Cholera toxin perturbs the paracellular barrier in the small intestinal epithelium of rats by affecting claudin-2 and tricellulin.

Cholera toxin is commonly known to induce chloride secretion of the intestine. In recent years, effects on epithelial barrier function have been reported, indicating synergistic co-regulation of transporters and tight junction proteins. Our current study focused on the analysis of cholera toxin effects on transepithelial resistance and on tight junction proteins, the latter known as structural correlates of barrier function. Ligated segments of the rat jejunum were injected with buffered solution containing cholera toxin (1 μg/ml) and incubated for 4 h. Subsequently, selfsame tissue specimens

Expression of claudin-11, -23 in different gastric tissues and its relationship with the risk and prognosis of gastric cancer.

Claudins play an important role in regulating the permeability of epithelial and endothelial cells and in the maintenance of cell polarity. We aimed to investigate expression of claudin-11, -23 in different gastric tissues and its relationship with clinicopathologic parameters and prognosis of gastric cancer. We compared their expression levels in the paired cancerous tissues versus those in the adjacent noncancerous tissues by real-time PCR, western blotting and immunohistochemistry. The results showed that the expression of claudin-11, -23 was greatly increased in paracancerous gastric tissu

Humanisation of a claudin-1-specific monoclonal antibody for clinical prevention and cure of HCV infection without escape.

OBJECTIVE: HCV infection is a leading cause of chronic liver disease and a major indication for liver transplantation. Although direct-acting antivirals (DAAs) have much improved the treatment of chronic HCV infection, alternative strategies are needed for patients with treatment failure. As an essential HCV entry factor, the tight junction protein claudin-1 (CLDN1) is a promising antiviral target. However, genotype-dependent escape via CLDN6 and CLDN9 has been described in some cell lines as a possible limitation facing CLDN1-targeted therapies. Here, we evaluated the clinical potential of th

Claudin-1 regulates pulmonary artery smooth muscle cell proliferation through the activation of ERK1/2.

Tumor necrosis factor alpha (TNF-α), a crucial inflammatory cytokine, is involved in the pathogenesis of pulmonary arterial hypertension (PAH). TNF-α can induce claudin-1 (CLDN1) expression and CLDN1 has been reported to be associated with the regulation of cellular functions including cell proliferation, migration. Thus, we aimed to explore whether CLDN1 participated in the etiology of PAH. Our study showed that CLDN1 expression was markedly increased in the lungs of rats with monocrotaline(MCT)-induced PAH, especially in the pulmonary arterial smooth muscle sections. We also found that CLDN1

Expression of claudin-5, -7, -8 and -9 in cervical carcinoma tissues and adjacent non-neoplastic tissues.

Recent data indicate that the tight junction proteins are abnormally regulated in several human cancers and the expression of these proteins is involved in the etiology and progression of cancer. To explore the expression distinction of the tight junction proteins claudin-5, -7, -8 and -9 in the adjacent non-neoplastic tissues and cervical carcinoma tissues, 72 cervical carcinoma tissues and the samples of non-neoplastic tissues adjacent to the tumors were examined for expression of claudin-5, -7, -8 and -9 by streptavidin-perosidase immunohistochemical staining method. The positive expression

Claudin 3, 4, and 15 expression in solid tumors of lung adenocarcinoma versus malignant pleural mesothelioma.

Epithelioid malignant pleural mesothelioma (MPM) can easily be confused with lung adenocarcinomas (ACAs). In serous effusion, claudin (cldn) 3 is shown to be useful in the diagnosis of mesothelioma vs ACAs. Cldn15 is reported to be overexpressed in epithelioid mesothelioma and absent in human airway epithelium. The aim was to assess the value of cldn3 and cldn4 compared to that of BerEp4 and thyroid transcription factor-1 (TTF1) in differentiating lung ACA from epithelioid MPM and to examine the expression of cldn15 in these tumors. The expression of cldn3, cldn4, cldn15, BerEp4, and TTF1 was

Postnatal changes in the expression of claudin-11 in the testes and excurrent ducts of the domestic rabbit (Oryctolagus cuniculus domesticus).

