Natural product scaffold-based bactericide discovery: novel β-carboline derivatives as potent reactive oxygen species inducers for crop disease control.

BACKGROUND: Natural β-carboline alkaloids have emerged as one of the most promising scaffolds in anticancer and fungicidal agent discovery. However, their application in antiphytopathogenic bacteria remains underexplored, primarily due to challenges in synthetic accessibility and insufficiently elucidated mechanisms of action. This gap has created a significant bottleneck in translating these natural products into practical agricultural applications. RESULTS: Basing on the natural β-carboline (βC) scaffold, 41 synthetic derivatives and six commercialized analogues were obtained to discuss the

A novel Zn chelate (TSOL) that moves systemically in citrus plants inhibits growth and biofilm formation of bacterial pathogens.

Ternary solution (TSOL) is a novel Zn chelate-based systemic antimicrobial formulation designed for treating citrus bacterial pathogens 'Candidatus Liberibacter asiaticus' and Xanthomonas citri subsp. citri. TSOL is a component of MS3T, a novel multifunctional surface/sub-surface/systemic therapeutic formulation. Antimicrobial activity of TSOL was compared with the antimicrobial compound ZnO against X. citri subsp. citri and 'Ca. L. asiaticus' surrogate Liberibacter crescens in batch cultures. X. citri subsp. citri and L. crescens were also introduced into microfluidic chambers, and the inhibi

Acylpiperidine Formononetin Derivatives as Antimicrobial Lead Compounds: Activity and Preliminary Mode-of-Action Studies.

Global agricultural production relies heavily on chemical pesticides, leading to resistance, residues, and pollution, threatening sustainable agriculture. There is an urgent need to develop novel antimicrobial agents from low-toxicity, easily degradable natural products. Using formononetin as a lead compound, acylpiperidine-containing derivatives were synthesized and evaluated against one oomycete, seven plant pathogenic fungi, and four pathogenic bacteria. C25 exhibited superior inhibitory effects against Phytophthora capsici and Xanthomonas oryzaepv.oryzae, with EC50 values of 3.38 and 1.67

Discovery of DNA Gyrase as a Promising Target against Phytopathogenic Bacteria.

The growing resistance of phytopathogenic bacteria highlights the need for new bactericides. DNA gyrase, essential in bacteria but absent in plants and animals, is an underexploited target for agricultural bactericides. Here, we modeled the Ralstonia solanacearum GyrA-GyrB-DNA complex using AlphaFold3 and conducted structure-based virtual screening of 1.27 million compounds. Among 100 tested candidates, GI-A14 showed potent, broad-spectrum antibacterial activity against R. solanacearum, Pseudomonas syringae pv tomato, and Xanthomonas oryzae pv oryzae, with minimum inhibitory concentrations of

Integrative analysis of sulfate transporters in Citrus sinensis reveals CsSULTR3;5 involved in pathogen stress response.

Selenium treatment has been shown to enhance plant resilience against both biotic and abiotic stresses; however, whether it can improve citrus resistance to bacterial diseases remains unexplored. In this study, we systematically identified sulfate transporter (Sultr) genes in the Citrus sinensis genome that are involved in selenate uptake. Analysis of gene structure, conserved motifs, and protein domains revealed a high degree of conservation among CsSultrs. Collinearity analysis between C. sinensis and Arabidopsis thaliana Sultr genes suggested possible evolutionary duplication and divergence

Increased risks of antibiotic resistant genes (ARGs) induced by chlorine disinfection in the reverse osmosis system for potable reuse of reclaimed water.

The reverse osmosis (RO) process has been applied extensively in wastewater reclamation for industrial and potable reuse. To prevent biofouling, chlorine disinfection was usually used in pretreatment. However, this study found that chlorine disinfection could significantly increase risks of antibiotic resistant genes (ARGs) in the RO system. With the increase of chlorine concentration from 0 to 5 mg/L, the accumulative relative abundance of 14 common ARGs in the membrane foulants increased by 49.6%. Among these ARGs, tolC, acrA and acrB (resistance to multiple drugs) showed the highest incream

First Discovery of Novel Pyrido[1,2-a]pyrimidinone Mesoionic Compounds as Antibacterial Agents.

