Alpha-synuclein (α-syn) mislocalization and accumulation in intracellular inclusions is the major pathological hallmark of degenerative synucleinopathies, including Parkinson's disease, Parkinson's disease with dementia and dementia with Lewy bodies. Typical symptoms are behavioural abnormalities including motor deficits that mark disease progression, while non-motor symptoms and synaptic deficits are already apparent during the early stages of disease. Synucleinopathies have therefore been considered synaptopathies that exhibit synaptic dysfunction prior to neurodegeneration. However, the mec

Preterm infant brain injury is a leading cause of morbidity and disability in survivors of preterm infants. Unfortunately, the effective treatment remains absent. Recent evidence suggests that GSK-3β inhibitor TWS119 has a neuroprotectiverole in adult brain injury by activation of Wnt/β-catenin signaling pathway. However, the role on neonatal brain injury is not yet explored. The study aims to evaluate the effect of TWS119 at 7 d after hypoxic-ischemic brain damage and investigate the mechanism that it regulates Wnt and Notch signaling pathways at 24 h after hypoxic-ischemic brain damage in ne

Alzheimer's disease (AD) is multifactorial with unclear etiopathology. Due to the complexity of AD, many attempted single therapy treatments, like Aβ immunization, have generally failed. Therefore, there is a need for drugs with multiple benefits. Naturally occurring phytochemicals with neuroprotective, anti-amyloidogenic, antioxidative, and anti-inflammatory properties could be a possible way out. In this study, the effect of Moringa oleifera (MO), a naturally occurring plant with high antioxidative, anti-inflammatory, and neuroprotective effects, was evaluated on hyperhomocysteinemia (HHcy)

Schistosomiasis is a tropical disease caused by a flatworm trematode parasite that infects over 200 million people worldwide. Treatment and control of the disease rely on just one drug, praziquantel. The possibility of drug resistance coupled with praziquantel's variable efficacy encourages the identification of new drugs and drug targets. Disruption of neuromuscular homeostasis in parasitic worms is a validated strategy for drug development. In schistosomes, however, much remains to be understood about the organization of the nervous system, its component neurotransmitters and potential for d

Glutamatergic system and mTOR signaling pathway have been proposed to be important targets for pharmacological treatment of major depressive disorder. Previous studies have shown that inosine, an endogenous purine, is able to exert a remarkable antidepressant-like effect in mice. Nevertheless, the role of glutamatergic system and mTOR in this effect was not previously determined. This study was designed to investigate the possible modulation of NMDA receptors (NMDAR), AMPA receptors (AMPAR) and mTOR complex 1 (mTORC1) signaling pathway in the inosine anti-immobility effect in the tail suspensi

Alternative splicing generates multiple isoforms of the microtubule-associated protein Tau, but little is known about their specific function. In the adult mouse brain, three Tau isoforms are expressed that contain either 0, 1, or 2 N-terminal inserts (0N, 1N, and 2N). We generated Tau isoform-specific antibodies and performed co-immunoprecipitations followed by tandem mass tag multiplexed quantitative mass spectrometry. We identified novel Tau-interacting proteins of which one-half comprised membrane-bound proteins, localized to the plasma membrane, mitochondria, and other organelles. Tau was

Agmatine is an endogenous neuromodulator that has been shown to have antidepressant-like properties. We have previously demonstrated that it can induce a rapid increase in BDNF levels after acute administration, suggesting that agmatine may be a fast-acting antidepressant. To investigate this hypothesis, the present study evaluated the effects of a single administration of agmatine in mice subjected to chronic unpredictable stress (CUS), a model of depression responsive only to chronic treatment with conventional antidepressants. The ability of agmatine to reverse CUS-induced behavioral and bi

Melatonin, an indoleamine hormone secreted into circulation at night primarily by the brain's pineal gland, has been shown to have a wide variety of actions on the development and physiology of neurons in the CNS. Acting via two G-protein-coupled membrane receptors (MT1 and MT2), melatonin modulates neurogenesis, synaptic functions, neuronal cytoskeleton and gene expression. In the present studies, we sought to characterize the behavior and neuronal biology of transgenic mice lacking both of these melatonin receptors as a way to understand the hormone's receptor versus non-receptor-mediated ac

Mutations in the X-linked gene, methyl-CpG binding protein 2 (Mecp2), underlie a wide range of neuropsychiatric disorders, most commonly, Rett Syndrome (RTT), a severe autism spectrum disorder that affects approximately one in 10,000 female live births. Because mutations in the Mecp2 gene occur in the germ cells with onset of neurological symptoms occurring in early childhood, the role of MeCP2 has been ascribed to brain maturation at a specific developmental window. Here, we show similar kinetics of onset and progression of RTT-like symptoms in mice, including lethality, if MeCP2 is removed p

Early odor preference learning in rats provides a simple model for studying learning and memory. Learning results in an enhanced output from mitral cells, which carry odor information from the olfactory bulb to the olfactory cortex. Mitral cell NMDA receptors (NMDARs) are critically involved in plasticity at the olfactory nerve to mitral cell synapse during odor learning. Here we provide evidence that L-type calcium channels (LTCCs) provide an additional and necessary source of calcium for learning induction. LTCCs are thought to act downstream of NMDARs to bridge synaptic activation and the t

