Zembrin® Mitigates Reserpine-Induced Motor Dysfunction and Oxidative Stress in Parkinson's Disease: In Vivo and In Silico Analyses.

Parkinson's disease (PD), impacting millions worldwide, leads to motor deficits and various non-motor symptoms. Although there is no cure, treatment primarily involves dopamine replacement therapy, especially L-dopa for motor symptoms, and additional drugs are required to address non-motor effects. This underscores the increasing demand for dual-acting drugs that can effectively target both symptom types in PD. This study explored the potential effects of a standardised Mesembryanthemum tortuosum extract, Zembrin®, in treating PD, utilising in vivo and in silico models. Zebrafish larvae were s

Sex-specific attenuation of photoreceptor degeneration by reserpine in a rhodopsin P23H rat model of autosomal dominant retinitis pigmentosa.

Inherited retinal degenerations (IRDs) constitute a group of clinically and genetically diverse vision-impairing disorders. Retinitis pigmentosa (RP), the most common form of IRD, is characterized by gradual dysfunction and degeneration of rod photoreceptors, followed by the loss of cone photoreceptors. Recently, we identified reserpine as a lead molecule for maintaining rod survival in mouse and human retinal organoids as well as in the rd16 mouse, which phenocopy Leber congenital amaurosis caused by mutations in the cilia-centrosomal gene CEP290 (Chen et al., 2023). Here, we show the therape

Evaluation of the therapeutic effect of mesoporous silica nanoparticles loaded with Gallic acid on reserpine-induced depression in Wistar rats.

BACKGROUND: The current study evaluates the free gallic acid (GA) and GA-loaded mesoporous silica nanoparticles (MSNs) antidepressant efficacy in a rat model of depression caused by reserpine. METHODS: By using a scanning electron microscope (SEM), dynamic light scattering (DLS), and zeta potential, MSNs and GA-loaded MSNs were characterized. The efficiency of encapsulation and the release of GA-loaded MSNs were also investigated. The effect of GA, either in its free form or loaded on (MSNs) on oxidative stress biomarkers and monoamine neurotransmitters levels (serotonin (5-HT), norepinephrin

Inability of a reserpine-based screen to identify strains overexpressing efflux pump genes in clinical isolates of Staphylococcus aureus.

Overexpression of efflux pump genes conferring multidrug resistance (MDR) in Staphylococcus aureus results in reduced susceptibility to select biocides, dyes and fluoroquinolones. Reserpine is commonly used as an inhibitor of MDR efflux pumps and previous work from our laboratory using a reserpine-based screen to identify clinical isolates with an efflux phenotype revealed that nearly one-half overexpressed norA-B-C, mepA or mdeA. The accuracy of reserpine in predicting efflux pump gene overexpression in clinical strains was examined in detail. Bloodstream isolates of S. aureus previously clas

Effects of neonatally administered chlorpromazine and reserpine on the responsiveness of rat hepatic drug-metabolising enzymes to testosterone in adult life.

The effects of neonatally administered chlorpromazine and reserpine on the response of rat hepatic drug-metabolising enzymes to testosterone in adult life have been investigated using the chlorinated cyclodiene substrate DME. Neonatal treatment with chlorpromazine and reserpine had effects on the metabolism of DME similar to, but not as pronounced as, those of castration when adult. The effects of adult castration of male rats on hepatic microsomal metabolism of DME were fully reversed by treatment with testosterone propionate, with metabolism being restored to that of a control intact male. Antidepressant-like Effects of Orthosiphon stamineus: Integrated Evidence from In Silico, In Vitro, and a Reserpine-Induced Depression Model.

BACKGROUND: Traditional antidepressants are associated with undesirable side effects, limited efficacy, and delayed therapeutic responses. Natural products have emerged as a potential alternative to these drugs. This research aimed to investigate the antidepressant-like activity and possible mechanism of Orthosiphon stamineus (OS) leaf ethanolic extract. METHODS: Molecular docking and in vitro studies were performed to assess the inhibitory activity of ethanolic extracts of OS leaves against the monoamine oxidase (MAO-A) enzyme. Behavioral tests were used to evaluate the antidepressant-like a

Exploring the therapeutic potential of hesperidin nanoparticles, carbidopa-levodopa, or their combined administration in a rat model of reserpine-induced Parkinsonism.

In the pursuit of novel and effective therapeutic strategies, this study investigates the effects of hesperidin nanoparticles (HSP NPs), either alone or in combination with Carbidopa-levodopa (CLD), in a reserpine-induced Parkinson's disease (PD) rat model. Rats were randomly assigned to five experimental groups: control, PD model, PD model + HSP NPs, PD model + CLD, and PD model + HSP NPs + CLD. Behavior was assessed by the open field and grip strength tests. Monoamine levels, oxidative stress, enzyme activity, tumor necrosis factor-α (TNF-α), and brain-derived neurotrophic factor (BDNF) were

Unveiling the therapeutic potential of anthocyanins in reserpine-induced fibromyalgia: A multi-dimensional study on behavioral, molecular, and pathological changes.

