Detection of aspartate kinase in rat liver and its activation by Ca++ and calmodulin.

The enzyme aspartate kinase (EC 2.7.2.4) has been detected in rat liver (animal tissue) for the first time. This enzyme, like the aspartate kinase from bacteria and plants is inhibited by lysine and threonine. Further, the activity of the enzyme is stimulated over two fold by Ca++ and calmodulin and inhibited by EGTA, a Ca++ chelator and trifluoperazine, an anti-calmodulin compound.

Does calmodulin participate as an intermediate in the process of glycogenolysis in rat submandibular gland?

A role of calmodulin in the process of the glycogenolysis induced by adrenaline was investigated in rat submandibular gland. When the slice was incubated in the Ca2+-free medium, the adrenaline-induced glycogenolysis was significantly inhibited. Moreover, the inhibition of glycogenolysis induced in the Ca2+-free medium was further enhanced by addition of 2 mM EGTA. Chlorpromazine, trifluoperazine and W-7, calmodulin antagonists, had no effect on the glycogenolysis induced by adrenaline. These results suggest that the adrenaline-induced glycogenolysis is dependent on the presence of Ca2+ and th

Phase shifting of the circadian conidiation rhythm in Neurospora crassa by calmodulin antagonists.

The effects of chemicals capable of antagonizing the functions of calmodulin, such as trifluoperazine, chlorpromazine, imipramine, alprenolol, W7, and W13, on the circadian conidiation rhythm of Neurospora crassa were examined. Trifluoperazine, at a 30-microM concentration, was most effective in shifting the phase of the conidiation rhythm and caused a maximum phase delay at circadian time (CT) 6 and maximum phase advance at CT 9. Chlorpromazine was less effective than trifluoperazine, and a 300-microM concentration of chlorpromazine was required for a similar phase shift. Imipramine, at a 1-m

The inhibitory effects of calmodulin antagonists on the endothelium-dependent relaxation in rabbit aorta.

Effects of trifluoperazine (TFP, 10(-5) M), trifluoperazine sulfoxide (TFP-SO, 10(-5) M), N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide (W-7-, 10(-4) M), N-(6-aminohexyl)-1-naphthalenesulfonamide (W-5, 10(-4) M) and verapamil (10(-5) M) on the endothelium-dependent relaxation of the contractions induced in the isolated rabbit aorta were studied. Carbachol (10(-6) - 10(-6) M) and A23187 (10(-8) M) relaxed the contraction due to norepinephrine (10(-6) M and histamine (10(-5) M) in the aorta with endothelium. TFP and W-7 completely inhibited the relaxation induced by carbachol and A23187. In

Effect of calmodulin antagonists on hypoxia and reoxygenation damage in isolated rabbit hearts.

The effect of calmodulin antagonists trifluoperazine (2.5 X 10(-7) M and 2.5 X 10(-6) M) and R 24571 (10(-8) M and 10(-7) M) on Langendorff-perfused rabbit hearts subjected to 180 min hypoxia and 30 min reoxygenation was studied. Coronary flow, force of contraction, oxygen consumption and release of lactate, noradrenaline and LDH were measured. Both drugs were found to reduce some of the deleterious consequences of hypoxia, i.e., they caused: marked reduction in the hypoxic LDH release; reduction in hypoxic contracture; increased recovery of active tension, oxygen consumption and coronary flow

Effect of calmodulin inhibitors on viability and mitochondrial potential of Plasmodium falciparum in culture.

Calmodulin inhibitors are toxic for a variety of protozoa. Chlorpromazine, calmidazolium, and trifluoperazine inhibited the incorporation of [3H]hypoxanthine and [3H]phenylalanine into Plasmodium falciparum organisms in cultures with 50% inhibitory concentrations varying from 5.1 microM (with calmidazolium) to 48 microM (with chlorpromazine), the former being more sensitive than the latter. However, these concentrations also immediately dissipated rhodamine 123 from the parasite mitochondrion. Similar concentrations inhibit other protozoa, as well as mammalian cells, and the possibility that m

Interaction of smooth muscle relaxant drugs with calmodulin and cyclic nucleotide phosphodiesterase.

