Suppression of pre-B cell colony formation by catecholamine oxidation.

The sympathetic nervous system regulates many aspects of lymphocyte function by stimulating adrenergic receptors with catecholamines. To determine the effects of β-adrenergic receptor (β-AR) signaling on B cell development, we utilized the pre-B cell colony forming unit (CFU-Pre-B) assay and found that isoproterenol (ISO), a β-AR selective catecholamine, potently suppresses pre-B cell colony formation. Further investigation using selective agonists and knockout mice showed that the colony suppression was independent of adrenergic receptor signaling. This colony suppression was replicated by tr

Megakaryocytopoiesis in polycythemia vera: characterization by megakaryocytic progenitors (CFU-Meg) in vitro and quantitation of marrow megakaryocytes.

Megakaryocytopoiesis in polycythemia vera (PV) was characterized by in vitro growth of marrow megakaryocytic progenitors (CFU-Meg) and quantitation of megakaryocyte numbers in marrow biopsy specimens in 14 patients with PV. Megakaryocyte numbers and CFU-Meg numbers in the 14 patients were variable, with values from the control range to markedly increased numbers. Nine of the 14 patients showed spontaneous CFU-Meg growth, and the presence or absence of spontaneous CFU-Meg growth was found to be related to the degree of marrow megakaryocyte increase, i.e., those with spontaneous CFU-Meg had high

Analysis of colony-stimulating factors and macrophage progenitor cells in mice immunized against Listeria monocytogenes by adoptive transfer.

Experiments were performed to elucidate the role of colony-stimulating factors in host defenses to the intracellular pathogen Listeria monocytogenes. Mice were protected against Listeria sp. by adoptive transfer of immune spleen cells and were then challenged with listeriae intravenously. Control mice were injected with spleen cells from uninfected mice. Adoptively immunized (immune) mice had significantly fewer listeriae in spleens and livers 2 and 4 days after Listeria challenge than did control mice. During acute infection, colony-stimulating activity in serum was increased earlier (10 h) i

Common pattern of two distinct types of colony-stimulating factor in human tissues and cultured cells.

Conditioned media prepared from human lung, placenta, peripheral leukocytes, cultured human pancreatic carcinoma cells, and cultured cervical carcinoma cells exhibit a common pattern of two distinct colony-stimulating factors (CSF) separable by isoelectrofocusing. Type I CSF appears polydisperse on isoelectrofocusing, showing multiple peaks with an isoelectric point of 3.6-4.7 and mol wt 50,000. It has a much higher activity in mouse than in human bone marrow, and stimulates the formation of predominately macrophage colonies. Type II CSF has an isoelectric point of 5.7 and mol wt 27,000 and ex

Lipoic Acid Attenuates Lipopolysaccharide- and Escherichia coli-Induced Reactive Oxygen Species Production and Neutrophil Extracellular Trap Formation Without Impairing Escherichia coli or Staphylococcus aureus Killing by Human Neutrophils.

Neutrophils are essential for antimicrobial defense through reactive oxygen species (ROS) production, tumor necrosis factor-alpha (TNF-α) release, and neutrophil extracellular trap formation. Lipoic acid, a redox-active antioxidant, modulates activation in human neutrophils. Neutrophils isolated from healthy donors were pretreated with lipoic acid and then exposed to lipopolysaccharide (LPS) or whole Escherichia coli, according to the specific assay. ROS production, NET formation, TNF-α release, bacterial killing, metabolic activity, and cell death were assessed using fluorometric assays, enzy

The effect of infection with human immunodeficiency virus on the anticryptococcal activity of lymphocytes and monocytes.

The effect of infection with human immunodeficiency virus (HIV) on the capacity of human lymphocytes and monocytes to inhibit and kill Cryptococcus neoformans in an 18-h assay was examined. In vitro infection of the Jurkat human T cell line with each of 3 HIV strains caused significant loss of anticryptococcal activity, which peaked 3-4 weeks after HIV infection. Lymphocytes from HIV-seropositive and -seronegative persons had similar activity, even if highly enriched for CD4 cells. The activity of lymphocytes from both seropositive and seronegative donors was increased by culture with interleu

Effect of combined infection with Salmonella and influenza virus on their respective proliferation in chicken embryonated eggs.

BACKGROUND: Salmonella is a major food-borne bacterial pathogen that causes food poisoning related to the consumption of eggs, milk, and meat. Food safety in relation to Salmonella is particularly important for eggs because their shells as well as their contents can be a source of contamination. Chicken can also be infected with influenza virus, but it remains unclear how co-infection of Salmonella and influenza virus affect each other. AIM: The potential influence of co-infection of Salmonella and influenza virus was examined. METHODS: Salmonella Abony and influenza virus were injected into

In Vitro and In Vivo Effects of the Polymyxin-Vorinostat Combination Therapy Against Multidrug-Resistant Gram-Negative Pathogens.

