Vinpocetine protects against chloroquine-induced cardiotoxicity by mitigating oxidative stress.

Chloroquine (CQ) and hydroxychloroquine (HCQ) are classical antimalarial drugs, and recently have been used for other applications including coronavirus disease 2019 (COVID-19). Although they are considered safe, cardiomyopathy may associate CQ and HCQ applications particularly at overdoses. The goal of the present study was to evaluate the potential protective effect of the nootropic agent vinpocetine against CQ and HCQ adverse effects with a specific focus on the heart. For this purpose, a mouse model of CQ (0.5 up to 2.5 g/kg)/HCQ (1 up to 2 g/kg) toxicity was used, and the effect of vinpoc

Oxidative stress contributes to flumioxazin-induced cardiotoxicity in zebrafish embryos.

Flumioxazin is a widely applied herbicide for the control of broadleaf weeds, including aquatic plants. Current evidence suggests that flumioxazin could induce cardiac defects (ventricular septal defects) in vertebrates, but the underlining mechanisms remain unclear. Because of the inhibitory effect of flumioxazin on polyphenol oxidase, the assumption is made that flumioxazin-induced cardiotoxicity is caused by oxidative stress. To verify whether oxidative stress plays an important role in flumioxazin-induced cardiotoxicity, we compared the differences in heart phenotype, oxidative stress leve

Cocaethylene cardiotoxicity in emergency department patients with acute drug overdose.

OBJECTIVES: Cocaine use results in over 500,000 emergency department (ED) visits annually across the United States and ethanol co-ingestion is reported in 34% of these. Commingling cocaine with ethanol results in the metabolite cocaethylene (CE), which is metabolically active for longer than cocaine alone. Current literature on the cardiotoxicity of CE compared to cocaine alone is limited and lacks consensus. This study aims to fill this gap in the literature and examine cardiovascular events in cocaine use as confirmed by urine toxicology versus CE exposure. METHODS: This was a secondary dat

Drug-induced QT Prolongation Atlas (DIQTA) for enhancing cardiotoxicity management.

Drug-induced prolongation of the QT interval is common in a variety of pharmaceutical treatments and can lead to serious clinical outcomes. Although substantial efforts have been made to prevent drug-induced QT interval prolongation, the lack of a centralized data source remains the main obstacle to further study of the underlying mechanism and the development of effective prediction strategies. To fill this gap, we propose a schema for stratifying the risk of marketed QT prolonging drugs based on US Food and Drug Administration (FDA)-approved drug labeling and developed a Drug-Induced QT Prol

Compound Danshen Dripping Pill inhibits doxorubicin or isoproterenol-induced cardiotoxicity.

Heart failure (HF) is the advanced heart disease with high morbidity and mortality. Compound DanShen Dripping Pill (CDDP) is a widely used Traditional Chinese Medicine for cardiovascular disease treatment. Herein, we investigated if CDDP can protect mice against doxorubicin (DOX) or isoprenaline (ISO)-induced HF. After 3 days feeding of normal chow containing CDDP, mice were started DOX or ISO treatment for 4 weeks or 18 days. At the end of treatment, mice were conducted electrocardiogram and echocardiographic test. Blood and heart samples were determined biochemical parameters, myocardial str

Isorhapontigenin protects against doxorubicin-induced cardiotoxicity via increasing YAP1 expression.

As an effective anticancer drug, the clinical limitation of doxorubicin (Dox) is the time- and dose-dependent cardiotoxicity. Yes-associated protein 1 (YAP1) interacts with transcription factor TEA domain 1 (TEAD1) and plays an important role in cell proliferation and survival. However, the role of YAP1 in Dox-induced cardiomyopathy has not been reported. In this study, the expression of YAP1 was reduced in clinical human failing hearts with dilated cardiomyopathy and Dox-induced in vivo and in vitro cardiotoxic model. Ectopic expression of Yap1 significantly blocked Dox-induced cardiomyocytes

Melatonin attenuates doxorubicin-induced cardiotoxicity through preservation of YAP expression.

There are increasing concerns related to the cardiotoxicity of doxorubicin in the clinical setting. Recently, melatonin has been shown to exert a cardioprotective effect in various cardiovascular diseases, including cardiotoxic conditions. In this study, we examined the possible protective effects of melatonin on doxorubicin-induced cardiotoxicity and explored the underlying mechanisms related to this process. We found that in vitro doxorubicin treatment significantly decreased H9c2 cell viability and induced apoptosis as manifested by increased TUNEL-positive cells, down-regulation of anti-ap

Heart slice culture system reliably demonstrates clinical drug-related cardiotoxicity.

