Feeding of Whitefly on Tobacco Decreases Aphid Performance via Increased Salicylate Signaling.

BACKGROUND: The feeding of Bemisia tabaci nymphs trigger the SA pathway in some plant species. A previous study showed that B. tabaci nymphs induced defense against aphids (Myzus persicae) in tobacco. However, the mechanism underlying this defense response is not well understood. METHODOLOGY/PRINCIPAL FINDINGS: Here, the effect of activating the SA signaling pathway in tobacco plants through B. tabaci nymph infestation on subsequent M. persicae colonization is investigated. Performance assays showed that B. tabaci nymphs pre-infestation significantly reduced M. persicae survival and fecundity

Chitotriosidase: the yin and yang.

The enzyme chitotriosidase (ChT), the human analogue of chitinases from non-vertebrate species, is one of the most abundant and indicative proteins secreted by activated macrophages. Its enzymatic activity is elevated in serum of patients suffering from Gaucher's disease type 1 and in some other inherited lysosomal storage disorders, as well as in diseases in which macrophages are activated. The last decade has witnessed the appearance of a substantial number of studies attempting to unravel its cellular functions, which have yet not been fully defined. A great deal of progress has been made i

Supercritical Fluid Extraction of Ergosterol from Lentinula edodes and Pleurotus ostreatus: Optimization and Synergistic Effects of Biomass Pre-Treatments.

Ergosterol (ERG) is a bioactive sterol found in fungal cell membranes with reported cholesterol-lowering, antioxidant, and antitumor properties. Supercritical Fluid Extraction (SFE) conditions were optimized for Lentinula edodes (shiitake) using Response Surface Methodology and subsequently applied to Pleurotus ostreatus (oyster). Optimized SFE (690 bar, 69.8 °C, no co-solvent) produced significantly more concentrated ERG extracts than Soxhlet extraction for both species-280.57 ± 10.80 mgERG/gextract for shiitake and 95.87 ± 7.18 mgERG/gextract for oyster-corresponding to a 107% and 65% increa

Active invasion of bacteria into living fungal cells.

The rice seedling blight fungus Rhizopus microsporus and its endosymbiont Burkholderia rhizoxinica form an unusual, highly specific alliance to produce the highly potent antimitotic phytotoxin rhizoxin. Yet, it has remained a riddle how bacteria invade the fungal cells. Genome mining for potential symbiosis factors and functional analyses revealed that a type 2 secretion system (T2SS) of the bacterial endosymbiont is required for the formation of the endosymbiosis. Comparative proteome analyses show that the T2SS releases chitinolytic enzymes (chitinase, chitosanase) and chitin-binding protein

Clonally expanded EOMES+ Tr1-like cells in primary and metastatic tumors are associated with disease progression.

Regulatory T (Treg) cells are a barrier for tumor immunity and a target for immunotherapy. Using single-cell transcriptomics, we found that CD4+ T cells infiltrating primary and metastatic colorectal cancer and non-small-cell lung cancer are highly enriched for two subsets of comparable size and suppressor function comprising forkhead box protein P3+ Treg and eomesodermin homolog (EOMES)+ type 1 regulatory T (Tr1)-like cells also expressing granzyme K and chitinase-3-like protein 2. EOMES+ Tr1-like cells, but not Treg cells, were clonally related to effector T cells and were clonally expanded

Chitotriosidase in the Pathogenesis of Inflammation, Interstitial Lung Diseases and COPD.

As a member of 18 glycosyl hydrolase (GH) family, chitotriosidase (Chitinase 1, CHIT1) is a true chitinase mainly expressed in the differentiated and polarized macrophages. CHIT1 is an innate immune mediator that digests the cell walls of chitin-containing eukaryotic pathogens, such as fungi. However, CHIT1 is dysregulated in granulomatous and fibrotic interstitial lung diseases characterized by inflammation and tissue remodeling. These include tuberclosis, sarcoidosis, idiopathic pulmonary fibrosis, scleroderma-associated interstitial lung diseases (SSc-ILD), and chronic obstructive lung dise

Structural Diversification and Biosynthesis of Pesticidal Pyrroline-Derived Alkaloid from Penicillium citrinum SCSIO DF147.

The pyrroline-derived alkaloid N-(2-methyl-3-oxodec-8-enoyl)-2-pyrroline (MOEP, 1) is a potential pesticide that remains to be structurally diversified for target screening and biosynthesis studies. Bioactivity-guided investigation of the marine-derived fungus Penicillium citrinum SCSIO DF147 yielded five classes of pyrroline-derived alkaloids (1-15), including five new compounds (2-4, 14, and 15). These compounds were first evaluated for chitinase (OfChtII and OfChi-h) inhibition, revealing that both the 3-pyrroline ring and the Δ8',9' double bond are key functional groups. Pesticidal and fun

Microbes as a tool for the bioremediation of fish waste from the environment and the production of value-added compounds: a review.

Fish are the most edible protein source worldwide and generate several remnants such as scales, viscera, head, bone, and skin. Fish wastes are not disposed of properly, which adversely affects the environment, especially the water bodies where fish processing industries dispose of their waste. Fish waste mainly contains nitrogen, oil, fat, salts, heavy metals, and organic compounds, which increase the biological oxygen demand and chemical oxygen demand. Fish waste can degrade in various ways, such as physicochemical or by enzymatic action, but using microbes is an environmentally friendly appr

Chitin hoops obtained from Ommatoiulus sabulosus (Diplopoda, Julidae) used for bovine serum albumin adsorption.

