The chaperonin CCTα is required for efficient transcription and replication of rabies virus.

Negri bodies (NBs) are formed in the cytoplasm of rabies virus (RABV)-infected cells and are accompanied by a number of host factors to NBs, in which replication and transcription occur. Here, it was found that chaperonin containing TCP-1 subunit alpha (CCTα) relocalizes to NBs in RABV-infected cells, and that cotransfection of nucleo- and phospho-proteins of RABV is sufficient to recruit CCTα to the NBs' structure. Inhibition of CCTα expression by specific short hairpin RNA knockdown inhibited the replication and transcription of RABV. Therefore, this study showed that the host factor CCTα is

The RNA polymerase of influenza a virus: mechanisms of viral transcription and replication.

The influenza A virus RNA genome segments are packaged in ribonucleoprotein complexes containing RNA polymerase and nucleoprotein. The ribonucleoprotein is involved in the transcription of viral genes and replication of the viral RNA genome in the nucleus of the infected cells, and represents the minimal transcriptional and replicative machinery of an influenza virus. During transcription, the viral RNA polymerase synthesizes capped and polyadenylated mRNA using 5΄ capped RNA primers. During replication, the viral RNA polymerase generates a complementary RNA (cRNA) replication intermediate, a

Nigericin is a potent inhibitor of the early stage of vaccinia virus replication.

Poxviruses remain a significant public health concern due to their potential use as bioterrorist agents and the spread of animal borne poxviruses, such as monkeypox virus, to humans. Thus, the identification of small molecule inhibitors of poxvirus replication is warranted. Vaccinia virus is the prototypic member of the Orthopoxvirus genus, which also includes variola and monkeypox virus. In this study, we demonstrate that the carboxylic ionophore nigericin is a potent inhibitor of vaccinia virus replication in several human cell lines. In HeLa cells, we found that the 50% inhibitory concentra

Infection and replication of avian influenza H5N1 virus in an infected human.

The highly pathogenic avian influenza H5N1 viruses usually cause severe diseases and high mortality in infected humans. However, the tissue tropism and underlying pathogenesis of H5N1 virus infection in humans have not been clearly elucidated yet. In this study, an autopsy was conducted to better understand H5N1 virus distributions in tissues of infected humans, and whether H5N1 virus can replicate in extrapulmonary tissues. We found that the lungs had the higher viral load than the spleen, whereas no detectable viruses in tissues of heart, liver, kidney, large intestine, small intestine, or b

Synergistic combination effect of cidofovir and idoxuridine on vaccinia virus replication.

In view of the potential menace of a terrorism attack with smallpox virus, an intensive search of chemotherapeutic agents active against orthopoxviruses is underway. We comparatively studied the antiviral activity of cidofovir (CDV) and idoxuridine (IUdR) against two vaccinia virus (VV) strains, Bratislava and RIIPD, in cell cultures of chick embryo fibroblasts (CEF). The investigations were carried out according to cytopathic effect (CPE) inhibition assay protocols. To determine the cytotoxicity of the compounds, maximal tolerated concentration (MTC) was calculated in CEF cell monolayers and

Inhibitory effect of furanonaphthoquinone derivatives on the replication of Japanese encephalitis virus.

Japanese encephalitis still occurs in endemic and epidemic forms over a wide area of Asia. Although the vaccine against Japanese encephalitis virus (JEV) is widely used, no antiviral drug has been reported. We used several different kinds of furanonaphthoquinone derivatives and found antiviral activity against JEV. Especially, 2-methylnaphtho[2,3-b]furan-4,9-dione (FNQ3) indicated the highest antiviral activity, followed by 2-(1-hydroxyethyl)-, 5(or 8)-hydroxy-, and 2-methyl-5(or 8)-hydroxy-analogs of naphtho[2,3-b]furan-4,9-dione. In the presence of 3 microg/ml FNQ3, the virus yields in Vero

In vitro activity of acetylsalicylic acid on replication of varicella-zoster virus.

