant (pKa) value in a series of the compounds. Effects on cat spinal reflex were also observed. These propanol derivatives revealed no selective depression of presynaptic depression or

Effect of papain-induced emphysema on permeability of rat lung to drugs.

To investigate the effect of a papain-induced emphysema-like condition on pulmonary absorption of drugs, rats were exposed to either papain aerosol or distilled water aerosol (control) intermittently for 2 wk, and rates of drug absorption from damaged and control lungs were compared. To measure absorption rates, 0.1 ml of drug solution (0.1-10 mM) was administered through a tracheal cannula to anesthetized animals, and after various times lungs were assayed for unabsorbed compound. In absorption experiments with the lipoid-insoluble compounds, mannitol, p-aminohippuric acid, and procaine amide

[The efficacy of local anaesthetics administered prior to castration of male suckling piglets].

Objective of this study was to examine the efficacy of various local anaesthetics (Procaine Hydrochloride, Procaine Hydrochloride + Epinephrin, Lidocaine Hydrochloride) used in castration of four to six day old male piglets. Animals of the control groups (I-IV) were treated analogous to the castration groups (V-IX) but not castrated. In groups I and V 15 minutes prior to castration/fixation the piglets were fixed as for injection but not medicated. The injection volume was standardised to 0.5 ml per testes. Local anaesthetics were applicated intratesticularly in five groups and intrascrotarly

Local anaesthetic blockade of neuronal nicotinic ACh receptor-channels in rat parasympathetic ganglion cells.

1 The effects of the local anaesthetics QX-222 and procaine on nicotinic acetylcholine (ACh)-evoked currents in cultured parasympathetic cardiac neurones of the rat were investigated by use of the whole-cell, perforated-patch, and outside-out recording configurations of the patch clamp method. 2 QX-222 and procaine, applied to the extracellular surface, reversibly inhibited the peak amplitude of the whole-cell nicotinic ACh-evoked current in a concentration-dependent manner, with half-maximal inhibitory concentrations (IC50) of 28 microM and 2.8 microM, respectively, at -80 mV. In these neuron

Preparation and evaluation of Eudragit gels. I: Eudragit organogels containing drugs as rectal sustained-release preparations.

New organogels containing salicylic acid, sodium salicylate, procaine, and ketoprofen were prepared by using Eudragit L and S, and propylene glycol. The physical properties (e.g., penetration and extensibility) of the organogels were affected considerably by the incorporation of a basic drug, such as procaine, rather than an acidic drug, such as salicylic acid. In in vivo evaluation using rabbits, it was found that 30% Eudragit L organogels containing 1.25% (w/w) salicylic acid or 2.5% (w/w) ketoprofen showed sustained plasma salicylic acid or ketoprofen levels and almost the same AUC as compa

Comparative studies of local anesthetic effects on artificial membranes and isolated cardiac tissues.

Drugs with local anesthetic properties like propranolol, tetracaine, lidocaine, procaine, induce concentration dependent potentials in artificial membranes. The size of the drug induced potential correlates with cardiodepressant activity, measured as decrease in force of contraction of guinea-pig atria and papillary muscles. Drugs inducing high potentials like propranolol and tetracaine, show low EC50 values in cardiac tissues. Propranolol in comparison to lidocaine diffuses more rapidly across the lipid membrane as derived from the time dependent decline in membrane potential and from radioac

Transmission in squid giant synapses: the importance of oxygen supply and the effects of drugs.

Synaptic transmission was studied in giant synapses of the stellate ganglion of the squid. When bathed in air-saturated sea water, the synapses deteriorate in 10 to 20 min.; if the sea water is saturated with 100 per cent oxygen, they function steadily for up to 12 hours. Optimal results probably require a medium with lower magnesium and higher calcium than the sea water used. Of eighteen compounds known to affect other synapses (Table I), none had stimulatory effects when applied to the preparation, but ten produced synaptic depression in concentrations of 10(-3) gm. per ml. or higher. The on

Local anesthetics inhibit muscarinic acetylcholine receptor-mediated calcium responses and the recruitment of β-arrestin in HSY human parotid cells.

OBJECTIVES: Local anesthetics act on G protein-coupled receptors (GPCRs); thus, their potential as allosteric modulators of GPCRs has attracted attention. Intracellular signaling via GPCRs involves both G-protein- and β-arrestin-mediated pathways. To determine the effects of local anesthetics on muscarinic acetylcholine receptors (mAChR), a family of GPCRs, we analyzed the effects of local anesthetics on mAChR-mediated Ca2+ responses and formation of receptor-β-arrestin complexes in the HSY human parotid cell line. METHODS: Ca2+ responses were monitored by fura-2 spectrofluorimetry. Ligand-in

A study of the mechanism by which some amphiphilic drugs affect human erythrocyte acetylcholinesterase activity.

The effects of the local anaesthetics procaine, tetracaine and lidocaine and of the antidepressant imipramine on human erythrocyte acetylcholinesterase were investigated. All four amphiphilic drugs inhibited enzymic activity, the IC50 (the concentration causing 50% inhibition) being about 0.40 mM for procaine, 0.05 mM for tetracaine, 0.70 mM for imipramine and 7.0 mM for lidocaine. Procaine and tetracaine inhibited acetylcholinesterase activity competitively at concentrations at which they did not perturb the physical state of the membrane lipid environment, as assessed by steady-state fluores

Effects of anticonvulsants on local anaesthetic-induced neurotoxicity in rats.

The effects of various anticonvulsants on local anaesthetics procaine- and lidocaine-induced convulsions were investigated in rats. Pretreatment with diazepam (2.5-5 mg/kg, intraperitoneally) and clonazepam (5-10 mg/kg, intraperitoneally) completely protected the rats against both local anaesthetic-induced convulsions. Phenobarbital (12.5-50 mg/kg, subcutaneously) also significantly decreased the incidence of both convulsions and prolonged their latencies. Carbabazepine (10-40 mg/kg, intraperitoneally) did not completely repress both convulsions, but it prolonged their latencies. Phenytoin (5-

The effects of local anaesthetics on the electrical and mechanical activity of the guinea-pig ureter.

The effects of local anaesthetic tertiary amines (procaine, lignocaine and tetracaine) as well as neutral (benzocaine) and permanently charged (QX-314) local anaesthetics were studied on the evoked electrical and mechanical activity of the ureter smooth muscle. 'Low' concentrations of procaine and lignocaine (0.1-1mM) at pH 7.4 increased the duration of the slow plateau component of the evoked action potentials. The amplitude of the phasic contraction was consequently increased within the first 5 min of exposure. Tetracaine 0.1 mM caused a transient increase in the duration of the plateau and