A new hoop shaped three dimensional chitin was obtained successfully from the body segment of a diplopod species (Ommatoiulus sabulosus) by following the procedure decolorization, demineralization and deproteinization. Purity of the hoop shaped three-dimensional chitin was proved by FT-IR analysis and chitinase digestive test. The important bands for α-chitin such as 1654.2, 1619.7 and 1552cm-1 were found after FT-IR analysis. And the chitinase digestive test revealed the purity of chitin (with digestion rate of 94.7%). SEM analysis showed that the chitin surface consisted of highly porous str

Nasal polyp chitinolytic activity associated with smoking or allergy.

BACKGROUND: Cells of the innate immune system that are implicated in allergy and immunity bind to chitin during tissue infiltration in a process negatively regulated by vertebrate chitinases. Both acidic mammalian chitinase (AMCase) and chitotriosidase (ChT) exert chitinolytic activity. The levels of activities of these enzymes in nasal polyps (NPs) of subjects who smoke or who suffer from allergies are unknown. In the present work, we measured the activities of AMCase and ChT in NPs of smokers and allergic subjects. METHODS: We report a prospective cohort study in a tertiary care facility. A

From Antifungal Hydrolases to Modern Plant Immunity: Revisiting the Legacy of Boller.

The 1988 papers by Mauch, Mauch-Mani, Hadwiger and Boller on pea antifungal hydrolases captured a simple but powerful idea: plants can attack pathogens by directly targeting the structural polymers that hold their cell walls together. At that time, chitinases and β-1,3-glucanases were known to accumulate after infection, wounding and ethylene treatment, but whether these proteins actually restricted fungal growth remained unclear. By resolving distinct hydrolase isoforms and showing that inducible chitinase and β-1,3-glucanase act synergistically to inhibit fungal growth and lyse hyphal tips,

Functional Analysis of Trichodysplasia Spinulosa-Associated Polyomavirus-Encoded Large T Antigen.

Trichodysplasia spinulosa-associated polyomavirus (TSPyV or human polyomavirus 8) was identified from patients with trichodysplasia spinulosa, a rare skin disease affecting the faces of immunocompromised patients. Like other polyomaviruses, the TSPyV genome encodes a large T antigen (LT). However, the expression and functions of TSPyV LT in infected cells remain largely unknown. In the present study, we cloned a full-length TSPyV LT cDNA from cells transfected with the full-length of TSPyV LT DNA. Transfection study using green fluorescence protein-tagged LT expression plasmids showed that TSP

Plasma chitotriosidase in health and pathology.

Chitotriosidase (Chit), a member of the mammalian chitinase family, structurally homologous to chitinases from other species, is synthesized and secreted by specifically activated macrophages. After discovery of a striking increase of plasma Chit activity in serum from Gaucher's disease type I patients, an increasing number of inherited and acquired conditions sharing macrophage activation have been associated with increased Chit activity. In addition to lipid storage disorders, where Chit activity has longer been used as a marker of disease activity and therapeutic response, elevation of plas

Molecular cloning and characterization of an estrogen-dependent porcine oviductal secretory glycoprotein.

A family of estrogen-dependent porcine oviductal secretory glycoproteins (POSPs) that exhibit structural similarities are synthesized and secreted into the oviductal lumen at proestrus, estrus, and metestrus. The objectives of this study were to clone the POSP cDNA, obtain the full-length cDNA and protein sequence, examine tissue specificity and species distribution, characterize its regulation, and establish its identity by comparison to other known protein, RNA, or DNA sequences. A full-length cDNA of 2022 base pairs was obtained with an open reading frame of 1581 nucleotides, coding for a d

Host gastric corpus microenvironment facilitates Ascaris suum larval hatching and infection in a murine model.

Ascariasis (roundworm) is the most common parasitic helminth infection globally and can lead to significant morbidity in children including chronic lung disease. Children become infected with Ascaris spp. via oral ingestion of eggs. It has long been assumed that Ascaris egg hatching and larval translocation across the gastrointestinal mucosa to initiate infection occurs in the small intestine. Here, we show that A. suum larvae hatched in the host stomach in a murine model. Larvae utilize acidic mammalian chitinase (AMCase; acid chitinase; Chia) from chief cells and acid pumped by parietal cell

Changes in gene expression during adaptation of Listeria monocytogenes to the soil environment.

Listeria monocytogenes is a ubiquitous opportunistic pathogen responsible for listeriosis. In order to study the processes underlying its ability to adapt to the soil environment, whole-genome arrays were used to analyse transcriptome modifications 15 minutes, 30 minutes and 18 h after inoculation of L. monocytogenes EGD-e in soil extracts. Growth was observed within the first day of incubation and large numbers were still detected in soil extract and soil microcosms one year after the start of the experiment. Major transcriptional reprofiling was observed. Nutrient acquisition mechanisms (pho

Marked increase of methylumbelliferyl-tetra-N-acetylchitotetraoside hydrolase activity in plasma from Gaucher disease patients.

Methylumbelliferyl-tetra-N-acetylchitotetraoside hydrolase activity was increased 53- to 484-fold in plasma from Gaucher disease patients and no activator could be found. High activity was also measured in other lysosomal storage diseases including Krabbe disease, Wolman disease, GM1-gangliosidosis and to a lesser extent Niemann-Pick disease type B, but the activities were lower than the lowest values in Gaucher patients. Kinetic properties of the high activity in Gaucher plasma were similar to those of controls. It is not known whether the increased activity represents intrinsic enzyme activi