Topical application of a mixture of acetylsalicylic acid (ASA) and diethyl ether is effective in the treatment of acute herpes zoster and postherpetic neuralgia. To study whether the other-than-analgesic effects of that treatment could be due to an antiviral activity of ASA the effects of the drug on the replication of varicella zoster virus (VZV) were assessed by the fluorescent focus assay on MRC5 and Vero cells. ASA caused a marked reduction in the spread of infection in MRC5 monolayers while in growing Vero cells the effective dose proved toxic. ASA concentrations (5-10 mM) which were effe

Entry and replication of Japanese encephalitis virus in cultured neurogenic cells.

The entry mode and growth pattern of Japanese encephalitis (JE) virus in mouse neuroblastoma N18TG2 cells and mouse neuroblastoma x rat glioma NG108-15 hybrid cells were studied by electron microscopy. At two minutes after inoculation, JE virions adsorbed onto and directly penetrated through the plasma membrane of the hybrid cells, whereas virions did not adsorb nor entered the neuroblastoma cells. Correspondingly, the hybrid cells showed assembling progeny JE virions in the cisternae of rough endoplasmic reticulum (RER) 1 day postinoculation (p.i.) although virions were rarely found on the fo

Terminal sequences of the replicative form of RNA of the Japanese encephalitis virus.

The 5' and 3' terminal sequences of the replicative form (RF) of RNA of a flavivirus, the Japanese encephalitis (JE) virus, strain Ja0ArS982, have been determined by in vitro labelling and mobility shift analysis. The plus strand sequence was 5'AGAAGUUUAUCUGUGUGAA...UCUOH3', while the minus strand sequence was 5'AGAUCCUGUGUUCUUCCUCA...UCUOH3'. These sequences were similar to those of West Nile (WN) virus being identical in 12 nucleotides at the 5'terminal of the minus strand, and in the 5'terminal dinucleotides, 5'AG3'. Somewhat more internal hexanucleotides 5'CUGUGU3' are conserved among 3 fl

Replication and dissemination of Rift Valley fever virus in Culex pipiens.

Following ingestion of 10(4.2) to 10(7.2) plaque-forming units (PFU) of Rift Valley fever (RVF) virus, 662 of 850 female Culex pipiens (78%) became infected. Those mosquitoes that became infected separated into two distinct groups: 1) those with a nondisseminated infection limited to the gut, and 2) those with a disseminated infection. The former group contained a median of 10(3.2) PFU, while those females with a disseminated infection contained a median of 10(5.5) PFU. Only those females with a disseminated infection were capable of transmitting virus by bite to a susceptible hamster. This is

Characterization of a non-syncytiogenic autonomously replicating variant of measles virus.

A variant, derived from both measles and subacute sclerosing panencephalitis virus preparations, incapable of producing intercellular fusion or syncytia typical of measles virus infections is described. This non-syncytiogenic (N-S) virus was easily detected using immunofluorescence, was genetically stable and replicated autonomously. The variant was defective in the production of both infectious virus and haemagglutinin in Vero and Hep2 cells. Persistent infections in Hep2 cells were readily established without the cooperation of defective interfering particles or temperature-sensitive mutants

Block to influenza virus replication in cells preirradiated with ultraviolet light.

Ultraviolet (uv) irradiation of CEF cells immediately before infection with influenza A (fowl plague) virus inhibited virus growth; no inhibition of the growth of a parainfluenza virus (Newcastle disease virus) could be detected in irradiated cells. The kinetics of inhibition after various doses of uv irradiation were multihit, with an extrapolation number of two. When irradiated cells were allowed to photoreactivate by exposure to visible light for 16 hr their capacity to support influenza virus replication was largely restored; this process was sensitive to caffeine, suggesting that it requi

Interferon gamma-induced MX1 and IFITM1 inhibit Varicella-Zoster Virus Replication.

UNLABELLED: Interferon gamma (IFN-γ) is a potent cytokine secreted following primary varicella-zoster virus (VZV) infection. Our previous studies demonstrated that pretreatment with IFN-γ completely inhibited VZV replication in lung fibroblast MRC-5 and retinal epithelial ARPE-19 but not in melanoma MeWo cells, suggesting that IFN-γ-stimulated protein(s) inhibit viral replication. Our microarray analysis revealed that 34 interferon-stimulated genes (ISGs) were upregulated by greater than 2.5-fold at 8 h post-treatment in both ARPE-19 and MRC-5 cells compared to those of MeWo cells. The depleti

Acyclic Nucleoside Phosphonate Prodrug LAVR-289 Inhibits Marek's Disease Virus Replication.

