Comparison of the effects of mercuric chloride and methylmercuric chloride on the isolated phrenic nerve and on the phrenic nerve-diaphragm preparation from the rat.

This study compared the effects of HgCl(2) and CH(3)HgCl on the compound action potential of the isolated rat phrenic nerve and the twitch contractions of rat phrenic nerve-diaphragm preparations during direct and indirect stimulation. Both HgCl(2) and CH(3)HgCl, at a concentration of 3.7 x 10(-5)m, inhibited the twitches during indirect stimulation. CH(3)HgCl was slightly more potent than HgCl(2). The inhibition might be explained by a simultaneous inhibition of the phrenic nerve. CH(3)HgCl caused an additional curare-like inhibition at the neuromuscular junction. HgCl(2) potentiated the indi

Regulation of chloride in quiescent sheep-heart Purkinje fibres studied using intracellular chloride and pH-sensitive micro-electrodes.

1. The intracellular Cl activity, alpha iCl was measured inside quiescent sheep cardiac Purkinje fibres, bathed in normal Tyrode at pH 7.40, buffered with approximately 22 mM-bicarbonate/approximately 5% CO2 + 95% O2. The measurements were made using liquid ion-exchanger Cl-sensitive micro-electrodes. 2. After internal Cl levels had been depleted by prolonged exposure to Cl-free media (glururonate-substituted) when external Cl was restored, there was a rapid re-accumulation of Cl inside the fibres to levels that were much higher than those expected for a passive Cl distribution. Such a process

Protective effects of dietary potassium chloride on hemodynamics of Dahl salt-sensitive rats in response to chronic administration of sodium chloride.

BACKGROUND: Dietary potassium supplementation decreases blood pressure and prevents strokes in humans, and prevents strokes and renal damage in Dahl salt-sensitive (DSS) rats. OBJECTIVE: To study the effects of various concentrations of dietary potassium chloride (KCl) on the hemodynamics of Dahl salt-resistant (DSR) and DSS rats receiving a 1% sodium chloride (NaCl) diet for 8 months, to determine whether there is an optimal dietary concentration of KCl that minimizes increases in blood pressure and causes least impairment of blood flow in the brain and kidneys. METHODS AND RESULTS: We foun

Activation of Thioglycosides with Ferric Chloride: Scope and Mechanism.

Previously, we disclosed a new method for the activation of armed and superarmed ethylthio glycosides using an inexpensive and abundant iron-(III) chloride. Reported herein are the next steps toward refining the reaction conditions, broadening the scope of this method to glycosidation of less-reactive glycosyl donors, and investigating the reaction mechanism. This reaction is swift and generally high-yielding. The completion of the reaction can be monitored by eye, and typically, the color change (start of the reaction) can be detected right after the addition of the glycosyl donor to the prem

CLN3 mediates chloride efflux from lysosomes.

Neurodegenerative diseases, which pose significant challenges for effective treatment, often involve risk variants of lysosomal gene products that disrupt lysosomal function, leading to the accumulation of indigestible materials and damage to brain cells. The lysosome is a degradative organelle and a signaling hub that senses nutrient availability. How lysosomal dysfunction contributes to neurodegenerative diseases is an important open question. In this study, we identified CLN3 (ceroid lipofuscinosis, neuronal 3), an endolysosomal protein that is linked to Batten disease, as an evolutionarily

The Role of Chloride in Cardiorenal Syndrome: A Practical Review.

Chloride, long considered a passive extracellular anion, has emerged as a key determinant in the pathophysiology and management of heart failure (HF) and cardiorenal syndrome. In contrast to sodium, which primarily reflects water balance and vasopressin activity, chloride exerts broader effects on neurohormonal activation, acid-base regulation, renal tubular function, and diuretic responsiveness. Its interaction with With-no-Lysine (WNK) kinases and chloride-sensitive transporters underscores its pivotal role in electrolyte and volume homeostasis. Hypochloremia, frequently observed in HF patie

A case of severe benzalkonium chloride intoxication in a cat.

BACKGROUND: Benzalkonium chloride (BAC) is a quaternary ammonium compound (QAC), that can be found in a wide variety of household products-from disinfectants to medicaments and home fragrances-but also professional products. In pets, cats have long been reported as more sensitive than dogs to QACs; in fact, signs of irritation such as oral ulcerations, stomatitis and pharyngitis can be observed after contact with concentrations of 2% or lower. In a review of 245 cases of BAC exposure in cats, reported by the Veterinary Poisons Information Service (United Kingdom) only 1.2% of the cases died or

Synthetic Receptors with Micromolar Affinity for Chloride in Water.

