Targeting SARS-CoV-2 main protease for the discovery of a broad-spectrum COVID-19 inhibitor by intensive multi-tiered validation.

SARS-CoV-2 and its emerging variants continue to pose a significant global public health threat. The SARS-CoV-2 main protease (Mpro) is a critical target for the development of antiviral agents that can inhibit viral replication and transcription. In this study, we identified chebulagic acid (CHLA), isolated from Terminalia chebula Retz., as a potent non-peptidomimetic and non-covalent Mpro inhibitor. CHLA exhibited intermolecular interactions and provided significant protection to Vero E6 cells against a range of SARS-CoV-2 variants, including the wild-type, Delta, Omicron BA.1.1, BA.2.3, BA.

Attenuation of quorum-sensing-dependent virulence factors and biofilm formation by medicinal plants against antibiotic resistant Pseudomonas aeruginosa.

Pseudomonas aeruginosa use small signaling molecules such as acyl homoserine lactones (AHLs), which play an important role in release virulence factors and toxin for further establishment of host infection. Thus, involving with the QS system would provide alternative ways of preventing the pathogenicity. In the present study, totally six medicinal plants (Terminalia bellerica, Celastrus paniculatus, Kingiodendron pinnatum, Schleichera oleosa, Melastoma malabathricum, Garcinia gummi-gutta) were screened for anti-QS activity using biomonitor strain of Chromobacterium violaceum CV12472. Study on Quality Markers of Chebulae Fructus Immaturus by the Integration of Network Pharmacology, UHPLC Fingerprint, and Serum Pharmacochemistry.

The chemical composition of Chebulae Fructus Immaturus, a traditional Chinese medicine, is complex. Its efficacy relies on the interaction of its components, making quality evaluation based on a single chemical insufficient. This study proposes a multi-dimensional feature network to enhance Chebulae Fructus Immaturus quality assessment, focusing on effectiveness, content, traceability, and specificity. The network pharmacological analysis is employed to elucidate the underlying mechanism by which chemical components in Chebulae Fructus Immaturus exert their therapeutic effects, specifically in

Exploring the Potential of Phytotherapeutics against Auto-inflammatory Disorders: A Review.

INTRODUCTION: Various inflammatory conditions, such as arthritis, neuralgia, soft tissue injuries, and neuropathic pain, significantly affect daily quality of life. Among these, Osteoarthritis (OA) and Rheumatoid Arthritis (RA) are the most common types, often leading to joint discomfort and reduced quality of life. Conventional treatments like NSAIDs, though effective for pain relief, are associated with adverse effects, including an increased risk of cardiovascular disease. This review aims to highlight the potential role of herbal medicines in managing arthritis and reducing inflammation.

Pre-Courtship Behavior of Proholopterus chilensis (Coleoptera: Cerambycidae) in a Nothofagus obliqua (Nothofagaceae) Forest.

The pre-courtship behavior of Proholopterus chilensis on Nothofagus obliqua trees was recorded for the first time, documenting a putative female "calling" behavior and the consequent male "oriented search," enabling the description of behavioral units, sequences, frequencies, and degrees of stereotypy. Post-resting activity in both sexes began with walking and grooming during the first hour of the scotophase, following a period of daytime inactivity. Subsequently, females extended their ovipositor both horizontally and vertically, alternating between contact with the tree substrate and, simult

Antibacterial activity of the endophytic fungal extracts and synergistic effects of combinations of ethylenediaminetetraacetic acid (EDTA) against Pseudomonas aeruginosa and Escherichia coli.

The growing threat of antibiotic resistance in bacteria is a critical public health concern. Combining natural compounds with antimicrobial agents is an alternative approach to improve the antibacterial efficacy and safety of these agents. The strategy is to restore the effectiveness of existing antibiotics while minimizing the required concentrations of antibiotics or antimicrobial agents. This study aimed to isolate the endophytic fungi from medicinal plants, including Lantana camara, Orthosiphon aristatus, Mansonia gagei, Terminalia bellirica, Oroxylum indicum, Elaeagnus latifolia, Talinum

Neurobiology of egg-laying behavior in Drosophila: neural control of the female reproductive system.

Egg-laying is one of the key aspects of female reproductive behavior in insects. Egg-laying has been studied since the dawn of Drosophila melanogaster as a model organism. The female's internal state, hormones, and external factors, such as nutrition, light, and social environment, affect egg-laying output. However, only recently, neurobiological features of egg-laying behavior have been studied in detail. fruitless and doublesex, two key players in the sex determination pathway, have become focal points in identifying neurons of reproductive significance in both central and peripheral nervous

The Drosophila serine protease homologue Scarface regulates JNK signalling in a negative-feedback loop during epithelial morphogenesis.

In Drosophila melanogaster, dorsal closure is a model of tissue morphogenesis leading to the dorsal migration and sealing of the embryonic ectoderm. The activation of the JNK signal transduction pathway, specifically in the leading edge cells, is essential to this process. In a genome-wide microarray screen, we identified new JNK target genes during dorsal closure. One of them is the gene scarface (scaf), which belongs to the large family of trypsin-like serine proteases. Some proteins of this family, like Scaf, bear an inactive catalytic site, representing a subgroup of serine protease homolo

Asynchronous leaf and cambial phenology in a tree species of the Congo Basin requires space-time conversion of wood traits.

BACKGROUND AND AIMS: Wood traits are increasingly being used to document tree performance. In the Congo Basin, however, weaker seasonality causes asynchrony of wood traits between trees. Here, we monitor growth and phenology data to date the formation of traits. METHODS: For two seasons, leaf and cambial phenology were monitored on four Terminalia superba trees (Mayombe) using cameras, cambial pinning and dendrometers. Subsequently, vessel lumen and parenchyma fractions as well as high-resolution isotopes (δ13C/δ18O) were quantified on the formed rings. All traits were dated and related to we
