Assuring the Biofunctionalization of Silicone Covalently Bonded to Rhamnolipids: Antibiofilm Activity and Biocompatibility.

Silicone-based medical devices composed of polydimethylsiloxane (PDMS) are widely used all over the human body (e.g., urinary stents and catheters, central venous catheters stents) with extreme clinical success. Nevertheless, their abiotic surfaces, being prone to microorganism colonization, are often involved in infection occurrence. Improving PDMS antimicrobial properties by surface functionalization with biosurfactants to prevent related infections has been the goal of different works, but studies that mimic the clinical use of these novel surfaces are missing. This work aims at the biofunc

Bacterial Degradation of Persistent Organic Pollutants: From Molecular Mechanisms to Synthetic Biology.

Persistent organic pollutants (POPs) are among the most recalcitrant environmental contaminants due to their chemical stability, hydrophobicity, toxicity, and capacity for long-range transport, yet mechanistic understanding remains fragmented across molecular, cellular, and systems scales, hindering integrated bioremediation strategies. This study synthesizes recent advances in bacterial POP degradation by integrating evidence from aerobic oxidative, anaerobic reductive, and cometabolic pathways across molecular, genomic, regulatory, metabolic, and multi-omics levels. The analysis reveals that

Bioprospecting Indoor Bacteria Producing Bioactive Metabolites.

New biomolecules are prospected for new antimicrobial, antiparasitic and cytostatic drugs. Here, we present bioassay-guided methods for prospecting indoor bacteria producing non-protein antibiotic molecules. Out of 650 bacterial colonies tested, 108 antimicrobial isolates representing 15 species and cryptic Actinobacteria, produced non-protein antibiotics affecting mammalian cells. Methanol extracts of the 108 strains tested with a battery of bioassays revealed four toxicity endpoints: (1) Cell death (CD). (2) Boar sperm motility inhibition (BSMI). (3) Inhibition of cell proliferation (ICP). (

Integrated Analysis of Paired Crude Oil and Produced Water Reveals Stress-Tolerant Hydrocarbon-Degrading Bacteria with Bioremediation Potential.

Oilfield microbiome studies have extensively characterized microbial community composition or isolated individual hydrocarbon degraders, but integrated exploration of cultivable functional bacteria from paired crude-oil and produced-water environments remains limited, particularly for strains combining hydrocarbon utilization with stress-tolerance and biosurfactant-related traits. In this study, paired crude-oil and produced-water samples from the Huoshaoshan Oilfield were investigated by combining 16S rRNA amplicon sequencing with culture-dependent isolation and functional screening. A total

Sophorolipids as anticancer agents: progress and challenges.

Sophorolipids (SLs) are considered effective biosurfactant for cancer treatment, which can efficiently inhibit the viability of various cancer types including breast, lung, liver, cervical and colon cancers. Their mechanism of action targets apoptosis and operates at the level of caspase enzymes, upregulation and downregulation of the B-cell lymphoma (Bcl)-family proteins, and changes in mitochondrial membrane permeability. The binding of SLs to the cancer cell receptors modulates the expression of Bax, APAF1, Bcl-2 and Bcl-x, and triggers the release of cytochrome c into the cytosol which fur

[Up to Date of Cyanobacterial Natural Products].

More than 2000 compounds have been reported from cyanobacteria. The most successful example is dolastatin 10, of which a related compound monomethylauristatin E is used as antibody-drug conjugate (ADC) for Hodgkin lymphoma and systemic anaplastic large cell lymphoma. Recently genome-based analyses by Piel led to the discovery of novel compounds from cyanobacteria. W. H. Gerwick found a potential as anti-SARS-CoV-2 agent in gallinamide A, which was reported as a cathepsin L inhibitor. In our group columbamides were isolated from the marine cyanobacterium Moorena bouillonii. The geometry of the

Bioballs carrying a syntrophic Rhodococcus and Mycolicibacterium consortium for simultaneous sorption and biodegradation of fuel oil in contaminated freshwater.

Nonpathogenic effective bacterial hydrocarbon degraders, Rhodococcus ruber S103, Mycolicibacterium parafortuitum J101 and Mycolicibacterium austroafricanum Y502, were isolated from mixed polycyclic aromatic hydrocarbon (PAH)-enriched river sediments. They possessed broad substrate specificities toward various PAHs and aliphatic compounds as sole carbon sources. These strains exhibited promising characteristics, including biosurfactant production, high cell hydrophobicity, biofilm formation and no antagonistic interactions, and contained genes encoding hydrocarbon-degrading enzymes. The mixed b

Phase Transfer of Palladized Nanoscale Zerovalent Iron for Environmental Remediation of Trichloroethene.

