Cyanide, a metabolic poison, is a rising chemial threat and ingestion is the most common route of exposure. Terrorist organizations have threatened to attack the USA and international food and water supplies. The toxicokinetics and toxicodynamics of oral cyanide are unique, resulting in high-dose exposures, severe symptoms, and slower onset of symptoms. There are no FDA-approved therapies tested for oral cyanide ingestions and no approved intramuscular or oral therapies, which would be valuable in mass casualty settings. The aim of this review is to evaluate the risks of oral cyanide and its u

Environmental chemicals contribute substantially to the global burden of chronic disease, yet for most compounds now detectable in human biological samples, causal links to specific health outcomes remain poorly established. HRMS can profile thousands of chemical features simultaneously in a single sample. What has not kept pace is study design. No investigation has yet combined external environmental source monitoring, multi-matrix internal biomonitoring, and toxicokinetic normalization within a single longitudinal framework. This Perspective argues that closing this design gap is the prerequ

Second-generation anticoagulant rodenticides (SGARs) have been used since the 1980s for pest management. They are highly efficient even in warfarin-resistant rodents. Nevertheless, because of their tissue persistence, nontarget poisoning by SGARs is commonly described in wildlife. Due to this major problem, a new generation of anticoagulants must be developed to limit this risk. This study proposes a method of developing a new generation of anticoagulant rodenticides by revisiting the old SGARs based on the concept of stereochemistry. Each current SGAR is a mixture of diastereomers. Diastereom

Homosalate (HMS) is an organic UV filter used in sunscreens and personal care products. Despite its widespread use and detection in environmental matrices, little is known regarding its exposure in humans. HMS is used as a mixture of cis- and trans-isomers, and we recently revealed major differences in human toxicokinetics, indicating the need to consider these isomers separately in exposure and risk assessments. In the course of these previous investigations of human HMS toxicokinetics, we identified two trans-HMS-specific and one cis-HMS-specific biomarker candidates. However, the latter lac

Parkinson's disease (PD) prevalence has been steeply increasing over the last years. Environmental pollution, including certain pesticides, is considered a significant contributor to this rise. Certain genetic factors may pose an individual or subpopulation at increased risk to develop pesticide-induced PD. To support New Approach Methodologies (NAMs)-based hazard and risk assessment while considering known sources of interindividual variability, the aim of the current work was to identify biological factors related to increased susceptibility towards pesticide-induced PD onset and progression

Exposure to organic solvents can affect the central nervous system. Propylene glycol ethers such as propylene glycol methyl ether (PGME) and propylene glycol butyl ether (PGBE) are organic solvents widely used in commercial products including cleaning products. PGME has an established Swiss occupational exposure limit and a biomonitoring limit value while PGBE does not, which is probably due to the absence of toxicokinetic data. Our aim was to (1) Characterize PGBE elimination toxicokinetics in healthy human participants (N = 11) exposed under controlled conditions; (2) Provide urinary elimina

Methods for both total and form-variable tin analysis in biological media are reviewed. While total tin analysis was common in the past, and in some cases still is, better understanding of the toxicology and toxicokinetics of organotins in biological systems requires knowledge of specific tin forms present. Both chromatographic and non-chromatographic methods for chemical speciation of organotins in various media have been described, the former appearing to offer the most promising approaches. High performance liquid chromatography (HPLC) in tandem with tin-specific metal detection presently a

Dimethoxyethyl phthalate (DMEP) is an industrial phthalates reagent exposed to human body. Its hydrolytic metabolite ethylene glycol methyl ether (EGME) and further oxidative metabolite methoxyacetic acid (MAA) have reproductive and developmental toxicity, and the latter is more toxic than the former. However, due to the lack of sensitive and reliable analytical method, the in vivo clearance and biotransformation of EGME and MAA is poorly understood. In order to evaluate their toxicokinetic profiles, a precolumn derivatization liquid chromatography-tandem mass spectrometry (LC-MS/MS) method wa

Since there are pathomorphological similarities between porcine mycotoxic nephropathy caused by ochratoxin A and Balkan endemic nephropathy (BEN), it has been suggested that the same aetiological agent has a role in BEN. Based on the results from several field and experimental studies carried out on pigs, an appropriate analytical method of monitoring possible human exposure to ochratoxin A was developed. The toxicokinetic properties of the toxin were species specific, although in all the animal species studied (with the exception of fish), as well as in humans, two binding proteins were found

Phomopsins are mycotoxins mainly contaminating lupin-derived food matrices posing safety concerns, with phomopsin A considered the most potent congener. Their toxicity is linked to disruption of microtubule dynamics, yet the shortage of toxicokinetic and toxicodynamic data prevents adequate human risk assessment, representing a critical gap for food safety. To bridge this data gap through New Approach Methodologies (NAMs), a computational 3D modelling pipeline, combining molecular dynamics simulations and binding free energy calculations, was applied to investigate the interactions of phomopsi

