A comparative analysis of MSCs and their exosomes in osteoarthritis therapy: complementary roles and clinical translation.

OBJECTIVE: There is presently no cure for osteoarthritis (OA). Despite the promise of mesenchymal stem cell (MSC) transplantation, issues such as low cell survival and engraftment have shifted research toward MSC-derived exosomes (MSC-EXOs). Because they mirror the immunomodulatory and tissue-regenerative actions of MSCs without the safety and logistical issues of cell-based therapies, MSC-EXOs constitute an emerging class of medicines. This paper compares and contrasts MSCs and MSC-EXOs for OA treatment. MATERIALS AND METHODS: A literature search was conducted in PubMed, Google Scholar, Web

Advances in Stem Cell Therapy for Brain Diseases via the Intranasal Route.

Stem cell therapy efficiently targets the brain for most neurological disorders like migraine, Parkinsonism, schizophrenia, Alzheimer's disease, and brain injury. The major obstacles in potential brain targeting are the physiological barriers like blood-brain-barrier and bloodcerebrospinal fluid barrier. The conventional injectable route or direct transplantation of stem cells results in brain injury and is less feasible clinically. The alternative to these invasive routes is the intranasal route that is non-invasive, easily repeatable technique and highly effective in brain targeting without

Stem cells in urology.

The shortage of donors for organ transplantation has stimulated research on stem cells as a potential resource for cell-based therapy in all human tissues. Stem cells have been used for regenerative medicine applications in many organ systems, including the genitourinary system. The potential applications for stem cell therapy have, however, been restricted by the ethical issues associated with embryonic stem cell research. Instead, scientists have explored other cell sources, including progenitor and stem cells derived from adult tissues and stem cells derived from the amniotic fluid and plac

Stem cell gene therapy for chronic renal failure.

Recently, stem cell research has attracted considerable attention because it could be used for the regeneration of damaged organs that are untreatable by conventional techniques, and several stem cells (or progenitor cells), such as endothelial stem cells and neural stem cells have been discovered. Following the progression of this field of research, the potential for stem cell gene therapy has increased and several therapeutic benefits have already been reported. Although this approach was originally investigated for fatal or hereditary diseases, chronic renal failure is also a candidate for

[Increasing clinical applications of stem cells from umbilical cord blood and consequences for the handling of this biomaterial].

Umbilical cord blood (UCB) of a newborn contains stem cells with a remarkably high differentiation and regeneration potential. They are therefore useful for application in regenerative medicine. In this review current clinical applications are summarised and the necessity for the storage of UCB stem cells is derived and discussed. A Medline search for publications regarding clinical application of UCB stem cells was carried out and other data bases were reviewed. The transplantation of UCB stem cells, a special class of adult stem cells, has not only been established successfully in a variety

Hematopoietic stem cell gene therapy: progress toward therapeutic targets.

The concept of hematopoietic stem cell gene therapy is as exciting as that of stem cell transplantation itself. The past 20 years of research have led to improved techniques for transferring and expressing genes in hematopoietic stem cells and preclinical models now routinely indicate the ease with which new genes can be expressed in repopulating stem cells of multiple species. Both modified murine oncoretroviruses and lentiviruses transmit genes into the genome of hematopoietic stem cells and allow expression in the host following transplantation. Using oncoretroviruses, therapeutic genes for

Combining Growth Factor and Stem Cell Therapy for Stroke Rehabilitation, A Review.

Stroke is a serious, life-threatening condition demanding vigorous search for new therapies. Recent research has focused on stem cell-based therapies as a viable choice following ischemic stroke, based on studies displaying that stem cells transplanted to the brain not only survive but also cause functional recovery. Growth factors defined as polypeptides that regulate the growth and differentiation of many cell types. Many studies have demonstrated that combined use of growth factors may increase results by the stimulation of endogenous neurogenesis, anti-inflammatory, neuroprotection propert

A Comprehensive Review of the Role of Stem Cells in Neuroregeneration: Potential Therapies for Neurological Disorders.

Stem cell research has emerged as a groundbreaking field with significant potential for advancing neuroregeneration and neurological disorder treatment. Neurological conditions such as Alzheimer's disease, Parkinson's disease, stroke, and spinal cord injuries pose severe challenges due to their impact on quality of life and the limited efficacy of current treatments, which primarily focus on symptom management rather than addressing the underlying damage. Neuroregeneration, the process of repairing and restoring damaged neural tissues, is crucial for improving patient outcomes, given the centr

Application of Bone Marrow Stem Cell Based Therapy in Bone Loss Diseases.

Hematopoietic bone marrow stem cell transplantation has been regularly used in clinical practice for over 40 years. Further research indicates that bone marrow stromal cells are also stem cells that are capable of self-renewal, proliferation and differentiation. With appropriate induction, the bone marrow stromal cells can differentiate into bone, cartilage, tendon, adipose tissue and fibrous tissue, which would potentially be applied for treating bone loss diseases. This review outlines research in understanding role of bone marrow stem cells and stem cells from other origins in bone repair,

Utilization of Modified Induced Pluripotent Stem Cells as the Advance Therapy of Glaucoma: A Systematic Review.

