Diagnosis of Taenia solium infections based on "mail order" RNA-sequencing of single tapeworm egg isolates from stool samples.

Combined community health programs aiming at health education, preventive anti-parasitic chemotherapy, and vaccination of pigs have proven their potential to regionally reduce and even eliminate Taenia solium infections that are associated with a high risk of neurological disease through ingestion of T. solium eggs. Yet it remains challenging to target T. solium endemic regions precisely or to make exact diagnoses in individual patients. One major reason is that the widely available stool microscopy may identify Taenia ssp. eggs in stool samples as such, but fails to distinguish between invasi

An antigenic recombinant serine protease from Trichinella spiralis induces protective immunity in BALB/c mice.

In this study, we report the cloning and characterization of a cDNA encoding a Trichinella serine protease gene (TspSP-1.3) from GenBank. The recombinant TspSP-1.3 protein (rTspSP-1.3) was expressed in an Escherichia coli expression system and purified with Ni-affinity chromatography. Real-time quantitative PCR analysis revealed that TspSP-1.3 was expressed at significantly higher levels in muscle larvae and adult worms than in newborn larvae. TspSP-1.3 was detected in excretory-secretory proteins of Trichinella spiralis with western blotting. Immunization with the rTspSP-1.3 antigen induced h

Morphological and molecular characterisation of echinostomatid larvae (Digenea: Echinostomatidae) infecting Biomphalaria aff. straminea from Argentina.

Echinostomes are a globally distributed group of digeneans with complex life cycles. In South America, many species are known only from definitive hosts, and in Argentina, most larval stages and adult records remain unlinked. Formal morphological descriptions and molecular data are essential for identifying larval stages; however, molecular reference sequences of larval digeneans remain scarce. We present an integrated morphological and molecular characterisation of echinostome cercariae from Biomphalaria aff. straminea collected between January 2017 and February 2018 in a rice field from Corr

Comparative microRNAs profile of Schistosoma japonicum male worms derived from single-sex and bisexual infections: Implications of the multifunctional role of microRNA.

Schistosoma japonicum is a dioecious parasite that requires constant pairing between male and female worms for female maturation and egg production. MicroRNAs (miRNAs) play crucial roles in regulating various biological processes, including parasite development and host-pathogen interactions, but their functions in schistosomes remain largely unexplored. This study aimed to investigate the differentially expressed miRNAs (DEMs) between mated male (MM) and single-sex male (SM) worms to gain insights into their regulatory roles in schistosome reproduction. Total RNA was extracted from 28-day-old

A new species of Trichobilharzia (Digenea: Schistosomatidae) from Mareca sibilatrix (Aves: Anatidae), an endemic South American duck.

Avian schistosomatids are blood flukes parasitizing a wide spectrum of aquatic birds. However, its research in the Neotropics is ongoing with several putative new taxa pending description. Although waterfowl represent the most important avian hosts for these flukes, only a small proportion of these birds have been assessed for schistosomatids. This study aimed to describe avian schistosomatids from two native ducks from the Southern Cone of South America. A total of 24 Chiloe wigeon (Mareca sibilatrix) and three Cinnamon teals (Spatula cyanoptera) from different localities in Chile and Argenti

The Human Blood Fluke, Schistosoma mansoni, Harbors Bacteria Throughout the Parasite's Life Cycle.

While symbiotic relationships between invertebrates and bacteria have been extensively described, studies of microbial communities inhabiting parasitic worms remain scarce. Exploring the microbiota associated with helminths responsible for major infectious diseases will inform on parasite biology, host-pathogen interactions, and disease pathophysiology. We investigated the presence of microorganisms inhabiting tissues of the human parasite Schistosoma mansoni. In situ hybridization using a pan-bacterial 16S rRNA gene probe revealed bacteria colonizing key developmental stages that were success

Redescription of Proctophantastes gillissi (Overstreet et Pritchard, 1977) (Trematoda: Zoogonidae) with discussion on the systematic position of the genus Proctophantastes Odhner, 1911.