This study aimed to evaluate the developmental and reproductive toxicity of HSYA across three segments (FEED, EFD, and PPND). HSYA was administered intravenously to Sprague-Dawley (SD) rats at doses of 25, 75, or 250 mg/kg. FEED: In the 250 mg/kg/day group, body weights of F0 males decreased during weeks 2, 3, and 4 post-administration. Several males in the high-dose group exhibited yellow-red ocular secretions from day one to week seven, which resolved thereafter. EFD: No significant differences were observed in the incidence of malformations or variations in the F1 generation. Fetal body len

INTRODUCTION: Metformin poisoning with lactic acidosis is an uncommon yet clinically serious condition related to the inhibition of normal aerobic metabolism. Toxicity may occur after an acute overdose although it is much more common after a systemic insult, such as acute kidney injury, in the setting of chronic use. Hemodialysis is currently the preferred extracorporeal treatment modality (Grade 1D evidence) although some patients may be too hemodynamically unstable to tolerate it. Continuous renal replacement therapy is considered an alternative if hemodialysis is unavailable but an evaluati

Evaluating the potential for methylmercury (MeHg) toxicity relies on accurately predicting the mercury (Hg) body burden that results from eating fish. Hg body burden is directly determined by the slow elimination kinetics of MeHg in the human body (kel = 0.014 days-1 or t1/2 =50 days). Existing studies on MeHg half-life in humans demonstrate a wide range values (t1/2 = 30 to >150 days) and has lead to uncertainty in the derivation of a regulatory standard for acceptable daily oral intake. The causes of variation in MeHg toxicokinetics in humans remain little explored. Here we characterize vari

INTRODUCTION: Bongkrekic acid (BA) has a unique mechanism of toxicity among the mitochondrial toxins: it inhibits adenine nucleotide translocase (ANT) rather than the electron transport chain. Bongkrekic acid is produced by the bacterium Burkholderia gladioli pathovar cocovenenans (B. cocovenenans) which has been implicated in outbreaks of food-borne illness involving coconut- and corn-based products in Indonesia and China. Our objective was to summarize what is known about the epidemiology, exposure sources, toxicokinetics, pathophysiology, clinical presentation, and diagnosis and treatment o

There is a wide variation in sensitivity to lead (Pb) exposure, which may be due to genetic susceptibility towards Pb. We investigated whether a polymorphism (rs1800435) in the δ-aminolevulinic acid dehydratase (ALAD) gene affected the toxicokinetics and toxicodynamics of Pb. Among 461 Chinese Pb-exposed storage battery and 175 unexposed workers, allele frequencies for the ALAD1 and ALAD2 alleles were 0.968 and 0.032, respectively. The Pb-exposed workers had a higher fraction of the ALAD1-2/2-2 genotype than unexposed workers (7.8% vs. 2.3%, p=0.01). The Pb levels in blood (B-Pb) and urine (U-

The risks to non-target birds and other wildlife from the use of vertebrate pesticides, including anticoagulant rodenticides, are determined to a significant extent by species' intrinsic susceptibility, and the toxicokinetics of the compounds used. Brodifacoum is highly toxic to birds and mammals. The acute toxicity of brodifacoum to birds in New Zealand varies from <1 mg/kg in pukeko (Porphyrio p. melanotus), the native swamp hen, to >20 mg/kg in the paradise shelduck (Tadorna variegata). Like other second-generation anticoagulants brodifacoum is strongly bound to vitamin K epoxide reductase

BACKGROUND: Carbamazepine (CBZ) is a widely used antiepileptic drug with a narrow therapeutic window. Its potential for severe toxicity in overdose is primarily due to its active metabolite, carbamazepine-10,11-epoxide (CBZ-E). In acute CBZ intoxication, sex-specific differences and toxicokinetics are poorly understood, especially the impact on neurological impairment, despite its clinical relevance.
OBJECTIVES: The present study aimed to explore the clinical, biochemical, and toxicokinetic data of patients with acute CBZ poisoning to assess the relationship between CBZ exposure and the level

Reports of microplastics (MPs) in human blood and tissues have intensified interest in the relationship between external exposure and internal measurements. This area of research remains poorly understood, with no clear resolution, as the current biomonitoring data still lack a quantitative linkage to environmental sources that could support risk-relevant interpretation. Here, we employ an illustrative pipeline spanning exposure synthesis, probabilistic intake modeling, compositional fingerprinting, and pharmacokinetic compatibility mapping to clarify what existing evidence can and cannot supp

Per- and polyfluoroalkyl substances (PFAS) are persistent organic pollutants with immunotoxicity as a primary health concern. Their in vivo fate has long been interpreted within a blood-centric toxicokinetic framework, whereas the lymphatic system, a key immune surveillance hub, has been overlooked as an exposure compartment. Given their structural similarity to long-chain fatty acids, PFAS may be absorbed via intestinal lymphatics, directly exposing resident immune cells, a hypothesis never experimentally verified. Here, using rat models with mesenteric lymph duct and jugular vein cannulation
