Meningioma microenvironment harbors a rich immune landscape that evolves with biological state.

BACKGROUND: Mechanisms driving aggressive meningiomas remain poorly understood. Given the pivotal role of the immune microenvironment in tumor progression, we developed a comprehensive atlas of the meningioma microenvironment, with a view toward identifying modifiable opportunities. METHODS: The immune microenvironment of 2,727 meningiomas was profiled using orthogonal methodologies, including 24 with mass cytometry, 24 single-cell RNAseq, 1,437 bulk RNAseq, 1,125 DNA methylation, 117 multiplex immunofluorescence, as well as that of 5 paired peripheral blood samples and 10 human meninges. Diagnostic biomarkers, prognostic signatures, and therapeutic resistance: shedding light on the dual role of autophagy in glioblastoma.

Glioblastoma (GBM) is an exceptionally aggressive brain malignancy with a dismal prognosis, characterized by a median survival of roughly 15 months despite standard radiotherapy and Temozolomide treatment. A primary driver of this therapeutic resistance is autophagy. While initially tumor-suppressive, autophagy transitions into a cytoprotective mechanism in established GBM, acting as a molecular survival switch that enables cells to withstand metabolic stress, hypoxia, and therapy-induced DNA damage. Crucially, this complex intracellular degradation process has emerged as a powerful tool for G

Decoding Senescence-Driven Heterogeneity in Early-Onset Colorectal Cancer for Prognostic and Therapeutic Stratification.

Early-onset colorectal cancer (EOCRC), diagnosed in patients under 50, is particularly aggressive, yet lacks targeted therapeutic strategies. This study aimed to explore the role of cellular senescence in driving EOCRC's malignancy and developed a senescence scoring system (EO-Senscore) to guide precision oncology. Through a multi-omics analysis of 2961 patients, we discovered that cellular senescence is more pronounced and varied in EOCRC and is not tied to chronological age. Two distinct senescence subtypes were identified: Cluster 1 (low-senescence tumors) showed prolonged survival, enhance

Neuroimmune Interactions in Pancreatic Cancer.

Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive primary malignancy, and recent technological advances in surgery have opened up more possibilities for surgical treatment. Emerging evidence highlights the critical roles of diverse immune and neural components in driving the aggressive behavior of PDAC. Recent studies have demonstrated that neural invasion, neural plasticity, and altered autonomic innervation contribute to pancreatic neuropathy in PDAC patients, while also elucidating the functional architecture of nerves innervating pancreatic draining lymph nodes. Research into

[Drivers license qualification for epileptics].

The question whether a person with epilepsy qualified for a driving licence must be examined from the point of view of the individual as well as that of the community. The general public should be protected against unduly high risks from epileptic drivers, whereas the patient has a right to live as normal a life as possible, which includes driving an automobile. Too rigid criteria for obtaining the license increase the number of persons who evade medical control and drive "illegally". To require physicians to report their epileptic patients to the authorities would be counterproductive; it wou

Reckless Self-Destructive Behavior and PTSD in Veterans: The Mediating Role of New Adverse Events.

The addition of self-destructive and reckless behavior as a symptom of posttraumatic stress disorder (PTSD) in DSM-5 has stimulated renewed interest in understanding relationships between these behaviors and trauma-related psychopathology. This study examined the relationship between reckless and self-destructive behaviors (RSDB), intervening exposure to new adverse events, and later PTSD severity in a sample of trauma-exposed veterans. At baseline, participants were assessed for RSDB (past 5 years) and current PTSD severity (N = 222). PTSD severity was then reassessed approximately 4 years la

[Problem of specific syndrome formation in psychosomatic disease situations. (Psychodynamics of rheumatic diseases)].

Three features of psychosomatic diseases with negative rheumafactor were psychoanalytically investigated (35 patients). Each of these diseases (the Palindromic Rheumatism, the Reiters Disease and the Psoriatic Arthritis), showed a characteristic syndrome. Corresponding with the patients with rheumafactor-positive Rheumatoid Arthritis was only the fact of inhibited aggressive impulses in the somatic field by the blockade of the muscle system. We believe that this study proves our earlier found thesis, that in psychosomatic diseases we have to work out the specific conflict, the specific psychod

Interplay of TGFβ signaling and microRNA in thyroid cell loss of differentiation and cancer progression.

Thyroid cancer has been rapidly increasing in prevalence among humans in last 2 decades and is the most prevalent endocrine malignancy. Overall, thyroid-cancer patients have good rates of long-term survival, but a small percentage present poor outcome. Thyroid cancer aggressiveness is essentially related with thyroid follicular cell loss of differentiation and metastasis. The discovery of oncogenes that drive thyroid cancer (such as RET, RAS, and BRAF), and are aligned in the MAPK/ERK pathway has led to a new perspective of thyroid oncogenesis. The uncovering of additional oncogene-modulated s

The YAP1 Signaling Inhibitors, Verteporfin and CA3, Suppress the Mesothelioma Cancer Stem Cell Phenotype.

Mesothelioma is an aggressive cancer that has a poor prognosis. Tumors develop in the mesothelial lining of the pleural and peritoneal cavities in response to asbestos exposure. Surgical debulking followed by chemotherapy is initially effective, but this treatment ultimately selects for resistant cells that form aggressive and therapy-resistant recurrent tumors. Mesothelioma cancer stem cells (MCS) are a highly aggressive subpopulation present in these tumors that are responsible for tumor maintenance and drug resistance. In this article, we examine the impact of targeting YAP1/TAZ/TEAD signal

Resolvin E1 and Maresin 1 Attenuate Nucleotide-Binding Oligomerization Domain-Like Receptor Protein 3-Associated Inflammation and Invasion in Oral Squamous Cell Carcinoma.

