It is well understood that cancer therapies including chemotherapy, tyrosine kinase inhibitors, immune checkpoint inhibitors, and radiation can increase the risk of cardiovascular disease in patients with cancer. This can manifest as a multitude of pathologies including left ventricular dysfunction, myocarditis, cardiomyopathy, accelerated atherosclerosis, and coronary vasospasm. Multimodal cardiac imaging plays a critical role in diagnosing such pathologies by relying on noninvasive tools including echocardiograms, cardiac magnetic resonance imaging, positron emission tomography, single-photo

Cell cycle checkpoint kinases play a pivotal role in protecting against replicative stress. In this study, valproic acid (VPA), a histone deacetylase inhibitor (HDACi), was found to promote breast cancer MCF-7 cells to traverse into G2/M phase for catastrophic injury by promoting PPP2R2A (the B-regulatory subunit of Phosphatase PP2A) to facilitate the dephosphorylation of Chk1 at Ser317 and Ser345. By contrast, VPA protected normal 16HBE cells from HU toxicity through decreasing PPP2R2A expression and increasing Chk1 phosphorylation. The effect of VPA on PPP2R2A was at the post-transcription l

INTRODUCTION: Pazopanib, a tyrosine kinase inhibitor, and nivolumab, an immune checkpoint inhibitor, are both considered to cause hepatotoxicity with different pathophysiology. We report a case in which a patient died of severe hepatotoxicity who was presumed to have been caused by the administration of nivolumab followed by pazopanib for metastatic renal cell carcinoma.
CASE PRESENTATION: A 74-year-old male with metastatic renal cell carcinoma was treated with nivolumab as a third-line treatment. However, nivolumab was subsequently discontinued, as it caused severe thyroiditis. About 2 month

The therapeutic management of metastatic breast cancers has benefited in the past few years from important progress linked to the development of new molecules, inhibiting poly adenosine diphosphate ribose polymerase (PARP), cyclin-dependent kinases (CDK4/6) or immune checkpoints. More recently, the development of antibody-drug conjugates (ADC) targeting tumor cells, allows for the delivery of cytotoxic agents, at low doses, while increasing their concentration in the tumoral microenvironment. Currently, two ADC are available for the management of HER2-negative metastatic breast cancers, sacitu

Chronic lymphocytic leukemia (CLL) is a biologically heterogeneous B cell malignancy in which non-malignant T lymphocytes constitute a critical component of the tumor microenvironment and significantly influence disease evolution and the therapeutic response. Growing evidence suggests that CLL-associated T cells not only participate in the antitumor response but also activate signals that promote the development of CLL subclones. Although novel targeted therapies, such as Bruton's tyrosine kinase (BTK) inhibitors, BTK degraders, B-cell lymphoma 2 (BCL-2) inhibitors, T cell engagers, immune che

Chemoresistance represents an ongoing challenge in treating patients with acute myeloid leukemia (AML), and a better understanding of the resistance mechanisms can lead to the development of novel AML therapies. Here, we demonstrated that low expression of the DNA damage response gene Schlafen 11 (SLFN11) correlates with poor overall survival and worse prognosis in patients with AML. Moreover, we showed that SLFN11 plays an essential role in regulating chemotherapy sensitivity in AML. AML cells with suppressed levels of SLFN11 do not undergo apoptosis in response to cytarabine because of aberr

Checkpoint kinase 2 (CHEK2) gene codes for an important mediator of DNA damage response pathway. Its mutations increase risk of several types of cancer. We analysed selected CHEK2 mutations in 631 Czech colorectal cancer (CRC) patients. The increased risk of CRC was associated with mutations in CHEK2 gene region involving fork head-associated domain [39/631 (6.2%) cases versus 19/683 (2.8%) controls; odds ratio (OR)=2.3; 95% confidence interval (CI)=1.3-4.0; p=0.003], and with the most frequent I157T mutation [30/631 (4.8%) cases versus 17/683 (2.5%) controls; OR=2.0; 95% CI=1.1-3.6; p=0.03].

Statins are widely prescribed lipid-lowering agents that also exert pleiotropic anticancer effects, such as induction of apoptosis and ferroptosis, modulation of autophagy, and remodeling of the tumor microenvironment. Consistent with these multifaceted actions, statins have demonstrated synergistic activity with several chemotherapeutic agents. Emerging evidence indicates that statins modulate the activity of key DNA damage response kinases, such as ataxia-telangiectasia (ATM) and checkpoint kinase 2 (CHK2), in colorectal cancer cells, suggesting a potential impact on DNA repair pathways. This paper reviews the functions of and connections between the presumed DNA damage sensors: poly(ADP-ribose) polymerase (PARP), DNA-dependent protein kinase (DNA-PK), the protein product of the ataxia telangiectasia mutated (ATM) gene, and the tumor suppressor, p53. Recognition of DNA damage is associated with the generation of alarm signals. The possible alarm signals include synthesis of poly(ADP-ribose) polymers and initiation of phosphorylation cascades by kinases complexed with the DNA damage sensors, DNA-PK and ATM; the role of other factors is discussed, among them BRCA1 and 2, IRF-1 a

