BACKGROUND: Ankylosing spondylitis, a systemic illness is a foundation of progressing joint swelling that for the most part influences the spine. However, it frequently causes aggravation in different joints far from the spine, and in addition organs, for example, the eyes, heart, lungs, and kidneys. It's an immune system ailment that may be activated by specific sorts of bacterial or viral diseases that initiate an invulnerable reaction that don't close off after the contamination is recuperated. The particular reason for ankylosing spondylitis is obscure, yet hereditary qualities assume a hu

An important property of the nociceptive system is its plasticity, ie, the ability to change in an experience-dependent manner, which is implicated in the transition from acute pain to chronic pathological pain. Disease-induced plasticity can occur at both structural and functional levels and manifests as changes in individual molecules, synapses, cellular function, and network activity. In this short review, the author discusses how synaptic plasticity may mediate pathophysiological alterations linked to chronic pain by virtue of shifting the balance between excitation and inhibition, with a

Osteoarthritis (OA) is a progressive disorder with high incidence in the ageing human population that still has no treatment currently. This disorder induces the breakdown of articular cartilage, leading to the exposure and damage of bone surfaces. For a global understanding of OA development, the systematic integration of known OA-related proteins with protein-protein interaction (PPI) networks is required. In this work, the OA-related interactome was reconstructed using multiple data sources to have the most up-to-date information on OA-related proteins and their interactions. We then combin

Over the past decade, it has become apparent that many G-protein-coupled receptors (GPCRs) generate signals that control cellular differentiation and growth, including stimulation of Ras family GTPases and activation of mitogen-activated protein (MAP) kinase pathways. The mechanisms that GPCRs use to control the activity of MAP kinases vary between receptor and cell type but fall broadly into one of three categories: signals initiated by classical G protein effectors, e.g., protein kinase (PK)A and PKC, signals initiated by cross-talk between GPCRs and classical receptor tyrosine kinases, e.g.

Microneedles have gained increasing attention as an ideal device for transdermal drug delivery; however, their application to proteins has been a challenging task because of their inadequate tip-loading and unsatisfactory stability. Herein, we report the development of a microgel-integrated microneedle array patch (MI-MAP) capable of not only localizing protein cargoes in the tip portion but also stabilizing them via protein-phenolic interactions. Incorporation of protein-capturing microgels allows enrichment of protein cargoes within the tip region of microneedles, resulting in a significant

MAPS technology represents an innovative approach for the development of polysaccharide-based vaccines, relying on the affinity interaction between biotin and rhizavidin (rhavi), with potential for enhanced coverage through the incorporation of pathogen-specific proteins. Due to its flexibility, this platform is particularly attractive for the development of multivalent vaccines, as it is required for Klebsiella pneumoniae, a multidrug-resistant bacterium highlighted as prevalent cause of neonatal sepsis in low- and middle-income countries, against which no vaccines are currently available. BACKGROUND: Colorectal cancer (CRC) is a major cause of morbidity and mortality, with chronic inflammation from inflammatory bowel disease (IBD) representing a well-established risk factor. Clarifying shared molecular mechanisms may facilitate early detection and prevention strategies.
METHODS: Proteomic data from the UK Biobank were analysed using the Olink proximity extension assay for seven CRC-associated proteins (TFF3, TFF1, AHCY, RETN, LCN2, SELE and CEACAM5) previously identified via machine learning. Expression levels in CRC and IBD cases were compared with controls. Multilayer intera

Cuproptosis is a novel form of regulated cell death driven by intracellular copper accumulation, leading to lipoylated protein aggregation and Fe-S cluster destabilization. Dysregulation of this process has been implicated in various pathological conditions, including cancers, neurodegenerative diseases and metabolic diseases. Despite rapidly growing interest in cuproptosis, a systematically curated intelligent agent dedicated to cuproptosis-related genes (CRGs) and their disease associations remains lacking. To address this, we constructed a cuproptosis-related artificial intelligence (AI) kn

Cross-linking mass spectrometry is a powerful method for structural analysis, but choosing between cleavable and non-cleavable cross-linkers remains challenging. We rigorously compared non-cleavable DSS with cleavable DSSO and found that DSS consistently yields more cross-link identifications from isolated protein complexes to bacterial lysates. The advantage of DSS diminishes as sample complexity increases. At the highest complexity tested-human cell lysate-the trend reverses, with DSSO outperforming DSS. The superior performance of DSS in less complex samples is likely explained by its longe