Plant bacterial diseases cause tremendous decreases in crop yield and quality, and there is a lack of highly effective and low-risk antibacterial agents. A series of novel pyrido[1,2-a]pyrimidinone mesoionic compounds containing vanillin moieties were synthesized, and the application of these mesoionic compounds as plant antibacterial agents was reported here for the first time. The bioassay results revealed that the mesoionic compounds had good antibacterial activity. Of these compounds, compound 11 showed excellent in vitro activity against Xanthomonas oryzae pv. oryzae, with an EC50 value o

Antimicrobial resistance in the intensive care unit.

Antimicrobial resistance remains a major clinical problem in the intensive care unit despite the introduction of potent new antibiotics and the application of infection control measures. Impaired antibiotic permeability, antibiotic inactivation and alteration of antibiotic target sites, typically in concert, are mechanisms by which this resistance occurs. Resistance traits may be chromosomally mediated, in which case they may be expressed constitutively or inducibly, or may be mediated by plasmids and/or transposons that may confer resistance to multiple drugs and facilitate spread to other or

Development of a xanthan gum film for the possible treatment of vaginal infections.

Bacterial vaginosis is a vaginal infection that affects 60% of women of reproductive age worldwide. It is mainly caused by the bacterium Gardnerella vaginalis and is a factor that increases the probability of getting sexually transmitted diseases. We aimed to develop a new pharmaceutical form for the treatment of vaginal infections. We employed the solving-casting method to fabricate a polymeric film with Xanthan gum, a natural polymer produced by the bacterium Xanthomonas campestris, and metronidazole, one of the most commonly used drugs for vaginal infections. In order to characterize the fi

Thermotherapy Treatments of Fruit Trees of the Genus Prunus Infected by Viruses and Bacteria.

The thermotherapy technique has been used to evaluate the viability of hot water treatments (HWTs) for the elimination of different pathogens that infect commercial fruit trees. The present project focuses on common pathogens of Prunus species at the nursery level when external propagation material is needed, which is not always in optimal sanitary conditions. Different combinations of temperature/duration treatments were applied to budwood of nectarines infected by Xanthomonas arboricola pv. pruni (Xap) and almonds infected by Prunus necrotic ringspot virus (PNRSV). These treated buds were th

Improved Biological Control of Bacterial Leaf Blight Using a Surfactant Complex of CO2 Micro-Nanobubbles Coated with Crude Ethyl Acetate Extract of Trichoderma polyalthiae UBZSN2-1.

The bacterium Xanthomonas oryzae pv. oryzae is an important pathogen that causes wilt leaf blight disease in rice (Oryza sativa L.), leading to a reduction in rice yield. Therefore, this study aimed to investigate the potential of a surfactant complex composed of CO2 nanobubbles (CO2-NBs) coated with sorbitan monostearate (Sp60) and a crude extract of Trichoderma polyalthiae as active ingredient delivery agents for controlling leaf blight under both laboratory and greenhouse conditions. The addition of Sp60 and crude extract as surfactants significantly influenced the size uniformity and stabi

cDNA-AFLP analysis of differential gene expression in the prokaryotic plant pathogen Erwinia carotovora.

For studies of differential gene expression in prokaryotes, methods for synthesizing representative cDNA populations are required. Here, a technique is described for the synthesis of cDNA from the potato pathogens Erwinia carotovora subsp. atroseptica (Eca) and Erwinia carotovora subsp. carotovora (Ecc) using a combination of short oligonucleotide (11-mer) primers that were known to anneal to conserved sequences in the 3' regions of enterobacterial genes. Specific PCR amplifications with primers designed to anneal to 14 known genes from either Eca or Ecc revealed the presence of the correspond

Phytomelatonin receptor PMTR1 is crucial for melatonin-mediated plant broad-spectrum disease resistance.

PMTR1 is essential for melatonin-improved plant broad-spectrum disease resistance to bacterial pathogens and viruses. Melatonin plays vital roles in various plant stress responses, and the identification of phytomelatonin receptor 1 (PMTR1) in recent decades further extends the phytomelatonin signaling pathway. However, whether and how PMTR1 participates in plant broad-spectrum disease resistance has remained unclear. Here, we confirmed that exogenous melatonin enhanced disease resistance to bacterial blight and mosaic disease, indicating the crucial role of melatonin in cassava broad-spectrum
