Burkholderia mallei cluster 1 type VI secretion mutants exhibit growth and actin polymerization defects in RAW 264.7 murine macrophages.

Burkholderia mallei is a facultative intracellular pathogen that causes severe disease in animals and humans. Recent studies have shown that the cluster 1 type VI secretion system (T6SS-1) expressed by this organism is essential for survival in a hamster model of glanders. To better understand the role of T6SS-1 in the pathogenesis of disease, studies were initiated to examine the interactions of B. mallei tssE mutants with RAW 264.7 murine macrophages. Results obtained by utilizing modified gentamicin protection assays indicated that although the tssE mutants were able to survive within RAW 2

Proteomic Evidence for Biofilm-Mediated Copper Stress Adaptation in Oleidesulfovibrio alaskensis G20.

Copper (Cu) is an essential cofactor for enzymes regulating key metabolic processes; however, excess Cu can inhibit bacterial growth. The ability of Oleidesulfovibrio alaskensis G20 (OA G20) to survive under heavy metal stress is compelling. Our previous study on OA G20 exposed to Cu suggests that bacteria might use biofilm formation to adapt to high levels of toxic metal ions. To elucidate the molecular mechanisms underlying this adaptation, we performed a comparative proteomic analysis of OA G20 stress-induced biofilms (30 μM Cu) versus control (no Cu) and their respective extracellular frac

Avian pathogenic Escherichia coli virulence protein Hcp2a induces incomplete autophagy in chicken HD11 cells.

The hemolysin co-regulator protein (Hcp) is a core virulence protein of the Type VI Secretion System (T6SS) in avian pathogenic Escherichia coli (APEC) and plays a critical role in host-pathogen interactions. While several bacterial effectors are known to subvert host immunity by modulating autophagy, whether and how the APEC Hcp2a protein influences autophagy in chicken macrophages (HD11) remains unclear. Here, we demonstrate that Hcp2a is efficiently internalized into HD11 cells and induces significant cytotoxicity. Notably, Hcp2a treatment elicited an autophagic response, as evidenced by el

Composition of microbiota from chicken caecal mucosa considerably differs from microbiota in the caecal lumen.

BACKGROUND: Despite the importance of microbiota at mucosal surfaces of the gut and their different composition in comparison to those colonising the gut lumen, only a few papers specifically addressed this topic. In this study, we therefore defined mucosa-associated and lumen-associated microbiota from the chicken caecum. RESULTS: To identify mucosa-adapted microbiota members, caeca from adult hens instead of chicks should be analysed. Mucosa-adapted microbiota included Treponema-like species, Brachyspira innocens, Mucispirillum sp., Helicobacter pullorum, Desulfovibrio sp. and Anaerobiospir

Copper is an intestinal habitat filter affecting the gut microbiota interactions with Salmonella Typhimurium.

BACKGROUND: Foodborne pathogens, including Salmonella enterica serovar Typhimurium (S. Typhimurium), pose a significant threat to both human health and livestock productivity. The pandemic S. Typhimurium ST34 clone acquired a genomic island (SGI-4) conferring high copper resistance, an adaptation relevant in the context of the widespread use of copper sulphate at therapeutic levels in pig farming. We investigated how high dietary copper influences the piglet gut microbiota and Salmonella-microbiota interactions that may explain the global spread of S. Typhimurium ST34. RESULTS: An on-farm stu

Glycosyltransferase-Like Toxin of Burkholderia gladioli Strain NGJ1 Is a Potent Antifungal Protein with Potential for Control of Sheath Blight Disease in Rice.

Sheath blight disease caused by the fungal pathogen Rhizoctonia solani poses a significant challenge for sustainable rice cultivation. It is important to develop environmentally friendly measures for its control. Previously, a rice-associated Burkholderia gladioli strain NGJ1 was shown to exhibit mycophagous and antifungal activity on R. solani. Here, we report that a B. gladioli glycosyltransferase-like 1 (BGT1) protein with a canonical D×D (aspartic acid × aspartic acid) motif that is homologous to the glycosyltransferase toxin of different bacteria is encoded in the antibacterial type VI se

Design, Synthesis, and Antibacterial Activity Evaluation and Mechanism of 1-H-Indole-3-pyrazolamide Derivatives Containing Piperazine.

