Disseminated Mucormycosis With Cerebral Involvement Owing to Rhizopus Microsporus in a Kidney Recipient Treated With Combined Liposomal Amphotericin B and Posaconazole Therapy.

Three months after a kidney transplant, a man experienced an internuclear ophthalmoplegia. Magnetic resonance imaging found a punctuate hyperintensity of the brainstem. Afterwards, the patient presented with peripheral facial paralysis. A complete morphologic assessment showed an increase of the brainstem lesion, together with an excavated pulmonary nodule. Combination therapy with high-dose liposomal amphotericin B and voriconazole was begun for the putative aspergillosis. Owing to its atypical clinical presentation and negative detection of Aspergillus galactomannan antigen on sera, a biopsy

Unravelling the antifungal mode of action of curcumin by potential inhibition of CYP51B: A computational study validated in vitro on mucormycosis agent, Rhizopus oryzae.

Like human, fungi too are known to share lot of structural similarities amongst their CYPs (Cytochrome P450 super family of enzymes) which allows antifungal 'azole' compounds to interact with CYPs of human. Clotrimazole, an 'azole' antifungal drug, is a known inhibitor of fungal CYP named CYP51B. Curcumin, a phytochemical obtained from Curcuma longa has the ability to interact with several different human CYPs to induce inhibition. The sequence and the structural similarities amongst both human and fungal CYPs suggest a strong possibility for curcumin to interact with fungal CYP51B to behave l

Germlings of Rhizopus arrhizus, rather than sporangiospores, lead to rhino-orbito-cerebral mucormycosis acquired intranasally in a murine model of uncontrolled diabetes.

Mucormycosis is a rapidly fatal, angioinvasive fungal infection. The disease presents as distinct clinical entities, with each being linked to specific risk factors and routes of transmission. Rhino-orbito-cerebral (ROC) manifestation remains the predominant form of mucormycosis in the developing world, especially in India amongst patients with uncontrolled diabetes. Limited literature is available on experimental animal models to study mucormycosis, and a murine model of ROC mucormycosis that recapitulates the risk factor (diabetes) and mode of acquisition (intranasal) of this disease is larg

A novel mannan-specific chimeric antigen receptor M-CAR redirects T cells to interact with Candida spp. hyphae and Rhizopus oryzae spores.

Invasive fungal infections (IFIs) are responsible for elevated rates of morbidity and mortality, causing around of 1.5 million deaths annually worldwide. One of the main causative agents of IFIs is Candida albicans, and non-albicans Candida species have emerged as a spreading global public health concernment. Furthermore, COVID-19 has contributed to a boost in the incidence of IFIs, such as mucormycosis, in which Rhizopus oryzae is the most prevalent causative agent. The effector host immune response against IFIs depends on the activity of T cells, which are susceptible to the regulatory effec

Gangrenous cutaneous mucormycosis caused by Rhizopus oryzae: a case report and review of primary cutaneous mucormycosis in China over Past 20 years.

Cutaneous mucormycosis is a rare opportunistic infection caused by zygomycetes that can be rapidly fatal if unrecognized. We describe the clinical, histopathological, fungal and molecular features of a case of gangrenous cutaneous mucormycosis. The patient presented with great necrosis on his right forearm at the site of detained intravenous cannula needle. He had type II diabetes and chronic renal insufficiency. KOH mount of black eschar showed many broad, aseptate fungal hyphae with right-angle branching. PAS staining of the tissue sample revealed similar broad hyphae in the dermis and cutis

Case Report: COVID-19-associated Rhinosinusitis Mucormycosis Caused by Rhizopus arrhizus: A Rare but Potentially Fatal Infection Occurring After Treatment with Corticosteroids.

COVID-19 first emerged in Wuhan, China, in December 2019. Since that time, the frequency of bacterial and fungal coinfections has been continuously increasing. Although invasive pulmonary aspergillosis is being increasingly recognized in association with COVID-19, there is limited information regarding COVID-19-associated mucormycosis. We describe a 50-year-old woman with uncontrolled diabetes who received systemic corticosteroids and remdesevir during her admission for COVID-19. A few days after discharge, the patient was readmitted because of facial swelling and numbness, and a diagnosis of

The expression of fungal CotH, human glucose-regulated protein 78 (GRP78), and predicted miRNAs in macrophages and diabetic mice infected with Rhizopus oryzae.

UNLABELLED: The spore coat protein homolog (CotH) has been identified among virulence factors of Mucorales that mediate fungal invasion by glucose-regulated protein 78 (GRP78) ligand on endothelial cells. The aim of this study was to examine the gene expressions of spore coat protein homolog 3 (CotH3) and GRP78, along with their target miRNAs, in infected human macrophages, mice models, and mucormycosis patients. To assess changes in the relative expressions of the GPR78 and CotH3 genes, this study used the real-time quantitative PCR method to quantify their target miRNAs in macrophages derive

Rhizopus-associated soft tissue infection in an immunocompetent air-conditioning technician after a road traffic accident: a case report and review of the literature.

Rhinocerebral or sinopulmonary mucromycosis is a well-recognized human fungal infection found among immunocompromised and diabetic patients. However, the infection is rare among immunocompetent hosts. We are reporting the case of an adult immunocompetent male patient working as an air-conditioning technician. The patient was a victim of a road traffic accident (RTA) and sustained multiple fractures in the proximal part of the left tibia, distal femur, and scapula. Two weeks postoperatively, Rhizopus microspores were isolated from an infected traumatic wound over the distal femur. Surgical debr

Generation of chemotactic factors by Rhizopus oryzae in the presence and absence of serum: relationship to hyphal damage mediated by human neutrophils and effects of hyperglycemia and ketoacidosis.