Since the 1970s, numerous antivirals targeting human herpesviruses have been developed, yet their application in veterinary medicine remains limited due to poor bioavailability and significant toxicity in animals. Recently, LAVR-289, a novel acyclic nucleoside phosphonate analogue, has demonstrated broad-spectrum antiviral activity against various DNA viruses. In the present study, we investigated the antiviral potential of LAVR-289 against Marek's disease virus (MDV), an avian herpesvirus responsible for a fatal lymphoproliferative disease in chickens and substantial economic losses to the po

Mechanistic insights into flavonoid-mediated regulation of human influenza virus replication.

Influenza A virus (IAV) remains a major global health concern due to its rapid mutation rate, antigenic variability, and the emergence of resistance to frontline antivirals such as oseltamivir and zanamivir. Despite available vaccines and therapeutics, their limited efficacy against emerging strains underscores the urgent need for new antiviral approaches. Therefore, plant-derived bioactive compounds, particularly flavonoids, have gained attention for their ability to target both viral and host pathways, yet their mechanisms of action remain incompletely understood. This review synthesises cur

Nucleotide analogue bemnifosbuvir inhibits hepatitis E virus replication in preclinical models.

BACKGROUND: Hepatitis E virus (HEV) infections remain a global health concern. Immunocompromised patients are at an increased risk of developing chronic HEV infection and thereby severe liver disease. Current off-label regimens are suboptimal with treatment failure being reported. Therefore, there is an urgent need for an effective anti-HEV treatment. OBJECTIVE: In this study, we aimed to identify potent inhibitors of HEV replication. DESIGN: We developed a rapid, image-based screening platform based on a full-length HEV fluorescence reporter virus and screened a nucleotide/nucleoside analog

Cellular lipids: fundamental host factors for monkeypox virus replication and pathogenesis.

The recent outbreaks of mpox and other poxvirus diseases worldwide have highlighted the critical need for effective antiviral treatments. Throughout its life cycle, MPXV, a member of the Orthopoxvirus genus, takes advantage of the host’s lipid metabolism. In addition to being essential for maintaining cellular homeostasis, lipids are also used by viruses for entrance, reproduction, assembly, and egress. Lipid rafts (LRs), membrane microdomains rich in cholesterol that are essential for viral entry and viral envelope structure, are a crucial component of this exploitation. This study summarizes

Garcinone C inhibits pseudorabies virus replication through EGF/PI3K/Akt axis.

INTRODUCTION: Pseudorabies virus (PRV), a significant pathogen of swine, causes substantial economic losses, and even poses an emergent public health concern due to its zoonotic potential. The continuous evolution of PRV has undermined the effectiveness of current vaccines and antiviral drugs. Consequently, there is an urgent need to develop novel strategies to curb its spread. METHODS: The inhibitory effect of garcinone C on PRV replication was assessed in vivo and in vitro. To determine the stage of antiviral action, treatments were administered at different time points: pre-treatment, co-t

Umbelliferone Inhibits Zika Virus (ZIKV) NS5 Activity and Replication In Vitro.

This study focused on the inhibitory effects of umbelliferone on Zika virus (ZIKV) NS5 activity and viral replication in vitro. Umbelliferone exhibited no detectable cytotoxicity and inhibited ZIKV replication while showing no activity against Japanese encephalitis virus (JEV), another member of the Flaviviridae family. Structure-activity relationship studies on structurally related derivatives of umbelliferone revealed that the hydroxyl group at the C7 position is critical for suppression of ZIKV replication. To identify the stage of the ZIKV life cycle targeted by umbelliferone, we performed

Brefeldin A inhibits duck plague virus replication by impairing virion envelopment.

Duck plague virus (DPV) is the causative agent of duck plague (DP), an acute and highly fatal disease that inflicts substantial economic losses in the waterfowl industry. The virus acquires its envelope from the endoplasmic reticulum (ER) and Golgi apparatus during virion assembly. Currently, there are no specific antiviral drugs available, making the development of novel anti-DPV drugs critically important. Brefeldin A (BFA) is an antibiotic known to disrupt endoplasmic reticulum-Golgi transport and inhibit the replication of various enveloped viruses. However, its inhibitory effect and mecha
