Nickel toxicity in embryos and larvae of the South American toad: effects on cell differentiation, morphogenesis, and oxygen consumption.

Nickel, a widely distributed heavy metal in the biosphere, produces systemic, carcinogenic, and teratogenic effects. The objectives of the present study are to report the acute, short-term chronic, and chronic toxicity of Ni in Rhinella arenarum embryos as well as the stage-dependent susceptibility to this heavy metal, including oxygen consumption, teratogenesis, and adverse effects on cell differentiation processes. The stages evaluated were blastula (S.7), gastrula (S.11), tail bud (S.17), fin circulation (S.22), and complete operculum (S.25), in this last case by means of toxicity profile c

The GTP-binding protein of rod outer segments. II. An essential role for Mg2+ in signal amplification.

The role of Mg2+ in the GTP hydrolytic cycle was investigated by using purified subunits (G alpha and G beta, gamma) of the GTP-binding protein isolated from Bufo marinus rod outer segments (ROS). Mg2+ markedly stimulated the rate of GTP and guanosine-5'-O-(3-thiotriphosphate) (GTP gamma-s) binding to G alpha. This effect was especially striking in the presence of very small quantities of illuminated ROS disc membranes. GTP hydrolysis could occur in the absence of Mg2+, and Mg2+ increased the rate of GTP hydrolysis only about 50%. These data indicate that Mg2+ plays a fundamental role in ampli

Age-related changes in carbonic anhydrase of toad lens.

Carbonic anhydrase in the toad lens is immunologically identical to the (high activity) enzyme form present in the erythrocyte. As with the erythrocyte carbonic anhydrase, electrofocusing of the freshly-extracted lens enzyme separated three variants with isoelectric points of 6.1, 5.7 and 5.4, all of which exhibited inhibition properties characteristic of the high activity type of carbonic anhydrase. No evidence of the low activity form of the enzyme was found. In contrast to the erythrocyte isoelectric variants, which are stable in vitro, the three variants of lens carbonic anhydrase exhibite

Fluoride inhibition of the hydro-osmotic response of the toad urinary bladder to antidiuretic hormone.

The hydro-osmotic response of the toad bladder to antidiuretic hormone and cyclic AMP was inhibited by the methoxyflurane metabolite, fluoride. The osmotic transfer of water in the absence of hormone was unaffected by fluoride as was the hydroosmotic response due to hypertonicity of the serosal bathing media. Osmotic water movements across N-ethylmaleimide-"fixed" vasopressin or cyclic AMP-stimulated bladders were likewise unchanged by fluoride, suggesting that fluoride is exerting an action subsequent to the endogenous formation of cyclic AMP but before the final effector mechanism. Fluoride

Changes in the water and sodium permeability of amphibian skin in the presence of the efferent plasma of "dehydrated dog kidney".

Two kidneys of the same dog were perfused separately with the same blood pool. After the control periods one of the kidneys was replaced by another one taken from a previously dehydrated dog. The presence of the "dehydrated kidney" resulted in a significant and almost immediate decrease in creatinin clearance of the contralateral perfused kidney. The Ccreat increased after the removal of the dehydrated kidney. Plasma samples were taken during the control, transplantation and recovery periods and their activity on the water permeability (osmotic water flow) and sodium transport was examined in

Chronic suppurative otitis media in cleft palate: microorganism etiology and susceptibilities.

The adverse effects of ultraviolet B radiation from 547.2 to 30,096 J/m2 on morphogenesis, cell differentiation, and lethality of amphibian embryos at six developmental stages were evaluated from 24 up to 168 h postexposure. The ultraviolet B radiation lethal dose 10, 50, and 90 values were obtained for all developmental stages evaluated. The lethal dose 50 values, considered as the dose causing lethality in the 50% of the organisms exposed, in J/m2 at 168 h postexposure, ranged from 2,307 to 18,930; gill circulation and blastula were the most susceptible and resistant stages, respectively. Vertebrate phylogeny of hydrogen sulfide vasoactivity.

