Evolutionary biology of antimalarial drug resistance: Understanding of the evolutionary dynamics.

Anti-malarial drug resistance poses a significant challenge to global malaria control efforts, necessitating a deeper understanding of the evolutionary dynamics underlying the emergence and spread of resistance. This study explores how evolutionary theory provides a framework for elucidating the molecular mechanisms and genetic variation within parasite populations that drive resistance evolution. Drawing on recent research findings, we discuss the role of natural selection, genetic diversity, and fitness costs in shaping resistance phenotypes. Additionally, we highlight the implications of ev

Dynamic Emissions and Secondary Organic Aerosol Formation from Real Personal Care Products: New Insights into Siloxane-Containing Molecules.

In urban atmosohere, volatile methyl siloxanes (VMSs) was treated as the important maker from pesonal care products (PCPs) of volatile chemical products (VCPs), yet the link between their realistic emission profiles and secondary organic aerosol (SOA) formation remains elusive. Moving beyond previous studies using pure standards, we systematically characterized the dynamic emissions and SOA evolution of four typical PCPs using an evaporation chamber coupled with an oxidation flow reactor and other online gas/aerosol mass spectrometry. VMS dominated the emissions (>95%) for most PCP products an

Cdk1 heterozygosity triggers premeiotic endoreplication and unreduced sperm formation via the Cdk2/Ccne1-Fzr1a pathway in loach, a teleost.

Polyploidy represents a major driver of eukaryotic genome evolution. In vertebrates, premeiotic endoreplication is considered as an important route to polyploidization, yet the molecular basis governing this process remains elusive. Here, we reported that heterozygous mutation of cdk1 in the loach, Misgurnus anguillicaudatus (Teleostei, Cobitidae), triggered premeiotic endoreplication in spermatogonia, resulting in the production of unreduced diploid sperms. Through integrated proteomic and phosphoproteomic profiling, we uncovered that Cdk1 heterozygosity led to widespread dysregulation of the

Pangenomic analysis of Wolbachia provides insight into the evolution of host adaptation and cytoplasmic incompatibility factor genes.

INTRODUCTION: The genus Wolbachia provides a typical example of intracellular bacteria that infect the germline of arthropods and filarial nematodes worldwide. Their importance as biological regulators of invertebrates, so it is particularly important to study the evolution, divergence and host adaptation of these bacteria at the genome-wide level. METHODS: Here, we used publicly available Wolbachia genomes to reconstruct their evolutionary history and explore their adaptation under host selection. RESULTS: Our findings indicate that segmental and single-gene duplications, such as DNA methyl

Val43 residue of NsrR is crucial for the nitric oxide response of Salmonella Typhimurium.

ABSTRACT: In pathogenic bacteria, the flavohemoglobin Hmp is crucial in metabolizing the cytotoxic levels of nitric oxide (NO) produced in phagocytic cells, contributing to bacterial virulence. Hmp expression is predominantly regulated by the Rrf2 family transcription repressor NsrR in an NO-dependent manner; however, the underlying molecular mechanism in enterobacteria remains poorly understood. In this study, we identified Val43 of Salmonella Typhimurium NsrR (StNsrR) as a critical amino acid residue for regulating Hmp expression. The Val43-to-Ala-substituted mutant NsrR isolated through ran

Tumor-Promoting Inflammation in Serrated Colorectal Neoplasia: Immune Ecosystems and Clinical Implications.

Serrated colorectal neoplasia represents a biologically distinct route to colorectal cancer that differs from the conventional adenoma-carcinoma sequence in its molecular alterations, epithelial programs, and microenvironmental evolution. Recent studies indicate that tumor-promoting inflammation and dynamic tumor-microenvironment interactions are integral to the evolution of serrated tumors. In this review, we discuss serrated neoplasia as a process shaped by heterogeneous immune ecosystems rather than a uniform inflammatory state. We propose two broad immune trajectories: an inflamed/antigeni

Analysis of the evolutionary forces shaping mitochondrial genomes of a Neotropical malaria vector complex.

Many vectors of human malaria belong to complexes of morphologically indistinguishable cryptic species. Here we report the analysis of the newly sequenced complete mitochondrial DNA molecules from six recognized or putative species of one such group, the Neotropical Anopheles albitarsis complex. The molecular evolution of these genomes had been driven by purifying selection, particularly strongly acting on the RNA genes. Directional mutation pressure associated with the strand-asynchronous asymmetric mtDNA replication mechanism may have shaped a pronounced DNA strand asymmetry in the nucleotid

Extent of Nonequilibrium Sets Relaxation Barriers in Dewetting Polymer Films.

Thin polymer films are archetypal nonequilibrium systems in which chain conformations are frozen by rapid preparation. The resulting molecular recoiling stress drives the system toward equilibrium and relaxes with time and temperature. We show that the activation energy governing this relaxation depends on the magnitude of the stress, revealing that polymers farther from equilibrium must overcome higher barriers, while stress release enables relaxation through progressively easier pathways. This stress-dependent evolution of activation barriers reflects a balance between energetic and entropic

A pivot mutation impedes reverse evolution across an adaptive landscape for drug resistance in Plasmodium vivax.

BACKGROUND: The study of reverse evolution from resistant to susceptible phenotypes can reveal constraints on biological evolution, a topic for which evolutionary theory has relatively few general principles. The public health catastrophe of antimicrobial resistance in malaria has brought these constraints on evolution into a practical realm, with one proposed solution: withdrawing anti-malarial medication use in high resistance settings, built on the assumption that reverse evolution occurs readily enough that populations of pathogens may revert to their susceptible states. While past studies

Experimental Evolution Identifies Vaccinia Virus Mutations in A24R and A35R That Antagonize the Protein Kinase R Pathway and Accompany Collapse of an Extragenic Gene Amplification.