The limited availability of human heart tissue and its complex cell composition are major limiting factors for the reliable testing of drug efficacy and toxicity. Recently, we developed functional human and pig heart slice biomimetic culture systems that preserve the viability and functionality of 300 μm heart slices for up to 6 days. Here, we tested the reliability of this culture system for testing the cardiotoxicity of anti-cancer drugs. We tested three anti-cancer drugs (doxorubicin, trastuzumab, and sunitinib) with known different mechanisms of cardiotoxicity at three concentrations and a

miR-451 Silencing Inhibited Doxorubicin Exposure-Induced Cardiotoxicity in Mice.

Oxidative stress and cardiomyocytes apoptosis were closely involved in the pathological process of doxorubicin- (Dox-) induced cardiac injury. MicroRNA-451 (miR-451) was mainly expressed in cardiomyocytes. However, the role of miR-451 in Dox-induced cardiac injury remained unclear. Our study aimed to investigate the effect of miR-451 on Dox-induced cardiotoxicity in mice. We established a Dox-induced cardiotoxicity model in the mice and manipulated miR-451 expression in the heart using a miR-451 inhibitor, which was injected every other day beginning at one day before Dox injection. Oxidative

Lycopene Attenuates Tulathromycin and Diclofenac Sodium-Induced Cardiotoxicity in Mice.

Recent experiments showed a potential cardiotoxic effect of the macrolide antibiotic (tulathromycin). This study was performed to investigate whether diclofenac sodium (DFS) potentiates the cardiotoxicity of tulathromycin and increases the cardioprotective effects of lycopene against DFS and tulathromycin. Seven groups (eight per group) of adult Swiss albino mice received saline (control), tulathromycin (a single subcutaneous dose of 28 mg/kg/bw on day 14), DFS (a single oral dose of 100 mg/kg/bw on day 14), tulathromycin plus DFS, or lycopene (oral, 10 mg/kg/bw daily for 15 d) combined with t

Sab mediates mitochondrial dysfunction involved in imatinib mesylate-induced cardiotoxicity.

Imatinib mesylate is an effective treatment for chronic myelogenous leukemia and gastrointestinal stromal tumors. Although imatinib mesylate is highly tolerable, it has been implicated in severe congestive heart failure in mouse models and patients. A hallmark of imatinib mesylate-induced cardiotoxicity is mitochondrial dysfunction. The mitochondrial scaffold Sab has been implicated in facilitating signaling responsible for mitochondrial dysfunction in a c-Jun N-terminal Kinase (JNK)-dependent manner. We examined the impact of Sab-mediated signaling on imatinib mesylate cardiotoxicity in H9c2

Lipid Emulsion, More Than Reversing Bupivacaine Cardiotoxicity: Potential Organ Protection.

Efforts to develop a treatment for bupivacaine cardiotoxicity led to the discovery that Intralipid, a popular brand of intravenous lipid emulsion, could be used not only as an effective treatment for anesthetic-induced cardiac arrest, but also as a means of reversing many other toxicities. Contradictory data exist regarding the mechanism of action of lipid emulsion, a combination of fatty acids traditionally used in parenteral nutrition. Some researchers attribute the effects to lipophilicity and the individual characteristics of the lipids, while other data demonstrate a direct empowering mec

Ataxia telangiectasia mutated in cardiac fibroblasts regulates doxorubicin-induced cardiotoxicity.

AIMS: Doxorubicin (Dox) is a potent anticancer agent that is widely used in the treatment of a variety of cancers, but its usage is limited by cumulative dose-dependent cardiotoxicity mainly due to oxidative damage. Ataxia telangiectasia mutated (ATM) kinase is thought to play a role in mediating the actions of oxidative stress. Here, we show that ATM in cardiac fibroblasts is essential for Dox-induced cardiotoxicity. METHODS AND RESULTS: ATM knockout mice showed attenuated Dox-induced cardiotoxic effects (e.g. cardiac dysfunction, apoptosis, and mortality). As ATM was expressed and activated

Flecainide cardiotoxicity precipitated by electrolyte imbalance. Caution with thiazide diuretics.