Myostatin (MSTN), encoded by the MSTN gene, is a critical negative regulator of skeletal muscle mass. This study aims to identify and characterize the miRNAs involved in the development of the double-muscling phenotype in MSTN-deficient rabbits. We performed high-throughput sequencing to analyze the miRNA expression profiles in gluteus maximus tissue from wild type (MSTN+/+) and MSTN-KO (MSTN+/- and MSTN-/- inclusive) rabbits. Differentially expressed miRNAs (DEmiRNAs) were identified, and their potential target genes were predicted. Functional enrichment analysis of these target mRNAs was con

Psychiatric disorders are closely linked to immune dysregulation, yet the genetic relationships between peripheral immune organs, particularly the spleen, and different psychiatric disorders remain poorly understood. This study aimed to explore these associations. Linkage disequilibrium score regression (LDSC) was used to evaluate the genetic correlations between spleen volume and schizophrenia, bipolar disorder, and depression. For the two traits showing significant genetic correlation, MAGMA gene-level analysis was further performed to identify significant overlapping genes as shared genes.

It is evident that a robust immune system keeps Candida albicans infection in check, but weakened immunity opens the door for shifting from a benign yeast form to an invasive hyphal form which leads to systemic candidiasis with high mortality rate. However, the crucial players contributing to the increased susceptibility of immune-deficient individuals to Candida infection remain obscure. To uncover the molecular differences between these conditions, blood-associated proteins from the NDEx database and differentially expressed genes from GEO datasets of immunocompetent and immune-deficient ind

BACKGROUND: Necroptosis is inflammatorily sparked and closely associated with sepsis, but the crosstalk between necroptosis and inflammation in sepsis has rarely been studied in depth. This study is designed to reveal the role of necroptosis in the pathogenesis and development of sepsis by screening and validating hub necroptotic septic genes.
METHOD: We obtained datasets from the Gene Expression Omnibus (GEO) database. We screened differentially expressed genes (DEGs) and intersected them with necroptotic genes from the literature. Intersected genes were organized into one protein-protein in

The CANT1 gene, encoding a nucleotidase linked to calcium signaling, is crucial for spinal development, with its deficiency causing skeletal developmental issues. However, the relationship between CANT1 and scoliosis remains unclear. This study aims to explore the role of CANT1 in scoliosis from a genetic perspective and its potential as a therapeutic target. This study employed a multi-method approach, including Mendelian Randomization (MR) analysis, colocalization analysis, mediation analysis, enrichment analysis, PPI network construction, and molecular docking. Osteoarthritis (OA) is a common progressive joint disorder marked by synovial inflammation, cartilage degeneration, the formation of osteophytes, though its underlying molecular mechanisms remain unclear. This study integrated bioinformatics and experimental validation to identify key genes in OA synovium and their association with immune infiltration. Analysis of the GSE82107 dataset (10 OA, 7 controls) revealed 909 differentially expressed genes (525 upregulated, 384 downregulated). WGCNA identified the "midnightblue" module, and its intersection with DEGs yielded 122 genes enriched in cytok

BACKGROUND: Lupus nephritis (LN) is the most common and severe clinical manifestation of systemic lupus erythematosus (SLE) with considerable morbidity/mortality and limited treatment options. Since kidney biopsy is a relative hysteretic indicator, it is indispensable to investigate potential biomarkers for early diagnosis and predicting clinical outcomes of LN patients. Extracellular proteins may become the promising biomarkers by the secretion into body fluid. Our study linked extracellular proteins with lupus nephritis to identify the emerging biomarkers.

Bioluminescence resonance energy transfer (BRET) saturation is a method of studying protein-protein interaction (PPI) upon quantification of the dependence of the BRET signal on the acceptor/donor (A:D) expression ratio. In this study, using the very bright Nluc/YFP BRET pair acquired respectively with microplate reader and automated confocal microscopy, we significantly improved BRET saturation assay by extending A:D expression detection range and normalizing A:D expression with a new BRET-free probe. We next found that upon using variable instead of fixed amount of donor molecules co-express