The redescription of Proctophantastes gillissi (Overstreet et Pritchard, 1977) (Trematoda: Zoogonidae) was made on specimens collected from Muraenolepis marmorata Günther, 1880 (Gadiformes) caught in the Ross Sea and the Amundsen Sea (Antarctic). The fish is a new host of this parasite. Phylogenetic relations of P. gillissi were inferred by Maximum Likelihood and Bayesian inference analysis of partial sequences from 28S rDNA. The findings from analysis of the molecular data are not consistent with the traditional point of view about the position of the genus Proctophantastes Odhner, 1911 in th

Malformations of the gill filaments of the ruffe Gymnocephalus cernuus (L.) (Pisces) caused by echinostomatid metacercariae.

In parasite surveys of fishes from Lake Balaton and its tributaries in Hungary, infections with metacercariae of a species of the digenean genus Echinochasmus (Trematoda: Echinostomatidae) were found in seven species of fish. In ruffe, Gymnocephalus cernuus, malformations of the gill filaments apparently caused by these infections were observed. These malformations were in the form of bifurcations of the filaments at about their mid-length. At the point where the filaments bifurcate, an Echinochasmus metacercaria was always embedded in the cartilaginous ray of the gill filament. All specimens

First insights into the ectoparasitic helminth fauna of freshwater fish on an Adriatic island.

Surface flow of freshwater on Adriatic islands is rare due to the extreme permeability of the karst terrain. Hence, most helminthological studies of freshwater fishes in the Adriatic drainage have focused on mainland freshwater systems, while data from islands are scarce. We collected minnow, Phoxinus lumaireul (Schinz, 1840), specimens in the Suha Ričina stream on Krk Island and screened them for helminth ectoparasites. Identification of metacercariae cysts (black spots) was carried out by sequencing part of the 28S rDNA gene, and a single monogenean worm was identified by sequencing part of

Multifunctional Roles of MicroRNAs in Schistosomiasis.

Schistosomiasis is a parasitic disease that is caused by helminths of the genus Schistosoma. The dioecious schistosomes mate and lay eggs after undergoing a complex life cycle. Schistosome eggs are mostly responsible for the transmission of schistosomiasis and chronic fibrotic disease induced by egg antigens is the main cause of the high mortality rate. Currently, chemotherapy with praziquantel (PZQ) is the only effective treatment against schistosomiasis, although the potential of drug resistance remains a concern. Hence, there is an urgent demand for new and effective strategies to combat sc

Cercarial dermatitis transmitted by exotic marine snail.

Cercarial dermatitis (swimmer's itch) is caused by the penetration of human skin by cercariae of schistosome parasites that develop in and are released from snail hosts. Cercarial dermatitis is frequently acquired in freshwater habitats, and less commonly in marine or estuarine waters. To investigate reports of a dermatitis outbreak in San Francisco Bay, California, we surveyed local snails for schistosome infections during 2005-2008. We found schistosomes only in Haminoea japonica, an Asian snail first reported in San Francisco Bay in 1999. Genetic markers place this schistosome within a larg

Meta-taxonomic analysis of prokaryotic and eukaryotic gut flora in stool samples from visceral leishmaniasis cases and endemic controls in Bihar State India.

Visceral leishmaniasis (VL) caused by Leishmania donovani remains of public health concern in rural India. Those at risk of VL are also at risk of other neglected tropical diseases (NTDs) including soil transmitted helminths. Intestinal helminths are potent regulators of host immune responses sometimes mediated through cross-talk with gut microbiota. We evaluate a meta-taxonomic approach to determine the composition of prokaryotic and eukaryotic gut microflora using amplicon-based sequencing of 16S ribosomal RNA (16S rRNA) and 18S rRNA gene regions. The most abundant bacterial taxa identified

RNA interference in vivo in Schistosoma japonicum: Establishing and optimization of RNAi mediated suppression of gene expression by long dsRNA in the intra-mammalian life stages of worms.

