Fibroblast pyruvate carboxylase is required for collagen production in the tumour microenvironment.

The aberrant production of collagen by fibroblasts is a hallmark of many solid tumours and can influence cancer progression. How the mesenchymal cells in the tumour microenvironment maintain their production of extracellular matrix proteins as the vascular delivery of glutamine and glucose becomes compromised remains unclear. Here we show that pyruvate carboxylase (PC)-mediated anaplerosis in tumour-associated fibroblasts contributes to tumour fibrosis and growth. Using cultured mesenchymal and cancer cells, as well as mouse allograft models, we provide evidence that extracellular lactate can

C3 and C4 plant systems respond differently to the concurrent challenges of mercuric oxide nanoparticles and future climate CO2.

Future climate CO2 (eCO2) and contamination with nano-sized heavy metals (HM-NPs) represent concurrent challenges threatening plants. The interaction between eCO2 and HM-NPs is rarely investigated, and no study has addressed their synchronous impact on the metabolism of the multifunctional stress-related metabolites, such as sugars and amino acids. Moreover, the characteristic responses of C3 and C4 plant systems to the concurrent impact of eCO2 and HM-NPs are poorly understood. Herein, we have assessed the impact of eCO2 (620 ppm) and/or HgO-NPs (100 mg/Kg soil) on growth, physiology and meta

Exercise-induced metabolic remodeling in type 2 diabetes mellitus: insights from metabolomics.

Type 2 diabetes mellitus (T2DM) is one of the most pressing global public health challenges. Exercise intervention, as a cost-effective and safe non-pharmacological strategy, exerts its metabolic benefits through coordinated multi-organ and multi-target regulation. Metabolomics technologies provide powerful tools for dissecting the molecular mechanisms underlying exercise intervention in T2DM. This narrative review summarizes current evidence on metabolic dysregulation in T2DM and the metabolic mechanisms underlying exercise intervention, with particular emphasis on insights provided by metabo

Fumarase deficiency in dichorionic diamniotic twins.

Fumarase deficiency is a rare autosomal recessive inborn error of metabolism of the Krebs Tricarboxylic Acid cycle. A heavy neurological disease burden is imparted by fumarase deficiency, commonly manifesting as microcephaly, dystonia, global developmental delay, seizures, and lethality in the infantile period. Heterozygous carriers also carry an increased risk of developing hereditary leiomyomatosis and renal cell carcinoma. We describe a non-consanguineous family in whom a dichorionic diamniotic twin pregnancy resulted in twin boys with fumarase deficiency proven at the biochemical, enzymati

[Catalytic properties of glutathione reductase from rats liver at norm and toxic hepatitis].

Homogeneous preparations of liver glutathione reductase (GR, EC 1.6.4.2.), obtained from control rats and animals with toxic hepatitis, catalytic properties of this enzyme have been investigated. A number of the enzyme properties did not change under conditions of hepatitis. These included electrophoretic mobility (R(f) = 0.23 +/- 0.01), molecular mass (104 +/- 5.20 kDa), pH-optimum (7.4 +/- 0.37). There was similarity in pK values of functional groups. At the same time, decrease in affinity of enzyme to a substrate and coenzyme and occurrence substrate-linked inhibition was observed under con

A role for mitochondrial enzymes in inherited neoplasia and beyond.

Mitochondrial defects have been associated with neurological disorders, as well as cancers. Two ubiquitously expressed mitochondrial enzymes--succinate dehydrogenase (SDH) and fumarate hydratase (FH, fumarase)--catalyse sequential steps in the Krebs tricarboxylic-acid cycle. Inherited heterozygous mutations in the genes encoding these enzymes cause predispositions to two types of inherited neoplasia syndromes that do not share any component tumours. Homozygous mutations in the same genes result in severe neurological impairment. Understanding this link between inherited cancer syndromes and ne

FT-IR and FT-Raman spectra, normal coordinate analysis and ab initio computations of Trimesic acid.