To develop antibacterial agents with novel mechanisms of action, a series of novel 3-pyrazolylindole derivatives incorporating piperazine moieties were designed and synthesized. The in vitro antibacterial activities of these compounds were evaluated against four phytopathogenic bacterial strains: Xanthomonas oryzae pv oryzae (Xoo), Xanthomonas axonopodis pv citri (Xac), Pseudomonas syringae pv actinidiae (Psa), and Ralstonia solanacearum (Rs). All of the synthesized derivatives exhibited moderate antibacterial activity. In particular, compound P16 demonstrated potent inhibitory activity agains

Genomic Analysis and Virulence Features of Vibrio cholerae Non-O1/Non-O139 Harbouring CARB-Type β-Lactamases From Freshwater Bodies, Argentina.

Vibrio cholerae is a globally distributed, free-living bacterium in aquatic ecosystems. While non-O1/non-O139 serogroups typically do not produce cholera toxin, they have the potential to cause diarrhoea. These strains may act as reservoirs of antibiotic resistance in rivers, lakes and oceans. Understanding their genetic resistance and virulence can shed light on their role in spreading antimicrobial resistance and their pathogenicity. In this study, we characterised 60 V. cholerae non-O1/non-O139 strains from 16 freshwater bodies located throughout the Province of Córdoba, Argentina. We found

Protozoal food vacuoles enhance transformation in Vibrio cholerae through SOS-regulated DNA integration.

Vibrio cholerae, the bacterial pathogen responsible for the diarrheal disease cholera, resides in the aquatic environment between outbreaks. For bacteria, genetic variation by lateral gene transfer (LGT) is important for survival and adaptation. In the aquatic environment, V. cholerae is predominantly found in biofilms associated with chitinous organisms or with chitin "rain". Chitin induces competency in V. cholerae, which can lead to LGT. In the environment, V. cholerae is also subjected to predation pressure by protist. Here we investigated whether protozoal predation affected LGT using the

Microfibrils: a cornerstone of extracellular matrix and a key to understand Marfan syndrome.

The extracellular matrix is made of collagen, reticular, elastic and oxytalan fibers, amorphous ground substance and adhesive proteins playing a structural role, such as fibronectin; the basement membrane is a specialized matrix compartment which adheres to non-connective tissues and is continuous with the remaining matrix thanks to reticular fibers, anchoring fibrils, collagen VI filaments and oxytalan fibers. Microfibrils are constituents of elastic and oxytalan fibers that confer mechanical stability and limited elasticity to tissues, contribute to growth factor regulation, and play a role

Deletion of cysteine 369 in lysyl hydroxylase 1 eliminates enzyme activity and causes Ehlers-Danlos syndrome type VI.

This study describes the relative contribution of the 10 cysteine residues in lysyl hydroxylase 1 (LH1) to enzyme activity. We have identified a novel mutation of a 15-bp deletion in exon 11 in one LH1 allele, that codes for amino acids 367-371 (DLCRQ), in two unrelated compound heterozygous patients with Ehlers-Danlos type VI. The mutations in their other alleles were a C1119T change (exon 10) and a predicted Q49X (exon 2). We confirmed that the loss of cysteine 369 in the deleted sequence contributed to the diminished enzyme activity by structure/function analysis of mutant LH1 constructs, i

The gut microbiota in Salmonella Typhi infection and translocation: Mechanisms of colonization resistance, pathogen subversion, and prospects for microecological intervention.