Some smooth muscle relaxant drugs with an unknown mechanism of action have been tested for their interaction with calmodulin and with calmodulin-induced cyclic nucleotide phosphodiesterase (PDE) activity. The affinity of these drugs for calmodulin does not parallel their inhibitory effect on the calmodulin activation of PDE. The lack of parallelism could be due to a binding of the drugs to different sites on calmodulin; furthermore a binding of papaverine, octylonium bromide and felodipine to PDE molecule might also be considered to explain their inhibitory effect on PDE basal activity. The my

Structural and functional properties of calmodulin from the eukaryotic microorganism Dictyostelium discoideum.

Calmodulin was purified from the eukaryotic microorganism Dictyostelium discoideum and characterized in terms of its nearly complete primary structure and quantitative activator activity. The strategy for amino acid sequence analysis took advantage of the highly conserved structure of calmodulin and employed a new procedure for limited cleavage of calmodulin that uses a protease from mouse submaxillary gland. Fourteen amino acid sequence differences between Dictyostelium and bovine calmodulin were identified unequivocally, as well as an unmethylated lysine at residue 115 instead of N epsilon,

Calmodulin: The Essential Biological Regulator Self-Regulates Its Activity, Structure and Function.

Calmodulin is an evolutionarily conserved critical controller protein found in animals, plants, fungi, and microbes. As the primary calcium sensor and effector in eukaryotes, calmodulin translates calcium signals into vital biological responses by binding to and regulating calmodulin-binding proteins. While it was previously shown to control several hundred proteins, evidence now indicates it binds to over 100 in its calcium-free state and more than 500 when activated by calcium ions. While calmodulin function is modulated by calcium levels, the final cytosolic levels of calcium are often unde

Calmodulin kinase II inhibition suppresses atrioventricular conduction by regulating intracellular Ca2+ homeostasis.

BACKGROUND: Ca2+/calmodulin-dependent protein kinase II (CaMKII) inhibition decelerates atrioventricular node (AVN) conduction, providing a potential treatment of tachycardia. However, the effectiveness of CaMKII inhibition on tachycardia and its underlying mechanism remains unclear. OBJECTIVE: We aimed to assess the effectiveness of CaMKII inhibition in reducing ventricular rates during atrial fibrillation and to elucidate the underlying mechanism in affecting AVN electrophysiology. METHODS: Cardiac CaMKII inhibition (AC3-I) mice were used. Transesophageal atrial pacing was performed to eva

Calmodulin-Cork Model of Gap Junction Channel Gating-One Molecule, Two Mechanisms.

The Calmodulin-Cork gating model is based on evidence for the direct role of calmodulin (CaM) in channel gating. Indeed, chemical gating of cell-to-cell channels is sensitive to nanomolar cytosolic calcium concentrations [Ca2+]i. Calmodulin inhibitors and inhibition of CaM expression prevent chemical gating. CaMCC, a CaM mutant with higher Ca2+-sensitivity greatly increases chemical gating sensitivity (in CaMCC the NH2-terminal EF-hand pair (res. 9-76) is replaced by the COOH-terminal pair (res. 82-148). Calmodulin colocalizes with connexins. Connexins have high-affinity CaM binding sites. Sev

Ca2+/calmodulin-dependent protein kinase II is essential in hyperacute pressure overload.

BACKGROUND: Activation of Ca2+/calmodulin-dependent protein kinase II (CaMKII) is established as a central intracellular trigger for various cardiac pathologies such as hypertrophy, heart failure or arrhythmias in animals and humans suggesting CaMKII as a promising target protein for future medical treatments. However, the physiological role of CaMKII is scarcely well defined. AIM & METHODS: To investigate the role of CaMKII in hyperacute pressure overload, we evaluated the effects of pressure overload induced by transverse aortic constriction (TAC) on survival, cardiac function, protein expr

Salvia miltiorrhiza causes tonic contraction in rat ileum through Ca²⁺-calmodulin pathway.