As our previous studies had shown that human neutrophils could kill Rhizopus oryzae hyphae in vitro, interactions of these hyphae with neutrophils and serum were further explored. Heated or fresh normal human sera suppressed hyphal metabolic activity as determined by [14C]uracil uptake, but severe ketoacidosis (8 X 10(-3) M beta-hydroxybutyric acid plus 2 X 10(-3) M acetoacetic acid at pH 7.0) negated this effect. Hyperglycemia (500 mg/dl) and severe ketoacidosis did not affect damage to hyphae by human neutrophils. Hyphae generated factors from sera which induced comparable chemotactic respon

Mechanistic elucidation of Rhizopus stolonifer-fermented orange peel in enhancing Saccharomyces cerevisiae growth and multi-stress tolerance: process optimization, metabolomics, and pathway analysis.

Saccharomyces cerevisiae, a pivotal organism in ethanol fermentation, frequently encounters challenges in high-stress industrial settings due to ethanol toxicity, acidic conditions, and thermal stress. This study aimed to optimize the fermentation of orange peel to enhance bioactive compounds and evaluate its impact on the stress tolerance of S. cerevisiae. The optimized conditions led to an increase in the total flavonoid content of the fermented orange peel (FOP). Supplementation with FOP significantly enhanced yeast tolerance; viability exceeded that of the control under 20% ethanol, improv

VOZ-dependent priming of salicylic acid-dependent defense against Rhizopus stolonifer by β-aminobutyric acid requires the TCP protein TCP2 in peach fruit.

Vascular plant one-zinc finger (VOZ) transcription factors (TFs) play crucial roles in plant immunity. Nevertheless, how VOZs modulate defense signaling in response to elicitor-induced resistance is not fully understood. Here, the defense elicitor β-aminobutyric acid (BABA) resulted in the visible suppression of Rhizopus rot disease of peach fruit caused by Rhizopus stolonifer. Defense priming by BABA was notably associated with increased levels of salicylic acid (SA) and SA-dependent gene expression. Data-independent acquisition proteomic analysis revealed that two VOZ proteins (PpVOZ1 and Pp

Size and Zeta Potential Clicked Germination Attenuation and Anti-Sporangiospores Activity of PEI-Functionalized Silver Nanoparticles against COVID-19 Associated Mucorales (Rhizopus arrhizus).

The SARS-CoV-2 infections in Indian people have been associated with a mucormycotic fungal infection caused by the filamentous fungi Rhizopus arrhizus. The sporangiospores of R. arrhizus are omnipresent in the environment and cause infection through inhalation or ingestion of contaminated air and foods. Therefore, the anti-sporangiospore activity of polyethyleneimine functionalized silver nanoparticles (PEI-f-Ag-NPs) with variable size and surface charge as a function of the molecular weight of PEI was explored. The results showed that both PEI-f-AgNP-1 and PEI-f-AgNP-2, potentially, attenuate

Development of a Monoclonal Antibody and a Serodiagnostic Lateral-Flow Device Specific to Rhizopus arrhizus (Syn. R. oryzae), the Principal Global Agent of Mucormycosis in Humans.

Mucormycosis is a highly aggressive angio-invasive disease of humans caused by fungi in the zygomycete order, Mucorales. Though a number of different species can cause mucormycosis, the principal agent of the disease worldwide is Rhizopus arrhizus, which accounts for the majority of rhino-orbital-cerebral, pulmonary, and disseminated infections in immunocompromised individuals. It is also the main cause of life-threatening infections in patients with poorly controlled diabetes mellitus, and in corticosteroid-treated patients with SARS-CoV-2 infection, where it causes the newly described diseas

Case report: Disseminated mucormycosis misdiagnosed as malignancy developed from allergic bronchopulmonary mycosis caused by Rhizopus microsporus following SARS-CoV-2 infection in a woman.

Mucormycosis has become more prevalent during the COVID-19 pandemic and is associated with a high mortality rate. However, concurrent host allergic reactions, invasive pulmonary mucormycosis, and disseminated mucormycosis are rarely reported. Herein, we describe a case of disseminated mucormycosis initially misdiagnosed as a malignancy that developed from allergic bronchopulmonary mycosis caused by Rhizopus microsporus in a woman with post-SARS-CoV-2 infection. The previously healthy patient presented with a sizeable mass in the right middle lobe and multiple lesions across the lungs, brain, s

Integrated omics-based analysis reveals the role of ferulic acid activating a co-production of 4-vinyl guaiacol and fumaric acid by Rhizopus oryzae.

The efficient valorization of lignocellulose-derived phenolics remains a major challenge in sustainable biomanufacturing. Here, an integrated omics approach was employed to elucidate how ferulic acid (FA) activates the co-production of 4-vinyl guaiacol (4-VG) and fumaric acid by Rhizopus oryzae. Co-supplementation of FA with glucose significantly enhanced metabolic performance, increasing fumaric acid yield by 16.9% and inducing simultaneous 4-VG synthesis. Metabolomic profiling revealed that FA reprograms cellular metabolism by reinforcing the tricarboxylic acid cycle and redox pathways while

Cutaneous mucormycosis in a critically ill nonagenarian patient caused by Rhizopus species treated with intravenous isavuconazole and topical amphotericin B along with wound debridement.

This case report highlights cutaneous mucormycosis in a 99-year-old patient who was effectively treated with intravenous isavuconazole and local amphotericin B after wound debridement. The patient showed significant improvement within two weeks, highlighting the importance of diagnosis and treatment for this rare fungal infection. It underscores the need for early recognition and suggests that combining antifungal treatments with surgical intervention may be an optimal strategy, even in patients lacking typical risk factors.
