[Study of renal osteodystrophy by bone biopsy. Age as an independent factor. Diagnostic value of bone remodeling markers].

The spectrum of bone disease in uremic patients on hemodialysis has changed in the last years. Undecalcified bone biopsy with histomorphometric measurements and tetracycline labelling remains the gold standard for diagnosis of the different forms of renal osteodystrophy. But because of its invasive nature and complicated laboratory processing a number of non-invasive biochemical parameters have been proposed. The aim of our study was to determine the prevalence of the different forms of renal osteodystrophy in our patients in hemodialysis. Moreover we analyse the correlation between several bi

Trabecular bone remodeling and bone mineral density in the adult cat during chronic dietary acidification with ammonium chloride.

Ammonium chloride (NH4Cl) is used as a urinary acidifier in the treatment and prevention of feline urologic syndrome. It is reported to cause alterations in calcium and bone metabolism in humans, dogs, and rats. Adult cats with normal renal function were fed 1.5% NH4Cl for 6 months to study the effects of chronic dietary acidification on trabecular bone remodeling of the iliac crest and bone mineral density (BMD) of lumbar vertebral trabecular bone and femoral cortex. Histomorphometric analyses of iliac crest biopsies were performed before and after treatment. Static and dynamic parameters of

A numerical investigation of stress, strain, and bone density changes due to bone remodelling in the talus bone following total ankle arthroplasty.

Total ankle arthroplasty is the gold standard surgical treatment for severe ankle arthritis and fracture. However, revision surgeries due to the in vivo failure of the ankle implant are a serious concern. Extreme bone density loss due to bone remodelling is one of the main reasons for in situ implant loosening, with aseptic loosening of the talar component being one of the primary reasons for total ankle arthroplasty revisions. This study is aimed at determining the performance and potential causes of failure of the talar component. Herein, we investigated the stress, strain, and bone density

Honokiol, a structurally defined natural product, promotes SIRT3-linked bone formation in osteoporotic bone remodeling and fracture repair.

CONTEXT: Honokiol is a structurally defined biphenolic natural product with reported mitochondrial and cytoprotective activities. Whether honokiol modifies pathological skeletal remodeling through sirtuin 3 (SIRT3)-linked mechanisms remains unclear. OBJECTIVE: This study investigated whether honokiol regulates bone cell lineage responses and improves osteoporotic bone remodeling and fracture repair through SIRT3-associated bone formation. MATERIALS AND METHODS: Mouse bone marrow-derived mesenchymal stem cells and RAW264.7 cells were used to assess osteogenesis, adipogenesis, and osteoclastog

The role of the neuro-immune-bone axis in osteoporosis: from bone remodeling imbalance to multi-system interactions.

Osteoporosis is a systemic metabolic bone disease characterized by decreased bone mass, destruction of bone microstructure, and increased risk of fractures. Traditional views mainly attribute it to an imbalance between osteoblast-mediated bone formation and osteoclast-mediated bone resorption, but increasing evidence suggests that the pathogenesis and progression of osteoporosis are also regulated by complex interactions among the nervous system, immune system, and skeletal system. The neuro-immune-bone axis provides an important framework for integrating the bidirectional regulation between i

Synergistic Effect of Poly(ethylenephosphoric Acid) and Cerium in Bone Substitute Composites on Tissue Response and Bone Remodeling.

To reduce the time of postoperative recovery and to prevent post-surgical complications, biocompatible synthetic materials with osteoconductive and osteoinductive properties are used as bone substitutes in large bone defect management. A simplified biomimetic approach to similar materials is based on the use of an inorganic filler, a polymer matrix, and a compatibilizer, mimicking the composition of the natural bone. Based on plate-like micro-sized carbonated hydroxyapatite (pCAp), we prepared compression-molded samples optionally containing an additional polyester component (poly(ε-caprolacto

Marginal Bone Loss Around the Implant: A Retrospective Analysis of Bone Remodeling Over Five Years of Follow-Up.

INTRODUCTION: Bone remodeling around implants in implant-supported rehabilitation is a continuous debate with no consensus in the literature. This study aimed to investigate the implant- and patient-specific factors contributing to marginal bone loss near the implant. MATERIALS AND METHODS: We included patients who had implant-supported prosthetic rehabilitation using one implant system, between 2014 and 2018, who had full follow-up documentation and orthopantomography over five years, and who had no unwell-controlled systemic pathologies that may influence bone metabolism. RESULTS: Eighty-o

An Additional Lrp4 High Bone Mass Mutation Mitigates the Sost-Knockout Phenotype in Mice by Increasing Bone Remodeling.

Pathogenic variants disrupting the binding between sclerostin (encoded by SOST) and its receptor LRP4 have previously been described to cause sclerosteosis, a rare high bone mass disorder. The sclerostin-LRP4 complex inhibits canonical WNT signaling, a key pathway regulating osteoblastic bone formation and a promising therapeutic target for common bone disorders, such as osteoporosis. In the current study, we crossed mice deficient for Sost (Sost-/-) with our p.Arg1170Gln Lrp4 knock-in (Lrp4KI/KI) mouse model to create double mutant Sost-/-;Lrp4KI/KI mice. We compared the phenotype of Sost-/-

In-vivo bone remodeling potential of Sr-d-Ca-P /PLLA-HAp coated biodegradable ZK60 alloy bone plate.