ETHNOPHARMACOLOGICAL RELEVANCE: Danshen, root of Salvia miltiorrhiza (SM), has been traditionally used in Chinese medicine for the treatment of heart, abdomen, gurgling in the intestines, and relieving fullness. However, the effects of SM on intestine have rarely been done to date. AIM OF THE STUDY: To investigate the contraction effect of SM on isolated rat ileum and its mechanisms involved. MATERIALS AND METHODS: The isometric contractions of ileum segments were investigated in organ baths for spontaneous activity and response to ethanolic extracts of SM. To determine the contraction mecha

Antiamnesic effect of B. monniera on L-NNA induced amnesia involves calmodulin.

Amnesia may result from ageing, chronic drug abuse or head injury and there are limited therapeutic strategies to such conditions. We have shown that Bacopa monniera, a memory enhancing drug can reverse both diazepam and scopolamine induced amnesia in mice. In order to understand the downstream effects of B. monniera, this study was designed to investigate how B. monniera antagonizes MK801, an NMDA receptor antagonist and N(omega)-Nitro-L-arginine (L-NNA), a nitric oxide synthase inhibitor. We compared the degree of reversal B. monniera imparts on MK801 and L-NNA induced anterograde amnesia in

Ca2+/calmodulin-dependent protein kinase II in the rat cranial sensory ganglia.

Immunohistochemistry for Ca(2+)/calmodulin-dependent protein kinase II (CaMKII) was performed on the rat cranial sensory ganglia. More than one half of neurons was immunoreactive for the enzyme in the trigeminal (60%), jugular (70%), petrosal (55%) and nodose ganglia (63%). These neurons were mainly small to medium-sized. The co-expression study demonstrated that one half of CaMKII-immunoreactive (ir) neurons was also immunoreactive for calcitonin gene-related peptide (CGRP) or the vanilloid receptor subtype 1 (VR1) in the trigeminal, jugular and petrosal ganglia. In the nodose ganglion, CaMKI

Ca2+/calmodulin-dependent protein kinase II phosphorylation regulates the cardiac ryanodine receptor.

The cardiac ryanodine receptor (RyR2)/calcium release channel on the sarcoplasmic reticulum is required for muscle excitation-contraction coupling. Using site-directed mutagenesis, we identified the specific Ca2+/calmodulin-dependent protein kinase II (CaMKII) phosphorylation site on recombinant RyR2, distinct from the site for protein kinase A (PKA) that mediates the "fight-or-flight" stress response. CaMKII phosphorylation increased RyR2 Ca2+ sensitivity and open probability. CaMKII was activated at increased heart rates, which may contribute to enhanced Ca2+-induced Ca2+ release. Moreover,

Physiological calcium concentrations regulate calmodulin binding and catalysis of adenylyl cyclase exotoxins.

Edema factor (EF) and CyaA are calmodulin (CaM)-activated adenylyl cyclase exotoxins involved in the pathogenesis of anthrax and whooping cough, respectively. Using spectroscopic, enzyme kinetic and surface plasmon resonance spectroscopy analyses, we show that low Ca(2+) concentrations increase the affinity of CaM for EF and CyaA causing their activation, but higher Ca(2+) concentrations directly inhibit catalysis. Both events occur in a physiologically relevant range of Ca(2+) concentrations. Despite the similarity in Ca(2+) sensitivity, EF and CyaA have substantial differences in CaM binding

Ca2+/calmodulin-dependent facilitation and inactivation of P/Q-type Ca2+ channels.

Trains of action potentials cause Ca(2+)-dependent facilitation and inactivation of presynaptic P/Q-type Ca(2+) channels that can alter synaptic efficacy. A potential mechanism for these effects involves calmodulin, which associates in a Ca(2+)-dependent manner with the pore-forming alpha(1A) subunit. Here, we report that Ca(2+) and calmodulin dramatically enhance inactivation and facilitation of P/Q-type Ca(2+) channels containing the auxiliary beta(2a) subunit compared with their relatively small effects on channels with beta(1b). Tetanic stimulation causes an initial enhancement followed by