Fibromyalgia (FM) is a chronic syndrome, associated with unidentified musculoskeletal pain, depression, neuropathic pain, memory impairment, and chronic fatigue. As a natural therapy with minimal risk, the multi-therapeutic benefits of Anthocyanins (ANs) are unfolded. This study evaluates efficacy of two doses of ANs, compared to a common treatment for FM; Neurontin (Neu), against reserpine (Res)-induced FM-associated biochemical, molecular and behavioral alterations in rats. Fibromyalgia is induced by Res (1 mg/kg, sc) for three consecutive days, followed by ANs (100, 200 mg/kg, po), and Neu

Aging accentuates decrease in tyrosine hydroxylase immunoreactivity associated with the increase in the motor impairment in a model of reserpine-induced parkinsonism.

Parkinson's disease (PD) is an age-related neurodegenerative disorder characterized by progressive dopaminergic neuron loss. Animal models have been used to develop a better understanding of the pathophysiologic mechanisms of PD. However, these models are usually conducted with young animals diverging of the age of PD patients, suggesting a bias in translational science. Thus, the aim of the study was to evaluate the effect of the age on rats in a progressive parkinsonism model induced by reserpine (RES). Adult (6 - 8 month-old) or elderly (18 - 24 month-old) male rats were assigned to six gro

Epalrestat improves motor symptoms by reducing oxidative stress and inflammation in the reserpine induced mouse model of Parkinson's disease.

BACKGROUND: Parkinson's disease (PD) is a progressive neurodegenerative disorder affecting a large number of elderly people worldwide. The current therapies for PD are symptom-based; they do not provide a cure but improve the quality of life. Muscular dysfunction is the hallmark clinical feature of PD and oxidative stress and inflammation play a critical role in its pathogenesis. Epalrestat is used for the treatment of diabetic neuropathy and is known to improve antioxidative defense mechanisms in the CNS. Therefore, in this study, we investigated the role of Epalrestat in the reserpine induce

Spontaneous pain-associated facial expression and efficacy of clinically used drugs in the reserpine-induced rat model of fibromyalgia.

Fibromyalgia-associated chronic pain occurring without organic causes exerts negative effects on patients' quality of life, thereby necessitating the development of superior drugs. Since non-organic pain in patients with fibromyalgia occurs without external stimuli, an endpoint measure that reflects patients' spontaneous pain should be implemented in preclinical research. The present study is the first to apply the rat grimace scale (RGS), a facial expression-dependent measure developed for quantifying spontaneous pain, to the rat with reserpine-induced myalgia, an animal model of fibromyalgia

Reserpine requires the D2-type receptor, dop-3, and the exoribonuclease, eri-1, to extend the lifespan in C. elegans.

Lifespan extension is an all systems encompassing event. Involvement of reduced insulin/IGF1 signalling is well worked out, first in the model organism Caenorhbaditis elegans followed by other systems including humans. But the role of neuronal component in lifespan extension is not well understood due to the refractory nature of neurons to small RNA interference (sRNAi) in C. elegans. Earlier, we have demonstrated that an antihypertensive drug, reserpine, extends lifespan through modulation of neurotransmitter release, especially, acetylcholine, in C. elegans. Intriguingly, the reserpine media

High concentrations of atrial natriuretic peptide and brain natriuretic peptide in rat pericardial fluid and their reduction by reserpine in vivo.

We determined concentrations and molecular sizes of natriuretic peptides in rat pericardial fluid and plasma by use of specific radioimmunoassays (RIA) and gel filtration HPLC. Our study shows that pericardial fluid forms a local extracellular storage of immunoreactive (ir) atrial natriuretic peptide (irANP) and brain natriuretic peptide (irBNP) near the heart where these peptides can be found in high concentrations in vivo. The concentrations of irANP, irBNP and NH2-terminal fragment of proANP (irNT-proANP) in pericardial fluid were 9.8 +/- 3.7, 0.49 +/- 0.47 and 28.9 +/- 11.8 nmol/l, respect

Studies on the gastroulcerogenic effects of pylorus ligation, hypothermic restraint stress, aspirin, indomethacin and reserpine in morphine dependent rats.

The gastric mucosal damage induced by pylorus ligation for 6 h, hypothermic restraint stress, aspirin, indomethacin and reserpine was studied in morphine dependent rats. Morphine tolerance and dependence were produced by administering the gradually increasing concentrations of morphine sulphate in drinking water for 21-24 days. The tolerance and dependence produced by morphine were confirmed by a decreased analgesic response to morphine in the hot plate test and by producing a naloxone precipitated withdrawal syndrome respectively. The intensity of gastric mucosal damage induced by pylorus lig

Therapeutic implications of hypertension-induced glomerular injury. Comparison of enalapril and a combination of hydralazine, reserpine, and hydrochlorothiazide in an experimental model.

Systemic hypertension does not always reflect concomitant glomerular hypertension. At similar levels of systemic hypertension, glomerular injury occurs only in kidneys that lack protective preglomerular vasoconstriction, which results in glomerular hypertension. indeed, glomerular hypertension and glomerular injury do not develop in rats with spontaneous hypertension that have effective preglomerular vasoconstriction. In the experiments reported herein, the normal adaptive response (afferent arteriolar dilation) to a reduction of one and five-sixths of the renal mass in rats with spontaneous h
