Toxin-induced conditioned disgust responses: Comparing emetine and scopolamine methyl nitrate (SMN) to lithium chloride (LiCl).

Chemotherapy patients often develop anticipatory nausea, a learned response produced by an environmental cue triggering nausea in the absence of the emetic stimulus. This can be modelled in rats by pairing the administration of an emetic treatment with exposure to a distinct context. Rats are a non-emetic species, but they are still capable of experiencing nausea, which is observable through the presence of the gaping response. The rat model of anticipatory nausea mainly utilizes lithium chloride (LiCl) to evoke the gaping response, yet various other putative emetic toxins and agents have rema

Green tea extract containing enhanced levels of epimerized catechins attenuates scopolamine-induced memory impairment in mice.

ETHNOPHARMACOLOGICAL RELEVANCE: Green tea has been used as a traditional medicine to control brain function and digestion. Recent works suggest that drinking green tea could prevent cognitive function impairment. During tea manufacturing processes, such as brewing and sterilization, green tea catechins are epimerized. However, the effects of heat-epimerized catechins on cognitive function are still unknown. To take this advantage, we developed a new green tea extract, high temperature processed-green tea extract (HTP-GTE), which has a similar catechin composition to green tea beverages. AIM O

Scopolamine impairs acquisition and facilitates consolidation of fear conditioning: differential effects for tone vs context conditioning.

Cholinergic antagonism impacts selected learning tasks. To understand where scopolamine exerts its action, learning tasks differentially sensitive to hippocampus and amygdala lesions were used. Hippocampal lesions prevent context fear conditioning without effect on tone conditioning. These lesions also produce a time-dependent retrograde deficit in context conditioning. The amygdala is necessary for both tone and context conditioning. To examine the possibility that cholinergic antagonism mimics hippocampal damage or amygdala damage, rats were given scopolamine (1 mg/kg) either before or after

Transdermally administered scopolamine vs. dimenhydrinate. I. Effect on nausea and vertigo in experimentally induced motion sickness.

The effect of transdermally administered scopolamine (TTS-scopolamine) (2.5 cm2 surface area, one and two patches) and dimenhydrinate (100 mg) on experimental motion sickness was examined in 16 healthy volunteers in a randomized double-blind study. Nausea was induced by Coriolis manoeuvre and vertigo by calorization of the ear. In all subjects, scopolamine was found in urine in concentrations indicating adequate absorption of the drug. One TTS-scopolamine, two TTS-scopolamine and dimenhydrinate caused a statistically significant reduction in nausea when compared with placebo. Dimenhydrinate wa

Aging Reduces Basal Cholinergic Reserve, but the Pattern of Scopolamine-Induced Memory Impairment Remains Comparable in Rats.

BACKGROUND: Experimental models of cognitive dysfunction are essential for investigating mechanisms of memory impairment and therapeutic strategies. The scopolamine-induced memory impairment (SIMI) model is widely used for inducing acute cholinergic dysfunction and related cognitive deficits. However, using young animals to study a disorder that predominantly affects older individuals can lead to misleading results. This study investigated whether age alters the behavioral and neurochemical outcomes of the acute SIMI model. METHODS: Young and aged (5 and 22 months old) male Wistar rats receiv

Targeted transdermal scopolamine delivery over the salivary glands for sialorrhea: Dose-response and systemic safety in rats.

Sialorrhea is a common and distressing symptom in patients with neurological disorders, often impairing quality of life and increasing the risk of aspiration. Although oral anticholinergic agents are widely prescribed, their systemic side effects frequently limit long-term adherence. This study investigated the dose-response relationship and systemic safety of transdermal scopolamine applied directly over the salivary glands in a rat model. Formulations containing 0.2%, 0.5%, 1%, and 5% scopolamine (0.05 g per rat) were compared with oral administration at a dose of 0.2 mg/kg. Transdermal appl

Chemical characterization of anti-amyloidogenic phytochemicals from Aconogonon alpinum: Attenuation of cognitive impairment in scopolamine-induced mice.

Neurodegenerative diseases are characterized by abnormal protein aggregation and oxidative stress, which contribute to neuronal damage and cognitive decline. Aconogonon alpinum, known as Shinga in Korea, is a perennial Polygonaceae plant traditionally used in folk medicine for its anti-inflammatory properties, yet its neurobiological potential remains poorly characterized. In this study, we investigated the amyloid-related processes and cognitive-enhancing effects of A. alpinum extract through bioactivity-guided isolation and pharmacological evaluation. In APPswe-transfected HeLa cells, A. alp

Piracetam reverses scopolamine-induced memory disorder in mice: an animal model using behavioral, oxidative, and cholinesterase biomarkers.

Memory impairment diseases have become a serious health problem worldwide. In this context, an animal model capable of recognizing substances with the ability to recover memory disorders would be welcome. Thus, the present work aims to evaluate the effect of piracetam on reversal of scopolamine-induced memory deficit in male Swiss mice (n = 6) after fifteen days of treatment. To achieve this objective, behavioral, oxidative and cholinesterase inhibition markers were used. Memory impairment was evaluated by object and social recognition and step-down inhibitory avoidance tests. The evaluation o

Fatal Datura poisoning: identification of atropine and scopolamine by high performance liquid chromatography/photodiode array/mass spectrometry.

A forensic method comprising solid phase extraction and HPLC analysis was developed for the detection and confirmation of atropine and scopolamine, the main toxic alkaloids of Datura stramonium and Datura ferox. This method allowed the direct coupling of an electrospray (ZMD) mass selective detector to the HPLC system. Under these conditions, atropine and scopolamine were well separated from other components and detected on the PDA (LOD = 1 microg/ml) and ZMD (LOD(atropine) = 10 pg/ml; LOD(scopolamine) = 100 pg/ml) detectors. Four geographically isolated populations of each of D. stramonium an