A man presented with recurrent syncope, weakness and fatigue. His ECG showed marked QRS widening and he had gross hyponatraemia and hypokalaemia. His medications included bendroflumethiazide (long term) and flecainide (started 2 months previously). This presentation was consistent with flacainide cardiotoxicity exacerbated by electrolyte disturbance. The syncopal episodes probably represented life-threatening arrhythmias. The ECG and symptoms resolved completely once the electrolytes were corrected. Increased cardiotoxicity with hypokalaemia is documented, but not widely recognised. Hyponatrae

Upregulation of YAP attenuates doxorubicin-induced cardiotoxicity through inhibiting ferroptosis.

BACKGROUND: Doxorubicin (DOX) is a widely utilized anthracycline chemotherapeutic agent in clinical settings; however, its application is significantly constrained by the cardiotoxic effects it induces, the underlying mechanisms of which remain inadequately understood. Ferroptosis, a recently identified form of iron-dependent programmed cell death, has been implicated in cardiovascular diseases, yet its involvement in doxorubicin-induced cardiotoxicity (DIC) requires further elucidation. Yes-associated protein (YAP), a pivotal component of the Hippo signaling pathway, plays a crucial role in m

Experimental evaluation of N-acetylcysteine against doxorubicin cardiotoxicity in rats.

BACKGROUND: Doxorubicin (DOX) is a widely used anthracycline antibiotic in the treatment of pediatric malignancies. However, its clinical application is significantly limited by its well-documented cardiotoxic side effects. The hypothesis of this study is that N-acetylcysteine (NAC), as an antioxidant agent, may reverse DOX-induced cardiotoxicity. Therefore, we aimed to evaluate the potential protective role of NAC against DOX-induced cardiotoxicity in rat heart tissue in this experimental study. METHODS: Thirty rats were randomly divided into three groups (n = 10 each): control, DOX, and DOX

Doxorubicin-Induced Cardiotoxicity: Comprehensive Pathway Insights and Advanced Preclinical Therapeutics.

Doxorubicin, a secondary metabolite of Streptomyces peucetius var. caesius and a member of the anthracycline family, exerts anticancer effects via DNA intercalation and topoisomerase II inhibition in tumor cells. However, its clinical application is limited by dose-dependent and cumulative cardiotoxicity. The mechanisms underlying doxorubicin-induced cardiotoxicity (DIC) include oxidative stress, lipid peroxidation, mitochondrial dysfunction, calcium dysregulation, disrupted iron homeostasis, nitric oxide release, and inflammatory mediator production. Emerging evidence highlights autophagy dys

Cardiotoxicity adverse outcome pathway network: towards mechanistic and quantitative modelling.

INTRODUCTION: Chemical-induced heart toxicity remains a major challenge in drug development and environmental safety, largely because current testing often relies on narrow, late-stage endpoints that miss the complex biological progression of the toxicities. To address this, we developed a comprehensive Adverse Outcome Pathway (AOP) network that maps how early (bio) chemical triggers evolve into organ-level dysfunction. METHODS: By integrating data from the OECD AOP-Wiki, we constructed a unified network of 64 biological events and 94 documented relationships that identifies the critical biol

Hesperidin attenuates cisplatin-induced cardiotoxicity in rats through regulating autophagy.

OBJECTIVE: Cisplatin is a well-known anticancer drug whose use has been limited due to a variety of side effects. Evidence has confirmed that cisplatin consumption is associated with cardiac toxicity. Unfortunately, no drug has yet been found to treat cardiac toxicity or reduce the side effects caused by cisplatin. However, hesperidin has been identified as an antioxidant agent which has protective effects on the heart tissue. This study explored the impact of hesperidin on cisplatin-induced cardiac toxicity through its antioxidant effect and adjustment of autophagy. MATERIALS AND METHODS: It

Immunologically Active Cardiac Fibroblasts expressing MHC-II promote Doxorubicin cardiotoxicity.

A common side effect of the chemotherapeutic anthracycline, doxorubicin, is cardiotoxicity and progression to heart failure. Patients treated with anthracyclines show increased circulating cytotoxic CD8+IFN-γ+ T cells. In mice, cumulative doses of doxorubicin also result in increased circulating and cardiac infiltrating CD8+IFN-γ+ T cells, and these cells are required for pathological fibrosis and contractile dysfunction. The present study demonstrates that cardiac fibroblasts become immunologically active and express major histocompatibility complex (MHC) class II on the onset of doxorubicin