The FT-IR and FT-Raman spectra have been recorded of Trimesic acid (1,3,5-benzenetricarboxylic acid, H3BTC). The molecular structure, conformational stability, geometry optimization, vibrational frequencies have been investigated. The total energy calculations of H3BTC were tried for various possible conformers. The spectra were interpreted with the aid of normal coordinate analysis based on ab initio Hartree-Fock (HF) and density functional theory (DFT/B3LYP) methods and 6-31+G(d,p) basis set level and was scaled using scale factors yielding good agreement between observed and calculated freq

Metabolic Regulation of Inflammation: Exploring the Potential Benefits of Itaconate in Autoimmune Disorders.

Itaconic acid and its metabolites have demonstrated significant therapeutic potential in various immune diseases. Originating from the tricarboxylic acid cycle in immune cells, itaconic acid can modulate immune responses, diminish inflammation, and combat oxidative stress. Recent research has uncovered multiple mechanisms through which itaconic acid exerts its effects, including the inhibition of inflammatory cytokine production, activation of anti-inflammatory pathways, and modulation of immune cell function by regulating cellular metabolism. Cellular actions are influenced by the modulation

Metabolomic Profiling in the Characterization of Degenerative Bone and Joint Diseases.

Osteoarthritis and inflammatory arthropathies are a cause of significant morbidity globally. New research elucidating the metabolic derangements associated with a variety of bone and joint disorders implicates various local and systemic metabolites, which further elucidate the underlying molecular mechanisms associated with these destructive disease processes. In osteoarthritis, atty acid metabolism has been implicated in disease development, both locally and systemically. Several series of rheumatoid arthritis patients have demonstrated overlapping trends related to histidine and glyceric aci

[Characteristics of carbohydrate metabolism in the rat liver in thiamine deficiency].

Thiamine deficiency in rats induced by oxythiamine is accompanied by an increase in the free NADP+/NADPH ratio in liver tissue, which results in multifold stimulation of the metabolite flux in the oxidation branch of the pentose cycle. The increase in the intracellular concentrations of isocitrate and alpha-ketoglutarate with a simultaneous decrease of malate in the liver of vitamin-deficient rats points to the inhibition of alpha-ketoglutarate dehydrogenase responsible for the anomalous metabolism under conditions of thiamine deficiency. The decrease of the functional activity of the tricarbo

Enhancing catalytic activity and pore structure of metal-organic framework-808 via ligand competition for biodiesel production from microalgal lipids at reduced temperatures.

Catalysts with hierarchical porous structures and increased active defects play a crucial role in catalyzing the conversion of microalgae lipids. However, the template methods used for pore expansion and the acidification process employed to enhance activity are cumbersome and prone to deactivation. It is necessary to propose a simple and versatile synthetic approach to overcome these challenges. By modulating N,N-dimethylformamide basicity with formic acid, MOF-808 exhibited enhanced coordination of benzene-1,3,5-tricarboxylic acid to Zr-clusters, creating three types of functional defects. Pneumococcal H₂O₂ reshapes mitochondrial function and reprograms host cell metabolism.

Streptococcus pneumoniae (Spn), a primary cause of pneumonia, induces acute lung parenchymal damage through a unique metabolic pathway generating hydrogen peroxide (H₂O₂) as a byproduct. This study demonstrates that Spn-derived H₂O₂, primarily produced by pyruvate oxidase (SpxB), inhibits key tricarboxylic acid (TCA) cycle enzymes (aconitase, glutamate dehydrogenase, and α-ketoglutarate dehydrogenase) in lung epithelial cells, leading to citrate accumulation and diminished NADH production for oxidative phosphorylation. RNA sequencing reveals SpxB-dependent upregulation of glycolytic genes (HIF

When copper turns killer: Decoding copper dyshomeostasis and cuproptosis in neurodegenerative pathogenesis and precision metal interventions.