Hydrogen sulfide (H(2)S) is a recently identified endogenous vasodilator in mammals. In steelhead/rainbow trout (Oncorhynchus mykiss, Osteichthyes), H(2)S produces both dose-dependent dilation and a unique dose-dependent constriction. In this study, we examined H(2)S vasoactivity in all vertebrate classes to determine whether H(2)S is universally vasoactive and to identify phylogenetic and/or environmental trends. H(2)S was generated from NaHS and examined in unstimulated and precontracted systemic and, when applicable, pulmonary arteries (PA) from Pacific hagfish (Eptatretus stouti, Agnatha),

Some aspects of the hypothalamic and pituitary development, metamorphosis, and reproductive behavior as studied in amphibians.

In this review article, information about the development of the hypothalamo-hypophyseal axis, endocrine control of metamorphosis, and hormonal and pheromonal involvements in reproductive behavior in some amphibian species is assembled from the works conducted mainly by our research group. The hypothalamic and pituitary development was studied using Bufo embryos and larvae. The primordium of the epithelial hypophysis originates at the anterior neural ridge and migrates underneath the brain to form a Rathke's pouch-like structure. The hypothalamo-hypophyseal axis develops under the influence of

Bioaccumulation and maternal transfer of mercury and selenium in amphibians.

Amphibian population declines have been documented worldwide and environmental contaminants are believed to contribute to some declines. Maternal transfer of bioaccumulated contaminants to offspring may be an important and overlooked mechanism of impaired reproductive success that affects amphibian populations. Mercury (Hg) is of particular concern due to its ubiquity in the environment, known toxicity to other wildlife, and complex relationships with other elements, such as selenium (Se). The objectives of the present study were to describe the relationships between total Hg (THg), methlymerc

Light-induced calcium release and re-uptake in toad rods.

Laser micromass analysis was used to investigate the effect of light on the Ca content of rod outer segments in the isolated retina and eyecup of the toad, Bufo marinus. Isolated retinas were incubated in Ringer for which most of the Ca (normally 97% 40Ca) was replaced with 44Ca, so that release of internal Ca (as 40Ca) could be distinguished from the uptake of 44Ca from the external medium. Continuous illumination produced a decline in outer segment 40Ca content. In bright light, the decrease in 40Ca was sigmoidal, beginning with a delay of at least 4 sec, reaching a maximal rate of 1-2 x 10(

Effects of calcium and guanosine-3',5'-cyclic-monophosphoric acid on receptor potentials of toad rods.

Membrane potential was recorded intracellularly from rod outer segments in the isolated retina of Bufo marinus. The composition of the solution flowing over the exposed receptor side of the retina was changed rapidly and ionophoretic injections were made into single rod outer segments. Lowering the external Ca2+ concentration, adding 3-isobutyl-1-methylxanthine (IBMX) to the bath, or ionophoretic injections of guanosine-3':5'-cyclic monophosphoric acid (cGMP) lead to depolarization of the rod membrane and an increase in the maximum peak amplitude of the receptor potential. Lowering the Ca2+ co

A single inhalation of vapor from dried toad secretion containing 5-methoxy-N,N-dimethyltryptamine (5-MeO-DMT) in a naturalistic setting is related to sustained enhancement of satisfaction with life, mindfulness-related capacities, and a decrement of psychopathological symptoms.

BACKGROUND: 5-methoxy-N,N-dimethyltryptamine (hereinafter referred to as 5-MeO-DMT) is a psychedelic substance found in the secretion from the parotoid glands of the Bufo alvarius toad. Inhalation of vapor from toad secretion containing 5-MeO-DMT has become popular in naturalistic settings as a treatment of mental health problems or as a means for spiritual exploration. However, knowledge of the effects of 5-MeO-DMT in humans is limited. AIMS: The first objective of this study was to assess sub-acute and long-term effects of inhaling vapor from dried toad secretion containing 5-MeO-DMT on aff

Cardiorespiratory effects of forced activity and digestion in toads.

Digestion and physical activity are associated with large and sometimes opposite changes in several physiological parameters. Gastric acid secretion during digestion causes increased levels of plasma bicarbonate ([HCO-3](pl)), whereas activity leads to a metabolic acidosis with increased lactate and decrease in plasma bicarbonate. Here we describe the combined effects of feeding and activity in the toad Bufo marinus to investigate whether the increased bicarbonate buffering capacity during digestion (the so-called alkaline tide) protects the acid-base disturbance during activity and enhances t

Spatial spread of light-induced sensitization in rod photoreceptors exposed to low external calcium.