UNLABELLED: Most new human infectious diseases emerge from cross-species pathogen transmissions; however, it is not clear how viruses adapt to productively infect new hosts. Host restriction factors represent one species-specific barrier that viruses may initially have little ability to inhibit in new hosts. For example, viral antagonists of protein kinase R (PKR) vary in their ability to block PKR-mediated inhibition of viral replication, in part due to PKR allelic variation between species. We previously reported that amplification of a weak PKR antagonist encoded by rhesus cytomegalovirus,

Hepadnavirus evolution and molecular strategy of adaptation in a new host.

In order to elucidate the mechanisms of hepadnavirus evolution in vivo and to trace the fate of known quasispecies in a single animal during the acute phase of infection, a woodchuck (Marmota monax) was infected with the hepadnavirus woodchuck hepatitis B virus (WHV). Woodchuck 197 (W197) was injected intravenously with pooled sera collected from a chronic carrier that had been infected originally with a molecular clone of known genome sequence (WHV7). Viral genome variants from both the inoculum and the follow-up sera from W197 were characterized for the presence of quasispecies related to th

PLA/PCL composites manufactured from commingled yarns for biomedical applications.

Composites manufactured with textiles weaved with commingled yarns using PLA (polylactic acid) and PCL (polycaprolactone) fibres are promising candidates for connective tissue engineering. In this work, textiles were fabricated using PLA/PCL commingled yarns in a ratio of 3 to 1, which were subsequently consolidated by compression moulding to produce solid composite plates. Specimens were extracted from the composite plates and submitted to degradation testing by immersion in PBS fluid (phosphate-buffered saline) at different periods. The dry mass, mechanical performance (tensile tests), therm

Generation and iterative affinity maturation of antibodies in vitro using hypermutating B-cell lines.

We show that iterative antigen-mediated selection of B-cell lines that constitutively hypermutate their immunoglobulin V genes during culture can be exploited to generate antibodies in vitro. From Ramos, a hypermutating human B-cell line expressing IgM of unknown specificity, we derived descendants that exhibit stepwise improved binding to streptavidin. Binding is initially conferred by mutations in complementarity-determining regions (CDRs), but maturation is due to strategic framework mutations. A more powerful system is provided by a hypermutating chicken B-lymphoma line, owing to its rapid

Evolutionary aspect of spike glycoprotein's conformational dynamics.

The conformational transition of the receptor binding domain (RBD) of the SARS-CoV-2 spike (S) protein from the "closed" to the "open" state is an important dynamic event influencing the infectivity of the virus. However, how accumulating mutations reshape these conformational dynamics during viral evolution remains to be clarified. In this study, we observed the motion of the RBD of fully glycosylated S protein trimers from wild type to Omicron (BA.2, BA.4&5) variants by using all-atomic molecular dynamics simulations. Our study indicates that, although a fully converged free energy landscape

Composition-mediated intermolecular interaction transition and vibrational reorganization in n-hexane-ethanol binary mixtures.

Mixing polar ethanol with nonpolar n-hexane provides a suitable liquid system for examining composition-dependent changes in local molecular organization and vibrational behavior. This study combines Raman spectroscopy and DFT calculations to clarify the molecular origin of these changes in n-hexane-ethanol binary mixtures with ethanol volume fractions from 0.1 to 0.9. As the ethanol content increased, the ethanol CCO band near 894 cm-1 exhibited a redshift-blueshift-redshift trend, whereas the n-hexane CH stretching bands at 2898 cm-1, 2923 cm-1, and 2950 cm-1 showed the opposite blueshift-re

Hepatitis B virus genetic diversity.

Hepatitis B virus (HBV) is a human DNA virus, which replicates through an RNA intermediate because of the reverse-transcriptase (RT) activity of its DNA polymerase. As a result, the mutation rate for HBV is higher than the rate observed for most DNA viruses. HBVs are classified into genotypes based on genomic sequencing, and antigenic subtypes based on the antigenic properties of its major surface glycoprotein, the HBV surface antigen (HBsAg). Subgenotypes have been identified within most of the HBV genotypes. The HBV groups defined by the different genotype-HBsAg subtype associations found ov

Maize BMS cultured cell lines survive with massive plastid gene loss.

As part of developing an ex planta model system for the study of maize plastid and mitochondrial gene expression, a series of established Black Mexican Sweet (BMS) suspension cell lines was characterized. Although the initial assumption was that their organelle biochemistry would be similar enough to normal in planta cells to facilitate future work, each of the three lines was found to have plastid DNA (ptDNA) differing from control maize plants, in one case lacking as much as 70% of the genome. The other two BMS lines possessed either near-wild-type ptDNA or displayed an intermediate state of

Bridging past and future: the evolution of genetic diagnosis in FSHD and the role of emerging technologies in a globalized framework.

Facioscapulohumeral muscular dystrophy (FSHD) is a neuromuscular disorder characterized by marked clinical and molecular heterogeneity. Over more than two decades, molecular diagnosis has relied on Southern blotting to resolve the complex D4Z4 macrosatellite at chromosome 4q35. While this approach remains a reference standard, its technical constraints and the need for complementary layers of information have become more evident as FSHD diagnostics is increasingly required to support patient stratification, genotype-phenotype correlation, and clinical trial readiness. In this context, the revi