A water-soluble coordination cage was obtained by reaction of Pd(NO3 )2 with a 1,3-di(pyridin-3-yl)benzene ligand featuring a short PEG chain. The cavity of the metal-organic cage contains one nitrate anion, which is readily replaced by chloride. The apparent association constant for chloride binding in buffered aqueous solution is Ka =1.8(±0.1)×105  M-1 . This value is significantly higher than what has been reported for other macrocyclic chloride receptors. The heavier halides Br- and I- compete with binding or self-assembly, but the receptor displays very good selectivity over common anions

Proton-Activated Chloride Channel: Physiology and Disease.

The maintenance of intracellular and extracellular pH relies on multiple ion transporters/channels. Proton-activated chloride channel (PAC) precisely regulates extracellular and early/late endosomal pH by transporting chloride ion (Cl-) across membranes and has been shown to be implicated in pH imbalance under hypoxic conditions, such as the acidic microenvironments of cancer and ischemia. In this article, the phenotypic characteristics, molecular mechanisms, physiology of PAC and its role in cancer, ischemic stroke and hypoxia will be discussed in order to provide some clues for developing po

CHRONIC EFFECTS OF CADMIUM CHLORIDE ON RAT EMBRYOGENESIS.

The purpose of this study was to analyze the effects of cadmium toxicity on rat embryogenesis when exposed to other heavy metal citrates. Despite the variety of scientific publications discussing the influence of cadmium on mammalian postnatal development, the effect of this metal on embryogenesis has not yet been sufficiently studied. In this experimental study, cadmium chloride was administered to experimental pregnant female Wistar rats at a daily dose of 1.0 mg/kg. Rats were allocated at random into groups receiving either cadmium chloride alone or additional zinc citrate, cerium citrate,

Structure-function relationships in the sodium chloride cotransporter.

The thiazide sensitive Na+:Cl- cotransporter (NCC) is the principal via for salt reabsorption in the apical membrane of the distal convoluted tubule (DCT) in mammals and plays a fundamental role in managing blood pressure. The cotransporter is targeted by thiazide diuretics, a highly prescribed medication that is effective in treating arterial hypertension and edema. NCC was the first member of the electroneutral cation-coupled chloride cotransporter family to be identified at a molecular level. It was cloned from the urinary bladder of the Pseudopleuronectes americanus (winter flounder) 30 ye

Combinatorial chloride and calcium channelopathy in myotonic dystrophy.

Myotonic dystrophy type 1 (DM1) involves misregulated alternative splicing for specific genes. We used exon or nucleotide deletion to mimic altered splicing of genes central to muscle excitation-contraction coupling processes in mice. Mice with forced-skipping of exon 29 in CaV1.1 calcium channel combined with loss of ClC-1 chloride channel function showed a markedly reduced lifespan, whereas other combinations of splicing mimics did not affect survival. The Ca2+/Cl- bi-channelopathy mice exhibited myotonia, weakness, and impairment of mobility and respiration. Chronic administration of the ca

Evidence for Oxonium Ions in Ethereal "Hydrogen Chloride".

Although solutions of hydrogen chloride in ethereal solvents like diethyl ether or dioxane are commonly employed in the laboratory, the solution structure of these reagents has yet to be firmly established. Here, we analyze solutions of ethereal hydrogen chloride or deuterium chloride in toluene, in dichloromethane, or in the absence of a co-solvent by in situ infrared spectroscopy. The resulting spectra are inconsistent with free HCl or often-proposed 1:1 HCl-ether complexes but closely match the predicted spectra of oxonium ions generated via protonation of diethyl ether. Effect of vinyl chloride exposure on cardiometabolic toxicity.

Vinyl chloride (VC) is an organochlorine mainly used to manufacture its polymer polyvinyl chloride, which is extensively used in the manufacturing of consumer products. Recent studies suggest that chronic low dose VC exposure affects glucose homeostasis in high fat diet-fed mice. Our data suggest that even in the absence of high fat diet, exposure to VC (0.8 ppm, 6 h/day, 5 day/week, for 12 weeks) induces glucose intolerance (1.0 g/kg, i.p.) in male C57BL/6 mice. This was accompanied with the depletion of hepatic glutathione and a modest increase in lung interstitial macrophages. VC exposure d