Palladium-doped nanoscale zerovalent iron (Pd-NZVI) has been shown to degrade environmental contaminants such as trichloroethene (TCE) to benign end-products through aqueous phase reactions. In this study we show that rhamnolipid (biosurfactant)-coated Pd-NZVI (RL-Pd-NZVI) when reacted with TCE in a 1-butanol organic phase with limited amounts of water results in 50% more TCE mass degradation per unit mass of Pd-NZVI, with a 4-fold faster degradation rate (kobs of 0.413 day(-1) in butanol organic phase versus 0.099 day(-1) in aqueous phase). RL-Pd-NZVI is preferentially suspended in water in b

Removing adsorbed heavy metal ions from sand surfaces via applying interfacial properties of rhamnolipid.

In this study, the interfacial properties of biosurfactant rhamnolipid were investigated and were applied to remove adsorbed heavy metal ions from sand surfaces with flushing operations. The surface tension-lowering activity, micelle charge characteristic, and foaming ability of rhamnolipid were identified first. For rhamnolipid in water, the negatively charged characteristic of micelles or aggregates was confirmed and the foaming ability at concentrations higher than 40 mg/L was evaluated. By using the rhamnolipid solutions in a batch washing approach, the potential of applying the interfacia

Ethyl methanesulfonate mutagenesis: Advancing bacterial genetics for sustainable agriculture.

Ethyl methanesulfonate (EMS) is a strong alkylating agent commonly used to induce random point mutations, particularly G: C to A: T transitions, by ethylating guanine bases in DNA. Its mutagenic properties, which stem from the transfer of ethyl groups to nucleophilic sites within cells, allow for the creation of various mutant libraries, aiding research in bacterial physiology, metabolism, and antibiotic resistance. This review briefly examines the mechanisms behind EMS mutagenesis and its applications in both forward and reverse genetics. In forward genetics, mutants generated by EMS with alt

Biologically active pigment and ShlA cytolysin of Serratia marcescens induce autophagy in a human ocular surface cell line.

BACKGROUND: The cellular process of autophagy is essential for maintaining the health of ocular tissue. Dysregulation of autophagy is associated with several ocular diseases including keratoconus and macular degeneration. It is known that autophagy can be used to respond to microbial infections and that certain microbes can exploit the autophagic process to their benefit. In this study, a genetic approach was used to identify surface-associated and secreted products generated by the opportunistic pathogen Serratia marcescens involved in activation of autophagy. METHODS: A recombinant human co

Diesel oil removal by Serratia sp. W4-01 immobilized in chitosan-activated carbon beads.

Serratia sp. W4-01 was immobilized in chitosan-activated carbon beads and used for diesel oil removal. The type and concentration of chitosan, activated carbon content, and bead diameter were investigated as factors affecting diesel oil removal. The results showed that 2% (w/v) squid pen chitosan beads modified with 1% activated carbon (w/v) and with a 3-mm diameter had a good spherical shape and strength as well as diesel oil removal capability. The immobilized W4-01 cells removed more than 40% of diesel oil after 7 days when the initial diesel oil concentration was 100 to 400 mg L-1, whereas

Interaction between Sophorolipids and β-glucan in Aqueous Solutions.

Skin disorders, including acne vulgaris, atopic dermatitis, and rosacea, are characterized by the presence of biofilms, which are communities of microorganisms. The mechanical stability of biofilms is attributed to one of their constituents-polysaccharides-which are secreted by microorganisms. Sophorolipids are biosurfactants with biofilm disruption and removal abilities and are expected to become alternatives for classical petrochemical-based surfactants in cosmetics. In this study, we investigated the influence of sophorolipids on β-glucan such as dispersion status, interaction mechanism, an

[The lactobacilli-protection system of pregnant women--efficient prevention of premature births by early detection of disturbances].

BACKGROUND: The most frequent cause of preventable early premature births (< 32 + 0 gestational weeks) is urogenital tract infection (UTI). Screening for UTI and Chlamydia are now standard examinations. Detection of ascending genital infections, especially bacterial vaginosis, plays an ever-increasing role. Lactobacilli, which produce, e. g., lactic acid, H2O2, biosurfactants and coaggregation molecules, are particularly important for the vaginal biosystem. Some evaluations suggest that early diagnosis and therapy within this "lactobacilli-protection system" apparently leads to better results