We previously found that the glutamate release was decreased in synaptosomes from rat cerebral cortex during the development of experimental autoimmune encephalomyelitis (EAE), the animal model of multiple sclerosis. Various other reports have shown a deficit in the expression of proteins associated with GABAergic neurotransmission in the neocortex of patients with multiple sclerosis and it was also demonstrated that the activation of GABAA receptors leads to an inhibition of glutamate release. Now, in order to evaluate the events that may affect the neuronal function in EAE synaptosomes, we a

BACKGROUND AND OBJECTIVES: Exposure to blue light emitted from electronic devices has been shown to impair cognitive performance; however, the mechanisms underlying these deleterious effects remain poorly understood. Orexin neurons in the hypothalamus, which play a key role in modulating cognitive processes and synaptic plasticity, project directly to the hippocampus, a brain region critical for learning and memory. Therefore, our study provides novel insights into the neural mechanisms underlying blue light-related cognitive dysfunction.
METHODS AND RESULTS: In this study, we evaluated cogni

B cells infiltrate the contralesional hippocampus following stroke, but whether lymphocytes modulate post-stroke plasticity and neuronal network function remains unknown. To identify immune cell mechanism(s) supporting remote plasticity, we examined the impact of B cell depletion on synaptic and neuronal activity in the hippocampal circuit following stroke. Basal synaptic transmission in the contralesional dentate gyrus (DG) following a stroke in adult male mice was decreased with B cell depletion. Expanding our studies to encompass the CA1 and DG regions of the hippocampal circuit in male and

Post-stroke depression (PSD) represents a complex neuropsychiatric challenge characterized by persistent neuroinflammation and synaptic dysfunction, yet effective therapeutic interventions are constrained by the blood-brain barrier (BBB) and the lack of specific targets. Using a multidimensional screening strategy for Chaihu-Jia-Longgu-Muli Decoction (CLM), we identified ginsenoside Rd (Rd) as a key blood-absorbed bioactive constituent associated with Epidermal Growth Factor Receptor (EGFR) signaling. To overcome the bioavailability bottleneck, a biomimetic nanodelivery system is engineered by

The inability of damaged neurons to regenerate poses a significant challenge in the repair of peripheral nerve injuries. Exogenous cell therapies, such as those involving neural stem cells or induced pluripotent stem cells, are limited by immune rejection and ethical concerns. Somatic cells offer a promising alternative, as they can be reprogrammed and differentiated into neuron-like cells, thereby promoting nerve repair. Satellite glial cells (SGCs) within the dorsal root ganglion (DRG) have the capacity to differentiate into multiple cell types. In this study, chemical small molecules were u

The mushroom bodies (MBs) in the insect brain serve sensory integration and memory formation. In the honeybee, they house two classes of intrinsic neurons: class I (spiny) and II (clawed) Kenyon cells (KCs). Both classes form postsynaptic elements in synaptic complexes (microglomeruli) comprising large axonal boutons from olfactory and visual projection neurons. To adapt their neuronal information processing systems, MB microglomeruli undergo age-, memory-, and environment-related structural plasticity. To analyze KC dendritic specializations and their connections with presynaptic boutons, we

Mild maternal immune activation (MIA) alone has been reported to exert minimal effects on the newborn brain, but when combined with postnatal stress (PS), it can trigger significant and, lasting neuro developmental changes. This study investigated the effects of MIA, alone and in combination with PS, on synaptic plasticity and molecular pathways in the prefrontal cortex, with a focus on potential links to neurodevelopmental disorders. Pregnant mice received an intraperitoneal injection of lipopolysaccharide (LPS, 50 μg/kg) or sterile normal saline on gestational day 17 (GD17). Offspring were e

The Diagnostic and Statistical Manual of Mental Disorders (DSM-5) categorizes postpartum depression (PPD) as a subtype of Major Depressive Disorder (MDD) with peripartum onset, generally arising within the initial trimester following delivery. This acute psychiatric condition is characterized by feelings of worthlessness, insomnia, extreme anxiety, or maternal neglect. Intranasal oxytocin (OT) and transcranial magnetic stimulation (TMS) have the potential to address impaired social cognition; nonetheless, their neuronal underpinnings, along with their safety and efficacy, are little comprehend

Ca2+/calmodulin-dependent protein kinase II α (CaMKIIα) and nitric oxide (NO) both play vital roles in learning and memory; however, the underlying mechanisms connecting them have remained elusive. To address this question, our study surprisingly observed that during learning and memory tasks, S-nitrosation of CaMKIIα, a key redox-based post-translational modification, significantly increased in mouse hippocampus. We then constructed mice with mutations in the major S-nitrosation sites of CaMKIIα (C280/289V) and found that the mutant mice exhibited remarkable cognitive impairments and attenuat

AIMS: Microglia, as resident macrophages in the brain, play an important role in depression. Heat shock protein 60 (HSP60), as a chaperone protein, plays a role in cell stress. However, the role of microglial HSP60 in depression remains unclear.
METHODS: CX3CR1-CreER was used to generate microglial-specific HSP60 knockout (HSP60 cKO) mice. Behavioral tests, western blotting, Golgi staining, biochemical assays, and proteomics were employed to assess depression-like symptoms, microglial activation, and synaptic changes.
RESULTS: HSP60 cKO male mice exhibited depressive-like behaviors, without
