Adolescent intermittent ethanol exposure induces sex-specific and time-dependent changes in affective behaviors and metabolomic profiles.

Adolescent binge drinking in humans is associated with adverse outcomes, here we examined sex- and withdrawal-dependent changes in affective behaviors and metabolomic profiles in male and female mice following AIE exposure. Male and female C57BL/6 J mice were exposed to intermittent ethanol vapor inhalation from postnatal (PND) 28-42, a model of intermittent binge-like ethanol exposure during adolescence. Affective behavior was assessed using the open field test (OFT), light/dark test (LDT), and tail suspension test (TST) during short-term withdrawal (PND 49-53) and again during long-term with

Impact of glyphosate-based herbicide exposure through maternal milk on offspring's antioxidant status, neurodevelopment, and behavior.

Glyphosate-based Herbicide (GBH) is a widely used pesticide that functions as a broad-spectrum, non-selective herbicide. Despite advanced research to describe the neurotoxic potential of GBH, the harmful effects on maternal behavior and neurodevelopment of offspring remain unclear. This study was conducted to highlight the effects of GBH on the antioxidant system, anxiety traits, social interaction, and cognitive and sensorimotor functions in pups exposed to 25 or 50 mg/l daily via their mother's milk. Concerning the biochemical biomarkers, GBH administered during the early stages of developme

Gamma-aminobutyric acid type A receptor β3 subunit forebrain-specific knockout mice are resistant to the amnestic effect of isoflurane.

BACKGROUND: β3 containing γ-aminobutyric acid type A receptors (GABA(A)-Rs) mediate behavioral end points of IV anesthetics such as immobility and hypnosis. A knockout mouse with targeted forebrain deletion of the β3 subunit of the GABA(A)-R shows reduced sensitivity to the hypnotic effect of etomidate, as measured by the loss of righting reflex. The end points of amnesia and immobility produced by an inhaled anesthetic have yet to be evaluated in this conditional knockout. METHODS: We assessed forebrain selective β3 conditional knockout mice and their littermate controls for conditional fear

Isolation and characterization of a serine proteinase with thrombin-like activity from the venom of the snake Bothrops asper.

A serine proteinase with thrombin-like activity was isolated from the venom of the Central American pit viper Bothrops asper. Isolation was performed by a combination of affinity chromatography on aminobenzamidine-Sepharose and ion-exchange chromatography on DEAE-Sepharose. The enzyme accounts for approximately 0.13% of the venom dry weight and has a molecular mass of 32 kDa as determined by SDS-PAGE, and of 27 kDa as determined by MALDI-TOF mass spectrometry. Its partial amino acid sequence shows high identity with snake venom serine proteinases and a complete identity with a cDNA clone previ

Sex difference in psychological behavior changes induced by long-term social isolation in mice.

Social isolation can induce psychological behavior changes. It is interesting to know whether there is sex difference in responding to social isolation or not. The present study compared the behavior difference between male and female mice isolated for 1-4 months. The results showed that the isolated male mice had higher accounts of locomotor activity than the isolated female and group-housed ones. Both isolated male and female mice spent shorter time in the dark box than the group-housed mice in the light/dark test, and isolated male mice spent less time in the closed arms than isolated femal

Brain injury and neurofunctional deficit in neonatal mice with hypoxic-ischemic encephalopathy.

Birth asphyxia accounts for the majority of developmental motor and cognitive deficits. Studies were undertaken to develop a reproducible murine model of perinatal hypoxic-ischemic encephalopathy (HIE) which would permit both anatomic and neurofunctional quantification of injury. Short-term neurofunctional outcomes consisted of three developmental reflexes (righting, cliff aversion and geotaxis) assessed in 7-day-old mouse pups 24 h after unilateral carotid artery ligation followed by inhalation of 8% oxygen. Cerebral infarct volume was dependent on duration of hypoxia, being approximately 2.5

Reversible analgesia, atonia, and loss of consciousness on bilateral intracerebral microinjection of pentobarbital.

Concussion, asphyxia, and systemically administered general anesthetics all induce reversible depression of the organism's response to noxious stimuli as one of the elements of loss of consciousness. This is so even for barbiturate anesthetics, which have only modest analgesic efficacy at subanesthetic doses. Little is known about the neural circuits involved in this form of antinociception, although for anesthetic agents, at least, it is usually presumed that the drugs act in widely distributed regions of the nervous system. We now report the discovery of a focal zone in the brainstem mesopon

Development of an animal model of solvent abuse for use in evaluation of extreme trichloroethylene inhalation.

