Oral self-administration of cocaine: chronic excessive intake by schedule induction.

Rats were exposed to daily 3-hr schedule-induced polydipsia sessions (fixed-time 1 min food-pellet delivery) with a cocaine hydrochloride solution as the available session fluid. Cocaine intake level (mg/kg) was a direct function of solution concentration (0.02-0.2 mg/ml). In a second experiment with 0.15 mg/ml cocaine solution available, mean daily session cocaine intake remained constant at about 40 mg/kg over the 5-week period of the experiment. Post-session serum samples of animals drinking either 0.15 or 0.2 mg/ml cocaine solution yielded serum cocaine values that were similar to those pr

Contrasting baseline-dependent effects of amphetamine, chlorpromazine and scopolamine on response switching in the pigeon.

Both amphetamine and scopolamine increase low rates and reduce high rates of responding. It has been suggested that the dependence of the effects of these drugs on control rate may be due to their non-specific disruptive cue properties rather than to specific pharmacological actions. To examine whether this possible non-specific disruption also applied to response-choice, pigeons were trained under a schedule in which 30 key-peck responses were required. This fixed-ratio could be completed by responding in any order on either or both of two keys, and then the first switch between the two keys

Behavioral determinants of drug action. The contributions of Peter B. Dews.

Peter B. Dews played a significant role in shaping the distinctive characteristics and defining the underlying principles of the discipline of behavioral pharmacology. His early and sophisticated use of schedules of reinforcement in the 1950s, incorporated from research in the experimental analysis of behavior and integrated into the discipline of pharmacology, provided tremendous insight, inspiration, and impetus to the newly emerging field of behavioral pharmacology. The experimental findings generated by Dews' research, blending the sophisticated use of behavior and pharmacological principl

Assessment of low level whole-body soman vapor exposure in rats.

We evaluated biochemical and behavioral effects of single, low-level exposures to the chemical warfare nerve agent soman (GD). Male Sprague-Dawley rats were trained on a variable-interval, 56-sec schedule of food reinforcement (VI56). The schedule specifies that a single lever press, following an average interval of 56 s, produces food reinforcement (i.e., a single food pellet). After training, rats received a single 60 min exposure to soman vapor at concentrations of 1.0-7.0 mg/m(3), or air control (n=8 for each treatment condition). Blood was sampled before and after the exposure. Following

Effects of cocaine, chlordiazepoxide, and chlorpromazine on responding of squirrel monkeys under second-order schedules of IM cocaine injection or food presentation.

Lever pressing by squirrel monkeys was maintained under second-order schedules of either food presentation or IM cocaine injection. Under one second-order schedule, every tenth response produced a brief (1-s) visual stimulus and the first brief stimulus presented after 30 min had elapsed was followed either by ten 300 mg food pellets or by a 3.0 mg IM injection of cocaine. Under another second-order schedule, the first response after 3 min produced the brief stimulus and the tenth brief stimulus was followed either by food or by cocaine. The two types of second-order schedules generated distin

N-acetylaspartylglutamate Inhibits Heroin Self-Administration and Heroin-Seeking Behaviors Induced by Cue or Priming in Rats.

Activation of presynaptic group II metabotropic glutamate receptors (mGluR2/3) inhibits drug reward and drug-seeking behavior, but the role of N-acetylaspartylglutamate (NAAG), an agonist of endogenous mGluR2/3, in heroin reward and heroin-seeking behavior remained unclear. Here, we aimed to explore the effects of exogenous NAAG on heroin self-administration and heroin-seeking behavior. First, rats were trained to self-administer heroin under a fixed ratio 1 (FR1) schedule for 10 days, then received NAAG (50 or 100 μg/10 μL in each nostril) in the absence or presence of LY341495 (1 mg/kg, i.p.

The neuropeptide-Y Y5 receptor antagonist L-152,804 decreases alcohol self-administration in inbred alcohol-preferring (iP) rats.

Neuropeptide-Y (NPY) is the most abundant and widely distributed peptide in the mammalian central nervous system and increases feeding behavior through actions at the Y5 receptor subtype. Recent pharmacological evidence indicates that NPY activity at this receptor subtype can modulate ethanol reinforcement. The purpose of this study was to determine if NPY Y5 receptor antagonism reduces ethanol self-administration and reinforcement in a rodent genetic animal model of alcoholism. Selectively inbred alcohol-preferring (iP) rats were trained to voluntarily consume ethanol (10% vol/vol) versus H2O

Specific antagonism of the behavioral effects of chlordiazepoxide and pentobarbital in the rat.

Stimulus control was established in rats with 5 mg/kg chlordiazepoxide and saline by employing a two-lever response choice task and an FR10 schedule of food reinforcement. Once trained, administration of chlordiazepoxide doses different from the training dose produced dose-responsive discrimination responding and the chlordiazepoxide discriminative cue was observed to transfer to pentobarbital. Pretreatment with the convulsant bemegride antagonized the pentobarbital transfer but not the chlordiazepoxide cue, whereas the specific benzodiazepine antagonist RO15-1788 decreased the chlordiazepoxid

Effects of psychoactive drugs on the conflict behavior under operant situation in Mongolian gerbils (Meriones unguiculatus).

