Enhanced gene transfection of macrophages by photochemical internalization: Potential for gene-directed enzyme prodrug therapy of gliomas.

BACKGROUND: Drawn by tumor synthesis of chemo-attractive factors, macrophages are frequently found in and around glioblastomas and play an important role both in augmenting as well as inhibiting tumor growth. Patient-derived macrophages have the potential, therefore, to act as targeted delivery vectors for a variety of anti-cancer treatments. Among these is ex vivo gene transfection and re-injection back into the patient of macrophages to target residual tumors. In this study, photochemical internalization (PCI) is investigated as a technique for the non-viral transfection of the cytosine deam

Solid Lipid Nanoparticles as Non-Viral Vectors for Gene Transfection in a Cell Model of Fabry Disease.

Here, we demonstrate the ability of solid lipid nanoparticle-based non-viral vectors to increase the α-galactosidase A levels of the IMFE1 cell line, an in vitro model for target cells in Fabry disease. For this purpose, vectors containing the pR-M10-αGal A plasmid, which encodes the α-galactosidase A enzyme, were prepared; the in vitro transfection efficacy was studied in IMFE1 cells, and the results were confirmed by RT-PCR. The cellular uptake of the vectors, intracellular disposition of the plasmid, and probable endocytosis pathways of the nanoparticles were also analyzed. The vectors used

Refining siRNA in vivo transfection: silencing SPHK1 reveals its key role in C5a-induced inflammation in vivo.

Источник пометил публикацию как отозванную

The transfection of siRNA in vivo is essential for the study of gene functions, target validation, and for gene therapy. However, the successful delivery of siRNA in whole organisms is still very difficult to achieve. A high-pressure delivery technique, called the "hydrodynamics" method, has been used for siRNA transfection in mice. However, it is a method based on a high-speed and high-volume of i.v. injection, which makes it very difficult to implement in vivo, due to vascular breakage. Here, we systematically investigated ways to optimize the siRNA delivery, in order to avoid strong side ef

Isolation, Cultivation, and Transient Transfection of Primary Human T Cells to Generate Chimeric Antigen Receptor (CAR) T Cells.

To assess the efficacy and potency of chimeric antigen receptor (CAR) T cells, it is essential to isolate, activate, expand, characterize, and genetically engineer primary human T cells. Compared to common T cell lines, primary cells can provide a more realistic model system, since they are highly variable, capable of T cell-mediated killing, and able to differentiate. Here, we provide a step-by-step protocol for all essential steps for the cultivation of primary T cells, including their isolation from blood products, activation, in vitro expansion, and transient transfection. We describe diff

A Quick Reverse Transfection and Dual-Imaging Protocol for Simultaneous Viability and Oligonucleotide Delivery Assessment in 4T1 Cells.

The 4T1 cell line is a widely used model for triple-negative breast cancer (TNBC) research due to its high metastatic potential and resemblance to clinical progression. However, its biological characteristics often limit the efficiency and reproducibility of conventional nucleic acid delivery protocols. Here, we present an optimized non-viral delivery framework for efficient cell-associated uptake in 4T1 cells. Five carrier systems-Xfect RNA polymer (Xfect), polyethyleneimine (PEI), chitosan (CHI), and gold nanoparticle conjugates (AuPEI and AuCHI)-were systematically evaluated using both stan

Engineering liposomes via click chemistry for multimodal biomedical applications: Evidence from eukaryotic transfection and antimicrobial light-triggered therapy.

Click chemistry is a powerful and versatile method for grafting various moieties onto a chemical backbone, enabling the integration of multiple functionalities into a single molecular scaffold. Here, a previously reported cationic lipophosphoramidate, BSV36, was employed in a series of liposomes functionalized with either a naphthalimide or a nitroaniline moiety. These modifications were introduced via aminolysis of phosphonodithioester click reaction, performed either before or after liposome formation. The various liposomes obtained could display - separately or in combination - multiple pro

Click-Chemistry Functionalized Lipid Nanoparticles for In Vitro and In Vivo Transfection of Primary Human CD4+ T Cells.

CD4+ T cells orchestrate adaptive immunity and strategies that enable their genetic reprogramming hold promise for treating cancer, immune dysregulation and infectious diseases such as HIV. However, current T cell engineering approaches rely heavily on ex vivo manipulation, which is labor-intensive, costly, and can alter cell phenotypes. Lipid nanoparticles (LNPs) offer a scalable nonviral alternative, yet conventional formulations show minimal interaction with lymphocytes and are rapidly cleared by the liver. In this study, we develop a modular DBCO-azide click-chemistry approach to functiona

Transfection of the free-living alga Chromera velia enables direct comparisons with its parasitic apicomplexan relative, Toxoplasma gondii.

Chromera velia is a photosynthetic, free-living alga closely related to the apicomplexan parasites, a phylum of intracellular pathogens responsible for many devastating diseases, including malaria, cryptosporidiosis and toxoplasmosis. With molecular and cellular landmarks clearly related to but distinguishable from those found in apicomplexan parasites, Chromera provides an opportunity to investigate the evolutionary origin of the structures and processes needed for intracellular parasitism. A transposon-based transient transfection system in CHO-K1 cells enables quality prediction of stable cell line proteins.

In biologics drug discovery, transient protein expression is widely used to rapidly produce biologics, thereby accelerating the identification of lead candidates. However, the accuracy and consistency of predicting further product quality in large-scale production needs to be considered, especially with respect to physicochemical properties and posttranslational modifications. With this in mind, a transient expression system utilizing Chinese hamster ovary K1 (CHO-K1) has been established, which integrates high expression capability with quality profiles similar to those of the protein produce

Sequentially conquering transfection obstacles of primary human T cells via non-classical lipid nanoparticles delivering reformative transposon system.

Chimeric antigen receptor (CAR)-T cells have shown promising efficacy in disease treatment. Considering the safety issues of viral vectors, non-viral vectors such as lipid nanoparticles (LNPs) or transposon systems have attracted growing attention in constructing CAR-T cells. However, the limited transfection efficiency of LNPs and the possible cellular damage via electroporation of transposon system restrict their applications. Herein, we propose to construct CAR-T cells with long-lasting CAR expression via non-classical LNPs delivering reformative piggyBac transposon system. The non-classica

Potentiation of anti-angiogenic eNOS-siRNA transfection by ultrasound-mediated microbubble destruction in ex vivo rat aortic rings.

Nitric oxide (NO) regulates vascular homeostasis and plays a key role in revascularization and angiogenesis. The endothelial nitric oxide synthase (eNOS) enzyme catalyzes NO production in endothelial cells. Overexpression of the eNOS gene has been implicated in pathologies with dysfunctional angiogenic processes, such as cancer. Therefore, modulating eNOS gene expression using small interfering RNAs (siRNAs) represents a viable strategy for antitumor therapy. siRNAs are highly specific to the target gene, thus reducing off-target effects. Given the widespread distribution of endothelium and th