We examined the expression of claudin-11 (CLDN11) in the testes and male reproductive tracts of rabbits. The rabbit CLDN11 cDNA sequences were nearly identical with human, mouse, and bovine CLDN11. The levels of CLDN11 mRNA and protein (22 kDa) were markedly increased in the testis during adult development. On postnatal day (PND) 10, CLDN11 was colocalized with ZO-1 at the lateral contacts between adjacent Sertoli cells and was perpendicular to the basal lamina. In adult testis on PND 180, CLDN11 was codistributed with ZO1, and the pattern of immunoreactivity consisted of wavy linear tracts pa

Epithelioid versus rhabdoid glioblastomas are distinguished by monosomy 22 and immunohistochemical expression of INI-1 but not claudin 6.

Epithelioid and rhabdoid glioblastomas are rare entities that share some overlapping morphologic features, but remain poorly characterized at the immunohistochemical and genetic level. We report 10 examples: 8 epithelioid glioblastomas (E-GBMs) and 2 rhabdoid GBMs (R-GBMs). E-GBMs tended to be superficially located, circumscribed, supratentorial tumors composed of monotonous, discohesive sheets of small rounded cells that mimicked metastatic malignant melanoma. R-GBMs showed tumor with classic rhabdoid features arising as a subpopulation of an otherwise classic GBM, fitting the definition of c

Monoclonal anti-claudin 1 antibodies prevent hepatitis C virus infection of primary human hepatocytes.

BACKGROUND & AIMS: Hepatitis C virus (HCV) infection is a challenge to prevent and treat because of the rapid development of drug resistance and escape. Viral entry is required for initiation, spread, and maintenance of infection, making it an attractive target for antiviral strategies. The tight junction protein claudin-1 (CLDN1) has been shown to be required for entry of HCV into the cell. METHODS: Using genetic immunization, we produced 6 monoclonal antibodies against the host entry factor CLDN1. The effects of antibodies on HCV infection were analyzed in human cell lines and primary human

The targeted overexpression of a Claudin mutant in the epidermis of transgenic mice elicits striking epidermal and hair follicle abnormalities.

Skin is one of the largest organs of the body, and is formed during development through a highly orchestrated process involving mesenchymal-epithelial interactions, cell commitment, and terminal differentiation. It protects against microorganism invasion and UV irradiation, inhibits water loss, regulates body temperature, and is an important part of the immune system. Using transgenic mouse technology, we have demonstrated that Claudin (Cldn)-containing tight junctions (TJs) are intricately involved in cell signaling during epidermal differentiation and that an epidermal suprabasal overexpress

Distribution of the tight junction proteins ZO-1, occludin, and claudin-4, -8, and -12 in bladder epithelium.

In mammals, the bladder stores urine without permitting the passage of urine contents into the bloodstream, a function, in part, of the uroepithelial-associated tight junction complex. The protein constituents that make up this high-resistance barrier in the bladder are currently unknown, although the claudins, a multigene family, are thought to govern paracellular transport in other epithelia. Reverse transcriptase-polymerase chain reaction analysis was used to define that mRNA for claudin-2, -4, -8, -12, and -13 was expressed in mouse bladder tissue. The localization of these claudins, as we

Reciprocal regulation of the junctional proteins claudin-1 and connexin43 by interleukin-1beta in primary human fetal astrocytes.

Vertebrate tissues use multiple junctional types to establish and maintain tissue architecture, including gap junctions for cytoplasmic connectivity and tight junctions (TJs) for paracellular and/or cell polarity barriers. The integral membrane proteins of gap junctions are connexins, whereas TJs are a complex between occludin and members of a recently characterized multigene family, the claudins. In normal brain, astrocytes are coupled by gap junctions composed primarily of connexin43 (Cx43), whereas TJs have not been detected in these cells. We now show that treatment of primary human astroc

Claudin-3 as a marker of intestinal permeability and its relation to inflammatory status in adolescents with functional gastrointestinal disorders.

BACKGROUND: Functional Gastrointestinal disorders (FGIDs) are highly prevalent among adolescents. Increased intestinal permeability plays a potential role in the pathogenesis of FGIDs. Claudin-3 (CLDN3) may serve as an indicator of gut barrier integrity. We aimed to assess serum level of CLDN3 as a noninvasive biomarker of intestinal permeability in adolescents with FGIDs, and its relation with inflammatory markers. METHODS: This study included 70 adolescents with FGIDs and 65 healthy controls. Serum levels of CLDN3 and inflammatory markers; C-reactive protein (CRP), interleukin-6 (IL-6), tum

Aquaporin 1 promotes proliferation and migration of tumor by up-regulating claudin-1 expression in colon cancer.