Effects of diazepam, pentobarbital, morphine, ethanol, caffeine, and nicotine on the conflict behavior established under a MULT FR 20/FR 20 punishment schedule of food reinforcement were investigated in Mongolian gerbils. Fourteen out of 17 gerbils learned lever-press response for food within 15 sessions of training under FR 20 schedule. Although high voltage of electric shock (125-225 V) was required for attaining lever-press suppression, all 14 gerbils exhibited stable conflict behavior, i.e., showing high response rate of 20-30 responses/min in the safety period and low response rate of 0-3

Comparison of the intravenous reinforcing effects of propofol and methohexital in baboons.

Propofol is a widely used intravenous anesthetic that can directly activate and positively modulate the GABA(A)-receptor. Propofol is not currently regulated under the USA Controlled Substances Act. The present study evaluated the intravenous reinforcing effects of propofol compared to the intravenous barbiturate anesthetic methohexital in baboons using a procedure in which doses of the test drug were substituted for a standard cocaine dose. Drug or vehicle was available for self-injection during daily 5.5-h sessions under a fixed-ratio 120 or 160 schedule of reinforcement. A 40-min timeout af

Prenatally malnourished female but not male rats show increased sensitivity to MK-801 in a differential reinforcement of low rates task.

A reduced behavioral sensitivity to drugs acting on central GABAergic, serotonergic, opioid and cholinergic systems has previously been identified in predominantly male malnourished animals. The present study sought to compare the effects of the non-competitive N-methyl-D-aspartate (NMDA) receptor antagonist MK-801 on responding maintained by a differential reinforcement of low rates (DRL-18s) operant schedule in two groups of rats with different prenatal nutritional histories (well-nourished and protein restricted). The schedule required that the rats space their responses at least 18 s apart

Interreinforcement time, work time, and the postreinforcement pause.

Six rats were trained with food deliveries contingent upon their pressing a lever and holding it down for fixed, cumulative durations. Hold requirements were varied from 7.5 seconds to 120 seconds. Lever holding was maintained reliably at hold requirements as long as 30 seconds to 105 seconds for different rats. At longer hold requirements, lever holding was erratic and tended to occur only early in sessions. At shorter and intermediate requirements, the patterns of lever holding resembled those of responding under fixed-ratio schedules for discrete responses, with breaks in responding immedia

Management of chronic rumination with a contingent exercise procedure employing topographically dissimilar behavior.

Walking was employed as a functionally incompatible activity in a contingent exercise procedure to manage chronic rumination in a 10 year old mentally retarded male. The high frequency ruminative behavior was rapidly reduced once the topographically dissimilar activity was introduced as a contingency. On the first day the contingent exercise procedure was used in conjunction with a differential reinforcement of other behavior (DRO) 30-sec schedule. Thereafter the contingent exercise was used alone. The ruminative behavior appeared to be under the control of the contingency and remained at esse

Differential effects on natural reward processing in rats after repeated heroin.

RATIONALE: Heroin users report reward deficits as well as reward enhancements (to drug stimuli). To better understand the causal relation between chronic heroin and alterations in natural reward processing, we used experimental techniques in animal models. METHODS: Separate groups of rats were trained in several food reward paradigms: conditioned place preference (CPP), food-reinforced lever pressing under a progressive ratio schedule of reinforcement, free feeding, and lever pressing with conditioned reinforcement. After training, the rats were subjected to 10 daily heroin (2 mg/kg) or salin

Effects of voucher-based intervention on abstinence and retention in an outpatient treatment for cocaine addiction: a randomized controlled trial.

The aims of this study were to assess whether voucher magnitude improved cocaine abstinence and retention in an outpatient treatment for cocaine dependence, and to determine the effectiveness of a contingency management intervention in a European cultural context. A randomized controlled trial was conducted in which 96 participants who were randomly assigned to 1 of 3 treatment conditions in a community setting: standard outpatient treatment, community reinforcement approach (CRA) plus low monetary value vouchers (each point earned was equivalent to 0.125 Euro, US$ 0.18), and CRA plus high mon

Self-administration of cocaine-antihistamine combinations: super-additive reinforcing effects.

Histamine H1 receptor antagonists have some behavioral effects that predict abuse liability. In the present study, diphenhydramine and cocaine each maintained i.v. self-administration under a progressive-ratio schedule in rhesus monkeys. When cocaine and DPH were combined in a 1:1 ratio of the ED50s, the combination was super-additive in all monkeys. The data predict that the combination of cocaine and histamine H1 receptor antagonists would have enhanced potential for abuse relative to either drug alone.

A clozapine-like effect of cyproheptadine on progressive ratio schedule performance.

The atypical antipsychotic drug clozapine has multiple pharmacological actions, some of which, including 5-hydroxytryptamine (5-HT₂) and histamine (H₁) receptor antagonist effects, are shared by the non-selective 5-HT receptor antagonist cyproheptadine. Atypical antipsychotics have a characteristic profile of action on operant behaviour maintained by progressive ratio schedules, as revealed by Killeen's (1994) mathematical model of schedule controlled behaviour. These drugs increase the values of a parameter that expresses the 'incentive value' of the reinforcer (a) and a parameter that is inv

A schedule induction probe technique for evaluating abuse potential: comparison of ethanol, nicotine and caffeine, and caffeine-midazolam interaction.

Previous research has shown that under daily 3h sessions of food schedule-induced drug solution polydipsia (on a fixed-time 1min (FT 1min) schedule, session probes at greater FT values induced comparatively more ml/pellet drinking of solutions of drugs possessing abuse potential (ethanol, cocaine, midazolam) than of vehicle or other drug solutions. The present study extended these findings to nicotine and caffeine, drugs that usually do not function as reinforcers in i.v. self-administration studies with animals, but are known to have reinforcing effects for humans.