Self-intoxication by inhalation of vapors of trichloroethylene (TCE) and other solvents is widespread. In order to develop exposure protocols which typify episodes of TCE "sniffing", male Wistar-Munich rats were exposed to TCE vapor levels ranging from 9000 to 16 000 ppm. TCE in concentrations of 14 000 ppm and greater quickly produced loss of righting reflex. Recovery from the narcosis was very rapid. Central nervous system (CNS) depression was found to be cumulative in rats subjected for 5 h to alternating periods of 5 min of 15 000 ppm TCE and 15 min of fresh air. Ethanol markedly potentiat

Naloxone-like actions of MIF-1 do not require the presence of the pituitary.

The actions of peripherally administered MIF-1 (Pro-Leu-Gly-NH2) and naloxone in blocking the effects of morphine in the tail-flick test were measured across a wide range of five doses in hypophysectomized and intact mice. The presence of the pituitary gland failed to influence the response to MIF-1 or naloxone. Both hypophysectomized and intact mice were significantly affected by these two compounds at doses of 0.01, 0.1, 1.0, and 10.0 mg/kg IP. The greatest effect of MIF-1 occurred at 100 mg/kg, but naloxone was lethal at this dose. Preliminary experiments with other tests showed that at 10

A PROPOSED COMMON MECHANISM OF ACTION FOR GENERAL AND LOCAL ANAESTHETICS IN THE CENTRAL NERVOUS SYSTEM.

Procaine and several general anaesthetics block production of action potentials in frog skeletal muscle fibres by a single mechanism of action, which suggests that there might be a common basic mechanism of action on neurones in the central nervous system. Procaine or cinchocaine given alone to intact white mice produced "excitement" and convulsions but when given 60 min after phenobarbitone they caused central nervous depression. Large, convulsant doses of procaine or cinchocaine abolished the righting reflex in mice previously treated with small, subanaesthetic doses of phenobarbitone. In co

Sestrin2 induced by hypoxia inducible factor1 alpha protects the blood-brain barrier via inhibiting VEGF after severe hypoxic-ischemic injury in neonatal rats.

Источник пометил публикацию как отозванную

OBJECTIVE: Hypoxic ischemic (HI) encephalopathy remains the leading cause of perinatal brain injury resulting in long term disabilities. Stabilization of blood brain barrier (BBB) after HI is an important target, therefore, in this study we aim to determine the role of sestrin2, a stress inducible protein which is elevated after various insults, on BBB stabilization after moderate and severe HI injuries. METHODS: Rat pups underwent common carotid artery ligation followed by either 150min (severe model) or 100min (moderate model) of hypoxia. 1h post HI, rats were intranasally administered with

Nocifensive reflex thresholds in rats: measures of central nervous system effects of barbiturates.

PURPOSE: To characterize the pharmacodynamic relationships between plasma pentobarbitone and thiopentone concentrations and nocifensive reflexes during emergence from anaesthesia. METHODS: Forty-nine rats were studied. Plasma barbiturate concentrations were measured with high performance liquid chromatography. Nocifensive reflexes were assessed with the hindlimb withdrawal latency (WL) to heat and the somatic motor response threshold (SMRT) to tail pressure. In Protocol I, SMRT, WL, sedation, and the presence of paw-licking and the righting reflex were assessed in unrestrained rats before and

Design of New Potent, Selective, and Long-Acting NOP Receptor Agonists through Multiple Sequential d-Amino Acid Substitutions of [Arg14 Lys15]N/OFQ(1-15)-NH2.

The nociceptin/orphanin FQ (N/OFQ) receptor (NOP) ligands are drug candidates for different diseases, but peptide ligands are often limited by rapid enzymatic degradation and short in vivo duration of action. Here, we applied a multiple D-amino acid substitution strategy to the peptide template [Arg14 Lys15]N/OFQ(1-15)-NH2 to improve metabolic stability while preserving receptor activity. Progressive substitutions revealed marked positional tolerance within the C-terminal address domain and identified [d-Lys13,15d-Arg14]N/OFQ(1-15)-NH2 (compound 1c) as the most promising compound. This led to

Synthetic modulation of brain-wide anesthesia-activated neural ensembles using a transgenic mouse system.