Schistosomes are parasitic platyhelminths that threaten over 600 million people globally. In recent years, RNA interference (RNAi) has been widely used as a molecular tool in research into the genomic function of parasites. We aim to develop effective protocols for application of RNAi technology in the intra-mammalian life stages of Schistosoma japonicum. In this work, the expression of the parasite gene encoding cathepsin B1 (SjCB1) was targeted by exposing the worms to 10 μg of long dsRNA dissolved in 0.1 ml of 0.7% NaCl injected into the tail vein of infected mice. This method was effective

Direct experimental manipulation of intestinal cells in Ascaris suum, with minor influences on the global transcriptome.

Ascaris suum provides a powerful model for studying parasitic nematodes, including individual tissues such as the intestine, an established target for anthelmintic treatments. Here, we add a valuable experimental component to our existing functional, proteomic, transcriptomic and phylogenomic studies of the Ascaris suum intestine, by developing a method to manipulate intestinal cell functions via direct delivery of experimental treatments (in this case, double-stranded (ds)RNA) to the apical intestinal membrane. We developed an intestinal perfusion method for direct, controlled delivery of dsR

TRICHINELLA MURRELLI POZIO AND LA ROSA, 2000 IN A GRAY FOX (UROCYON CINEREOARGENTEUS) FROM PENNSYLVANIA: A NEW HOST RECORD FOR THIS ZOONOTIC NEMATODE.

Trichinella murrelli Pozio and La Rosa, 2000, is the primary sylvatic trichinellid encountered in temperate North America. During a survey for Sarcocystis in wild canids, a single worm matching the morphology of encapsulated Trichinella was observed in a muscle tissue squash from a gray fox male originating from Pennsylvania. The worm was photographed and then separated from the host tissue by artificial digestion, and genomic DNA was extracted from the worm. This DNA was subjected to species-specific multiplex PCR and short-read genomic sequencing. The banding pattern of the multiplex PCR ind

Transcriptomes and pathways associated with infectivity, survival and immunogenicity in Brugia malayi L3.

BACKGROUND: Filarial nematode parasites cause serious diseases such as elephantiasis and river blindness in humans, and heartworm infections in dogs. Third stage filarial larvae (L3) are a critical stage in the life cycle of filarial parasites, because this is the stage that is transmitted by arthropod vectors to initiate infections in mammals. Improved understanding of molecular mechanisms associated with this transition may provide important leads for development of new therapies and vaccines to prevent filarial infections. This study explores changes in gene expression associated with the t

Molecular cloning of a member of the Fasciola hepatica saposin-like protein family.

A 436-bp complementary DNA (cDNA) was isolated from an adult Fasciola hepatica cDNA expression library by screening with the serum from a rabbit infected with F. hepatica for 4 wk. The deduced amino acid sequence encoded by this cDNA is an 11.5-kDa polypeptide that has significant homology to F. hepatica NK-lysin protein, to several members of saposin-like or NK-lysin protein families, as well as 3 amoebapore precursors of Entamoeba histolytica. The most striking feature observed within this protein, denoted FhSAP-2, is the presence of 6 conserved cysteine residues arranged within 5 amphipathi

Primary evaluation of immune responses in mice after a single mRNA immunization with Echinococcus granulosus Eg95 and Coenurus cerebralis TM16.

Echinococcus granulosus and Taenia multiceps cause zoonotic cystic diseases. Studies have shown that Eg95 from E. granulosus is highly conserved, exhibits excellent immunogenicity, and serves as a critical candidate antigen. In recent years, the TM16 recombinant protein antigen has been one of the most studied antigens for preventing C. cerebralis and demonstrates optimal experimental immunogenicity. This study developed both recombinant protein and mRNA-LNP vaccines targeting the Eg95 and TM16 antigens. Immunization of BALB/c mice showed that both vaccines significantly increased specific ant