With the stagnancy of antibiotics development, polymyxins have become the last defense for treatment of multidrug-resistant (MDR) Gram-negative bacteria, whereas the effect of polymyxin monotherapy is limited by resistance. The objective of this study was to evaluate the effects of polymyxin B (PMNB)-vorinostat (SAHA) combination therapy against Gram-negative pathogens in vitro and in vivo. The antibacterial activities of PMNB and SAHA were evaluated by susceptibility testing. The synergistic effect was assessed by checkerboard tests and time-killing kinetics experiments. CSF-1 deficiency in the op/op mouse has differential effects on macrophage populations and differentiation stages.

Osteopetrosis and the absence of colony-stimulating factor 1 (CSF-1) in op/op mice are associated with decreased cellularity of the bone marrow (to one tenth of the normal), a very significant reduction in the number of cells recovered from peritoneal, pleural, and alveolar lavages, moderate leukopenia, and a slight decrease in the number of cells per spleen and thymus. Furthermore, op/op mice possess deficiencies in the number of macrophages in various organs. These cells are apparently absent in the bone marrow, severely reduced (5%-15% of the normal number) in peritoneal and pleural cavitie

Development of multifunctional BP@MoS2 two-dimensional hybrid nanomaterials for label-free surface-enhanced Raman spectroscopy detection and synergistic light-conversion depredation of pathogenic bacteria.

Infectious diseases caused by pathogenic bacteria pose a significant challenge to global public health, underscoring the urgent need for early detection and on-site sterilization technology. In this study, we proposed a multifunctional two-dimensional BP@MoS2 nanocomposites, which not only enabled sensitive bacterial detection but also demonstrated efficient sterilization capabilities. Specially, the unique flower-like surface microstructure of the nanocomposites promoted target enrichment efficiency, while its hybridized band structure facilitated powerful charge transfer, thereby facilitated

Combination therapy strategies against multidrug resistant bacteria in vitro and in vivo.

Exploring effective combination antibacterial therapies has become a research focus. This study selected seven common antibiotics to perform a series of tests on different Gram-negative bacteria isolated from clinical samples of chronic obstructive pulmonary disease patients. More than 70% of the strains exhibited multidrug resistance but remained sensitive to polymyxin B. The checkerboard assay revealed a significant synergistic effect between polymyxin B and tetracycline against different resistant strains, with fractional inhibitory concentration index values consistently below 0.5. Development of two real-time polymerase chain reaction assays to detect Actinobacillus pleuropneumoniae serovars 1-9-11 and serovar 2.

Two real-time, or quantitative, polymerase chain reaction (qPCR) assays were developed to detect Actinobacillus pleuropneumoniae serovars 1-9-11 (highly related serovars with similar virulence potential) and serovar 2, respectively. The specificity of these assays was verified on a collection of 294 strains, which included all 16 reference A. pleuropneumoniae strains (including serovars 5a and 5b), 263 A. pleuropneumoniae field strains isolated between 1992 and 2009 in different countries, and 15 bacterial strains other than A. pleuropneumoniae. The detection levels of both qPCR tests were eva

Effect of bacterial virulence on IL-18 expression in the amnion infected with Escherichia coli.

PROBLEM: The upregulation of inflammatory substances threatens pregnancy. Interleukin-18 (IL-18) is elevated in women who miscarried. The purpose of this study was to develop a pig model of chorioamnionitis to study the effect of bacterial virulence on IL-18 response in experimentally infected amnion. METHOD OF STUDY: A total of 20,000 colony-forming units of Escherichia coli (an enteropathogenic O55 strain, EPEC or O86 non-pathogenic strain) were administered into the amniotic cavity of pig fetuses at 70% of gestation for 10 hr. Fetal amniotic fluid samples were analyzed for IL-18 levels by

Primary immune system responders to nucleus pulposus cells: evidence for immune response in disc herniation.

Although intervertebral disc herniation and associated sciatica is a common disease, its molecular pathogenesis is not well understood. Immune responses are thought to be involved. This study provides direct evidence that even non-degenerated nucleus pulposus (NP) cells elicit immune responses. An in vitro colony forming inhibition assay demonstrated the suppressive effects of autologous spleen cells on NP cells and an in vitro cytotoxicity assay showed the positive cytotoxic effects of natural killer (NK) cells and macrophages on NP cells. Non-degenerated rat NP tissues transplanted into wild

In vitro study of pesticide hematotoxicity in human and rat progenitors.

Some pesticides are hematotoxic and cause aplastic anemia, agranulocytosis, neutropenia, and thrombopenia. In order to evaluate hematopoietic progenitor cultures in the exploration of pesticide hematotoxicity, human and rat colony-forming unit-granulocyte and macrophage (CFU-GM) were cultured in the presence of different concentrations of pesticides, known to be either hematotoxic or innocuous for blood cells. The results were compared to the control culture of the same sample. Four insecticides (lindane, azinphos, mevinphos, parathion methyl), three herbicides, (2,4,5, T, bromacil, MCPA), and