BACKGROUND: With the rising incidence of colon cancer, several studies have indicated that aquaporin 1 (AQP1) expression is associated with the development of colon cancer. This study aims to elucidate the potential molecular mechanisms between them. METHODS: We screened data from The Cancer Genome Atlas (TCGA) database and retrospectively examined AQP1 protein expression in 127 colon cancer patients to analyze the relationship between AQP1 expression and pathological stages, prognosis. We created stable colon cancer cell lines with differential AQP1 expression, the effect of AQP1 expression

Matriptase proteolysis selectively drives disassembly and internalization of the claudin-7 complex while preserving E-cadherin.

Claudin-7 is a basolaterally enriched tight junction protein that participates in membrane complexes with EpCAM and Trop-2, linking epithelial adhesion to barrier organization. Although proteolytic processing of EpCAM/Trop-2 has been proposed to regulate this complex, its functional consequences for claudin-7 remain unresolved. Here, we define the role of matriptase in regulating claudin-7 dynamics in HaCaT human keratinocytes. Under steady-state conditions, matriptase knockout had only modest effects on claudin-7 distribution, suggesting a limited role in initial complex formation. In contras

Inflammation-Induced Claudin-2 Up-Regulation Limits Pancreatitis Development by Enhancing Pancreatic Ductal Transport.

BACKGROUND & AIMS: Pancreatitis is influenced by environmental and genetic factors. Genome-wide association studies identified polymorphisms at the X-linked CLDN2 locus as risk factors for chronic pancreatitis. CLDN2 encodes claudin-2 (CLDN2), a paracellular cation-selective channel at tight junctions. However, its role in pancreatitis susceptibility remains unclear. We aimed to determine the role of CLDN2 in pancreatitis onset and progression. METHODS: Human pancreatic tissue, pancreatitis mouse models, pancreatic ductal epithelial organoids, and isolated mouse pancreatic ducts were used. CL

Correlation and Overlap Between Claudin 18.2 and FGFR2b Overexpression: A Tissue Microarray Study With 1,538 Gastric Carcinomas.

PURPOSE: Claudin 18.2 (CLDN18.2) and fibroblast growth factor receptor 2b (FGFR2b) have recently emerged as promising therapeutic targets for advanced gastric cancer (GC). Before integrating CLDN18.2 and FGFR2b into routine practice, for optimal treatment planning, it is important to consider whether there exists an overlap between these biomarkers. MATERIALS AND METHODS: We evaluated CLDN18.2 expression in many patients with GC (n=1,538) using tissue microarrays that had been previously used to evaluate FGFR2b overexpression. We investigated the overlap between CLDN18.2 and FGFR2b expression

High Claudin 18.2 Expression Predicts Favorable Outcomes in Pancreatic Ductal Adenocarcinoma After Curative Resection.

BACKGROUND/AIM: Pancreatic ductal adenocarcinoma (PDAC) exhibits aggressive clinical behavior and poor prognosis, primarily due to late-stage diagnosis, early metastasis, and treatment resistance. Current therapeutic approaches, including operation, chemotherapy, and radiotherapy, demonstrate limited efficacy, underscoring the urgent need for novel targeted approaches. Claudin 18.2 (CLDN18.2) has emerged as a promising monoclonal antibody target, such as zolbetuximab; however, limited studies have examined its role in PDAC. This study investigated the association between CLDN18.2 expression an

Claudin-4 as a dual regulator of genome stability and immune evasion in high grade serous ovarian cancer.

High-grade serous carcinoma (HGSC) thrives in an immune-suppressive microenvironment marked by poor T-cell infiltration and blunted anti-tumor responses, driving immune evasion, tumor progression, and therapy resistance. Regulation of genomic instability, which restricts cytoplasmic DNA accumulation, is essential to prevent immune activation. Claudin-4, often overexpressed in HGSC, is strongly linked to therapy resistance and plays a key role in regulating initiation and resolution of genomic instability, shaping the tumor's potential ability to evade immune surveillance and withstand treatmen
