UNLABELLED: Streptococcus mutans (S. mutans) has a superior ability to rapidly metabolize sucrose into exopolysaccharides (EPS, mainly glucans), which serve as a critical virulence factor related to dental caries. Despite extensive research on sucrose-dependent EPS at the molecular and macroscale levels, however, the mechanisms underlying EPS effects at the microscale level remain poorly understood. Here, by employing bacteria tracking and fluorescence staining techniques, we investigated the role of sucrose-dependent EPS during biofilm development of S. mutans at the microscale level for both

Dental caries remains a major oral health problem, necessitating effective natural antimicrobial alternatives targeting Streptococcus Mutans. Chewing sticks from Mangifera indica and pongamia pinnata contain bioactive compounds with established antiseptic and bactericidal properties. Aqueous extracts (5%, 10% and 50% w/v) were prepared and tested against Streptococcus mutans using agar diffusion to assess zones of inhibition. Mangifera indica showed maximum antimicrobial efficacy at 50% concentration, followed by lower but notable effects at 10% and 5%. Thus, we show Mangifera indica as a pote

BACKGROUND: Dental caries, primarily driven by Streptococcus mutans (S. mutans) biofilms, remains a formidable clinical challenge due to the protective extracellular polymeric substance (EPS) matrix and the limited efficacy of conventional antibiotics.
METHODS: To address this, we report a biomimetic photothermal nanoplatform (FWA NPs) featuring a hierarchical, urchin-like multi-spiked architecture, synthesized via the co-assembly of ferrocene-tryptophan conjugates and in situ biomineralized gold nanoparticles.
RESULTS: Benefiting from this unique topological structure, FWA NPs maximize inte

Streptococcus mutans, is considered the main microbial etiological agent of dental caries, therefore it has been proposed as a useful predictor of caries risk as well as a target for caries prevention strategies. We aimed to compare the genomic characteristics of S. mutans strains isolated from individuals with high and low caries risk, in order to determine their genotypic features related to dental caries in adults. A total of 25 S. mutans isolates, obtained from the saliva of 13 volunteers with high dental caries activity and 12 caries-free individuals, were analysed using whole-genome sequ

Streptococcus mutans, a Gram-positive pathogen, is a primary causative agent of dental caries. It modifies the oral biofilm architecture on tooth enamel and, like other bacteria, requires transition metal ions such as Zn(II), Cu(II), and Ni(II) for survival and virulence. Physiological salivary Zn(II) levels are insufficient for optimal bacterial growth, prompting S. mutans to develop a specialized ABC transport system comprising AdcA, AdcB, and AdcC. Among these, the lipoprotein AdcA plays a pivotal role in Zn(II) acquisition. In this study, we examined two probable Zn(II)-binding sites in Ad

The purpose of this study was to evaluate the gene expression pattern of the caries-inhibiting effect of Cudrania tricuspidata (C. tricuspidata) extract on cariogenic bacteria Streptococcus mutans (S. mutans). We examined bacterial growth, tooth surface attachment, biofilm formation, acid production, free calcium release, and toxicity gene expression. The major components of the extract were investigated by UPLC-Q-TOF-MS analysis. Exposure to C. tricuspidata inhibited bacterial growth and attachment at concentrations of ≥15 μg/mL. Inhibition effects on biofilm formation, acid production, and f

Staphylococcus aureus, an opportunistic bacterium found in the oral cavity, has been reported as a causative agent of infective endocarditis and pneumonia. Salt is an essential mineral for cell maintenance in the human body. This study was conducted to clarify how salt affects the formation of biofilms by S. aureus and Streptococcus mutans, pathogens implicated in dental caries. Bacteria were cultivated with various concentrations of NaCl on a 96-well microtiter plate in tryptic soy broth with 0.25% sucrose or 0.25% glucose (TSBs and TSBg, respectively) for 16 h. The effects of glucosyltransfe

The lactoperoxidase (LPO) system shows promise in the prevention of dental caries, a common chronic disease. This system has antimicrobial properties and is part of the non-specific antimicrobial immune system. Understanding the efficacy of the LPO system in the fight against biofilms could provide information on alternative strategies for the prevention and treatment of caries. In this study, the enzymatic system was modified using four different (pseudo)halide substrates (thiocyanate, thiocyanate-iodide mixture, selenocyanate, and iodide). The study evaluated the metabolic effects of applyin

Microbial adhesion to oral splints may lead to oral diseases such as candidiasis, periodontitis or caries. The present in vitro study aimed to assess the effect of novel computer-aided design/computer-aided manufacturing (CAD/CAM) and conventional manufacturing on Candida albicans and Streptococcus mutans adhesion to oral splint resins. Standardized specimens of four 3D-printed, two milled, one thermoformed and one pressed splint resin were assessed for surface roughness by widefield confocal microscopy and for surface free energy by contact angle measurements. Specimens were incubated with C.

Streptococcus mutans is known to be a major causative agent of dental caries, and strains expressing the cell surface collagen-binding Cnm protein contribute to the development of several systemic diseases. A relationship between tonsillar immunity and glomerulonephritis has been recognized in IgA nephropathy (IgAN), and specific pathogens may have effects on tonsillar immunity (mucosal immunity). Here, we present findings showing a relationship between the presence of Cnm-positive S. mutans strains in the tonsils of IgAN patients and IgAN condition/pathogenesis. Analyses of tonsillar specimen

BACKGROUND: Dental caries and periodontal disease are most common oral diseases. Streptococcus mutans are considered to be the major pathogens in initiation of dental caries. Evidence shows that periodontal disease and caries share a number of contributory factors. Thus in view of these findings it would be worthwhile to examine whether Streptococcus mutans persist within the saliva and subgingival environment of the periodontitis patients and to determine whether there is any association between Streptococcus mutans colonization, pH of saliva and sub-gingival plaque pH in periodontal diseases

Streptococcus mutans is a major pathogen of dental caries. Collagen-binding proteins (CBPs) (approximately 120 kDa), termed Cnm and Cbm, are regarded as important cell surface antigens related to the adherence of S. mutans to collagenous tissue. Furthermore, CBP-positive S. mutans strains are associated with various systemic diseases involving bacteremia, such as infective endocarditis. Endodontic infection is considered to be an important cause of bacteremia, but little is known regarding the presence of S. mutans in dental pulp tissue. In the present study, the distribution and virulence of

Streptococcus mutans, a pathogen responsible for dental caries, is occasionally isolated from the blood of patients with bacteremia and infective endocarditis (IE). Our previous study demonstrated that serotype k-specific bacterial DNA is frequently detected in S. mutans-positive heart valve specimens extirpated from IE patients. However, the reason for this frequent detection remains unknown. In the present study, we analyzed the virulence of IE from S. mutans strains, focusing on the characterization of serotype k strains, most of which are positive for the 120-kDa cell surface collagen-bind

BACKGROUND: Clinical studies in which antimicrobial mouthrinses were shown to have significant antiplaque activity most frequently have used gingivitis as the clinically relevant endpoint. However, there is evidence to suggest that mouthrinses containing active agents effective against Streptococcus mutans, such as chlorhexidine, may also have a role in inhibiting dental caries. This clinical study was conducted to determine the effect of 2x-daily rinsing with an essential oil-containing antiseptic mouthrinse (Listerine Antiseptic) on levels of recoverable S. mutans and total streptococci in s