Glaucoma is an optic neuropathy disease that causes cupping of the optic disc and decreased visual field. Glaucoma is still the second leading cause of blindness globally, with a worldwide prevalence of more than 76 million people in 2020. However, no therapy can cure glaucoma completely, especially when optic nerve damage has occurred. Available treatments only play a role in keeping the intraocular pressure stable This research aims to determine the potential use of modified stem cell therapy to treat intraocular damage in glaucoma cases. Literature research was conducted by involving seven

Stem cell-based therapeutic strategies for down syndrome and Alzheimer's disease.

BACKGROUND: Down syndrome (DS) and Alzheimer's disease (AD) are two distinct yet interconnected neurological conditions that share overlapping pathological features, including amyloid-beta plaque accumulation, neuroinflammation, and progressive neurodegeneration. Individuals with DS are at increased risk of developing AD-like dementia owing to the overexpression of the amyloid precursor protein-encoding gene on chromosome 21. Despite significant research efforts, effective disease-modifying treatments remain unavailable for both conditions, necessitating the exploration of novel therapeutic ap

Clinical Trials of Stem Cell Therapy in Japan: The Decade of Progress under the National Program.

Stem cell therapy is a current world-wide topic in medical science. Various therapies have been approved based on their effectiveness and put into practical use. In Japan, research and development-related stem cell therapy, generally referred to as regenerative medicine, has been led by the government. The national scheme started in 2002, and support for the transition to clinical trials has been accelerating since 2011. Of the initial 18 projects that were accepted in the budget for preclinical research, 15 projects have begun clinical trials so far. These include the transplantation of retin

Homing and Migration of Umbilical Cord Mesenchymal Stromal Cells in Clinical Applications: Molecular Mechanisms, Translational Barriers, and Therapeutic Optimization.

Umbilical cord-derived mesenchymal stromal cells (UC-MSCs) offer distinct advantages for clinical translation, including accessibility, scalability, and broad immunomodulatory capacity. However, the efficacy of systemically delivered UC-MSCs is constrained by suboptimal in vivo trafficking. Intravascular administration is limited by pulmonary first-pass sequestration, inefficient endothelial recruitment, blood-mediated inflammatory injury, and poor retention. While UC-MSCs homing is often conceptualized through a leukocyte adhesion paradigm, this model incompletely describes culture-expanded U

Role of Stem-Cell Transplantation in Leukemia Treatment.

Stem cells (SCs) play a major role in advanced fields of regenerative medicine and other research areas. They are involved in the regeneration of damaged tissue or cells, due to their self-renewal characteristics. Tissue or cells can be damaged through a variety of diseases, including hematologic and nonhematologic malignancies. In regard to this, stem-cell transplantation is a cellular therapeutic approach to restore those impaired cells, tissue, or organs. SCs have a therapeutic potential in the application of stem-cell transplantation. Research has been focused mainly on the application of

Clonal Evolution of Stem Cells in the Gastrointestinal Tract.

The field of gastrointestinal epithelial stem cells is a rapidly developing area of adult stem cell research. The discovery of Lgr5(+) intestinal stem cells has enabled us to study many hidden aspects of the biology of gastrointestinal adult stem cells. Marked by Lgr5 and Troy, several novel endodermal stem cells have been identified in the gastrointestinal tract. A precise working model of stem cell propagation, dynamics, and plasticity has been revealed by a genetic labeling method, termed lineage tracing. This chapter introduces the reidentification of crypt base columnar cells as Lgr5(+) s

Stem cell induced inflammatory hypertrophy of the cauda equina.

Stem cell therapy can present clinicians with challenging clinical scenarios, as access to such treatments outpaces the research into their efficacy and safety due to the burgeoning trend of international travel to acquire stem cell therapy, or "stem cell tourism." Treatment of neurologic conditions remains an enticing potential application of stem cell therapy, often administered intrathecally. In response to such therapy, multiple adverse events have been described in the literature, including neoplasms, demyelinating disease, and seizures, among others. We present a case of symptomatic infl

Human dignity and human tissue: a meaningful ethical relationship?

Human dignity has long been used as a foundational principle in policy documents and ethical guidelines intended to govern various forms of biomedical research. Despite the vast amount of literature concerning human dignity and embryonic tissues, the majority of biomedical research uses non-embryonic human tissue. Therefore, this contribution addresses a notable lacuna in the literature: the relationship, if any, between human dignity and human tissue. This paper first elaborates a multidimensional understanding of human dignity that overcomes many of the shortcomings associated with the use o

Translational insights into stem cell preconditioning: From molecular mechanisms to preclinical applications.

Cell-based therapy (CBT) is a revolutionary approach for curing a variety of degenerative diseases. Stem cell-based regenerative medicine is a novel strategy for treating tissue damages regarding stem cells unique properties such as differentiation potential, paracrine impacts, and self-renewal ability. However, the current cell-based treatments encounter considerable challenges to be translated into clinical practice, including low cell survival, migration, and differentiation rate of transplanted stem cells. The poor stem cell therapy outcomes mainly originate from the unfavorable condition
