Prevention of precipitated withdrawal symptoms by activating central cholinergic systems during a dependence-producing schedule of morphine in rats.

Previous studies in this and other laboratories have suggested an important role for central cholinergic neurons in the expression of morphine withdrawal symptoms. This study was designed to determine whether the symptoms of withdrawal could be mitigated by normalization of the effect of morphine on cholinergic neurons. Since this effect is generally inhibitory, we used centrally acting cholinergic agonists to augment central cholinergic tone during chronic morphine infusion. Rats were made dependent following the intra-arterial (i.a.) infusion of increasing concentrations (35-100 mg kg(-1) da

Potencies and channel properties induced by semirigid agonists at frog nicotinic acetylcholine receptors.

Structure-activity relationships were investigated in a series of semirigid nicotinic agonists. Three of the agonists, (-)-ferruginine methiodide, arecoline methiodide, and its ketonic analogue arecolone methiodide, were cyclic analogues of anatoxin-a, a potent, naturally occurring, bicyclic alkaloid. Two other cyclic agonists, (-)-cytisine and (+/-)-muscarone, and the simplest agonist, the tetramethylammonium ion, were also tested. Arecolone methiodide and (-)-ferruginine methiodide have been tested as nicotinic agonists for the first time. Relative potency was assayed by contracture on the r

The relative potencies of some agonists at M2 muscarinic receptors in guinea-pig ileum, atria and bronchi.

The effects of some agonists on isolated preparations of guinea-pig ileum, atria and bronchial muscle have been compared with those of carbachol. The concentrations producing comparable responses were used to estimate the equipotent molar ratio relative to carbachol. Arecaidine propargyl ester was 4 to 5 times as active as carbachol on the ileum but more than 10 times as active as carbachol on atrial rate or atrial force, so the results confirm that this compound has a 2 to 3 fold selectivity for receptors in atria. Ethoxyethyltrimethylammonium iodide was one-quarter to one-third as active as

Chronic exposure to areca alkaloids at environmentally relevant concentrations alters neurobehavior, gut microbiota and brain metabolism in adult zebrafish.

Trace-level neuroactive contaminants in aquatic environments are increasingly recognized as ecological hazards, yet the neurotoxic potential of areca alkaloids remains largely unexplored. In this study, adult zebrafish were chronically exposed to arecoline or arecaidine for 90 days to investigate long-term neurobehavioral toxicity and associated biological alterations. Behavioral assessments revealed persistent impairments in social interaction, reduced aggression, and altered exploratory activity following alkaloid exposure. Concomitantly, areca alkaloids induced intestinal injury characteriz

Functional characterization of a muscarinic receptor stimulating gastrin release from rabbit antral G-cells in primary culture.

In previous studies carbachol-induced stimulation of gastrin release from antral G-cells in primary culture suggested the presence of muscarinic acetylcholine receptors on this cell type. Therefore, we attempted to pharmacologically characterize the muscarinic acetylcholine receptor subtype involved. Enzymatically isolated rabbit antral mucosal cells (0.8% G-cells) were separated by counterflow elutriation yielding a fraction (1.7% G-cells) that was placed in culture on collagen-coated well plates. After 24-36 h of culture 13.0 +/- 2.4% of total adherent cells were immunoreactive for gastrin a

Central muscarinic receptor subtypes and carrageenin-induced paw oedema in rats.

In an earlier report from this laboratory, it was demonstrated that the central cholinergic system exerted a modulatory pro-inflammatory effect on carrageenin-induced paw inflammation in rats. In this study, the role of the central muscarinic receptors in cholinergic modulation of peripheral inflammation was investigated by using several M1 and M2 receptor agonists and antagonists. The M1 receptor agonists aceclidine and arecholine and the nonspecific muscarinic receptor agonist oxotremorine augmented carrageenin oedema, an effect attenuated by the M1 receptor antagonist scopolamine. Serological diagnosis and post-treatment monitoring of Echinococcus granulosus in experimentally infected dogs using crude and recombinant fibronectin antigens.

Echinococcus granulosus is a zoonotic helminth responsible for cystic echinococcosis, a significant public health concern. The diagnosis of E. granulosus infections in definitive hosts, such as dogs, is challenging due to the absence of clinical signs. This study aimed to evaluate the diagnostic performance of crude (EgSCA) and recombinant (rEgFN162) antigens for the detection of E. granulosus infection in dogs using ELISA and Western blot assays. Additionally, it sought to identify the most suitable antigen and method for population-based screening and post-treatment monitoring. Adult E. gran

Human M1-, M2- and M3-muscarinic cholinergic receptors: binding characteristics of agonists and antagonists.

The muscarinic acetylcholine receptors were identified in membrane preparations from human tissues by the specific binding of 1-[benzilic-4,4'-3H] quinuclidinyl benzilate. Saturation binding isotherms of this radioligand yielded a total amount of receptors of 435 +/- 208, 159 +/- 65 and 913 +/- 89 fmol/mg protein, respectively, in the hippocampus, pons and submandibular gland. Non linear least squares analysis of competition binding studies with the antagonists pirenzepine and AF-DX 116 indicates that the majority of receptors are of the M1-type in the hippocampus (83%, high affinity for piren

MicroRNA let-7a mitigates the progression of oral submucous fibrosis by targeting high-mobility group AT-hook 2.