Copper is an essential cofactor for neuronal metabolism, enzymatic functions, and neurotransmission. However, copper dyshomeostasis-induced redox activity makes the brain vulnerable to oxidative and proteostatic stress. Cuproptosis, a recently characterized form of programmed cell death, is triggered by copper binding to lipoylated enzymes of the tricarboxylic acid cycle, resulting in proteotoxic stress, mitochondrial dysfunction, and cell death. Given that mitochondria are central to copper handling and the primary site of cuproptosis, we examine mitochondrial pathways and key cuproptosis-rel

SUCNR1 coordinates metabolic flux, mitochondrial function, and nutrient-dependent adaptation in hepatocytes.

Succinate, a mitochondrial metabolite, also functions as an extracellular signal through its receptor succinate receptor 1 (SUCNR1), coordinating responses to nutrient availability. The physiological role of SUCNR1 within hepatocytes, however, is unclear. We show that hepatic succinate levels and Sucnr1 expression are dynamically regulated by nutritional status. Mice lacking Sucnr1 in hepatocytes [Hep-Sucnr1 knockout (KO)] exhibit a fasting-like phenotype characterized by enhanced gluconeogenesis, elevated amino acids, and impaired metabolic flexibility. Mechanistically, loss of Sucnr1 comprom

α-Ketoglutarate dehydrogenase is a therapeutic vulnerability in acute myeloid leukemia.

Perturbations in intermediary metabolism contribute to the pathogenesis of acute myeloid leukemia (AML) and can produce therapeutically actionable dependencies. Here, we probed whether α-ketoglutarate (αKG) metabolism represents a specific vulnerability in AML. Using functional genomics, metabolomics, and mouse models, we identified the αKG dehydrogenase complex, which catalyzes the conversion of αKG to succinyl coenzyme A, as a molecular dependency across multiple models of adverse-risk AML. Inhibition of 2-oxoglutarate dehydrogenase (OGDH), the E1 subunit of the αKG dehydrogenase complex, im

Systemic metabolite kinetics mirror skeletal muscle energy metabolism during acute aerobic exercise.

Acute exercise increases energy demand in skeletal muscle and releases metabolic intermediates into circulation, yet the serum kinetics of exercise-mobilized metabolites remain poorly characterized. By applying high-frequency serial blood sampling and targeted metabolomics in a longitudinal exercise trial with 12 young, healthy adults (6 females, 6 males), we assessed temporal alterations in energy-related metabolites during acute aerobic exercise and after 1 h of recovery. We provide evidence for 42 exercise-responsive metabolites, including end products of glycolysis, tricarboxylic acid cycl

Comparative Metabolomics of Clostridium acetobutylicum ATCC824 and its Engineered Strain, C. acetobutylicum DG1.

Clostridium acetobutylicum, a strict gram-positive anaerobe, plays a pivotal role in biotechnological applications, particularly in the biosynthesis of 1,3-propanediol, a critical biofuel component and monomer for bioplastic production. This study introduces C. acetobutylicum DG1, a metabolically engineered strain designed to enhance the 1,3-propanediol pathway. Despite its development, comprehensive metabolic comparisons between the parent and modified strains remain unexplored. Our research addresses this gap by employing gas chromatography coupled with time-of-flight mass spectrometry to de

Effect of Symmetry and Increasing Hydrophobicity on the Self-Assembly and Function of Benzoylurea Derivatives.

A novel series of benzoylurea derivatives containing benzoic acid, m-dibenzoic acid, and benzene 1,3,5-tricarboxylic acid were designed with increasing hydrophobicity. The aggregation behavior of the derivatives was studied by several spectroscopic methods. The porous morphology of the resulting aggregates was examined by polar optical microscopy and field emission scanning electron microscopy. From X-ray single-crystal analysis, it is observed that N,N'-dicyclohexylurea containing compound 3 lost C3 symmetry and adopted a "bowl"-shaped conformation and self-assembles to form a supramolecular
