Identifying typical trajectories in longitudinal data: modelling strategies and interpretations.

Individual-level longitudinal data on biological, behavioural, and social dimensions are becoming increasingly available. Typically, these data are analysed using mixed effects models, with the result summarised in terms of an average trajectory plus measures of the individual variations around this average. However, public health investigations would benefit from finer modelling of these individual variations which identify not just one average trajectory, but several typical trajectories. If evidence of heterogeneity in the development of these variables is found, the role played by temporal

Black Americans With Sickle Cell Disease (SCD) Demonstrate Accelerated Epigenetic Pace of Aging Compared to Black Americans Without SCD.

BACKGROUND: Sickle cell disease (SCD) is a chronic medical condition characterized by red blood cell sickling, vaso-occlusion, hemolytic anemia, and subsequently, end-organ damage and reduced survival. Because of this significant pathophysiology and early mortality, we hypothesized that patients with SCD are experiencing accelerated biological aging compared with individuals without SCD. METHODS: We utilized the DunedinPACE measure to compare the epigenetic pace of aging in 131 Black Americans with SCD to 1391 Black American veterans without SCD. RESULTS: SCD patients displayed a significant

Translational immunology: Applying fundamental discoveries to human health and autoimmune diseases.

Studying the human immune system is challenging. These challenges stem from the complexity of the immune system itself, the heterogeneity of the immune system between individuals, and the many factors that lead to this heterogeneity including the influence of genetics, environment, and immune experience. Studies of the human immune system in the context of disease are increased in complexity as multiple combinations and variations in immune pathways can lead to a single disease. Thus, although individuals with a disease may share clinical features, the underlying disease mechanisms and resulti

Factors affecting log-transformed muscle 137Cs concentrations in wild boars in Fukushima Prefecture over 14 years.

The March 2011 Fukushima Daiichi Nuclear Power Plant accident resulted in extensive radiocesium contamination of forest ecosystems. Wild boars (Sus scrofa) are a key indicator species because of their high radiocesium accumulation; however, long-term spatiotemporal patterns and biological drivers of contamination have not been fully evaluated using a prefecture-wide dataset. We analyzed monitoring data from 3,609 wild boars collected across Fukushima Prefecture over 14 years (FY2011-FY2025). By integrating individual-level measurements with spatial soil deposition data, we fitted three mixed-e

Toward causal artificial intelligence for biologic treatment response in rheumatoid arthritis: current evidence and future directions.

Rheumatoid arthritis (RA) is a heterogeneous autoimmune disease with substantial variation in response to biologic therapy. Artificial intelligence (AI) is increasingly used to predict treatment response, but most existing models are association-based and offer limited interpretability for clinical decision-making. To critically review the role of causal artificial intelligence in predicting and explaining biologic treatment response in RA and assess its potential to improve interpretability and individualized treatment selection. A narrative review with an integrative approach was conducted,

Isolating single cycles of neural oscillations in population spiking.

Neural oscillations are prominent features of brain activity, characterized by frequency-specific power changes in electroencephalograms (EEG) and local field potentials (LFP). These oscillations also appear as rhythmic coherence across brain regions. While the identification of oscillations has primarily relied on EEG and LFP, they are also present in neuronal spiking. However, several questions remain unanswered: How do spiking oscillations relate to field potential oscillations? How are spiking oscillations correlated across brain regions? And how are they connected to other physiological a

Molecular Functionality of Cytochrome P450 4 (CYP4) Genetic Polymorphisms and Their Clinical Implications.

Enzymes in the cytochrome P450 4 (CYP4) family are involved in the metabolism of fatty acids, xenobiotics, therapeutic drugs, and signaling molecules, including eicosanoids, leukotrienes, and prostanoids. As CYP4 enzymes play a role in the maintenance of fatty acids and fatty-acid-derived bioactive molecules within a normal range, they have been implicated in various biological functions, including inflammation, skin barrier, eye function, cardiovascular health, and cancer. Numerous studies have indicated that genetic variants of CYP4 genes cause inter-individual variations in metabolism and d

Venous thromboembolic disease genetics: from variants to function.

Venous thromboembolic disease (VTE) is a prevalent and potentially life-threatening vascular disease, including both deep vein thrombosis and pulmonary embolism. This review will focus on recent insights into the heritable factors that influence an individual's risk for VTE. Here, we will explore not only the discovery of new genetic risk variants but also the importance of functional characterization of these variants. These genome-wide studies should lead to a better understanding of the biological role of genes inside and outside of the canonical coagulation system in thrombus formation and

Taking into account the influence of climatic factors and age in the mathematical modeling and analysis of the dynamic transmission of malaria.

Malaria is the most widespread and deadly parasitic disease in the world. This disease is transmitted from an infected individual to a healthy individual via the female Anopheles mosquito. Children under the age of 5 are most at risk from this infectious disease. Sub-Saharan Africa is most affected by malaria in the world. The geographical distribution and seasonality of the malaria cases observed are closely related to the climatic factors (temperature and precipitation) that influence its transmission dynamics. To control this disease, the use of insecticide treated nets and education of the

Genetic variation in the human brain dopamine system influences motor learning and its modulation by L-Dopa.

Dopamine is important to learning and plasticity. Dopaminergic drugs are the focus of many therapies targeting the motor system, where high inter-individual differences in response are common. The current study examined the hypothesis that genetic variation in the dopamine system is associated with significant differences in motor learning, brain plasticity, and the effects of the dopamine precursor L-Dopa. Skilled motor learning and motor cortex plasticity were assessed using a randomized, double-blind, placebo-controlled, crossover design in 50 healthy adults during two study weeks, one with

Brain maintenance biomarkers from structural and functional interactions in aging and neurodegeneration.