BACKGROUND/PURPOSE: Oral submucous fibrosis (OSF) is an irreversible fibrotic disorder of the oral cavity with a high potential for malignant transformation. MicroRNA lethal-7a (let-7a) has been recognized as a key antifibrotic regulator, but its specific role in OSF remains unknown. Therefore, this study aimed to elucidate the functional significance and the molecular mechanism of let-7a in OSF progression. MATERIALS AND METHODS: The expression of let-7a was quantified by real-time quantitative polymerase chain reaction in fibrotic buccal mucosal fibroblasts (fBMFs) isolated from OSF lesions

Areca nut alkaloids induce irreparable DNA damage and senescence in fibroblasts and may create a favourable environment for tumour progression.

BACKGROUND: Oral submucous fibrosis (OSMF) is a pre-malignant condition that is strongly associated with the areca nut alkaloids, arecoline (ARC) and arecaidine (ARD). The condition is characterised by the presence of senescent fibroblasts in the subepithelial mesenchyme which have the potential to promote malignancy in the neighbouring epithelial cells. We tested the hypothesis that areca nut alkaloids induce senescence in oral fibroblasts and promote the secretion of invasion-promoting transforming growth factor β (TGF-β) and matrix metalloproteinase-2 (MMP-2). METHODS: Two oral fibroblast

Hemicholinium-3 impairs spatial learning and the deficit is reversed by cholinomimetics.

The effects of hemicholinium-3 (HC-3) on spatial discrimination learning were studied. Rats were equipped with indwelling cannulae in the right lateral ventricle and, following recovery, were trained on a two platform spatial discrimination task in a water maze. In this task a visible escape platform remains in a fixed position in the pool during a single training session, whilst the location of an identical "float" (which affords no escape) is randomly varied. For each session the location of the fixed escape platform was changed and the rats were retrained to criterion following pretreatment

Immunological and molecular tools for environmental surveillance of canine echinococcosis: Steps toward a sustainable diagnostic algorithm.

Canine echinococcosis (CaEc) surveillance has evolved from necropsy and arecoline purgation to the detection of coproantigens (cELISA) and genomic copro-DNA (cPCR and cLAMP). Each technique has advantages and disadvantages regarding biosafety, ethics, and costs. In Argentina, there is no consensus on CaEc surveillance tools or their suitability for low- and medium-complexity laboratories. The aim of this work was to compare the performance of techniques with different targets for CaEc surveillance, including cELISA, nested cPCR, and two cLAMPEGSL (2.0 and 3.0). Environmental canine fecal sampl

Affinities of muscarinic drugs for [3H]N-methylscopolamine (NMS) and [3H]oxotremorine (OXO) binding to a mixture of M1-M4 muscarinic receptors: use of NMS/OXO-M ratios to group compounds into potential agonist, partial agonist, and antagonist classes.

The relative affinities of various muscarinic drugs in the antagonist ([3H]N-methyl scopolamine ([3H]NMS)) and agonist ([3H]Oxotremorine-m ([3H]OXO-M)) binding assays using a mixture of tissues containing M1-M4 receptor subtypes have been determined. [3H]NMS bound with high affinity (Kd = 25 +/- 5.9 pM; n = 3) and to a high density Bmax = 11.8 +/- 0.025 nmol/g wet weight) of muscarinic receptors. [3H]OXO-M appeared to bind to two binding sites with differing affinities (Kd1 = 2.5 +/- 0.1 nM; Kd2 = 9.0 +/- 4.9 microM; n = 4) and to a different population of binding sites (Bmax1 = 5.0 +/- 0.26 n

Silver-Bacterial Cellulose-Areca catechu Extract Nanocomposite Biofilm for Sustainable Food Packaging: Synthesis, Characterization, and Antibacterial Activity.

Bacterial cellulose (BC) is a low-cost, biodegradable biopolymer with excellent mechanical strength and water-holding capacity. It is an attractive material for sustainable food packaging; however, native BC lacks intrinsic antibacterial activity. In this study, silver-bacterial cellulose-Areca catechu nanocomposite biofilms were synthesized through in situ reduction of Ag+ on BC films, followed by impregnation with A. catechu fruit extract at concentrations of 0-30 mg/mL. The resulting films were characterized by scanning electron microscopy-energy-dispersive X-ray spectroscopy (SEM-EDX), Fou

miR-134-3p Deficiency-Induced Methionine Metabolism Promotes Fibrosis via SLC25A33 in OSF.

BACKGROUND: Fibrosis is a disease characterized by excessive collagen deposition by fibroblasts, leading to tissue and organ dysfunction. Fibroblasts are the primary effector cells, and their functional phenotype is regulated by various factors, with metabolic reprogramming being a crucial one. Previous studies have shown that microRNAs induce hypoxia, abnormal energy metabolism, and promote fibrosis. Our research aimed to identify the miRNA associated with oral submucous fibrosis (OSF) pathogenesis and to preliminarily explore its role in the phenotypic transformation of fibroblasts. METHODS