Magnesium and its alloys are widely applied biomaterials due to their biodegradability and biocompatibility. However, rapid degradation and hydrogen gas evolution hinder its applicability on a commercial scale. In this study, we developed an Mg alloy bone plate for bone remodeling and support after a fracture. We further coated the Mg alloy plate with Sr-D-Ca-P (Sr dopped Ca-P coating) and Sr-D-Ca-P/PLLA-HAp to evaluate and compare their biodegradability and biocompatibility in both in vitro and in vivo experiments. Chemical immersion and dip coating were employed for the formation of Sr-D-Ca-

Role of osteogenic Dickkopf-1 in bone remodeling and bone healing in mice with type I diabetes mellitus.

Type 1 diabetes mellitus (T1DM) is associated with low bone mass and a higher risk for fractures. Dickkopf-1 (Dkk1), which inhibits Wnt signaling, osteoblast function, and bone formation, has been found to be increased in the serum of patients with T1DM. Here, we investigated the functional role of Dkk1 in T1DM-induced bone loss in mice. T1DM was induced in 10-week-old male mice with Dkk1-deficiency in late osteoblasts/osteocytes (Dkk1f/f;Dmp1-Cre, cKO) and littermate control mice by 5 subsequent injections of streptozotocin (40 mg/kg). Age-matched, non-diabetic control groups received citrate

Molecular Signaling Pathways and Essential Metabolic Elements in Bone Remodeling: An Implication of Therapeutic Targets for Bone Diseases.

Bone is one of the dynamic tissues in the human body that undergoes continuous remodelling through subsequent actions of bone cells, osteoclasts, and osteoblasts. Several signal transduction pathways are involved in the transition of mesenchymal stem cells into osteoblasts. These primarily include Runx2, ATF4, Wnt signaling and sympathetic signalling. The differentiation of osteoclasts is controlled by M-CSF, RANKL, and costimulatory signalling. It is well known that bone remodelling is regulated through receptor activator of nuclear factor-kappa B ligand followed by binding to RANK, which eve

Bone-targeted delivery of TGF-β type 1 receptor inhibitor rescues uncoupled bone remodeling in Camurati-Engelmann disease.

Camurati-Engelmann disease (CED) is a genetic bone-modeling disorder mainly caused by mutations in the gene that encodes transforming growth factor-β1 (TGF-β1). Symptoms of CED include bone pain, fractures, and dysplasia. Currently, effective therapies for bone fracture and dysplasia in CED are urgently needed. We have demonstrated that TGF-β1 is a coupling factor for bone remodeling and is aberrantly activated in CED. Daily injection of TGF-β type 1 receptor inhibitor (TβR1I) attenuated CED symptoms, but this systemic administration caused serious side effects. In this study, we created a con

Osteoblast retraction induced by adherent neutrophils promotes osteoclast bone resorption: implication for altered bone remodeling in chronic gout.

Bone destruction in chronic gout is correlated with deposits of monosodium urate (MSU) crystals. Bone with MSU tophi were histopathologically shown to have altered remodeling and cellular distribution. We investigated the impact of neutrophils in bone remodeling associated with MSU and demonstrated that neutrophils, through elastase localized at their surface, induced retraction of confluent osteoblasts (OBs) previously layered on calcified matrix. This OB retraction allowed osteoclasts to resorb cell-free areas of the matrix. This neutrophil effect was concentration dependent and time depende

High-dose therapy improves the bone remodelling compartment canopy coverage and bone formation in multiple myeloma.

Bone loss in multiple myeloma (MM) is caused by an uncoupling of bone formation to resorption trigged by malignant plasma cells. Increasing evidence indicates that the bone remodelling compartment (BRC) canopy, which normally covers the remodelling sites, is important for coupled bone remodelling. Loss of this canopy has been associated with bone loss. This study addresses whether the bone remodelling in MM is improved by high-dose therapy. Bone marrow biopsies obtained from 20 MM patients, before and after first-line treatment with high-dose melphalan followed by autologous stem cell transpla

In vivo monitoring of bone architecture and remodeling after implant insertion: The different responses of cortical and trabecular bone.

The mechanical integrity of the bone-implant system is maintained by the process of bone remodeling. Specifically, the interplay between bone resorption and bone formation is of paramount importance to fully understand the net changes in bone structure occurring in the peri-implant bone, which are eventually responsible for the mechanical stability of the bone-implant system. Using time-lapsed in vivo micro-computed tomography combined with new composite material implants, we were able to characterize the spatio-temporal changes of bone architecture and bone remodeling following implantation i

[Is there a bone-preserving bone remodelling in short-stem prosthesis? DEXA analysis with the Nanos total hip arthroplasty].

BACKGROUND: It has been suggested that the use of a short-stem prosthesis could conserve proximal bone by proximal load transfer. Proximal stress shielding should be reduced, a phenomenon that has been associated with bone resorption around traditional stems. Bone remodelling of a metaphyseal fixed stem (Nanos, Smith & Nephew Int.) was analysed by the dual-energy x-ray absorptiometry. PATIENTS AND METHOD: This study included 36 patients undergoing the total hip replacement using the Nanos short stem in comparison to 36 patients operated by a traditional long-stemmed femoral stem (Alloclassic)

Roles of gut metabolites on bone mineralization and remodeling: impact of metabolic dysregulation on the gut-immune-bone axis.

Gut microbiota, the community of microorganisms in the gastrointestinal (GI) tract, plays many critical roles in the maintenance of an individual's health by regulating bone metabolism. Bone mass has been linked to the GI system because nutrients (minerals) are absorbed to maintain bone homeostasis. Dysbiosis of gut microbiota due to metabolic deregulation associated with obesity, fatty liver, and inflammatory status of an individual promotes altered gut microbial diversity, gut-epithelial integrity, releases of harmful gut metabolites those could mediate malabsorption of nutrients such as cal