Inflammation critically contributes to the progression of oral squamous cell carcinoma (OSCC), a common and aggressive oral cancer. Excessive nucleotide-binding oligomerization domain (NOD)-like receptor protein 3 (NLRP3) inflammasome activation drives tumor-associated chronic inflammation, which oral pathogens can trigger to enhance invasion and metastasis. The present study investigated the effects of resolvin E1 (RvE1) and maresin 1 (MaR1) on NLRP3-associated inflammation and evaluated whether promoting inflammation resolution limits tumor aggressiveness in OSCC. RvE1, MaR1, or vehicle was

Divergent Evolution of Malignant Subclones Maintains a Balance between Induced Aggressiveness and Intrinsic Drug Resistance in T-cell Cancer.

UNLABELLED: Evolution and outgrowth of drug-resistant cancer cells are common causes of treatment failure. Patients with leukemic cutaneous T-cell lymphoma have a poor prognosis because of the development of drug resistance and severe bacterial infections. In this study, we show that most patients with leukemic cutaneous T-cell lymphoma harbor multiple genetically distinct subclones that express an identical clonal antigen receptor but display distinct phenotypes and functional properties. These coexisting malignant subclones exhibit differences in tissue homing, metabolism, and cytokine expre

A Comprehensive multi-network analysis of ceRNAs and transcription factors for papillary thyroid carcinoma diagnosis and prognosis.

Papillary thyroid carcinoma (PTC) is a malignancy with an ambiguous etiology. The competitive endogenous RNA (ceRNA) hypothesis provides a framework for clarifying the molecular mechanisms that drive carcinogenesis. In this study, we constructed a novel ceRNA network to identify reliable diagnostic and prognostic indicators applicable across all stages of PTC. Transcriptome analysis was performed to identify stage-specific hub genes using the MCC, IVI, and MCODE algorithms. A novel five-layer ceRNA network and its associated regulatory network (DE-TF) were constructed. Receiver operating chara

Development of a stress response therapy targeting aggressive prostate cancer.

Oncogenic lesions up-regulate bioenergetically demanding cellular processes, such as protein synthesis, to drive cancer cell growth and continued proliferation. However, the hijacking of these key processes by oncogenic pathways imposes onerous cell stress that must be mitigated by adaptive responses for cell survival. The mechanism by which these adaptive responses are established, their functional consequences for tumor development, and their implications for therapeutic interventions remain largely unknown. Using murine and humanized models of prostate cancer (PCa), we show that one of the

The role of transcription factors in prostate cancer progression.

Prostate cancer is one of the most common malignancies in men, with most cases initially responding to androgen deprivation therapy. However, a significant number of patients eventually develop castration-resistant prostate cancer, an aggressive form of the disease. Although androgen receptor (AR) pathway inhibitors target AR signaling, and have extended survival in patients with castration-resistant prostate cancer, prolonged treatment can lead to the emergence of neuroendocrine prostate cancer (NEPC), a lethal subtype characterized by the expression of neuroendocrine markers and reduced AR a

Combination of immunotherapy and other targeted therapies in advanced cutaneous melanoma.

Cutaneous Melanoma (CM) is an aggressive cancer whose incidence is increasing worldwide. However, the knowledge of its biology and genes driving cell growth and survival allowed to develop new drugs that have improved PFS and OS of advanced disease. Both BRAF targeting agents and immune checkpoint inhibitors (ICIs) have been adopted for the treatment of metastatic disease and the adjuvant setting. Several melanoma patients show innate or acquired drug-resistance and thus new strategies are required for overcoming this complication. New ICIs have been developed, and strategies of combination or

UBE2T promotes papillary thyroid carcinoma progression by activating the JAK/STAT3 pathway via negative regulation of SOCS2.

Papillary thyroid carcinoma (PTC) exhibits aggressive behaviors such as tumor invasion and lymph node metastasis that critically influence prognosis, yet reliable predictors of invasiveness remain elusive. This study investigated the molecular mechanisms through which ubiquitin-conjugating enzyme E2T (UBE2T) drives PTC progression. Bioinformatics analysis of TCGA/GEO datasets and validation with institutional clinical samples revealed UBE2T overexpression correlated with advanced clinicopathological features. Functional experiments demonstrated that UBE2T overexpression enhanced PTC cell invas

Dysregulation of PRMT5 in chronic lymphocytic leukemia promotes progression with high risk of Richter's transformation.

Richter's Transformation (RT) is a poorly understood and fatal progression of chronic lymphocytic leukemia (CLL) manifesting histologically as diffuse large B-cell lymphoma. Protein arginine methyltransferase 5 (PRMT5) is implicated in lymphomagenesis, but its role in CLL or RT progression is unknown. We demonstrate herein that tumors uniformly overexpress PRMT5 in patients with progression to RT. Furthermore, mice with B-specific overexpression of hPRMT5 develop a B-lymphoid expansion with increased risk of death, and Eµ-PRMT5/TCL1 double transgenic mice develop a highly aggressive disease wi

The many pathways driving liver inflammation in MASH.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease worldwide, affecting one-third of the global population. Most patients exhibit simple steatosis, whereas up to 20% develop metabolic dysfunction-associated steatohepatitis (MASH), potentially culminating in liver cirrhosis and hepatocellular carcinoma. Diverse parallel mechanisms contribute to the development of MASH, which are fueled by hepatic lipotoxicity, intestinal dysbiosis, and pro-inflammatory diets shaping innate and adaptive immune responses. Moreover, adipose tissue is drivin