Light sensitizes rods that have been desensitized by exposure to low external calcium. Yoshikami and Hagins [Biophys. Soc. Abstr. 15, 169a (1975)] suggested that desensitization in low external calcium results from exposure of intracellular calcium binding sites subsequent to depletion of internal calcium, and that background light sensitizes in this situation by releasing calcium to occupy those binding sites. In this view, it might be expected that light-induced sensitization would be spatially restricted to the illuminated region of the outer segment. However, in the present experiments, ba

Energetics of sodium transport in the urinary bladder of the toad. Effect of aldosterone and sodium cyanide.

Experiments were designed to determine whether the stimulatory effect of aldosterone on sodium transport involves an increase in tissue ATP. Urinary bladders that were removed from toads presoaked in 0.6% saline for 48-72 h, mounted as sacs, and maintained in open circuit except for brief observation of short circuit current every 30 min responded to 100 nM aldosterone added to the serosal bath with an increase in short circuit current to 170% of control hemibladders, which plateaus at 2-3 h. Tissue (ATP)/(ADP) X (Pi) measured in perchloric acid extracts increased to a maximum of 208% of contr

Effects of metabolic and transport inhibitors on sodium transport and CO2 production of toad bladders: use of an uncoupling agent to distinguish between effects on transport and metabolism.

Agents that directly inhibit sodium transport secondary reduce the rat of metabolism. Conversely, agents that directly inhibit energy metabolism cause a secondary inhibition of active transport. The aim of the present investigation was to distinguish between these two effects with an uncoupling agent, 2,4-dinitrophenol (DNP). The rates of sodium transport (TNa) and CO2 production (QCO2) by paired urinary hemibladders from Dominican toads were simultaneously measured. DNP alone inhibited TNa, but stimulated QCO2 of the hemibladders, consistent with its action as an uncoupling agent. Ouabain, wh

Action of vasopressin, ouabain, and cyanide on the volume of isolated toad bladder epithelial cells.

Toad bladder epithelial cells were isolated under mild conditions in a calcium-free medium; they were found to exclude trypan blue, to consume oxygen, and to respond to vasopressin with an increased rate of oxygen consumption. Since isolated toad bladder epithelial cells are mostly spherical in shape, the cell diameter can be accurately measured with an ocular micrometer of an inverted microscope. Epithelial cells swelled by 29+/-3% in the presence of KCN. This cyanide-induced swelling of cells was prevented by amiloride or, alternatively, by replacing NaCl by equiosmotic amounts of mannitol i

Effect of lactate on depolarization-induced Ca(2+) release in mechanically skinned skeletal muscle fibers.

It is unclear whether accumulation of lactate in skeletal muscle fibers during intense activity contributes to muscle fatigue. Using mechanically skinned fibers from rat and toad muscle, we were able to examine the effect of L(+)-lactate on excitation-contraction coupling independently of other metabolic changes. We investigated the effects of lactate on the contractile apparatus, caffeine-induced Ca(2+) release from the sarcoplasmic reticulum, and depolarization-induced Ca(2+) release. Lactate (15 or 30 mM) had only a small inhibitory effect directly on the contractile apparatus and caused ap

Detection of changes in near-membrane Ca2+ concentration using a novel membrane-associated Ca2+ indicator.

A Ca2+ indicator has been synthesized and characterized which can be used to monitor rapid changes in the free Ca2+ concentration ([Ca2+]) immediately adjacent to cell membranes. This indicator, referred to as C18-Fura-2, consists of a Fura-2 molecule conjugated to a lipophilic alkyl chain which will insert into cell membranes. When associated with cell membranes in low concentrations, C18-Fura-2 exhibits an excitation spectrum with a large Stokes shift and a single isobestic point, thus [Ca2+] can be calculated ratiometrically. The apparent Ca2+ dissociation constant of cell-associated C18-Fu