Anesthetics are commonly used to produce unconsciousness, but the neural substrates of unconsciousness remain poorly understood. Here we test the hypothesis that general anesthesia works by targeting discrete cell types and neural circuits distributed brain-wide, rather than non-specific binding, using a mouse transgenic system. We take advantage of a mouse that expresses chemogenetic designer receptors exclusively activated by a designer drug (DREADDs) in an inducible Cre-dependent manner driven by Fos immediate early gene expression. Since Fos expression peaks with metabolic activity, we use

The activity of cholinergic neurons in the basal forebrain interferes with anesthesia-arousal process of propofol.

Previous research has demonstrated that basal forebrain (BF) regulates arousal during propofol anesthesia. However, as the BF comprises cholinergic neurons alongside two other types of neurons, the specific role of cholinergic neurons has not been definitively elucidated. In our study, calcium signal imaging was utilized to monitor the real-time activities of cholinergic neurons in the BF during propofol anesthesia. Additionally, we selectively stimulated these neurons to investigate EEG and behavioral responses during propofol anesthesia. Furthermore, we specifically lesioned cholinergic neur

Conceptualization and standardization of a non-invasive closed head injury model using directed shockwave to mice.

Traumatic brain injury (TBI) is a leading cause of death and disability worldwide, with closed head injury (CHI) being one of the most common forms of TBI. Preclinical modeling of TBI is challenging due to confounding factors like craniectomy and poorly controlled injury severity. This study proposes a non-invasive CHI model using directed shockwaves. The mice heads were exposed to the shockwave and accommodated together following the implantation of RFID tags for automated neurocognitive assessment. Following a 13-days paradigm, mice underwent a digital gait analysis and subsequent classical

Comparison of N-methyl-2-pyrrolidone (NMP) and the "date rape" drug GHB: behavioral toxicology in the mouse model.

N-methyl-2-pyrrolidone (NMP) and γ-hydroxybutyrate acid (GHB) are synthetic solvents detected in the recreational drug market. GHB has sedative/hypnotic properties and is used for criminal purposes to compromise reaction ability and commit drug-facilitated sexual assaults and other crimes. NMP is a strong solubilizing solvent that has been used alone or mixed with GHB in case of abuse and robberies. The aim of this experimental study is to compare the acute pharmaco-toxicological effects of NMP and GHB on neurological signs (myoclonia, convulsions), sensorimotor (visual, acoustic, and overall

KLF4 Exerts Sedative Effects in Pentobarbital-Treated Mice.

KLF4 is a zinc-finger transcription factor that plays an essential role in many biological processes, including neuroinflammation, neuron regeneration, cell proliferation, and apoptosis. Through effects on these processes, KLF4 has likely roles in Alzheimer's disease, Parkinson's disease, and traumatic brain injury. However, little is known about the role of KLF4 in more immediate behavioral processes that similarly depend upon broad changes in brain excitability, such as the sleep process. Here, behavioral approaches, western blot, and immunohistochemical experiments were used to explore the

Increased Alcohol Consumption in Mice Lacking Sodium Bicarbonate Transporter NBCn1.

The previous reports on an addiction vulnerability marker in the human SLC4A7 gene encoding the Na/HCO3 transporter NBCn1 suggest that this pH-regulating protein may affect alcohol-related behavior and response. Here, we examined alcohol consumption and sensitivity to the sedative effects of alcohol in male NBCn1 knockout mice. These mice displayed lower pH in neurons than wildtype controls, determined by intracellular pH in hippocampal neuronal cultures. Neurons from knockout mice had a higher action potential threshold and a more depolarized membrane potential, thus reducing membrane excitab

Investigation of sedative and hypnotic effects of Amygdalus communis L. extract: behavioral assessments and EEG studies on rat.

Amygdalus communis L. (almond) has been traditionally used as a natural medicine in the treatment of various diseases. The present research studied the sedative and hypnotic effects of the aqueous fraction of seeds of almond in rats. In order to investigate these effects, a combination of behavioral methods (open field test and loss of righting reflex test) as well as quantitative and analytic methods (EEG and EMG) were applied. The results of the open field test showed that a dose of 400 mg/kg of the almond extract significantly inhibited the locomotion activity of rats compared to normal. Th