Checkpoint kinase 2 (CHEK2) encodes a serine/threonine kinase involved in the DNA damage response through ATM-Chk2-p53 signaling. Its function in maintaining genomic stability classifies it as a tumor suppressor. Heterozygous germline pathogenic variants in CHEK2 are associated with a moderate increase in lifetime risk of breast and prostate cancer. This study assessed the prevalence of CHEK2 variants globally, with a focus on East Asian and Korean populations, for which data have remained limited. We analyzed 125,748 exomes from the Genome Aggregation Database (gnomAD), including 9,197 East A

Microcephalin-1 (MCPH1) exists as 2 isoforms that regulate cyclin-dependent kinase-1 activation and chromosome condensation during mitosis, with MCPH1 mutations causing primary microcephaly. MCPH1 is also a tumor suppressor protein, with roles in DNA damage repair/checkpoints. Despite these important roles, there is little information on the cellular regulation of MCPH1. We show that both MCPH1 isoforms are phosphorylated in a cyclin-dependent kinase-1-dependent manner in mitosis and identify several novel phosphorylation sites. Upon mitotic exit, MCPH1 isoforms were degraded by the anaphase-p

Ribonucleotide Reductase (RNR) is a rate-limiting enzyme in the production of deoxyribonucleoside triphosphates (dNTPs), which are essential substrates for DNA repair after radiation damage. We explored the radiosensitization property of RNR and investigated a selective RRM2 inhibitor, 3-AP, as a radiosensitizer in the treatment of metastatic pNETs. We investigated the role of RNR subunit, RRM2, in pancreatic neuroendocrine (pNET) cells and responses to radiation in vitro. We also evaluated the selective RRM2 subunit inhibitor, 3-AP, as a radiosensitizer to treat pNET metastases in vivo. Knock

Synthetic cobalt Schiff bases have recently emerged as promising metal-based anticancer candidates. In this study, we investigated the biological effects of a mononuclear octahedral cobalt(III) Schiff base complex [CoL3] (L = 2-((allylimino) methyl) - 6-methoxyphenol)) on the cell cycle progression and apoptosis of the HT-29 cells, a human colorectal cancer cell line. Cytotoxic activity of [CoL3] was evaluated using the MTT assay. Then, its impact on apoptosis was assessed via annexin V-FITC/propidium iodide (PI) staining. To elucidate the underlying mechanisms, we employed flow cytometry, rea

Although tyrosine kinase inhibitor and immune checkpoint inhibitor (TKI-ICI) combination therapy has emerged as a promising treatment for hepatocellular carcinoma (HCC), reliable biomarkers for predicting long-term survival remain underexplored. Here, we conducted metabolomics and lipidomics profiling in baseline plasma samples from 58 patients with HCC who received TKI-ICI therapy in a prospective phase II trial. Prognostic features were identified using an integrated machine learning framework combining random forest survival analysis, LASSO regression, and Cox modeling. Untargeted metabolom

BACKGROUND: Biliary carcinomas are aggressive cancers with a high mortality rate. When metastatic, biliary cancers are associated with a short survival and low response to treatments. The first line therapy of metastatic biliary carcinomas consists of a platinum doublet chemotherapy combination with an immune checkpoint inhibitor and results in a median overall survival in the range of approximately 12-13 months, with 20% to 25% of patients surviving at 2 years. Second line chemotherapy options based on fluoropyrimidines are associated with a median survival of less than 6 months. Genomic stud

Breast cancer remains the most common cancer among women worldwide, characterized by significant molecular heterogeneity. Advances in targeted therapy and immunotherapy have transformed treatment approaches, offering promising new options for patients. Targeted therapies are designed to inhibit the key pathways that drive tumor growth and survival. The cyclin-dependent kinase 4/6 (CDK4/6) inhibitors, phosphatidylinositol-3 kinase (PI3K) inhibitors, mammalian target of rapamycin (mTOR) inhibitors, and other treatments have significantly advanced treatment, delaying progression and extending sur

PURPOSE OF REVIEW: To summarize recent updates in the classification, clinical trial evidence, and evolving treatment strategies for nonclear cell renal cell carcinoma (nccRCC).
RECENT FINDINGS: The 2022 WHO classification eliminated the type 1/2 papillary paradigm, refined molecularly defined tumors such as fumarate hydratase (FH)-deficient and anaplastic lymphoma kinase-rearranged RCC, while acknowledging indolent tumors like clear cell papillary renal cell tumors. Prospective and retrospective data increasingly support the use of immune checkpoint inhibitor (ICI)-based regimens, particular