The systemic establishment of the strictly human-adapted pathogen Salmonella Typhi critically depends on functional interplay between its virulence effectors and the host gut microbiome. Organized within a "colonization-toxicity-immunity- microecological intervention" framework, this review synthesizes recent molecular and multi-omics evidence to delineate key host-pathogen-microbiota dynamics. During colonization, a healthy gut microbiota exerts multi-layered colonization resistance through nutrient and niche competition, as well as via microbial metabolites such as short-chain fatty acids an

The Madagascar hissing cockroach as a novel surrogate host for Burkholderia pseudomallei, B. mallei and B. thailandensis.

BACKGROUND: Burkholderia pseudomallei and Burkholderia mallei are gram-negative pathogens responsible for the diseases melioidosis and glanders, respectively. Both species cause disease in humans and animals and have been designated as category B select agents by the Centers for Disease Control and Prevention (CDC). Burkholderia thailandensis is a closely related bacterium that is generally considered avirulent for humans. While it can cause disease in rodents, the B. thailandensis 50% lethal dose (LD50) is typically ≥ 104-fold higher than the B. pseudomallei and B. mallei LD50 in mammalian mo

[Therapy for systemic metabolic disorders based on the detection of basic corneal landmarks in childhood].

Many systemic lysosomal storage disorders show basic corneal opacities already in childhood. The lysosome is a cell organelle, produced by Golgi's apparatus, that is surrounded by a membrane and contains hydrolytic enzymes that break down food molecules, especially proteins and other complex molecules. The ophthalmologist's precise diagnosis of corneal clouding at the slit-lamp may reveal the correct interpretation of the specific lysosomal storage disorder. It is very important to diagnose such diseases as soon as possible because today the development of systemic enzymatic therapies has broa

Spatiotemporal coordination of virulence, metabolism, and stress responses shapes infection dynamics of Xanthomonas perforans.

Plants host diverse microbial communities across spatially distinct foliar niches. For hemibiotrophic pathogens, the epiphytic leaf surface and the apoplast impose contrasting physiological constraints that shape infection strategies. We investigated how these environments program the transcriptional responses of a tomato pathogen, Xanthomonas perforans (Xp), and how virulence, metabolism, and regulation are coordinated across space and time during disease progression. By selectively enriching and profiling the transcriptome of a pathogen from the leaf surface and apoplast, we uncovered pronou

Identification and differential expression analysis of Hfq-bound novel small RNAs in Serratia marcescens by nitrocellulose-based partitioning.

BACKGROUND: The rising prevalence of multidrug-resistant bacteria poses a looming threat to healthcare and necessitates novel therapeutic strategies beyond conventional antibiotics. One such organism is Serratia marcescens, which possesses a broad range of virulence factors and has been listed as a high-priority pathogen by the WHO due to its increasing antibiotic resistance. Small RNAs are promising targets against antibiotic-resistant bacteria, as they are key regulators of bacterial gene expression. This study aimed to identify novel sRNA and elucidate their roles in regulating virulence-as

Molecular epidemiology of Campylobacter jejuni associated with Guillain-Barré syndrome.

Campylobacter jejuni is the leading cause of bacterial gastroenteritis and its post-infectious sequelae, Guillain-Barré syndrome (GBS). Disease epidemiology of campylobacteriosis differs between high- and low-income countries. The molecular epidemiology of GBS-associated C. jejuni strains has not been thoroughly studied to understand its population dynamics and unique genomic features. We investigated a genome-based epidemiological analysis of 90 GBS-associated C. jejuni strains from 10 different countries to elucidate bacterial genomic features and epidemiological patterns related to GBS indu

Transcriptional profiles of Burkholderia pseudomallei reveal the direct and indirect roles of Sigma E under oxidative stress conditions.

BACKGROUND: Burkholderia pseudomallei, the causative agent of melioidosis, is a Gram-negative bacterium widely distributed in soil and water in endemic areas. This soil saprophyte can survive harsh environmental conditions, even in soils where herbicides (containing superoxide generators) are abundant. Sigma factor E (σE) is a key regulator of extra-cytoplasmic stress response in Gram-negative bacteria. In this study, we identified the B. pseudomallei σE regulon and characterized the indirect role that σE plays in the regulation of spermidine, contributing to the successful survival of B. pseu