Brain maintenance may help explain why some individuals remain cognitively resilient despite aging, but its biological basis is not well understood. Here, we show that brain maintenance can be quantified from the relationship between brain structure and function. Using structural MRI and resting-state functional MRI from 1280 older adults, we built a model based on young adults to estimate the functional capacity supported by preserved brain structure, and defined brain maintenance as the difference between predicted and observed function. Brain maintenance was most evident in prefrontal, cing

Neuronal morphology enhances robustness to perturbations of channel densities.

Biological neurons show significant cell-to-cell variability but have the striking ability to maintain their key firing properties in the face of unpredictable perturbations and stochastic noise. Using a population of multi-compartment models consisting of soma, neurites, and axon for the lateral pyloric neuron in the crab stomatogastric ganglion, we explore how rebound bursting is preserved when the 14 channel conductances in each model are all randomly varied. The coupling between the axon and other compartments is critical for the ability of the axon to spike during bursts and consequently

Long-term glucocorticoids in relation to the metabolic syndrome and cardiovascular disease: A systematic review and meta-analysis.

The striking link of Cushing's syndrome with the metabolic syndrome (MetS) and cardiovascular disease (CVD) suggests that long-term exposure to extremely high cortisol levels catalyzes cardiometabolic deterioration. However, it remained unclear whether the findings from the extreme glucocorticoid overabundance observed in Cushing's syndrome could be translated into more subtle variations in long-term glucocorticoid levels among the general population, for example, due to chronic stress. Here, we performed a systematic review (PROSPERO: CRD42023425541) of evidence regarding the role of subtle v

SARS-CoV-2 biological clones are genetically heterogeneous and include clade-discordant residues.

Defective genomes are part of SARS-CoV-2 quasispecies. High-resolution, ultra-deep sequencing of bulk RNA from viral populations does not distinguish RNA mutations, insertions, and deletions in viable genomes from those in defective genomes. To quantify SARS-CoV-2 infectious variant progeny, virus from four individual plaques (biological clones) of a preparation of isolate USA-WA1/2020, formed on Vero E6 cell monolayers, was subjected to further biological cloning to yield 9 second-generation and 15 third-generation sub-clones. Consensus genomic sequences of the biological clones and sub-clone

Selection for universal facial emotion.

Facial expression is heralded as a communication system common to all human populations, and thus is generally accepted as a biologically based, universal behavior. Happiness, sadness, fear, anger, surprise, and disgust are universally recognized and produced emotions, and communication of these states is deemed essential in order to navigate the social environment. It is puzzling, however, how individuals are capable of producing similar facial expressions when facial musculature is known to vary greatly among individuals. Here, the authors show that although some facial muscles are not prese

Coding, modifier, and regulatory effects shape circulating APOL1 levels.

APOL1 variants G1 and G2 are major genetic risk factors for chronic kidney disease in people with recent sub-Saharan African ancestry, yet the mechanisms linking genotype to disease remain poorly defined. Circulating APOL1 is likely to play a critical role, although how specific genotypes and regulatory variants influence circulating APOL1 levels is incompletely understood. We analysed Olink-measured APOL1 NPX (Normalized Protein eXpression) levels from 43 587 UK Biobank participants. Associations between APOL1 NPX and known genotypes were assessed in individuals with recent sub-Saharan Africa

Investigation of ComBat Harmonization on Radiomic and Deep Features from Multi-Center Abdominal MRI Data.

ComBat harmonization has been developed to remove non-biological variations for data in multi-center research applying artificial intelligence (AI). We investigated the effectiveness of ComBat harmonization on radiomic and deep features extracted from large, multi-center abdominal MRI data. A retrospective study was conducted on T2-weighted (T2W) abdominal MRI data retrieved from individual patients with suspected or known chronic liver disease at three study sites. MRI data were acquired using systems from three manufacturers and two field strengths. Radiomic features and deep features were e

Nanomechanical properties of lipid bilayer: asymmetric modulation of lateral pressure and surface tension due to protein insertion in one leaflet of a bilayer.

The lipid membranes of living cells form an integral part of biological systems, and the mechanical properties of these membranes play an important role in biophysical investigations. One interesting problem to be evaluated is the effect of protein insertion in one leaflet of a bilayer on the physical properties of lipid membrane. In the present study, an all atom (fine-grained) molecular dynamics simulation is used to investigate the binding of cytotoxin A3 (CTX A3), a cytotoxin from snake venom, to a phosphatidylcholine lipid bilayer. Then, a 5-microsecond [corrected] coarse-grained molecula

Brain imaging, genetics and emotion.

This paper reviews the published evidence on genetically driven variation in neurotransmitter function and brain circuits involved in emotion. Several studies point to a role of the serotonin transporter promoter polymorphism in amygdala activation during emotion perception. We also discuss other polymorphisms (e.g. the COMT val158met polymorphism, tryptophan hydroxylase-2 -703 G/T) and putative effects on affective processing in cortical and limbic regions. A different line of research concerns studies with genetic disorders. Although at a less fine-grained level, studies with individuals wit

Conceptualizing disparities and differences in the psychobiology of traumatic stress.

Understanding biological pathways that mediate trauma-related psychopathology is a major goal for traumatic stress studies. There is growing interest in studying differences in neural, physiological, and behavioral correlates of traumatic stress across demographic groups (e.g., sex/gender, race/ethnicity). However, challenges remain in how to appropriately conceptualize the source, mechanisms, and practical utility of between-group variation. The present brief conceptual review discusses ethnicity, race, and sex/gender-related variability relevant to understanding the psychobiology of traumati
